Claims (1)
- 【特許請求の範囲】 1. 少なくとも7、好ましくは約7−24個のアミノ酸残基を有し、実 質的に、式 V−D−J の”VDJ”領域に相当するペプチド(式中、”V”は配列A−Sのジペプチド を含み、”D”は好ましくは2−5のアミノ酸残基を有し、配列L−Gのジペプ チドを含み、”J”は配列N−Q−Dのトリペプチド又はこの塩若しくは作用的 誘導体を含む。)。 2. ”V”はトリペプチド配列A−S−Sを含む請求項1記載のペプチ ド。 3. ”D”はトリペプチド配列L−G−G、トリペプチド配列R−L− G又はペンタペプチド配列L−G−L−G−A(配列番号(SEQ ID NO ):4の残基4−8)のいずれかを含む請求項1記載のペプチド。 4. ”V”のジペプチド配列A−Sは”D”に隣接するトリペプチドの 一部である請求項1−3のいずれか1項に記載のペプチド。 5. ”J”のトリペプチド配列N−Q−Dは”D”に隣接している請求 項1−5いずれか1項に記載のペプチド。 6. 前記”V”はVβ遺伝子によりコードされるタンパク質のC−末端 の少なくとも一部を含むVβセグメントを含む請求項1−5のいずれか1項に記 載のペプチド。 7. 前記VβセグメントのC−末端トリペプチド配列は配列A−Sのジ ペプチドを含む請求項6記載のペプチド。 8. 前記セグメントはVβ遺伝子によりコードされるタンパク質のC− 末端の1から約10までのアミノ酸残基を含む請求項6記載のペプチド。 9. 前記Vβ遺伝子はVβ6、Vβ8、Vβ12及びVβ16からなる 群から選択される請求項6記載のペプチド。 10.”J”はJβ遺伝子によりコードされるタンパク質のN−末端の少 なくとも一部を含む”Jβ”セグメントを含む請求項1−3のいずれか1項に記 載のペプチド。 11.前記JβセグメントのN−末端トリペプチド配列はN−Q−Dであ る請求項10記載のペプチド。 12.前記セグメントはJβ遺伝子によりコードされるタンパク質のN− 末端の1から約10個のアミノ酸残基を含む請求項10記載のペプチド。 13.前記Jβ遺伝子はJβ2.5である請求項10記載のペプチド。 14. 配列A−S−S−L−G−G−N−Q−D(配列番号(SEQ ID NO):1の47−55の残基)を含む約24個までのアミノ酸残基を有 するペプチド。 15.配列A−S−R−L−G−N−Q−D(配列番号(SEQ ID NO):3)を含む約24個までのアミノ酸残基を有するペプチド。 16.配列A−S−S−L−G−L−G−A−N−Q−D(配列番号(S EQ ID NO):4)を含む約24個までのアミノ酸残基を有するペプチド 。 17.配列A−S−S−L−G−A−N−Q−D(配列番号(SEQ ID NO):16)を含む約24個までのアミノ酸残基を有するペプチド。 18.請求項1−3又は14−17のいずれか1項のペプチド又はその補 体をコードするポリヌクレオチド配列を含むDNA構造。 19.請求項1−3又は14−17のいずれか1項のペプチドを含む薬剤 組成物。 20.ワクチンである請求項19の薬剤組成物。 21.請求項1−3又は14−17のいずれか1項のペプチドを含む抗− p277T細胞の認識に関与する抗−イディオタイプT細胞の存在を検出する薬 剤。 22.検出ラベルと接合している請求項21記載の薬剤。 23.請求項1−3又は14−17のいずれか1項のペプチドと第2分子 とを含む複合体。 24.前記第2分子はポリペプチドである請求項23の複合体。 25.前記第2分子は低有機分子である請求項23の複合体 26.個体の抗−イディオタイプT細胞の活性を調節する方法であって、 前記活性は抗−イディオタイプT細胞が抗−p277T細胞を認識する能力に関 連し、前記抗−イディオタイプT細胞の活性を調節するのに有効な請求項1−3 又は14−17のいずれか1項記載のペプチドの一定量を個体に投与する方法。 27.前記活性が増強されている請求項26記載の方法。 28.請求項1−3又は14−17のいずれか1項のペプチドをコードす るDNA配列の少なくとも一部に相補的なポリヌクレオチドを含むオリゴヌクレ オチド。 29.請求項1−3又は請求項14−17のいずれか1項記載のペプチド に対して同原T細胞を活性化し、これを患者に再投与するインシュリン−依存性 糖尿病の予防及び治療方法。 30.請求項1−3又は請求項14−17のいずれか1項記載のペプチド とインビトロ系で接触することにより増殖又はサイトカイン生産性の試験を行な う対象である血清のT細胞の測定を行なうIDDMの診断若しくはステージング の方法、又はIDDMの治療過程の監視方法。
