【発明の詳細な説明】
高められた活性を有する組換シュードモナス外毒素本発明は毒性及び治療能を高
めるように改良された組換シュードモナス(Pseudomanas)由来の毒
素の製造及び利用に関する。より詳しくは、本発明はシュードモナス外毒素のド
メインIa及び一定のドメイン■配列の除去で表わされるアミノ酸配列の中の欠
損を含んで成る外毒素に関する。
発明の背景
成長因子、抗体及びその他の細胞標的分子に付加された毒素は特異的なレセプタ
ー又は抗原を抱える存置な細胞を殺傷するのに利用されうる(Pastanら、
C[!I+ 47 : 641 (1986)及びVitetta ら、5ci
ence 238 : 1098 (1987))。有効な治療的毒素にとって
の−の見込みのある起源はシュードモナス外毒素Aである。シュードモナス外毒
素A (PE)はシュードモナスアエルギノーザ(Pseudomonasae
ruginosa)により分泌される非常に活性な七ツマータンパク質(分子量
66KO)であり、これはADPリボシル化を触媒すること(酸化NADのAD
Pリボシル成分の伸長因子2 (EF−2)への転移を触媒)によるEF−2の
不活性化を通じて真核細胞におけるタンパク質合成を阻害する。
この毒素は、協同で細胞障害を引き起こすように作用する3つの構造ドメインを
含む。ドメインIa(アミノ酸1−252)は細胞結合を媒介する。ドメイン■
(アミノ酸253−364)は細胞質ゾルへのトランスロケーションにとって重
要であり、そしてドメイン■(アミノ酸400−613)は伸長因子2のAPP
リボシル化を媒介し、これはタンパり質を不活性化し、そして死をもたらしめ
る。ドメインIb(アミノ酸365−399)の機能は不明であり続けているが
、しかしその主要部、即ちアミノ酸365−380は細胞障害性の損失を伴わな
いで削除できる。

1991)を参照のこと、PEは、Pa5tan and FitzGeral
d、 5cience 254 : 1173−1177 (1991)に記載
の通り、様々な細胞膜タンパク質を有する細胞を選択的に標的にできうる毒素を
作り上げるために、成長因子、抗体又はCD4と組合されている。
天然のPEは特徴的には肝臓不全に基づく死をもたらす。PEを有する免疫毒素
も肝臓を攻撃し、そしてもっと大量(20〜250倍多く)の用量で付与すると
肝R毒性に基づく死を招いてしまいうる。宿主における非特異的毒性が下がり、
且つ治療効能が高まったPEの改良形態が非常に所望されている。細胞結合性ド
メインIaを欠<PEの変異性は、非特異的な毒性の程度を下げながら有効であ
ることが見い出されている。例えば米国特許第4.892,827号を参照のこ
と。
発明の概要
本発明は従来述べられている分子よりも高い活性を示す改良組換えシュードモナ
ス外毒素分子を開示する。更に、ここに記載の発見は、サイズにおいてより小さ
く、免疫原性が低い傾向にあり、標的細胞の細胞質ゲルに侵入でき、そして腫瘍
の内部に浸透し易いPE分子を作り出すことを可能にする。
細胞障害性とするには、天然PEは細胞内でタンパク質分解的に切断されねばな
らない(Ogata ら、J、 Biol、 Chew、 265 : 206
7B−20685(1990))、この切断はアミノ酸279と280との間で
起こる。この切断の重要性は、この部位で切断することのできないPEの突然変
異形態が無毒であることを示唆する結果で説明されている。Oga ta ら、
前掲、しかしながら、細胞による切断はあまり効率的でない0本発明は細胞によ
る切断を必要としないPE誘導体を構築することにより非効率的な切断の問題を
解消することを狙いとする。かかる[事前切断型JPE分子の高められた能力を
有し、その理由は細胞質ゾルに至る活性毒素フラグメントの輸送の効率が高まっ
ているからである。
本発明は組換シュードモナス外毒素分子を含み、そのドメインIaは欠損してお
り、そしてそのドメイン■のアミノ末端から最初の27個のアミノ酸より多くは
欠損していない、好適なPE分子はドメインHのアミノ酸位置280におけるメ
チオニンで始まり、ドメインIbのおよそのアミノ酸365から380までの欠
損を含んで成り、そしてアミノ酸位置287においてシスティンの代わりとなる
セリンの置換を含んでいる。好適な分子には、その分子のカルボキシル末端にお
いて、REDIJ (Seq、 10 No、 14)、 REDL (Seq
、 ID No、 15)及びKDEL(Seq、 10 No、 16)より
成る群から選ばれるアミノ酸配列を有するものも含まれる。典型的なPE分子は
ほぼアミノ酸280から613より本質的に成るか、又はほぼアミノ酸280か
ら364及び381から613より本質的に成る。
このPE分子はリガンド結合因子、例えば抗体又はその結合性フラグメント、成
長因子、ホルモン、サイトカイン等に融合されていてもよい、このリガンド結合
因子は好ましくはほぼアミノ部位607の後に挿入し、そしてアミノ酸604−
613をリガンドのC末端に置く。
我々は、ドメイン■の中の一定の配列のみが結合性タンパク質を細胞質ゾルの中
にトランスロケートさせるのに必要であることを示したため、本発明のPE分子
は様々なペプチドを細胞へと輸送するのに利用されうる。ドメイン■はPE分子
から削除してよく、そしてワクチンとして又は遺伝子治療用途に利用するための
その他のペプチドで置き換えられてよい。
このPE分子は、ドメインlaの欠損及びドメインHのアミノ末端の中に欠損を
有することによっても特徴付けられ、この分子量よ細胞障害アッセイにおいてP
E40 (以下に説明)よりも標的細胞に対して少なくとも20倍細胞障害性で
ある。ここで、本明細書記載のPE40及び組換PE分子の標的細胞に対する細
胞障害性は、その標的細胞に対して、その標的細胞に特異的なりガント結合因子
に融合したPE40及びこの標的細胞に特異的なりガント結合因子に融合した組
換PE分子をアッセイすることにより測定する。
このPE分子のアミノ酸配列をエンコードする核酸配列を含んで成るヘクター及
びこの分子を発現する宿主細胞も考慮する。更Gこ、ここに記載の新規分子を有
する、癌及びその他の症状を処置するための薬理製剤及び方法を含む。

)によるA431細胞からの(”I) −EGFの放出(displaceme
nt)を示す、 A431細胞に結合している(1!5t)−EGFをdpmと
して測定し、そして毒性抜きでインキュベートした細胞のdpsの%とじて表わ

