PURPOSE: To obtain the title compound useful as an intermediate for rifampicin which is a tuberculostatic, readily in high yield, by reacting rifamycin S or SV with a primary amine and a methylene halide.
CONSTITUTION: Rifamycin S or SV is reacted with a primary amine formula I (R1 is alkyl group) and a methylene halide of formula II (X1 and X2 are halogens) in a solvent, e.g. DMF or DMSO, at room temperature to the boiling point of the solvent to give the 3-iminomethyl derivative of rifamycin S of formula III. The use of an oxidizing agent, e.g. manganese or lead dioxide, provides a high yield in a short time.
