【発明の詳細な説明】
近赤外レーザ吸収分光分析法を用いる診断試験発明の背景
本発明は一般に医療上の診断試験に関し、そしてより特定的には同位体存在比(
isotope ratio)を使用する代謝呼吸試験に関する。
或る種の医療診断試験は同位体で置換されて識別された化合物を患者に投与し、
ついで識別された化合物の代謝生成物について患者の呼気を監視することに頼っ
ている。在来の代謝呼気試験は14Cで置換されて識別された基質(サブストレ
ート)を患者に投与し、ついで呼吸に伴って吐き出される14CO2の濃度を測
定することに頼っている。14Cは放射性であるので、それは安価な放射監視装
置で容易に検出される。あいにく、この放射性はまた、患者に対して健康上の危
害を与えるために、問題がある。14C同位体を13Cのような非放射性の同位
体で置き換えるという方法が試みられてきた。これはICに曝されることに伴う
危害を除くけれども、それは新しい問題、すなわち呼気に伴う12CO2をどの
ようにして検出するかという問題を引き起こす。この問題は、13Cが約1%と
いう量で天然に比較的豊富に存在し、しかもこの値がかなり変動することも知ら
れているという事実によって、一層深刻となっている。在来の幾つかの方法は1
3Cを監視するために存在していて、同位体存在比質量分析法および核磁気共鳴
分光測定を包含しているけれども、連合した計装は非常に費用がかかるので、1
3Cで置換されて識別された化合物を診断試験で広汎に使用することが制限され
る。
リー(Lee)およびマイコフスキー(Majkowski)(米国特許第4,
684.805号)は、このために低温に冷却された、同調できる赤外鉛塩レー
ザダイオード(cryogenically cooled tunablei
nfrared lead−salt laser diode)ばかりでなく
、人の息に伴う或る種の分子を監視することによるその他の医療試験の使用を考
えている。関心が持たれている最強の吸収線は4μmないし5μmの範囲にあっ
たので、3μmないし30μmの範囲で発する鉛塩レーザダイオードは当然の選
択であった。しかし、鉛塩レーザダイオードと、それらを連合させた検出装置は
液体窒素温度で作動する。さらに、それらの出力は一般にマルチモードであって
、一般に1ミリワツトよりも小さい。
発明の要約
本発明は患者が有毒化合物および危険な化合物に曝されるのを監視するために、
また成る種の薬剤の濃度について検査するために、幾つかの病気および代謝障害
に対して患者をかばうことができる一部の簡単で、健康を損わない、迅速な医療
診断試験を提供する。
要するに本発明は、呼気試料中にある目標物質の量を測定するために赤外レーザ
ダイオード源のRF変調分光分析法(RF modulation 5pect
roscopy)を使用する。この型の周波数変調ではレーザ出力は変調周波数
で変調されて、レーザキャリアから置き換えられた側波帯を生ずる。変調された
ビームは側波帯を差別的に吸収するガス試料を通過する結果、レーザ周波数変調

。
成る種の試験において、目標物質は特定の同位体であって、その濃度は同位体存
在比として測定される。第一および第二の種類の同位体の存在を測定するために

本発明の一つの局面によれば、吸収は近赤外で測定される。co2に関する最も
強い吸収線は4μmないし5μmの領域にあるけれども、1. 6μmの領域の
中にはこれよりも遥かに弱い一部の線がある。1. 6μm領域中の12co、
吸収の測定は通常中心からはずれたものとして精々認識され、そしてIc○2吸
収の測定は不可能なものとして認識されるという事実にもかかわらず、本発明者
等はこの範囲内の幾つかの線が十分な精度をもって測定できることを見出した。
これは、優れた単一モードの1. 6μmレーザダイオードが市場で入手できる
ので、特に有利である。これらの装置は信頼性があって寿命が長く、そしてより
重要なことには、好都合な温度範囲、すなわち0〜50℃にわたって作動する。
したがって、本発明の技術は比較的安価で携帯に便利な計測器で実行することが
できる。特定の成る具体例においては、変調された赤外放射が試料容器と対照容
器を通過し、そしてそれぞれの吸収を表わす信号を互に関連させる。弱い吸収か
らみて、試料容器には多重通路容器(multi−pass cell)のデザ

特定の実施態様の説明
周波数変調分光分析法(FMS)および波長変調分光分析法(wMs)の概説F
