Claims (1)
- 【特許請求の範囲】 1.Bordetella菌株からの凝集原製剤の製造方法において、 (a)Bordetella菌株の細胞ペーストを得る段階 (b)該細胞ペーストから線毛性凝集原を選択的に抽出して、該凝集原を含む 第1の上清と第1の残渣沈殿物を得る段階 (c)前記第1の残渣沈殿から第1の上清を分離する段階; (d)前記第1の上清を、線毛性凝集原を含む透明上清と非線毛性凝集原汚染 物を含む第2の沈殿物を得るだけの温度および時間でインキュベーションする段 階; (e)前記透明上清を濃縮して、粗線毛性凝集原溶液を得る段階; (f)前記粗線毛性凝集原溶液から線毛性凝集原を精製して、線毛性凝集原製 剤を得る段階 を有してなる方法。 2.前記インキュベーション段階(d)を、約75℃〜約85℃の温度で実施 する請求項1記載の方法。 3.温度を約80℃とする請求項2記載の方法。 4.前記インキュベーション段階(d)を、約10分間〜約60分間実施する 請求項2記載の方法。 5.時間を約30分間とする請求項3記載の方法。 6.線毛性凝集原の段階(b)での選択的抽出を、細胞ペーストを約1M〜約 6Mの尿素を含む緩衝液に分散させることで行う請求項2記載の方法。 7.前記第1の上清を濃縮してから、インキュベーション段階(d)を行う請 求項2記載の方法。 8.濃縮段階(e)を、前記透明上清から線毛性凝集原を沈殿させ、沈殿した 線毛性凝集原を得られた上清から分離し、沈殿した線毛性凝集原を可溶化するこ とで行う請求項7記載の方法。 9.前記沈殿形成を、前記透明上清にポリエチレングリコールを加えることで 行う請求項8記載の方法。 10.前記沈殿形成を、前記透明上清に分子量約8000のポリエチレングリ コールを約3%〜約5%まで加えて、前記透明上清から前記凝集原を沈殿させる ことで行う請求項8記載の方法。 11.ポリエチレングリコールの濃度を約4.3〜約4.7重量%とする請求 項10記載の方法。 12.前記凝集原を、カラムクロマトグラフィーによって粗線毛性凝集原溶液 から精製する請求項1記載の方法。 13.前記カラムクロマトグラフィーで、セファデックス6Bおよび/または PEIシリカカラムクロマトグラフィーを行う請求項12記載の方法。 14.前記精製段階で、前記カラムクロマトグラフィー精製からの溶出液の滅 菌によって無菌線毛性凝集原製剤を得る請求項12記載の方法。 15.前記無菌線毛性凝集原製剤を、無機塩アジュバントに吸収させる請求項 14記載の方法。 16.前記無機塩アジュバントがミョウバンである請求項15記載の方法。 17.Bordetella菌株がBordetella pertussis菌株である請求項1記載の方法 。 18.実質的に凝集原1を含まない線毛性凝集原2(Agg2)および線毛性 凝集原3(Agg3)を含むBordetella菌株からの線毛性凝集原製剤。 19.線毛性Agg2の線毛性Agg3に対する重量比が、約1.5:1〜約 2:1である請求項18記載の製剤。 20.請求項1記載の方法によって製造される請求項19記載の線毛性凝集原 製剤。 21.請求項18、19または20に記載の凝集原製剤を含有する免疫原性組 成物。 22.Bordetellaによって生じる疾患からワクチンによって免疫感作された宿 主を保護するためにin vivoで使用されるワクチンとして製剤された請求項21 記載の免疫原性組成物。 23.1以上の他のBordetella抗原をさらに含有する請求項22記載の免疫原 性組成物。 24.前記1以上の他のBordetella抗原が、線維状ヘマグルチニン、69kD a外膜蛋白、アデニル酸シクラーゼ、Bordetellaリポオリゴ糖、外膜蛋白ならび に百日咳毒もしくはそのトキソイドからなる群から選択される請求項23記載の 免疫原性組成物。 25.約2:1:1の重量比で、B.pertussisの百日咳トキソイド、線維状へ マグルチニンおよび線毛性凝集原を含有する請求項24記載の免疫原性組成物。 26.前記重量比が、ヒト1回用量中に百日咳トキソイド約10μg、線維状 ヘマグルチニン約5μgおよび線毛性凝集原約5μgを含有させることで得られ ている請求項25記載の免疫原性組成物。 27.約10:5:5:3の重量比で、B.pertussisの百日咳トキソイド、線 維状ヘマグルチニン、69kDa外膜蛋白および線維状凝集原を含有する請求項 24記載の免疫原性組成物。 28.前記重量比が、ヒト1回用量中に百日咳トキソイド約10μg、線維状 ヘマグルチニン約5μg、69kDa蛋白約5μgおよび線毛性凝集原約3μg を含有させることで得られている請求項27記載の免疫原性組成物。 29.約20:20:5:3の重量比で、B.pertussisの百日咳トキソイド、 線維状ヘマグルチニン、69kDa蛋白および線毛性凝集原を含有する請求項2 4記載の免疫原性組成物。 30.前記重量比が、ヒト1回用量中に百日咳トキソイド約20μg、線維状 ヘマグルチニン約20μg、69kDa蛋白約5μgおよび線毛性凝集原約3μ gを含有させることで得られている請求項29記載の免疫原性組成物。 31.1以上の非Bordetella免疫原をさらに含有する請求項24記載の免疫原 性組成物。 32.前記非Bordetella免疫原が、ジフテリアトキソイド、破傷風トキソイド 、Haemophilusの莢膜多糖類、Haemophilusの外膜蛋白、B型肝炎表面抗原、小児 麻痺、おたふくかぜ、はしか、および風疹からなる群から選択される請求項31 記載の免疫原性組成物。 33.ヒト1回用量中に、ジフテリアトキソイドを約15Lfと破傷風トキソ イドを約5Lfの量でさらに含有する請求項29記載の免疫原性組成物。 34.さらにアジュバントを含む請求項23記載の免疫原性組成物。 35.前記アジュバントが、リン酸アルミニウム、水酸化アルミニウム、クイ ル(Quil)A、QS21、リン酸カルシウム、水酸化カルシウム、水酸化亜鉛、 糖脂質類縁体、アミノ酸のオクタデシルエステルおよびリポ蛋白からなる群から 選択される請求項34記載の免疫原性組成物。 36.Bordetellaによって引き起こされる疾患に対して宿主を免疫感作する方 法において、免疫学的に有効な量の請求項21記載の免疫原性組成物を宿主に対 して投与する段階を有してなる方法。 37.宿主がヒトである請求項36記載の方法。
