Abstract

PURPOSE: To obtain the title heat-resistant enzyme useful as medical enzyme formulas and reagents for biochemical studies, industrially advantageously, by crushing bacterium bodies of Thermus thermophilus HB8 strains cultured in a medium, and by separating and collecting the enzyme.
CONSTITUTION: Thermus thermophilus HB strains, which belong to the genus Thermus, are cultured in a medium containing 0.5% of peptone, 0.4% of a yeast extract, 0.2% of NaCl, and 0.1% of glucose at 75°C for about 6hr, and collected in the latter logarithmic growth phase. The bacterium bodies thus obtained are crushed by sound waves, etc. The crushed bodies are purified by centrifugation, dialysis, ion exchange resin treatment, ultrafiltration, etc. to give the desired cytochrome oxidase, having the heat resistance and a residual activity of about 75% after heat-treatment at 78°C for 20min.