MSおよびWMSと称する2つの型のレーザ吸収分光分析法を説明する。レーザ
吸収分光分析法においては、関心が持たれている信号が調査フィールドと試料と
の相互作用から生ずる。このフィールドはレーザビームであって、その波長は試
料の吸収線とほぼ共振している。調査フィールドの波長を正弦曲線に沿って変調
させると同時に、関心が持たれている共振特徴を通して調査フィールドの平均波
長を同調させることによって、共振の強さに比例している大きな信号が現れる。
図IA−Dは種々のFMSレジーム(regime)の成る特性を図解している
。外部電気光学変調器によるか、またはレーザダイオードの場合のように注入電
流(injection current)を変調させるかのいずれかによって
、FMSは高い周波数(数百MHzまたはGHz範囲)てレーザの変調を必要と
する。変調されたレーザビームは試料を通過し、そして転送されたビームは高速
光検出器および適当な処理電子装置を用いて変調される。
図IAおよび図IBは単一の変調周波数を使用する単調(single−t。
ne)FMS (STFMS)としてどのようなものが知られているかというこ
とに関する。周波数空間において、変調されたレーザフィールドはレーザの自然
放出周波数(natural emission frequency)である
キャリア周波数と、変調周波数のインテグラル マルチプル(integral
multiple)によってキャリアから変位された側波帯からなる。弱い変調
極限(modulation I 1m1t)(周波数変調指数β〈く1)にお
いて、レーザスペクトルは側波帯の最初のセットで近似させることができる(図
IA)。吸収が無い場合、キャリアと上方の側波帯から出たビート信号はキャリ
アと下方の側波帯から出たビート信号を丁度相殺するが、これはそれらが同じ振
幅を持ち、しかも位相が丁度180°ずれているためである。しかし、このレー
ザが吸収からはずれると、ビート信号の間の微妙なバランスは乱されて、吸収線
形(absorpt ion l 1neshape)の−次導関数(firs
t derivative)に似た信号が生ずる(図IB)。復調は普通変調周
波数で遂行されるけれども、それは変調周波数の調波て遂行することがてきる。
STFMSは直接吸収技術よりも優れた利点を提供する。直接吸収法は信号をレ
ーザ強度の変化の形で検出して、大きな1/f雑音成分が存在する周波数スペク
トルの領域でそれの検出帯域幅を有する。FMSは大部分のレーザが極めて僅か
なl/f雑音を示す変調周波数で信号を検出する。FMSにおける信号は側波帯
の差分吸収(differential absorption)から生ずるの
で、最高の感度のためには変調周波数は吸収のライン幅(I ine widt
h)に匹敵しなければならない。これは、空中で拡げられた吸収のためには変調
周波数が1〜3GHzの範囲に存在しなければならないことを意味している。こ
れは、変調周波数における検出が非常に高速の検出器を必要としていることを意
味しているので、問題である。一般に高帯域幅赤外検出器は広く利用できず、非
常に高価であり、そして極めて小さく、かつ損傷に感じ易い活性領域を有し、特
に中間赤外領域で光学的調整をかえって扱いにくくするという事実がある。
検出器の帯域幅を小さくするという要求は、高いけれども互に密接している2つ
の周波数、vlおよびV2 (図IC)でレーザの変調を要求している2トーン
(two−tone)FMS (TTFMS)の開発を導いた。TTFMSは、
信号が二三MHzのオーダーになり得る分離周波数VI V2で現在検出される
基本差(fundamental difference)とともに、STFM
Sの基準特性(basic characteristics)の大部分を保持
している。TTFMS信号は周波数変調指数β、振幅変調指数Mおよびそれらの
位相差ψによって左右され、そして線形の二次導関数に類似している(図jD)
。
小さな吸収についてはFM信号は吸収の大きさと比例している。復調は変調周波
数の相違またはこの相違の調波に基づいて遂行できる。
この技術は原則として量子またはショットノイズによってしか制限を受けないけ
れども、実際には残留振幅変調(RAM) 、エタロン フリンジ(etal。
n fringe)、レーザ過多または1/f雑音および光フィードバックのよ
うな幾つかの雑音源によって制限され、モしてレーザダイオードがこれらの雑音
源に感じ易いことは周知である。エタロン フリンジの影響を低減させ、またR