詳細な説明
本発明は高められた細胞障害活性を有する組換シュードモナス外毒素に関連し、
それにおいてはドメインHのアミノ末端の一部が欠損している。この分子は別の
標的分子に結合又は融合されており、従ってその改良型細胞毒素は所望の細胞を
標的とするようになっている。
天然PHは配列10表No、1に示している0本明細書に記載されている全ての
アミノ酸配列の位置は、対照のフレームとしてこの配列表を利用している0例え
ば、「本質的にほぼアミノ#!280〜613より成るJPE分子は、配列In
表No、1上のそれに実質的に対応する位置のアミノ酸を有する分子を意味して
いる。配列ID表No、1を対照のフレームとして利用している配列についての
欠損又は置換を示すためにその他の一般的な対照を本明細書では用いている。記
号「Δ」の利用は、その記号の後方のアミノ酸の欠失を意味する0例えば、「Δ
365−380」とは、PE分子からのアミノ酸365〜380の欠損を意味し
ている。アミノ酸置換はかっこにより示すことができ、例えば’ (set28
7) Jとは、アミノ酸位置287にセリンを有する分子を意味している。アミ
ノ酸はここでは時折り、IUPAC−TUB生化学命名協会により推奨されてい
る単文字コードによって表わしている。
本発明の数多くのPE分子は、標的細胞に特異的なりガント結合因子と複合され
ているとき、標的細胞に対するその高められた細胞障害性により固存に特徴付け
られる。この高められた細胞障害性は、天然分子又はドメインHに有意義な欠損
がないもの、例えば下記の実施例の章並びに引用することで本明細書に組込む一
般譲渡されたU、 S、 S、 N、 07/459.635及びU、 S、
S、 N、 071522,182に記載のPE(4E)又はPE40の使用に
比して認められる。細胞障害性の上昇を決定するためのアッセイは、課題のPE
分子とリガンド結合因子とを含んで成る融合タンパク質を、対照のPE分子、例
えばPE40と前記のりガント結合因子とを含んで成る融合タンパク質と比較す
るアッセイである。次に対応の融合タンパク質を、リガンド結合因子に対して特
異的な細胞に対する細胞障害アッセイで試験する。得られるIDs*(以下に定
義)は、リガンドレセプターから50%のラベル化リガンドを放出させる毒素の
濃度を、リガンドレセプターを抱える細胞上の組換毒素のID3゜で除すること
によってその値を調整することにより、細胞障害指数を得るように調整されうる
。次に各PE分子についての細胞障害指数を比較する。リガンド結合因子として
のTGFα及びEGF レセプターを抱えるA431細胞を利用する典型的なア
ッセイを下記に捷供する実施例に記載している。PE40又はドメインHの欠損
を有さないことが認められたその他のPE分子よりも約20倍以上、好ましくは
約60倍以上、そして最も好ましくは約300倍以上の適正な細胞障害指数を有
するPE分子が所望される。ドメインIaを欠<PE分子は、ドメインn、Ib
及び■を発現するプラスミドpJDにより発現されうる。プラスミドpJH8は
引用することで本明細書に組入れる米国特許第4,892,827号に記載され
ており、そしてRockville。

チドと競合する能力を測定する。
好適なPE分子はドメインIaが欠損しており、そしてドメインHのアミノ末端
から最初の27個より多くのアミノ酸が欠損していないものである。これは実質
的にはアミノMlから27aの欠損を示す。
この欠損によりもたらされる細胞障害性の利点は、下記の欠損があると大いに低
下する: 1−281 ; 1−283 ; l−286;及び314−380
、ドメイン■のアミノ末端からの、29.31.33又は36のアミノ酸では

本発明の組換PE分子はリガンド結合因子に、当業者に知られ、且つ利用されて
いる任意の方法により融合又はそうでなければ結合させることができうる。これ
らの2つの成分は任意の公知の様々な化学手順によって互いに化学結合させるこ
とができうる。例えば、その結合はへテロニ価架橋剤、例えばSP叶、カルボジ
イミド、グルタルアルデヒド等を介してよい。様々な免疫毒素の製法が当業界に
知られており、そして例えば、共に引用することで本明細書に組入れる、’Mo

出せうる。組換PE分子を抗体と一緒に用いるためには、アミノ酸位置287に
おいてシスティンを有するPE分子が、その毒素を抗体又はその他のりガントに
システィンのチオール成分を介して複合させるのに好ましい。
このPE分子は、E、コリ(大腸菌)の中での一本鎖抗体の生産を通しての如き
の組換手段によってリガンド結合因子に融合させてもよい。タンパク質をエンコ
ードする遺伝子は、当業者に知られる任意のクローニング手順によってcDNA
又はゲノム形態においてクローンさせることができうる。例えばSambroo

(1989)を参照のこと。
リガンド結合因子、特にTGFα(形質転換成長因子α)の如きの小さめの因子
は、PE分子のドメイン■内の位置に挿入するのが所望される。最も好ましくは
、このリガンド結合因子はPE分子のほぼアミノ酸位置607と604 との間
に融合させる。このことは、そのリガンド結合因子がこの分子のほぼアミノ酸6
07の後ろに挿入されており、そしてPEの適当なカルボキシル末端が、PEの
アミノ酸はぼ6o4−613をこの結合因子の後ろに置くことにより再生された
ことを意味している。従って、このリガンド結合因子は組換PE分子の中でほぼ
アミノ酸607の後ろに挿入され、そしてドメイン■のアミノ酸6o4−613
がそれに続く。所望の抗体由来の■、及びVHrtl域もドメイン■内に一本鎖
形態で挿入してよい。

当業者は、リガンド結合因子及びPE遺伝子に追加の改良、欠損、挿入等を施し
てよいことを認識するであろう、特に、標的細胞に対する融合タンパク質の細胞
障害性を高めるために、又は抗体についての抗原を有さない細胞に対する非特異
的な細胞障害性を低めるために、PEの中に、又は抗体遺伝子をPEに連結する
リンカ−の中に、欠損又は変更を施すことができうる。かがる構築体は全て、当
業者によく知られている遺伝子工学の方法により作られることができ、そしてそ
れらを様々な臨床又は生物系的用途にとって極めて適当なものに仕上げる親和性
、特異性、安定性及び毒性の様々な性質を有するタンパク質を産生せしめうる。
PE分子を含む本発明の融合タンパク質は様々な宿主細胞、例えばE、コリ、そ
の他の細菌宿主、酵母、並びにその他の高等真核細胞、例えばCO5,(:HO
及びHeLa細胞系及びミエローマ細胞系において発現されうる。その組換タン
パク質遺伝子は各宿主にとって適切な発現コントロール配列に作動連結されてい
るであろう。E、コリにとっては、これにはプロモーター、例えばT7. Tr
p又はラムダプロモーター、リポソーム結合部位及び好ましくは転写終結シグナ
ルが含まれる。真核細胞にとっては、そのコントロール配列にはプロモーター、
及び好ましくはイムノグロブリン遺伝子、SV40、サイトメガロウィルス等に
由来するエンハンサ−1並びにポリアデニル化配列が含まれ、そしてスプライス
ドナー及びアクセプター配列が含まれうる0本発明のプラスミドはよく知られて
いる方法、例えばE、コリにとっての塩化カルシウム形質転換並びに哺乳類細胞
にとってのリン酸カルシウム処理又はエレクトロポレーションによって特定の宿
主細胞の中に移入することができうる。プラスミドにより形質転換された細胞は
、プラスミド上に含まれるamp、 gpt、 neo及びhyg遺伝子の如き
の遺伝子により授けられる抗生物質に対する耐性により選別されうる。
発現したら、この組換融合タンパク質は当業界の標準の手順、例えば硫酸アンモ
ニウム沈殿、アフィニティーカラム、カラムクロマトグラフィー、ゲル電気泳動
等に従って精製されうる(一般的には、R,5copes、 Protein
Purification、 Springer−Verlag、 N、 Y、
(1982)を参照のこと)、少なくとも約90〜95%の均一性の実質的に純
粋な組成物が好ましく、そして98〜99%又はそれより高い均一性が薬理用途
にとって最も好ましい0部分的に、又は所望の均一性にまで精製したら、そのポ
リペプチドは治療的に使用されうる。
本発明の組換融合タンパク質及び薬理製剤は非経口投与、例えば静脈内投与、又
は身体の腔もしくは器官の内腔への投与にとって特に有用である。投与のための
製剤は一般に薬理学的に許容される担体、好ましくは水性担体の中に溶解されて
いるPE分子融合タンパク質の溶液を含んで成るであろう。様々な水性担体、例
えば緩衝食塩水等が使用されうる。これらの溶液は無菌であり、そして一般に不
要の物質を含まない、これらの製剤は慣用のよく知られた除菌技術によって除菌
されうる。これらの製剤は、生理学的条件に近づけるための必要な薬理学的に許
容される補助物質、例えばpH18!整及び緩衝剤、毒性調整剤等、例えば酢酸
ナトリウム、塩化ナトリウム、塩化カリウム、塩化カルシウム、乳酸ナトリウム
等を含みうる。これらの製剤の中での融合タンパク質の濃度は幅広く変えること
ができ、そして主に流体容量、粘度、体重等に基づき、選ばれた投与の特定の態
様及び患者の要望に応じて選ばれるであろう。
従って、静脈内投与にとっての典型的な薬理製剤は1日当り、患者当りで約0.
1〜10mgであろう、1日当り、患者当りで0.1〜約100mgの用量は、
特にその薬剤を血管ではなく隔離部位、例えば身体の腔又は器官の内腔に投与す
るときに利用されうる。非経口投与用製剤を調製するための実際の方法は当業者
に公知又は自明であり、そしてRemington’s Pharmaceut
ical 5cience第15版、Mack PublishingComp
any、 Easton、 Penn5ylvania (1980)の如きの
公開物に詳しく記載されている。
本発明の融合タンパク質又はそのカクテル(即ち、その他のタンパク質との)を
含む製剤は治療的処置のために投与されうる。治療的用途においては、製剤は疾
患に苦しむ患者に、その疾患を及びその合併症を治癒させる又は少なくとも部分
的に緩和させるのに十分な量で投与される。これを達成させるのに適当な量を「
治療的に有効な量」と定義する。この用途にとって有効な量は疾患の症度及びそ
の患者の健康の一般的な状態に依存するであろう。
該製剤の一回又は複数回の投与を、患者に必要とされ、且つ許容される用量及び
頻度に応じて投与せしめてよい、あらゆる事態においても、この製剤はその患者
を効果的に処置するのに十分な量の本発明のタンパク質を供すべきである。
本発明の組換融合タンパク質の用途の中にはとりわけこのタンパク質の毒性作用
によって排除されうる特定のヒト細胞により引き起こされる様々な疾患症状が含
まれる。−の好適な用途は例えばりガント結合因子としてのTGFαの利用によ
る癌の処置、又はT及びB細胞により生ずる自己免疫症状、例えば移植片一対一
宿主疾患、器官移植拒絶、I型糖尿病、多発性硬化症、リウマチ様関節炎、全身
性エリテマトーデス、重症筋無力症等の処置である。この融合タンパク質はイン
ビトロで、例えば移植する前での骨髄からの存寄な細胞の排除にも利用されうる
。融合タンパク質のりガント結合因子部分は意図する用途に従って選ぶ、結合因
子にとっての標的として働きうるT細胞の膜上のタンパク質にはCD2 (Tl
l)、 CD3. CD4及びCD8が含まれる。標的として働きうるB細胞上
に主に見い出せるタンパク質にはC[110(CALLA抗原) 、CD19及
びCD20が含まれる。 CD45はリンパ球様細胞上に幅広く認められる、可
能性のある標的である。結合因子にとってのこれら及びその他の可能性のある標
的リンパ球抗原は、Leucocyte Typing fir、 A、 J、
McMichael W、 0xfordtlniversity Pres
s、 1987に記載されている。この結合因子にとっての標的として働きうる
癌細胞上に見い出せる抗原には、癌胎児性抗原(CEA)、トランスフェリンレ
セプター、P−塘タンパク譬、C−erbB2 、及び癌についてのモノクロー
ナル抗体免疫抱合体の第3回国際会議(Third Internationa

