Abstract
PURPOSE: Partially hydrolyzed starch is hydrolyzed by two kinds of soluble enzymes to a high level and the product is made to act on an immobilized glucoamylase to produce glucose in high yield.
CONSTITUTION: Starch is hydrolyzed preferably by the use of α-amylase to prepare a starch hydrolyzate with 5W30 glucose equivalent (A). The product is hydrolyzed using a soluble glucoamylase, which is prepared from a mold of Aspergillus, and a soluble pluranase, e.g., obtained from Klebisella aerogenes described in USP 3490955, to give another starch hydrolyzate (B) containing 50W80wt% of the total saccharides, especially 65W85wt% of glucose. Then, the hydrolyzate (B) is adjusted to 30W50, preferably 35W40W/V% in the solid concentration and an immobilized glucoamylase is made to act on the hydrolyzate at a pH of 3.5W6, preferably 4W 5, at a temperature of 40W60, preferably 45W55°C, to produce glucose in high yield.
