Abstract

PURPOSE: To obtain the title substance useful as a raw material for preparing sprironolactone in high yield, by reacting androst-4-ene-3,17-dione with a metal acetylide, by reducing the reaction product after pH adjustment, followed by crystallizing the product from a mixed solvent of an alcohol and water.
CONSTITUTION: Androst-4-ene-3,17-dione is reacted with a metal acetylide in excess in a reaction medium, e.g. hydrophilic organic solvent, and the reaction mixture is then reacted with an aqueous weak acid of pKa≤2 at 50°C or below to give a pH 1W13, thus forming an 17-ethynyl-5-ene derivative. The resulting derivative is reduced with a reducing agent, e.g. a metal hydrogen complex, in an amount of 1W100 times that of the derivative at -30W70°C to form crude 3,17-dihydroxypregn-5-ene-20-yne, which is crystallized from a mixed solvent of water and an alcohol with water content of 5W95% to give the desired substance.
