Claims (1)
- 【特許請求の範囲】 1.特異的結合リガンドの測定用の分析要素であって、 (i) 多孔質展開区画;および (ii) 前記多孔質展開区画と液体接触状態にある少なくとも1つの追加の区画 を含んで成り、前記区画のうちの少なくとも1つに、着目の特異的結合リガンド のためのレセプターに特異的に結合するバナジウムブロモペルオキシダーゼ標識 免疫反応体を含有することを特徴とする分析要素。 2.前記バナジウムブロモペルオキシダーゼがアスコフィルム・ノドサム(As cophyllum nodosum )、セラミルム・ルブルム(Ceramirum rubrum)、ラミナリ ア・サッカリナ(Laminaria saccharina)、フカス・ディスチクス(Fucus dis tichus )、コラリナ・ピルリフェラ(Corallina pilurifera)、コラリナ・オフ ィシナリス(Corallina officinalis)またはマクロシスチス・ピリフェラ(Ma crocystis pyrifera )から得られる、請求項1に記載の要素。 3.前記バナジウムブロモペルオキシダーゼがアスコフィルム・ノドサム(As cophyllum nodosum )から得られる、請求項2に記載の要素。 4.(iii) 1または複数の前記区画と液体接触状態である吸収材料を更に含ん で成る、請求項1に記載の要素。 5.特異的結合リガンドの測定用の多層分析要素であって、 非孔質支持体を含んで成り、その上に液体接触状態において次の層: (i) 第一試薬または緩衝剤層; (ii) 着目の特異的結合リガンドと特異的に結合する未標識の免 疫反応体を含んで成る下塗層;および (iii) バナジウムブロモペルオキシダーゼ標識免疫反応体を含む多孔質展開層 を有することを特徴とする多層分析要素。 6.前記バナジウムブロモペルオキシダーゼがアスコフィルム・ノドサム(As cophyllum nodosum )、セラミルム・ルブルム(Ceramirum rubrum)、ラミナリ ア・サッカリナ(Laminaria saccharina)、フカス・ディスチクス(Fucus dis tichus )、コラリナ・ピルリフェラ(Corallina pilurifera)、コラリナ・オフ ィシナリス(Corallina officinalis)またはマクロシスチス・ピリフェラ(Ma crocystis pyrifera )から得られる、請求項5に記載の多層要素。 7.前記バナジウムブロモペルオキシダーゼがアスコフィルム・ノドサム(As cophyllum nodosum )から得られる、請求項6に記載の多層要素。 8.前記多孔質展開層がビーズ型展開層である、請求項5に記載の多層要素。 9.(iv) 1または複数の前記層と液体接触状態である吸収材料を更に含んで 成る、請求項5に記載の多層要素。 10.特異的結合リガンドの測定方法であって、 (A)特異的結合リガンドを含むと思われる液体試料を、 (i) 多孔質展開区画;および (ii) 前記多孔質展開区画と液体接触状態にある少なくとも1つの追加の区 画 を含んで成る分析要素であって、前記区画のうちの少なくとも1つに、前記リガ ンドに特異的に結合するバナジウムブロモペルオキシダーゼ標識免疫反応体を含 有する分析要素と接触せしめ; (B)前記分析要素に洗浄溶液を適用して、前記バナジウムブロモペルオキシダ ーゼ標識免疫反応体の反応形と前記バナジウムブロモペルオキシダーゼ標識免疫 反応体の未反応形とを分離し、ここで前記洗浄溶液は (a) バナジウムブロモペルオキシダーゼの酵素活性に応じてシグナルを与 える化学発光シグナル発生試薬であって、2,3−ジヒドロ−1,4−フタラジ ンジオン誘導体であるシグナル発生試薬; (b) ハロゲン、ハロゲン、擬ハロゲン、ハロゲン供給源または擬ハロゲン 供給源;および (c) ペルオキシドまたはペルオキシド生成試薬組成物 を含んで成る、約6.5 〜約10のpHを有する組成物を含み;そして (C)前記特異的結合リガンドの測度として、前記バナジウムブロモペルオキシ ダーゼ標識免疫反応体の未反応形または反応形のいずれかを検出する ことを含んで成る方法。