AMおよびレーザ過多の雑音を減する幾つかの案がかなりの成功を収めて実行さ
れてきた。量子雑音極限またはFM系において、検出できる最小の吸収は普通1
O−7ないし10−’の範囲にある。この水準における感度は種々のレーザ系で
示されてきた。実際、典型的な近赤外レーザビームについて雑音抑圧機構がない
場合、10−’の二三部のオーダーで感度を期待することができる。
波長変調分光分析法(WMS)は前記のFMS技術よりも先んじていて、同調可
能なダイオードレーザ源とともに1960年代末期以来使用されてきた。核磁気
共鳴(NMR)および電子常磁性共鳴(EPR)分光分析法で信号の感度を増強
させるために最初用いられたのは変調技術の自然の結果である。これらの方法は
NMRおよびEPRにおいて1950年代中に使用されていた。NMRおよびE
PRにおいて調査フィールドは試料の核スピンまたは電子スピンとほぼ共振する
RF電磁フィールドである。WMS(およびFMS)においてこのフィールドは
レーザビームであって、その波長は試料の吸収線とほぼ共振する。一般に変調周
波数の全ての高調波における共振から信号が回収される。通常第一および第二の
調波信号だけが記録され、そしてそれらは共振線形の一次および二次導関数に比
例している(図IEおよびIF)。
WMSにおいて、kHz領域の変調周波数は高感度分光分析検出のため伝統的に
使用されてきた。しかしながら、1980年代における周波数変調分光分析法(
FMS)の発達に伴って、MHz領域における変調および検出の利点が認識され
てきて、MHz周波数におけるWMSの利点が立証されてきた。FMSとWMS
との差異は僅かである。FMSにおいて、レーザの変調指数は小さいが、変調周
波数対吸収特性(absorption feature)の幅の比は大きい。
その結果、関心が持たれている吸収特性は単離された1個の側波帯または側波帯
の小さなセットで調査される。WMSにおいては変調周波数対吸収特性の幅の比
は小さいが、変調指数は大きい。その結果、吸収特性は多数の側波帯によって調
査される。したがってFMSおよびWMSは比較的普遍的な変調分光分析法の限
られた場合とみなすことができる。
同位体存在比測定用装置
図2は試料中の目標物質の濃度を測定するための装置10の高準位光兼電気ブロ
ック図である。本発明の特定の適用は呼気試料中に通常微量に存在している目標
物質の測定であって、そのためにこの装置は試料容器12Sおよび対照容器12
Rを含んでいる。特定の適用において、目標物質は”co、に対して測定される
同位体積”co2である。レーザダイオード イルミネーション サブシステム
15は2種の波長(λ1およびλ2)の光を供給して、各容器を通る両方の波長
を含むビームを指示する。この2種の波長は2種の同位体積に関する吸収線に対
応している。下記に述べるように、レーザ イルミネーション サブシステムは
吸収線の分離および相対強度に従って、単一のレーザダイオードまたは一対のレ
ーザダイオードを含むことができる。
連合した容器を通って伝送される光の強さを表す信号を提供するための検出器2
0Rおよび20Sが対照容器12Rおよび試料容器12sと結合している。制御
サーキットリー30はレーザを駆動させ、個々のレジームに従った駆動電流を変
調し、吸収線全体にわたってレーザ波長を同調させ(走査い、そして個々のレジ
ームに従った検出器信号を復調させる。変調および復調のサーキットリーは個々
のFMSレジームに従って変化する。
図3Aはイルミネーション サブシステム15が第一および第二の吸収線を含む
波長範囲にわたって同調できる単一のレーザダイオード40を含んでいる実施態
様を示す光概要図である。レーザダイオードから出る前面および背面の出力は2
つの容器に向かう2つのビームを提供する。その代りに、単一面の出力をビーム
スプリッタに向けて2つのビームを供給することもできる。好ましい実施態様に
おいて、レーザダイオードは近赤外レーザダイオードであって、これは典型的に
は0〜50℃の範囲の温度で運転される。これは一般に室温に近い運転を許容し
、それによって低温による冷却の必要が回避される。波長は温度によって変化す
るので、レーザダイオードは、例えば熱電的に制御された金属板上に取り付けら
れるような方法によって、温度制御されるのが好ましい。
対照容器は既知の、そして好ましくは高濃度の目標物質を有するガス試料を含ん
でいるので、単光路容器(single−pass cell)であってもよい
