Claims (1)
- 【特許請求の範囲】 1.親ポリペプチドが、極性はあるが帯電はしていない側鎖を持つ少なくとも 1種のアミノ酸残基を有するものである、該親ポリペプチドの合成類似体にして 、該残基の少なくとも1つがヒスチジン残基により置換されていることを特徴と する、上記の合成類似体。 2.前記親ポリペプチドと少なくとも略同等の生物活性を示す、請求の範囲第 1項に記載の類似体。 3.極性はあるが帯電はしていない側鎖を有する前記アミノ酸残基がグルタミ ン、アスパラギン及びトレオニンより成る群から選ばれる、請求の範囲第1項に 記載の類似体。 4.極性はあるが帯電はしていない側鎖を有する前記アミノ酸残基がグルタミ ンである、請求の範囲第3項に記載の類似体。 5.治療上有効な量の請求の範囲第1、2、3又は4項に記載の類似体及び生 理学的に許容できるキャリア又はその賦形剤を含んで成る製剤組成物。 6.請求の範囲第1項に記載の類似体を含んでいるドナーレザバーを有する電 気輸送デリバリーディバイス。 7.親ポリペプチドがグルタミン、トレオニン及びアスパラギンより成る群か ら選ばれる少なくとも1種の残基を有するものである、製剤用親ポリペプチド剤 の、向上した体表面通過電気輸送流量を示す合成類似体にして、該残基の少なく とも1つがヒスチジン残基により置換されていることを特徴とする、上記の合成 類似体。 8.前記親ポリペプチドと少なくとも略同等の生物活性を示す、請求の範囲第 7項に記載の類似体。 9.前記親ポリペプチド中の全てのGln残基が前記類似体中でHisで置換 されている、請求の範囲第7項に記載の類似体。 10.前記類似体の総電荷が約5乃至6の範囲のpHでは正であるが、pH7 .4においては実質的に等電性である、請求の範囲第7項に記載の類似体。 11.前記類似体が約5乃至6の範囲のpHにおいて前記親ポリペプチドより 大きな正の電荷を有している、請求の範囲第7項に記載の類似体。 12.請求の範囲第7項に記載の類似体を含んでいるドナーレザバーを有する 電気輸送デリバリーディバイス。 13.電気輸送送出性を高めるべく修飾された製剤用ポリペプチド剤にして、 pH約5−6において該ポリペプチド剤の電荷より大きい総電荷を有する類似体 を形成すべく、少なくとも1つのグルタミン残基がヒスチジン残基で置換されて いることを特徴とする該ポリペプチド剤を含んで成る組成物。 14.請求の範囲第13項に記載の組成物を含んでいるドナーレザバーを有す る電気輸送デリバリーディバイス。 15.製剤用親ポリペプチド剤の1つ以上のグルタミン残基をヒスチジン残基 で置換して該親ポリペプチド剤の、pH約3.5−8において該親ポリペプチド 剤の電荷より大きい総電荷を有する合成類似体を形成することを特徴とする、製 剤用親ポリペプチド剤を修飾して体表面を通る電気輸送を向上させる方法。 16.製剤用親ポリペプチド剤の少なくとも1つの非ヒスチジンアミノ酸残基 がヒスチジン残基(His)で特徴とする、製剤用親ポリペプチド剤を修飾して 体表面を通る電気輸送を向上させる方法。 17.前記親ポリペプチド剤の残基がグルタミン(Gln)、トレオニン(T hr)及びアスパラギン(Asn)より成る群から選ばれる、請求の範囲第16 項に記載の方法。 18.前記親ポリペプチド剤がグルタミン(Gln)を含んで成る、請求の範 囲第16項に記載の方法。 19.前記類似体が前記親ポリペプチド剤と少なくとも略同等の生物活性を示 す、請求の範囲第15項又は第16項に記載の方法。
