Abstract

PURPOSE: To prepare the title compound which is an antibacterial in a high yield without requiring addition of an excess primary amine, by reacting Rifamycin S with 1, 3, 5-tri-tert-butyl-hexahydro-s-triaine.
CONSTITUTION: One mole of Rifamycin S is reacted with 2/3 mole of 1, 3, 5-tri- tert-butyl hexahydro-s-triazine in the presence of an oxidizing agent, e.g. manganese dioxide, etc. and a catalytic amount of water in an inert organic solvent, e.g. pyridine, DMF, etc. at room temperature to the boiling point of the solvent to give 3-tert-butyliminomethylrifamycin S of formula (R is tert-butyl group) which is useful as an antibacterial or its intermediate.
