Abstract

PURPOSE: To assimilate fibrous substances and to obtain a heat-resistant cellulase having strong activity, by cultivating a heat-resistant cellulase producing strain belonging to a genus of Talaromyces in a medium.
CONSTITUTION: A microorganism capable of producing a heat-resistant cellulase, e.g., Talaromyces.sp.F-29(FERM-P No.4904), is cultivated in a medium. In the medium, rice hulls as a carbon source, ammonium sulfate as a nitrogen source, and KH2PO4 as an inorganic salt used respectively. The cultivation is carried out by solid or liquid cultivation. In the case of liquid culture, for example, a cultivation temperature of 40W55°C and a pH of 3W9 make cellulase activity maximum in 2W5 days. Isolation of cellulase after culture is made by a conventional method and the cultured substance is extracted with a buffer or a saline solution to give a raw enzyme solution, which may be purified by a conventional method, if necessary.
