Claims (1)
- 【特許請求の範囲】 1.低緊縮条件下で図5に示されるヌクレオチド配列からなるプローブまたは その相補体とハイブリッド形成するオピオイドレセプターをコードするヌクレオ チド配列を含有する単離および精製または組換え体DNA分子。 2.上記ヌクレオチド配列がヒトデルタオピオイド、ヒトカッパオピオイドレ セプター、ヒトミューオピオイドレセプター、ネズミ(murine)デルタオピオイ ドレセプターまたはネズミミューオピオイドレセプターをコードする請求項1に 記載のDNA分子。 3.宿主に導入したとき、宿主内でオピオイドレセプターを産生し得る発現系 を含むDNA分子であって、該発現系は、上記宿主内でオペラブルな(operable )異種制御配列にオペラブルに(operably)結合された上記オピオイドレセプタ ーをコードするヌクレオチド配列を含んでおり、その際上記オピオイドレセプタ ーは低緊縮条件下で図5のヌクレオチド配列またはその相補体とハイブリッド形 成するヌクレオチド配列によってコードされるものとして定義される。 4.上記オピオイドレセプターがヒトデルタオピオイドレセプター、ヒトカッ パオピオイドレセプター、ヒトミューオピオイドレセプター、ネズミ(murine) デルタオピオイドレセプターまたはネズミミューオピオイドレセプターである請 求項3に記載のDNA分子。 5.請求項3または請求項4に記載の発現系を含有するように修正された組換 え体宿主細胞。 6.コード化DNAを発現させてオピオイドレセプタータンパク質を産生させ る条件下で請求項5に記載の細胞を培養し、そして培養物から上記レセプタータ ンパク質を回収することを含む上記オピオイドレセプタータンパク質の生産方法 。 7.組換え体細胞表面にオピオイドレセプターが現れる組換え体細胞の産生方 法であって、該方法はコード化DNAを発現させて上記レセプタータンパク質を 上記細胞表面に産生させる条件下で請求項5に記載の細胞を培養することを含む 。 8.請求項7に記載の方法で調製した組換え体細胞。 9.オピオイドアゴニストまたはアンタゴニスト活性について候補物質をスク リーニングする方法であって、該方法は: 上記活性を検出するのに適当な条件下で候補物質の存在下および不存在下で請 求項8に記載の細胞をインキュベートし、そして 上記活性の存在、不存在または量を検出する、 ことを含む。 10.請求項6に記載の方法で産生されたオピオイドレセプター。 11.請求項6に記載の方法で産生されたオピオイドレセプターに対し免疫活 性の抗体を含む、赤血球不含抗体組成物。 12.脊椎動物におけるオピオイドレセプターの解剖的位置の決定方法であっ て、該方法は: 請求項11に記載の抗体組成物を脊椎動物対象に投与し; 上記抗体組成物が上記レセプターとコンプレックスを形成するのに十分な時間 待機し;そして 上記対象内で上記コンプレックスの位置を検出する、ことを含む。 13.オピオイドとオピオイドレセプターとの相互作用の阻止方法であって、 該方法は: 上記レセプターを請求項11に記載の抗体組成物と接触させ;そして 上記組成物を上記レセプターに結合させる、 ことを含む。 14.オピオイドレセプターをコードするDNAの発現調節方法であって、該 方法は請求項1に記載のDNAが該DNAに相補的な標的DNAとハイブリッド 形成する条件下で請求項1に記載のDNAに相補的な DNAを用いて上記発現を行い得る細胞を処理することを含む。