1988年)の誓約書に記載の抗原が含まれる。当業者は、上記の細胞以外のそ
の他のタイプ上で発現されるレセプター、例えば脱糖タンパク質又は成長因子又
はホルモンレセプター、例えば表皮成長因子レセプター等に結合するりガント結
合因子を選択できうることを理解しているであろう。
ここに記載のPE分子、そして最良には以下に記載のPE37及びPE35によ
り代表されるものは、遺伝子治療用途におけるシグナル配列として、又は例えば
ワクチンの使用に伴うときのようにシグナル配列が有用となるその他の用途にお
けるシグナル配列としても働くであろう。かかる用途においては、PE分子のド
メイン■のかなりの欠損が所望の抗原に置き換えられていてよい、「ドメイン■
のかなりの欠損」とは、ドメインの大部分が欠損している、即ちそのドメインの
機能が不活性化されている、又は破壊されていることを意味する。
この分子におけるドメイン■のほぼアミノ酸604−613の保持は非常に所望
される。
例えば、AIDS又は癌を処置するワクチンを作るためには、所望のタンパク質
の一部をドメイン■の・場所に挿入してよく、そしてリガンドは組換タンパク質
を抗原表示細胞に結合させるように所望のタンパク質とPEのカルボキシル末端
との間に挿入する。遺伝子治療にとっては、DNA配列はドメイン■の場所に挿
入してよい。

「組換」とは、課題の産物が遺伝子の新たな又は非天然的な組合せに至る操作の
結果物であることを意味する。
「ベクター」とは、DNA配列、典型的にはプラスミド又はウィルス形態におけ
るDNA配列であり、これは宿主の中で複製することが可能である。ベクターは
DNA配列を輸送又は操作するのに利用できうる。「発現ベクター」には、その
中に含まれているDNA配列を発現させてタンパク質産物を産生ずることのでき
るベクターが含まれる。コード配列はその発現を及ばしめることのできる別の配
列、例えばプロモーター及びエンハンサ−に連結している。
「有意義な毒性抜きで」なる表現は、本発明の融合タンパク質が、標的でない細
胞の機能を任意の感知できる程度に、又は任意の異常なレベルにまで影響を及ぼ

49が全てグルタミン酸に置き換えられ、TGFαがアミノ酸607の後ろに置

TGFαの後に置くことにより再生されている突然変異した不活性の天然結合ド
メインを有する全長PEである。
Hut 102細胞はT、 Waldw+an、 Leonardらからの贈呈

他の細胞系は全てAmericanType Cu1ture Co11ect
ion (Rockville、 MD)に由来する。
B、増幅−オリゴヌクレオチドC1,C2,C7及びC8は表1に詳細しており
、そしてこれらはDNA合成装置(Applied BiosysLe+ss、

Foster C1ty、 CA)を用いて構築した。ポリメラーゼ連鎖反応(
PCR)の反応は鋳型としての10ngのpCT4 (下記の参照)及び製造者

TCCNo、 が付与されている。プラスミドCT4 (pCT4)は、プラス
ミド4735/4Eの551 bp (塩基対)のBamHI −EcoRIフ
ラグメントと同一の[lNAフラグメントと、プラスミドDPIの3.6 kb
のBawHI −EcoRI脱リン酸化フラグメントとのりゲートによって作っ
た。プラスミドCT4はPH37/TGFα(PE280−613/TGPα)
と呼ぶタンパク質をエンコードする。
PE37欠損突然変異体は、プラスミド叶1の中に見い出せるNde 1−5a
cU制限部位へのNde I −Sac U消化PCRフラグメントの挿入によ
り作り上げた。プラスミドCT2は位置282のメチオニン、及び位置287を
除く天然PEのアミノ酸283−613をエンコードする。プラスミドCT3は
位置284のメチオニン、及び位W287を除く天然PEのアミノ酸285−6
13をエンコードする。プラスミドCT14は位置287のメチオニン、及び天
然PEのアミノ酸288−613をエンコードする。
PE37に由来するアミノ酸314−380の内部欠損を含むプラスミドCT8
を、アニールさせたオリゴヌクレオチドS3及びS4 (表1)の、プラスミド
DPIの3.9 kbの5ail−^palルミlフラグメント入により作った
。4つのプラスミド全ての配列をDNA配列決定により確認した。全ての突然変
異プラスミドをBamHI及びEcoRIで制限消化し、そしてプラスミドVC
4735/4Eの551 bpのBan+)I T −EcoRIフラグメント
と同一のDNAフラグメントにリゲートさせて突然変異プラスミドCT2/T、
pCT3/T、 pcT14/T及びpCT8/Tを作り上げた。これらのプ
ラスミドを制限分析により確認したところ、PE282−613/ TGPα、
PE284−613/ TGFα、PE287−613/ TGFα及びPE3

質のために用意した。 TGFαドメインを含むタンパク質に関しては、融合タ
ンパク質はKrei twaanら、前掲に記載の通り封入体から精製した。
グアニジンで抽出し、そしてPBSへの急速希釈により再生させたペリプラズマ
又は封入体を、Phar+ncia LKB Biotechnolgy、 I
nc。
(Piscataway、 NJ) FPLCを用い、Qセファロース、モノQ
HR515(Pharmacia−LKB、 Inc、、 Piscataw
ay、 NJ)又はPorous A/F (PerceptiveBiosy

