Abstract

PURPOSE: To obtain the titled compound in high purity and yield simply without the desalting step in a short operation time, by bringing crude coenzyme A into contact with an ion unexchangeable porous resin, and eluting the coezyme A with water.
CONSTITUTION: A culture fluid of Brevibacterium ammoniagenes IAM1641 (FERM-P No.1471) as a starting material is heated, treated with active carbon and an organic solvent, and vacuum-dried to give crude coenzyme A, which is dissolved in distilled water, reduced with 2-mercaptoethanol, passed through a column of an ion unexchangeable porous resin, e.g. Amberlite XAD-2, and then eluated with water of pH4. The components of the respective eluate fractions are separated and determined by paper chromatography to give an elution curve. Pure coenzyme A (CoA) is then obtained from the respective fractions, and concentrated to afford the aimed compound.
