Abstract

NEW MATERIAL:A tetrazole derivative of formula I [R1 is lower alkyl; A is lower alkylene; R2 is OH, lower alkoxy, or group formula -N(R3)R4 (R3 and R4 are H, lower alkyl, or cycloalkyl)] and its salt.
EXAMPLE: 4-(5-Methyl-1,2,3,4-tetrazol-1-yl)butyric acid.
USE: Antiulcer agent and antiphlogistic agent.
PROCESS: For example, reaction of an aminocarboxylic acid of formula (2) with a lower alcohol of formula (3) (R5 is alkyl) gives an aminocarboxylic acid ester of formula (4), which is made to react with a carboxylic acid halide of formula (5) (X is halogen) in the presence of a dehydrohalogenating agent to afford a compound of formula (6). The compound is made to react with PCl5, and the resulting imine derivative is made to react with HN3 without separating from the reaction system to obtain the compound of formula (10). Hydrolysis of the compound of formula (10) affords the compound of formula Ib, which can be converted to the compound of formula Ic by reacting with an amine of formula (7).