使用により精製した。引用することで組入れるLae++nli、 Natur
e 227 : 680−685(1970)に記載の5O5−PAGEをカラ
ム画分を分析するために用いた。
PE含有タンパク質の同定は、ポリクローナルウサギ抗−PE抗血清及びVec
tastainキット(Vector Labs+ Burlingame+
Ca)を用いるイムノブロッティングにより確認した。
E、タンパク質合成■害アッセイータンパク質合成の阻害は引用することで本明
細書に組入れるPr1orら、Ce1l 64 : 1017−1023(19
91)に記載の通りに実施した。細胞を毒素添加の前に、96穴プレートの中で
各ウェル当り15.000細胞において24時間平板培養した。
0.2%のBSA−PBS(生血清アルブミンーリン酸緩衝食塩水)の中に希釈
した毒素又はコントロールを200μl/ウエルの最終容量にまで加えた。37
°Cで16〜20時間のインキュベーションの後、各ウェルに〔コH]−ロイソ
ン(Amersham Corp、、 ArliArlln Heights、
IL :1μCiを0.2%のBSA−PBSの中に10μlまで希釈)で2
時間パルスに付した。凍結後、その細胞をガラスファイバーフィルターで集め、
そしてタンパク質の中への放射活性の取り込みをBetaplateシンチレー
ノヨンカウンター(Pharmacia、 LKB)で定量した。結果は、毒素
抜きでインキュベートした細胞の取り込みcps’(分当りのカウント数)の%
とじて算定した。競合アンセイを2μg/mlのEGFの存在下で細胞に付加し
た毒素を利用して行った。
F、 ADP−リボシル化アッセイ−タンパク質サンプルのADP−リボシル化
活性を、伸長因子2に冨む小麦胚芽を用いてCo11ierとKandelの手
順により測定した。引用することで本明細書に組入れるJ、 Biol。

G、(”’I)−EGF放出研究−^431細胞(ヒト表皮癌細胞)を24穴プ
レートの中で1mlの培地中で、ウェル当り8,000個の細胞において平板培

、0.8.4.20又は100 pmolの毒素のいづれかと組合せた(+zs
l)−EGF (New England Nuclear+ Inc、、Bo
ston、MA) 0.5Hg (0,05μCi)を含む結合バッファ−20
0μmで処理した。4°Cで口・フカ−上で90分平衡にした後、それらの細胞
を結合バッファーで洗い、0.5%のsos及び1mHのEDTAを含む10m
Mのトリス−HCl、 pH7,4で溶解し、そして結合リガンドをガンマ−検
出器で計測した。
H131KOのカルボキシル−末端フラグメントのデザイン−PHの37KOの
カルボキシル−末端フラグメント(PE37)が細胞質ゾルへとトランスロケー
トされ、そしてタンパク質合成を抑制しているかどうかを調べるため、我々は位
置280においてグリシンの代わりにメチオンで始まり、それに天然PEのアミ
ノ酸281−613が続くタンパク質をエンコードするプラスミドDPIを構築
した。天然PEにおいて、メチオニンによるgly 280の交換は細胞障害性
を低めない、更に、システィン287をセリンに変え、不適切なジスルフィド結
合の数を減らした。その結果、PH37はドメインrbにおいて位W372及び
379に位置する2個のシスティン残基のみを有する。プラスミド叶lのDNA
配列決定はそれが所望の配列を有することを確証せしめた。プラスミドDFIの
中に存在しているT7プロモーターを用いることにより、PE37はBL21
(λDE3)の中で発現され、そして細胞画分を5O5−PAGEにより分析し
たとき、ベリプラズマとスフェロプラストとに均等に分布していることが認めら
れた。毒素をペリプラズマから、アニオン交換クロマトグラフィー及びゲル濾過
を利用して〉90%の均一性にまで精製した。その精製タンパク質は5OS−P
AGE上で予測の分子量(37KD’)を有し、そして抗−PE抗体と免疫反応
性であった(図2及び3)。タンパク質の配列決定は14個のアミノ末端アミノ
酸を確証した(MWEQLEQSGYPVQR) 、 ADP リボシル化活性
は、天然PEと同一のADP リボシル化活性を有する分子PE40のそれと同

8B)を参照のこと、いくつかの細胞系で試験したとき、PE37はその標的細
胞に結合できないために非常にわずかな細胞障害活性しか有していなかった。
1、 PE37/ TGFαのデザイン及び活性−PE37をそのアミノ末端を
改変することなく細胞を標的化できるようにするため、TGFαをエンコードす
るcDNAを挿入せしめてTGFαがPE37のアミノ酸607の後ろに置かれ
、且つPHのカルボキシル末端が、TGFαの後にPEのアミノ酸(604−6
13)を置くことにより再生されているプラスミドを構築した。この位置に挿入
され、且つ不活性細胞結合ドメインをも含む毒素分子PE (4E) /TGF
α(PE664G”−TGFαとしても知られる)はEGP レセプターを抱え
る細胞に対しては非常に細胞障害性である。
Krei tmanら、前掲を参照のこと。
構造を図1に示す融合タンパク質PE37/TGFαをBL21 (λDE3)
の中で発現させ、そしてほとんど独占的に封入体の中で見い出された。
このタンパク質を7Mのグアニジンの中に溶かし、リン酸緩衝食塩水で再生し、
そしてその他のTGFα含有組換タンパク質について先に記載した通りにアニオ
ン交換及びゲル濾過で〉90%の均一性にまで精製した(Kreitmanら、
前掲) 、 5os−PAGE上での予測の分子量(43KD )で泳動した精
製タンパク質はPEに対する抗体と免疫反応性であり(図2及び3)、そしてそ
の他のPE分子と比べたときに完全たるADP リボシル化活性を有していた(
表2)、その組換タンパク質を様々な数のEGPレセプクーを発現する細胞系に
対して細胞障害性であり、そしてその細胞障害作用は存在するレセプターの数に
相関していた(表3)。更に、EGFレセプターを有さないHUT 102細胞
はPE37/TGFαの毒性作用に耐性であった。A431ヒト表皮癌細胞に及
ぼす細胞障害性は過剰のEGFを用いることで完全に阻害され、PE37/TG
FαはEGF レセプターを介して特異的に結合することを示唆する。 A43
1細胞に対するPE37/TGFαのI[lS。は、ドメイン■全体を含む分子
PE (4F) /TGFαより低く、従って細胞質ゾルにトランスロケートさ
れる前にプロセスされねばならない(図4)。このデーターはPE37/TGF
αが非常に活性であり、且つ特異的な組換毒素であることを示唆する。