。代表的な対照試料は”co2と” CO2とのl:1混合物である。試料容器
は、他方では僅かな濃度の目標物質を有する試料を含んでいるので、道理にかな
って物理的な長さを保つために多重光路容器(multi−pass cell
)であるのが好ましい。特定の実施態様において、容器はヘリオツド(Herr
iot)デザインでできていて、リエントラント(re−entrant)法で
作動する。試料容器には入口ガスポート47aおよび出口ガスポート47bが設
けられていて、これらはそれぞれの真空制御ソレノイド弁48aおよび48bに
よって制御される。被検者の呼気試料を集めるために風船またはその他の容れ物
が使用され、その後これは入口ポートと結合される。
図3Bはイルミネーション サブシステムが2つの吸収線に対応するそれぞれの
波長で作動する第一および第二のレーザダイオード40aおよび40bを含んで
いる実施態様を示す光構成図である。この実施態様においては、両方のレーザビ
ームがビームスプリッタ52に向かい、そしてこのビームスプリッタ52は各ビ
ームを2つの成分に分割し、そしてこれらの成分を両方の波長で各容器の中に向
けるように作動する。
図4Aは単一のレーザおよびTTFMSを用いる実施態様のための制御サーキッ
トリーのブロック図である。この実施態様において、単一のレーザは2つの吸収
線を含む範囲にわたって走査される波長を有する。与えられた検出器から出た信
号はそれぞれの間隔の間に抽出され、そしてその間隔においてレーザの波長がそ
れぞれの吸収線にわたって走査される。タイミング装置は、マイクロプロセッサ
の制御の下で試験手順を順番に配列するための信号を提供する。このため、それ
はランプ ゼネレータ(ramp generator)62を制御して、アナ
ログデータモジュール63Rおよび63Sにリセットおよび試料ゲート信号を提
供する。ランプ ゼネレータはレーザ駆動サーキットリー65に信号を供給して
波長の走査を制御する。レーザ駆動電流は低域フィルター67およびRFバイア
ス回路網68を通る。アナログモジュール63Rおよび63Sは検出器20Rお
よび20Sから復調された信号を受ける。
一対の水晶発振器70および72は2つの変調周波数において信号を供給し、1
96MHzおよび200MI(zがその代表的なものである。各信号は分割され
、そして信号の各第一部分は合体され、そして帯域フィルター73を通ってRF
バイアス回路網68に至る。発振器信号の他の部分はミクサ80のRFおよびL
O大入力適用され、そこからIF出力信号は低域フィルター85を通って2つの
周波数の差(この場合4MHz)に基ついて信号を発生する。差信号(diff
erence signal)は、それぞれのRF大入力検出器信号を受け取っ
て、変調された信号をそれぞれのIF比出力供給する一対のミクサ90Rおよび
90SのLO大入力適用される。これらの信号は低域で濾波されてアナログモジ
ュール63Rおよび63Sに伝達される。アナログ信号はアナログ−ディジタル
(A/D)変換サーキソトリ−92に伝達され、そしてディジタル化された値は
処理のためコンピュータ95に送られる。
図4Bは2つのレーザおよびTTFMSを用いる実施態様のための制御サーキソ
トリーのブロック図である。図4Aにおける参照数字に対応する参照数字が使用
されるが、レーザダイオード40aおよび40bのために回路要素に対応する添
字が付けられている。この実施態様においては、各レーザは2つの吸収線のうち
の単一の吸収線を含む範囲にわたって走査された波長を有する。この実施態様に
おいては、各レーザは一対の周波数において変調されるが、一方のレーザに対す
る2つの周波数の差は他方のレーザに対するものと異っている。例えば、第一の
レーザは196MHzおよび200MHzで変調できるのに対して(図4Aの実
施態様に相当している)、第二のレーザは200MHzおよび206MH2で変
調できる。したがって各検出器は変調周波数の2つのセットに対応する信号を供
給する。各検出器信号が4MHzおよび6MHzで復調されるように(特定の例
について)変調、復調およびレーザ駆動装置回路が重複している点を除いて、サ
ーキットリーは単一レーザの実施態様のそれと基本的に類似している。
図5Aは単一レーザおよびWMSを用いる実施態様のための制御サーキットリー
のブロック図である。この実施態様は、異なる変調および復調レジームが用いら
