Abstract

PURPOSE: To prepare an N-substituted aletheine useful as pharmaceuticals or their intermediates, easily, economically, in high yield and purity, by reacting an N- substituted-β-alanine with cysteamine or cystamine in the presence of a phosphorus amide derivative.
CONSTITUTION: An N-substituted aletheine is prepared by the direct condensation of an N-substituted-β-alanine with cystamine, cysteamine or their salts, in the presence of a phosphorus amide compound of formula (R1 is alkoxy, aryloxy, etc.; R2 is R1, or R2 and R1 together form a ring; R3 is alkyl, aryl; R4 is R3, or R3 and R4 together form a ring), e.g. 2-diethylamino-4-methyl-1,3,2-dioxaphosphorane. The N-substitution group is an amino-protecting group such as acyl group. The reaction is carried out at a temperature between 0°C and the boiling point of the solvent (e.g. dichloroethane, etc.).