変異体−PE37/TGFαのアミノ末端部分の調査は、PEの37KDのフラ
グメントがBアルファーヘリンクス(aa 287−308)へと導かれる負帯
電リーダー配列を形成する7つのアミノ末端アミノ酸(MWEQLEQ)を含む
ことを示した。 PE37のアミノ末端が活性にとって必須であるかを決定する
ため、一連の欠損突然変異体を構築した。それにおいては2.4又は7個のアミ
ノ酸がアミン末端から欠損している(図1及び表1)、第四の突然変異体を構築
し、これは通常のアミノ末端を、しかしながら大きめの内部欠損(アミノ酸31
4−380)を含む、 EGF レセプターを含む細胞系に及ぼす細胞障害性に
ついて試験することができるようにするため、TGFαをこれらの組換タンパク
質金てのカルボキシル末端の近くに置いた(図1)。
TGFαを含む全ての突然変異タンパク質をBL21 (ス[IE3)の中の封
入体として発現させ、そして前述の通りに精製した。〉90%の均一性にまで精
製され、5O5−PAGE上で適切な分子量で泳動した突然変異タンパク質はウ
サギ抗PE抗体に対して免疫反応性であり(回2及び3)、そして全ADP リ
ボシル化活性を有していた(表2)。A431及びMCF7細胞に対する■D、
。を表2並びに図4及び5に詳細する。PE37/TGFα(PE280−61
3/ TGFα)は活性が12〜25分の1に低下している2又は4個のアミノ
末端アミノ酸の欠損した最も活性な分子であった。7個の末端アミノ酸の欠損は
更に活性を低下させ、そして内部欠損は大幅な細胞障害の損失をもたらした。
細胞障害性の低下は組換毒素のEGFレセプターへの低められた結合能に原因し
うる。 TGFαは3本のジスルフィド結合を有するため、様々な不適切な折り
たたみ形態が再折りたたみプロセスの際に生じうる。 Kreit霧an、前掲
、EGFレセプター結合における相違がPE37/TGFα及びその他の組換毒
素間での10.。の相違に原因するかどうかを決定するため、(”5r)−EG
F放出アッセイを行った(図6)、各毒素の細胞障害活性をEGFレセプター結
合における相違について補正した。補正した細胞障害指数値は、EGF レセプ
ターから50%の(”’[)−EGPを放出させる毒素の濃度(nM)をへ43
1細胞に対する組換毒素の10.。で除することによって算定した。この指数は
、PE37/TGFαの、ここに記載のその他の突然変異体及びPE(4E)
/TGFαに比しての優れた細胞障害活性を強調せしめる。
上記の37KDタンパク質(PE37と呼ぶ)は好適な態様である。アミノ末端
メチオニンはアミノ酸位280にあるため、この分子はそれが細胞質ゾルにトラ
ンスロケートされるのにタンパク質分解又はジスルフィド結合還元のいづれに付
される必要もないであろう0位置280でのグリシノのメチオニンへの置換は細
胞障害性を下げない、 PE37を標的とするために−キメラ分子を、τGFα
をエンコードするcDHAを用いて作り上げた。 PE37/TGFαは標的細
胞を特異的に殺傷するように働き、なぜなら細胞障害性は過剰のEGFにより阻
害され、一方、EGF レセプターを欠< 1IUT102細胞はPE31/T
GFαに非感受性であったからである。アミノ酸253−280はタンパク質分
解プロセスを助長するためのみの毒素機能にとって明らかに必要である。
標的細胞によるPEの代謝の分析は細胞結合PE分子の約10%がプロセスを受
けたことを示す、Ogata ら、J、 Biol、 Chem、 265 :
20678−20685 (1990)を参照のこと、このことは、タンノぐ
り質分解プロセスがPEの作用において律速段階でありうることを示唆する0表
2番よ結合PE37/TGFαの方が、PE (4E) /TGFαよりも20
倍効率的に細胞質ゾルに到達することを示唆する。トランスロケートするPH(
4E)/TGFαのフラグメントはPE37/TGFαとほとんど同一であるた
め、実際の脱トランスロケーション段階は両分子に関して類(以してし)る。
従って、タンパク質分解プロセスはPR(4B) /TGFαの細胞障害性を制
約する傾向が強いようである。
事前の研究(Siegall ら、Biochem 30 : 7154−71
59 (1991))4よ、アミノ酸346−380は活性の損失を伴うことな
りPEから欠損されうるが、しかし位置345での追加の突然変異は有害である
ことを示している。残5314−380を欠< PE37/TGFα突然変異体
の劣る活性番よ314−346の間にある残基がトランスロケーションにかかわ
ってし)ることを示唆する。この要件は37KOの活性フラグメントを作り上番
ヂるのに必要不可決である。従って、トランスロケーションにとって重要である
ドメイン■の部分は位11280−345にある66個のアミノ酸のようである
。ドメインIb及び■(アミノ酸364−613)もトランスロケーションにと
って必要であり、なぜならそれらはPr1orら、Bioche+wistry
31 : 3555−3559 (1992)に記載の通りリボヌクレアーゼ
ノ\ルナーゼで置き換えられて細胞障害性分子を作り上げること力(できる力・
要である。 PE37/TGFαのアミノ末端からの2,4又番よ7個のアミノ
酸の欠損を有する突然変異タンパク質はPE37/TGFαよりも活性力(弱い
。EGF レセプターに対する結合について補正したとき、12〜25イ合の細
胞障害活性の損失が、2又は4個のアミノ末端アミノ酸を欠く突然変異体におい
て認められた。これらのアミノ末端欠損突然変異体はそれぞれ1個のグルタミン
及び正味の負帯電を保持している。
補正した細胞障害性において更に10倍の低下が7個のアミノ酸の欠)員により
認められた。
11.35KDのカルボキシル−末端PRフラグメント及び抗体−PE融合タン
パク質の構築。
A、材料及び細胞系−何らかのことわりのない限り、材料及び細胞系は先の実施
例のもとで記載したものと同一の起源より入手した。
B、増幅−オリゴヌクレオチドC9及びC2(表1参照のこと)をON八へ成装
置(^pplied BjosysLes+s)を用いて構築した。ポリメラー
ゼ連鎖反応(PCI+)は鋳型としてのLongのプラスミド叶1(上記)及び
製造者の仕様書(Gene Asp : Perkin−Elmer Cetu
s Instruments。
Norwalk、 CT)に従う試薬を用い、5%のホルムアミド(Fluka
Chemika)及び100 pIIIolのプライマー09と02の存在下で
実施した。各PCR反応は94°Cで1分の変性、42°Cで90秒のアニーリ
ング及び72℃で2分の重合、それに伴う各サイクルにおける10秒の伸長より
構成される全部で30のサイクルとした。増幅フラグメントを1.5%の低融点
アガロース(Sea Plaque ; FMCCarp、、 Rocklan
d+ ME)で精製した。
C9細菌株及びプラスミド−上記の)18101をプラスミドの増殖のために用
いた。誘発性T7 RNAポリメラーゼをプロファージ上に保有するBL21
(λDε3)(これも上記)を融合タンパク質発現のための宿主として用いた。
プラスミドCTl32は、Nde I −5acI[消化PCRフラグメン)
(C9及びC2を用いて増殖)の、上記のプラスミドDPIの脱リン酸化3.6
kbのNde I −Sac IIフラグメントへの挿入により作った。プラ
スミドCT11は、ドメイン1bからのアミノ酸365−380の欠損を含むP
E突然変異体をエンコードするプラスミドC3lO(Siegallら、Bio
chem、 30 : 7154−59 (1991)) と同一のDNA配列
を含むプラスミドの515 bpのSal I −BasHIフラグメントの、
プラスミドCT132ノ3.7 kbのSal I−Bawl脱リン酸化フラグ
メントとのリゲーションによって作り上げた。プラスミドCTIIをDNA配列
決定により確認し、そしてメチオニン及び天然PEのアミノ酸281−364.
381−613より成るPE35と称するタンパク質をエンコードする。
D9組換融合タンパク質の発現及び精製−シュードモナス外毒素タンパク質の発
現を、先の実施例に記載の通り、宿主BL21 (λDE3)を用いて行った。
細胞を、IPTGでの誘発に続いて90分インキュベートした。そのペリプラズ
マ画分をPE35及びN1ys PE38の精製のために用意した。 N1ys
PE38は、容易に誘導化されるリジンを含む11個のアミノ酸のペプチドが
先行するPEのaa 253−364.381−613を含む突然変異PEタン
パク質である。N1ys PE38は、先の実施例に記載のPhar+5aci
a LKB Biotechnology Inc、 FPLCを用いるQセフ
ァロース、モノQ (HR515; Pharvacia)及びTSK−250
(Tosohaas。
Montgomeryville、 PA)カラムの連続使用により精製した。
PE35を含むペリプラズマをQセファロースから、20mMのトリスpH7
,4中の0.26〜0.30μのNaC1でのT8離より精製した。その溶離液
を、1MのNaC1を含む50mMのトリス−アセテートpH7,0中のImg
/mlのCu5Oaで50%飽和となったキレート化性セファロースカラム(P
harllacia)上に注入した。そのフロースルーはほぼ純粋なPE35を
含み、それは10mM+71EDTA及び10mM(7)DTTを含むPBS中
でのTSK−250カラムにおいて七ツマ−として精製された。
Lae腸mli 、前掲の方法を用いる5O3−PAGEをカラム画分を分析す
