Abstract

PURPOSE: The continuous cultivation of a thermophilic bacterium such as bacillus stearothermophilus is conducted under a specified dilution to produce cell bodies of high pyruvate kinase content per unit body.
CONSTITUTION: A thermophilic bacterium in bacillus capable of producing pyruvate kinase, e.g., bacillus stearothermophilus ATCC 7953, ATCC 8005, ATCC 10149 or NCA 1503, is cultured wherein the dilution D is kept so as to satisfy the equation[D is dilution (1/hr); μmax is the maximum growth rate of the bacterium under the conditions of the continuous cultivation (1/hr)]. The culture medium is a usual enriched one containing glucose, meat essence and other nutritients and the cultivation is effected aerobically at about 40W75°C. After the cells are crushed, the resultant enzyme solution is purified by fractionation and adsorption to isolate pyruvate kinase.
