CONSTITUTION: UK788 strain (FERM P 5141) belonging to "Bacillus stearothermophilus" is cultivated especially by liquid culture in a general medium comprising common untrient sources of bacteria, e.g., carbon source such as glucose, molasses, etc., nitrogen source such as urea, ammonium sulfate, inorganic salts such as K, Na, Cu, etc. In the case of batch cultivation, the culture is preferably carried out until the latter period of logarithmic propagation, and in the case of continuous cultivation, continuous culture of the substance circulation type is adopted, and the rate of dilution is controlled in the vicinity of the maximum rate of the relative propagation of UK788 strain. Molds are collected from the culture by centrifuging, etc. Heat-resistant glucokinase is isolated and purified by a conventional method, e.g., adding an organic solvent, etc. to a supernatant liquid obtained by grinding the molds and centrifuging to attain purification through fractionation, adsorbing the molds on a carrier followed by purification, etc.