るために用いた。 PE含をタンパク質の同定はPEと反応性のウサギ血清を用
いるイムノプロンティングにより確認した。対応の抗体及び基質をVectaキ
ット(Vector Labs)を用いて供した。突然変異PEタンパク質濃度
を280 nmでの吸収により、1.2 ml/++g−cmの励起係数と仮定
して決定した。
E、免疫毒素の構築−1s+HのEDTAを含む0.2 Mのリン酸ナトリウム
(pH8,0)中のN1ys PE38(6〜13mg/+wl)を5倍過剰量
のイミノチオランで誘導し、そして37°Cで10分インキュベートした。タン
パク質を5ephadex G−25(POLO; Pharmacia)で未
反応の架橋剤から分けた。誘導化は典型的には、EIl+aan試薬(Ellm
an、 Arch、 Biochem。
Biophys、 82 : 70−77 (1959)を用いて測定し、N1
ys PE38のモル当り0.5モルのチオールを導入した。同様に、111M
のEDTAを含む0.2Mのリン酸ナトリウム(pH= 7 )中の旧ysPE
38(6〜13++g/+sl )を3倍過剰量のSMCC(スクシニミジル−
4−(N−マレイミドメチル)シクロヘキサン−1−カルボキシレート)で誘導
し、そして22°Cで1時間インキュベートした(Yoshitake ら、J
、 Biochem、 92 : 1413−1424 (1982)) 、タ
ンパク質を5ephadex G−25上で反応体から分離させた。誘導化は典
型的にはN1ys PE38のモル当り0.5個の反応性基を導入した。 PE
35を1mMのEIITA及び5f)+MのDTTを含む0.2 Mのリン酸ナ
トリウム(pH7、o)で保存し、そして抗体にカップリングする前に5eph
adex G−25でDTTから分離させた。モノクローナル抗体(阿^b)
B3は、数多くのヒト腫瘍上で見い出せる多糖抗原に対して反応性であり、そし
て前記の無血清培養培地から精製した。引用することで本明細書に組入れるPa
s tanら、Cancer Res、 51 : 3781−7(1991)
。門^b HB21はヒトトランスフェリンレセプターに対して特異的であり、
そして前述の1(B21を抱えるヌードマウスの腹水から精製した。 MAb濃
度を280 nmでの吸収により、1.4 ml/B−c■の励起係数と仮定し
て決定した。lff1MのEDTAを含む0.2Mのリン酸ナトリウム(p11
7.0)中のB及びHB21(4〜8mg/+*I)を2倍及び4倍過剰量のS
MCGそれぞれと反応させ、そして22゛cで1時間インキュベートした。誘導
化MAbを5ephadex G−25を用いて反応体がら分離させた。
B3及びHB21はそれぞれ分子当り0.83及び1.0個の測定反応性基をこ
れらの条件下で有していた。 Yoshitakeら、前掲を参照のこと、1m
M(7)EDTAを含む0.2Mのリン酸f ト’J ラム(1))18.0)
中(7)B3及び1(B21(4〜5mg/ml)もそれぞれ2倍又は3倍過剰
量の5PDPと反応させ、そして22゛Cで30分インキュベートした。誘導化
MAbを5ephadex G−25を用いて反応体から分離させた。B3及び
HB21はそれぞれ分子当り0.79及び0,56個の測定反応性基をこれらの
条件下で有していた(Carlssonら、BiocheIIl、 J、 17
3 : 723−737 (197B)) 、 5PDP又はSMCCのいづれ
かで誘導化せしめたB3又はII 821をそれぞれ2つのプールに分け、そし
てイミノチオラン又は還元PE35で誘導せしめた2〜3倍過剰量のN1ys
PE38と22°Cで16時間反応させた。反応はヨードアセトアミド(Sig
ma Chemical Co、、 SL、 Louis+ Mo、)の1mM
の最終濃度に至る添加により停止させた。更に、B3をイミノチオランで誘導さ
せた。 0.2 Mノリン酸ナトリ’yL (pH8,0)中(7)83 (5
〜10mg/vs l )を2倍過剰量のイミノチオランと37°Cで1時間イ
ンキュベートした。誘導化抗体を5ephadex G−25を用いて反応体か
ら分離させた。B3にはこれらの条件のもとて1.0個の反応性基が導入された
(Ellman、前掲)。このl’lAbを、DTNB(5,5’−ジチオ−ビ
ス−(2−二トロ安息香酸)で記載の通り(Fitz Gerald、 Met
h、 Enzymol。
151 : 139−145 (1987))に誘導化しておいたPH35と混
合した。22°Cで2時間のインキュベーション後、反応をシスティン(Pie
rce)の0.2 mMの最終濃度に至る添加により停止させた。同様に、イミ
ノチオランで誘導させたB3を22°Cで2時間、SMCCで誘導化しておいた
N1ys PBS8と反応させた。その反応を1mMのヨードアセトアミドで停
止させた。免疫毒素を41Q (HR515)及びTSK−250カラムの連続
使用により、FPLCを用いて単一ピークとして精製した。
F、 ADP−リボシル化アッセイ−タンパク質サンプルのADP−リボシル化
活性を、先の実施例に記載の通りに、伸長因子2に冨む小麦胚芽抽出物を用いて
Co11ierとKandelの手順により測定した。
G、タンパク質合成阻害アッセイータンパク質合成の阻害は先の実施例に記載の
通りに実施した。細胞を毒素添加の前に、96穴プレートの中で各ウェル当り1
5.000細胞において24時間平板培養した。
0.2%のBSA−PBSの中に希釈した毒素又はコントロールを200μl/
ウエルの最終容量にまで加えた。37°Cで16〜20時間のインキュベーショ
ンの後、各ウェルに〔3H〕−ロイシン(Amersham ; 1 uciを
0.2%のBSA−PBSの中に10μIまで希釈)で2時間パルスに付した。
凍結後、その細胞をガラスファイバーフィルターで集め、そしてタンパク質の中
への放射活性の取り込みをBetaplateシンチレーシランカウンター(P
harmacia)で定量した。結果は、毒素抜きでインキュベートした細胞の
取り込みCpIll(分当りのカウント数)の%として算定した。競合アッセイ
を2Mg/mlのEGFの存在下で細胞に付加した毒素を利用して行った。
H,35KDのカルボキシル−末端フラグメントのデザイン−我々は有用な免疫
毒素を作り上げるため、PEの35KDのカルボキシル末端フラグメント(PE
35と称す)をモノクローナル抗体に抱合せしめることができるかどうかを調べ
た。P E 3’5をエンコードするプラスミドをプラスミドCT132を用い
て構築した。
プラスミドCT132を、PE37の位置287においてシスティンを再導入せ
しめるPct?フラグメントを用いて構築した。 DNA配列決定はこの突然変
異が存在していることを確証せしめた。プラスミドCTIIは、プラスミドCT
132のPEをエンコードするDNA配列を、ドメインIbのアミノ酸365−
380の欠損を含むPEをエンコードするDNA配列に置き換えることにより構
築した。この欠損はPEタンパク質の細胞障害性には影響しない、従って、プラ
スミドCTII(f+T)はT7プロモーターと、位置280の曽et、それに
続く天然PEのアミノ酸281−364゜381−613をエンコードするDN
^とを含む、このプラスミドによりエンコードされるタンパク質(PE35)は
位置287において単独のシスティン残基を含む(図7 ) 、 PE35をB
L21 (λ[IF5)において発現させ、そして5O5−PAGEでペリプラ
ズマとスフ二口プラストとに均等に分布していることが見い出せた。〉95%の
均一性に至るベリプラズマからの精製をアニオン交換及びキレート化クロマトグ
ラフィー及びゲル濾過を用いて行った。PE35は50S−PAGE上で予測の
分子量(35KO)であることが認められ、そしてPEに反応性であるウサギ血
清と免疫反応性であった。 ADP リボシル化活性は、天然PEと同一のへ〇
P−リボシル化活性を有する分子であるlys PE40 (Collierと
Kandel、前掲)のそれと同一である。チオール基の数はEl1man試薬
を用いて測定したときにモル当り1個見つけられた。細胞系の数に基づいて試験
したとき、PH35は標的細胞に特異的に結合できないために非常にわずかな細
胞障害活性を存していた。
1、IF21を用いる免疫毒素のデザイン及び活性−PE35が細胞を特異的に
標的とすることができるかどうがを調べるため、それを、ヒトトランスフェリン
レセプターを認識する抗体MAb IF21 (ATCC)に抱合させた。 P
E35をチオエーテル及びジスルフィド結合の両者を用いて複合させた。 PE
35を基礎とする免疫毒素と、事前に作られた分子、旧ys PE38とを比較
するため、接近可能なりジン残基(図7)を含む11個のアミノ酸ペプチドが先
行しているPEのアミノ酸253−364゜381−613を含む分子をイミノ
チオランで誘導化させて遊離スルフヒドリル基を作り上げ、そしてSMCC(チ
オエーテル結合を供するため)又は5PDP (ジスルフィド結合を供するため
)のいづれがで誘導化させたMAbに複合させた。4つの免疫毒素それぞれをア
ニオン交換クロマトグラフィー及びゲル濾過を用いて〉95%の均一性にまで精
製した。それらは190.000 KDの分子量で泳動し、抗体と毒素との1対
1の比を示唆する。還元505−PAGEは抗体及び毒素フラグメントの予測の
パターンをもたらした。チオエーテル結合を介してPE35に抱合させた1(B
21 (IF21−5−C−PH35)を還元してPAGEにかけたとき、それ
は阿^b重鎖(50KD)及びMAb軽鎖(20KD)並びにPE35に結合し

