Description
Claims (1)
- 【特許請求の範囲】 1.増感剤に共有結合したランタニドキレート化剤を含むランタニドキレートで あって、該キレートが、少なくとも106M-1の平衡定数でランタニドと結合出来 、該キレート及び該ランタニドとの錯体が、ランタニド発光を高める事が出来、 そして該増感剤が、一般式、 (ここで、Xは、周期律5又は6族の原子を含む)の、多核異節環芳香族化合 物を含むキレート。 2.該増感剤の第1の位置2〜8炭素原子が、二重共有結合を介して酸素原子で 置換され、該第1の位置2〜8炭素原子と異なる該増感剤の第2の位置2〜8炭 素原子が、該増感剤を該キレート化剤に共有結合する連結基で置換される、請求 項1記載のキレート。 3.該第1の位置2〜8炭素原子が、位置2又は4炭素原子であり、該第2の炭 素原子が、位置7炭素原子である、請求項2記載のキレート。 4.該第1及び第2の位置2〜8炭素原子と異なる、該増感剤の第3の位置2〜 8炭素原子が、置換される、請求項2記載のキレート。 5.該第3の位置2〜8炭素原子が、位置4炭素原子であり、炭化水素又はハロ ゲン置換炭化水素で置換される、請求項4記載のキレート。 6.該増感剤が、2又は4−キノロン又は、2又は4−クマリンから成る、請求 項1記載のキレート。 7.該増感剤が、カルボスチリル124(7−アミノ−4−メチル−2−キノロ ン)、クマリン120(7−アミノ−4−メチル−2−クマリン)、クマリン1 24(7−アミノ−4−(トリフルオロメチル)−2−クマリン)、又はアミノ メチルトリメチルプソラレンから成る、請求項1記載のキレート。 8.該キレート化剤がDTPAから成る、請求項1記載のキレート。 9.試料部分に存在スルカモ知れない被検体を、発光で検出する方法であって、 以下の工程: 試料部分を、請求項1記載のランタニドキレートと、少なくとも106M-1の 平衡定数で該キレートと結合する事の出来るランタニドとの発光錯体と接触させ る工程であって、該キレートが、該被検体に、選択的に結合する事のできる試薬 に共有結合する工程、 該試薬が該被検体と選択的に結合出来る条件下で、該試料部分を培養する工 程、 該試料部分を、該キレートに第1の電子転移を誘発する事のできる第1の波 長の光に曝露する工程、 該試料部分からの光の発光を、第2の波長で検出する工程であって、該第2 の波長が、該第1の波長より長く且つ該キレートでの第2の電子転移から得られ る工程、 を含み、光の該発光の検出が、該被検体試料部分の存在と相関関係にあること を特徴とする方法。 10.試料部分において、第1の位置と第2の位置との間の距離を、発光ランタニ ドキレートドナーと、有機共鳴エネルギーアクセプターとを使用して共鳴エネル ギー移動で検出する方法であって、以下の工程: 該第1の位置に配置された該ドナー及び該第2の位置に配置された該アクセ プターを含む試料部分を、該ドナーでの第1の電子転移を誘発する事のできる第 1の波長の光に曝露する工程であって、該ドナーが、請求項1記載のランタニド キレート及び少なくとも106M-1の平衡定数で該キレートを結合する事の出来る ランタニドとの錯体を含み、該ドナーの発光とアクセプターの吸収とのスペクト ル重複が、ドナー発光強度又は寿命の検出可能な消滅、又はアクセプター発光強 度又は寿命の検出可能な増加によって測定した場合に、該ドナーから該アクセプ ターへのエネルギー移動を可能とするのに十分である工程、 第2の波長での該試料部分からの光の第1の発光強度であって、該第2の波 長が、該第1の波長より長く、該ドナーでの第2の電子転移から得られるもので あり、光の該第1の発光強度が、該試料部分の該第1及び第2の位置の間の 距離と逆の相関関係にある強度、及び 第3の波長での該試料部分からの光の第2の発光強度であって、該第3の波 長が、該第1の波長より長く、該アクセプターでの電子転移から得られるもので あり、光の該第2の発光強度が、該試料部分の該第1及び第2の位置の間の距離 と相関関係にある強度、の少なくとも1つを検出する工程、 を含む方法。
