Abstract

PURPOSE: To prepare a highly pure acylated product, economically, with simple post-treatment, by acylating a reactive derivative of carboxylic acid and a compound having an active methylene group or methyne group in a hydrous organic solvent, using a safely handleable base as a condensing agent.
CONSTITUTION: A reactive derivative of a carboxylic acid, e.g. acid halide, active acid amide, etc., and a compound having an active methylene group or active methyne group, e.g. dimethyl malonate, ethyl acetoacetate, etc., are acylated in a hydrous organic solvent in the presence of a base such as potassium carbonate, at room temperature or an elevated temperature, to obtain an acylated product useful as an intermediate of medicines, etc. The organic solvent is pref. methyl isobutyl ketone, tetrahydrofuran, etc.
EFFECT: The reaction proceeds without using a dangerous metal compound condensing agent such as metallic sodium, or a highly toxic crown ether catalyst.