N1ys PE38にチオエーテル結合を介して抱合させたIF21を同様に分
析すると、それは重及び軽鎖、並びにN1ys PE38に結合している重及び
軽鎖をもたらした。ジスルフィド結合を介して旧ys PE38に抱合させた)
1821 (IF21−5−5−PE3B)は還元されたMAb重鎖及び軽鎖、
並びに遊離毒素(38KD)をもたらした、同様に、ジスルフィド結合を介し−
CPE35 ニ抱合された8821 (IF21−3−5−PH35) ハMA
b重鎖及び軽鎖、並びに遊離毒素(35KO)をもたらした、 PEと反応性で
あるポリクローナルウサギ血清を用いての還元免疫毒素のウェスタンブロッティ
ングは、遊離毒素(ジスルフィドコンジュゲートの場合)又は抗体の重鎮及び軽
鎖に結合している毒素(チオエーテルコンジュゲートの場合)の存在を確証せし
めた。
次にコンジュゲートをその活性を決定するためにA431及びMCF?細胞に基
づいて試験した。 PE35に対するジスルフィド結合を採用するコンジュゲー
トはA431及びMCF?細胞に対して最も活性であった(表4及び図8 )
、 Ntys PE38コンジユゲートはコンジュゲーシヨンの方法に関係なく
ほぼ同一の細胞障害性を示したが、しかしA431細胞に対してはPE35に対
するジスルフィド結合を含むものよりも活性が5分の1であった。PE35を用
いて作ったチオエーテルコンジュゲートはPE35を含むジスルフィドコンジュ
ゲートよりも100分の1以下の活性であった。ヒトトランスフェリンレセプタ
ーを含まないマウスL929細胞はIF21を含む免疫毒素の毒性作用に耐性で
あった。更に、A431ヒト11表皮癌細胞系に及ぼす細胞障害性は10μg/
n+lのIF21を用いて阻害され、この免疫毒素はヒトトランスフェリンレセ
プターを介して特異的に結合することを示唆する。
J、B3を用いる免疫毒素のデザイン及び活性:PE35がヒト癌細胞を特異的
に標的とすることができるかどうかを調べるため、数多くのヒト癌上に見い出せ
る多Ii類抗原を認識するMAbであるMAb 83にそれを抱合させた(Pa
stan、 Cancer Res、 、前掲) 、 PE35をDTNBで活
性化させ、そしてジスルフィド結合を介して、イミノチオランで誘導化させてお
いたB3に複合させた。比較のため、N1ys PE38 (SMCCで誘導化
させておいたもの)を83 (イミノチオランで誘導化させておいたもの)に複
合させることにより作った免疫毒素を使用した。各免疫毒素をアニオン交換クロ
マトグラフィー及びゲル濾過を用いて〉95%の均一性にまで精製した。それら
は各々約210,000 KOの分子量で泳動し、抗体と毒素との1対1の比を
示唆する。還元5O5−PAGEは抗体及び毒素フラグメントの予測のパターン
をもたらした。チオエーテル結合を介してN1ys PE38に抱合させたB3
(B3−5−C−PE3B)は還元されてMAb重鎮(50KO)及び軽tj
f (20K[l)、並びにPE35に結合しているMAb重鎖及び軽鎖をもた
らした。ジスルフィド結合を介してPE35に抱合させたB3 (B3−5−S
−PE35)は還元されてMAb重鎮及び軽鎖、並びにi離毒素(35K[l)
をもたらした。還元された免疫毒素のPEに対するポリクローナルウサギ血清を
用いるウェスタンブロンティングは遊離毒素(ジスルフィドコンジュゲートの場
合)又はMAbの重鎮及び軽鎖(チオエーテルコンジュゲートの場合)の存在を
確証せしめた。ジスルフィド結合を含む免疫毒素はA431細胞に対して2倍の
活性であり、そしてMCF7細胞に対しては若干活性が強かった(表4及び図9
)。KB細胞は両毒素の毒性作用に対して耐性であった(表4)、KB細胞はヒ
ト類表皮癌腫に由来し、そしてATCCから獲得した。同様に、MCP7細胞に
対する免疫毒素の活性は900μg/+wlの83により完全に阻害され、免疫
毒素が83抗原に特異的に結合することを示唆する。
11Abがイミノチオランで誘導化され、そしてN1ys PE3BがSMCC
で誘導化されている83とN1ys PE38とのチオエーテルコンジュゲート
を、同一のタンパク質であるが誘導剤を逆にしたものを用いて作った同様のコン
ジュゲートと比較した。興味深いことに、SMCCで誘導化されたB3は、B3
がイミノチオランで誘導化されている同一の免i毒素よりも6〜8分の1の活性
であった。同様に、ジスルフィド結合を介してB3に抱合されているPE35を
含む免疫毒素は、B3が5popで誘導化されているときの方が、B3がイミノ
チオランで誘導化されているときよりも9分の1の活性であった。 IF21を
含む免疫毒素の活性に対する誘導化剤の有意義な作”用は認められなかった。
PE35はPE37の固有の特徴を有し、そして抗体に容易に抱合させることが
できる。これは任意287に1個のシスティン残基を含み、従ってチオエーテル
又はジスルフィド結合のいづれかを介して抗体に確実に複合させることができる
。我々はPE35を用いて作った免疫毒素を、遊離スルフヒドリル基を作り上げ
るためにイミノチオランで誘導化させたN1ys PE38を用いて構築したも
のと比較した。 SMCC又は5PDPのいづれかで誘導化されているMAb
IF21を2つのプールに分け、そしてチオエーテル又はジスルフィド結合それ
ぞれのいづれがを採用してコンジュゲートを作り上げるために平行で各毒素と反
応させた。誘導化は1対1の比で抗体と毒素とを含む免疫毒素の優勢を確実なも
のとするために行った。精製した1対1の免疫毒素のみをここで行った分析のた
めに利用した。
予測通り、HB21に対するジスルフィド又はチオエーテル結合のいづれかを用
いて作ったN1ys PE38コンジユゲートは同程度の毒性を有していた。
PE38を含む免疫毒素は細胞死をもたらしめるために細胞質ゾルに到達できる
ようカルボキシル末端フラグメントを遊離させる2つの重要なプロセシング段階
を、コンジュゲーシゴンの方法に関係なく必要とする。・・・(1)アミノ酸2
79と280との間でのタンパク質分解プロセシング、及び(2)アミノ酸26
5と287とにわたるジスルフィド結合の還元、対照的に、ジスルフィド結合を
介してPH35に抱合されたHB21はPE38コンジユゲートよりもA431
細胞に対して5倍活性であった。細胞質ゾルに到達する各免疫毒素の部分は類似
するため(PEのアミノ酸280−264.38L 613)、タンパク質分解
プロセスはこれらの細胞に対するPE38含有免疫毒素の作用の律速段階があり
うる。しかしながら、HB21−5−5−PE35は、N1ys PE38を含
むコンジュゲートに比してヒト乳癌MCF7細胞系に対する高められた細胞障害
性を示さなかった。MCF7細胞はPE38をタンパク質分解することにおいて
A431細胞よりも効率的である可能性がある。それ故、PE突然変異性のタン
パク質分解はこれらの細胞における律速段階ではないであろう。PE35及びP
E38がこの細胞系に対して類似の非特異的毒性を存する事実は(それぞれ20
0 ng/+wl、対、300 ng/ml)、MCF7細胞がN1ys PE
38をPE35とほぼ同様にプロセスする考えを裏付けする。
PE35はPEをプロセスする哺乳動物細胞により認識されるタンパク質分解部
位を含まないため、チオエーテル結合を介してHB21に結合しているPE35
を含む免疫毒素はかなり不活性である。観察される低い度合いの活性はMAb又
はPE35内のその他の部位で起こるタンパク質分解プロセシング及び得られる
フラグメントの非効率的なトランスロケーションに寄与しうる。
ジスルフィド結合を介してPH35に抱合されたB3を含む免疫毒素も、N1y
s PE38に対するB3チオエーテルコンジュゲートよりも活性であった。し
かしながら、バイパス式タンパク質分解プロセシングの効果の程度は8821コ
ンジユゲートで観察されるものより低かった。興味深いことに、MAbを誘導化
するのにSMCCを用いて作ったB3コンジュゲートは、MAbがイミノチオラ
ンで誘導化され、そしてN1ys PE38がSMCCで誘導化されている同じ
免疫毒素よりも活性が低かった。これらの試薬は共にアミノ基と反応するが、そ
れらは極性において相違する(イミノチオラン> 5PDP > SMCC)。
非極性反応体SMCCは固有のりジン残基を誘導化し、そして誘導過程の際に8
3の重要な結合特性を妨げる。同様に、B3を誘導化するためにイミノチオラン
を用いて作ったPE35ジスルフイドコンジユゲートはB3を誘導化するのに5
PDPを用いたものよりも10倍有能であった。
K、 B5−5−5−PE35によるインビボ結果B5−5−S−PH35を5
μgのレベルでマウスに静脈内注射した。免疫毒素の血清レベルを20時間にわ
たり、その血清をA431細胞とインキュベートし、そして前記の通りにタンパ
ク質合成に対する作用を測定することより決定した。標準曲線をコントロールマ
ウス血清に希釈したB5−5−5−PE35で作った。図10参照のこと。
ヌードマウスにおける皮下A431腫瘍の増殖に及ぼすB5−3−5−PE35
の効果を決定した。マウスには0日目にて2,000,000個のA431細胞
を、そして5日目にて25μgの83−5−5−PE35の単独静脈内投与を与
えた。立方暢−で測定した腫瘍増殖に基づく経時的な結果を図11に示す、免疫
毒素は腫瘍の完全な退化をもたらした。

