Abstract
PURPOSE: To provide a simple and highly sensitive means for the determination of lipid peroxide, by reacting a specimen containing lipid peroxide with glutathione peroxidase in the presence of reduced glutathione, and converting the produced oxidized glutathione to a fluorescent material.
CONSTITUTION: A highly sensitive micro-determination of th degree of peroxidation of a lipid peroxide in the living body is carried out by (1) reacting the lipid peroxide with glutathione peroxidase in the presence of reduced glutathione (GSH) to convert GSH to oxidized glutathione (GSSG), (2) reacting and eliminating the residual unreacted GSH with an SH reagent (e.g. N-ethylmaleimide), (3) making the reaction mixture to alkaline state, (4) reacting GSSG with o-phthalaldehyde, (5) exciting the produced fluorescent material with light having a wavelength of 343nm and measuring the intensity of the emitting light at a wavelength of 420nm, and (6) determining the concentration of the lipid peroxide in the specimen from a calibration curruve prepared by using specimens having known concentrations.