膀胱癌と診断された患者は、カルボキシル末端配列KDELを有するPE35/
TGFαで、60−1の希釈液中のそのタンパク質を6週間にねたって週に一度
カテーテルにより注入することによって処置されうる。
この分子はTP40よりも活性が高く、且つ小さいものであり、そしてB3抗原
(乳、類表皮、胃及び前立腺の癌腫を含む)を抱える腫瘍と診断された患者は、
それらの者をカルボキシル末端配列KDELを有するB3FvとのPE35融合
タンパク質を構成するPIEで、1日当りにて患者当り1〜100 mgの用量
で静脈内投与することにより処置されうる。
r B3Fν」は、重鎖Fvドメインのカルボキシル末端及び軽1iFvドメイ
ンのアミノ末端において開始する、柔軟リンカ−(Gly、 5er)により連
結されたMab B3の重及び軽鎖領域を含む配列を称する。これは全て引用す
ることで本明細書に組入れる一般譲渡されたU、 S、 S、 N。
07/767.331号に記載されている。このタンパク質をエンコードする遺
伝子はPH35遺伝子と融合されている。

Claims (26)
- 1.組換シュードモナス外毒素分子であって、ドメインIaが欠損しており、そ してドメインIIのアミノ末端から最初の27個より多くのアミノ酸が欠損して いない分子。
- 2.前記分子がドメインIIのアミノ酸位280にあるメチオニンで開始する、 請求項1記載の組換PE。
- 3.前記分子がドメインIbのほぼアミノ酸365〜380の欠損を含んで成る 、請求項1記載の組換PE。
- 4.前記分子がほぼアミノ酸280〜613より本質的に成る、請求項1記載の 組換PE。
- 5.前記分子がほぼアミノ酸280〜364及び381〜613より本質的に成 る、請求項1記載の組換PE。
- 6.前記分子がアミノ酸287においてシステインの代わりにセリンの置換を含 む、請求項1記載の組換PE。
- 7.前記分子がその分子のカルボキシル末端において、REDLX,REDL及 びKDELより成る群から選ばれるアミノ酸配列を更に含む、請求項1記載の組 換PE。
- 8.前記分子がドメインIIIの実質的な欠損を更に含んで成る、請求項1記載 の組換PE。
- 9.ドメインIIIのほぼアミノ酸604−613が保持されている、請求項8 記載の組換PE。
- 10.前記分子が、欠損したドメインIIIの代わりにPEに融合したリガンド 結合因子を更に含んで成る、請求項9記載の組換PE。
- 11.前記分子がリガンド結合因子に融合した、請求項1記載の組換PE。
- 12.前記リガンド結合因子がほぼアミノ酸位607の後に融合されており、そ してドメインIIIのアミノ酸604−613がそれに続いている、請求項11 記載の組換PE。
- 13.前記リガンド結合因子がTGFαである、請求項11記載の組換PE。
- 14.前記リガンド結合因子が抗体又はその結合性フラグメントである、請求項 11記載の組換PE。
- 15.前記リガンド結合因子がホルモンである、請求項11記載の組換PE。
- 16.前記リガンド結合因子が成長因子である、請求項11記載の組換PE。
- 17.前記リガンド結合因子が癌細胞レセプターに特異的に結合する、請求項1 1記載の組換PE。
- 18.アミノ酸280〜364及び381〜613より本質的に成り、この組換 PE分子内においてアミノ酸607の後にTGFαが挿入されており、そしてそ れにドメインIIIのアミノ酸604−613が続いている、請求項1記載の組 換PE。
- 19.前記PE分がその分子のカルボキシル末端において細胞質内保持配列を含 む、請求項11記載の組換PE分子。
- 20.組換シュードモナス外毒素(PE)分子であってドメインIIのアミノ末 端において欠損を有し、これによりその分子は細胞障害アッセイにおいて標的細 胞に対する細胞障害性がPE40よりも少なくとも20倍高くなっており、ここ でPE40及びこの組換PE分子の標的細胞に対する細胞障害性は、この標的細 胞に対して特異的なリガンド結合因子に融合させたPE40及びこの標的に対し て特異的なリガンド結合因子に融合させたこの組換PE分子を、その標的細胞に 対してアッセイすることにより測定する、組換PE分子。
- 21.請求項1記載のアミノ酸配列をエンコードする核酸配列を含んで成るベク ター。
- 22.請求項6記載のアミノ酸配列をエンコードする核酸配列を含んで成るベク ター。
- 23.請求項1記載の配列を発現する宿主細胞。
- 24.請求項6記載の配列を発現する宿主細胞。
- 25.請求項1記載の分子及び薬理学的に許容される担体を含んで成る薬理製剤 。
- 26.患者における腫瘍増殖を阻害するための方法であって、その患者に、その 身体腔の中へと又は器官の内腔の中へと、組換シュードモナス外毒素分子であっ てドメインIaが欠損されており、且つドメインIIのアミノ酸末端から最初2 7個のアミノ酸より多くが欠損されていない分子に融合させた腫瘍細胞に対して 特異的なリガンド結合因子を投与することを含んで成る方法。