れるが、制御サーキットリーおよびデータ取得サーキットリーが一般に同一であ
る図4Aの実施態様と相違している。同一の要素(例えばタイミング装置60、
ランプ ゼネレータ62およびレーザ駆動型サーキソトリ−65)については同
し参照数字が使用される
水晶発振器100は単一変調周波数において信号を供給し、lOMHzがその代
表的なものである。信号の一部はRFバイアス回路網68に伝達されてレーザ駆
動電流を変調させる。前述のように、一般に基本調波または二次的調波のいずれ
かで遂行される。単一レーザの実施態様の場合、基本調波または二次的調波のい
ずれかを好ましく使用する理由は多分少ししかないであろう。図示された特定の
実施態様において、対照検出器信号は基本調波(IOMH2)で復調されるのに
対して、試料検出器信号は二次的調波(20MH2)で復調される。したがって
、発振器信号の一部は周波数2倍器102および帯域フィルタ105を通過する
のに対して他の成分は直接使用される。lOMHzおよび20MHzの信号は一
対のミクサllORおよびlIO3のLO大入力適用され、そしてこれらのミク
サはそれぞれのRF大入力検出器信号を受け取って、復調された信号をそれぞれ
のIF比出力供給する。信号は図4Aの実施態様と同様に低域で濾波され、そし
てアナログモジュール63Rおよび63Sに伝達される。
図5Bは2つのレーザおよびWMSを用いる実施態様のための制御サーキットリ
ーのブロック図である。図4Bの実施態様の場合と同様に、各レーザは2つの吸
収線のうちの単一吸収線を含む範囲にわたって走査された波長を有する。この実
施態様においては、各レーザは異なる周波数、例えば8MHzおよびlOMHz
において変調される。各検出器信号が2つの周波数において復調されるように変
調、復調、およびレーザ駆動回路が重複している点を除いて、サーキットリーは
単一レーザの実施態様のそれと基本的に類似している。図5Aで使用された参照
数字に対応する参照数字が使用されたが、レーザダイオード40aおよび4゜b
のための回路要素に対応する添字aおよびbが付けられている。レーザが吸収線
に固定された波長を有する場合、対照検出器信号は吸収線で零交差を有し、これ
がレーザ波長をサーボ機構で制御するのを一層容易にするので、少なくともこの
対照検出器信号を基本波で復調するのが好ましい。
実験結果
AFGL HITRANデータベースによれば、1.6μmスペクトル領域にお
ける最も強い12CO2トランジションは2xlO−” cmのオーダーのライ
ンストレングス(l inestrength)を有し、そして最も強い目CO
,トランジションは2XIO−’″’cmのオーダーのラインストレングスを有
する。しかしAFGL一覧表はラインストレングスに因数分解された相対的な自
然存在比(relative natural abundance)を有する
。それ故CO2の特定の同位体混合物から吸収係数を算出するに当っては、相対
的な自然存在比でAFGLラインストレングスを割るのが重要である。以下の説
明はこれらの補正されたラインストレングスのみを使用する。
レーザにまたDC電流を供給するレーザダイオード制御器(メレスーグリオット
(Me I Ies−Gr 1ot)型番号06DLDOO3型)によって温度
が制御される熱電冷却器(メレスーグリオット型番号06DTCOO3型)上に
、1.6μm領域で作動する単一形式のレーザダイオードが据え付けられた。2
つのRFゼネレータ(HP型番号8350Cおよび3336B)によってレーザ
にRF駆動が供給され、そしてそれはレーザに容量結合された。大部分の実験に
使用された変調周波数は335MHzおよび345MHzであり、これらはこの
スペクトル領域におけるCO2吸収の175MHzドツプラー半値幅を超える。
レーザの波長はレーザ電流を500Hzの速度でランプ(ramp)させること
によってレーザの波長を吸収領域にわたって迅速に同調させた。1.55μmに
ついて被覆されたレンズアセンブリによってレーザの出力を平行にし、ついでリ
エントラント様式で作動するヘリオツドデザインの多重光路容器に通した。ヘリ
オツド容器の全容量は2.5リツトルであり、容器の中の光路長さの合計は90
3cmに調整された。
He−Ne可視レーザビームを最初のアラインメントのための赤外ビームととも
に伝播させた。ヘリオツド容器から出た出力ビームを高帯域幅InGaAs検出
器(三菱 PD7002)の上に集中させた。lOMHzのRF倍信号濾波し、
そして70dbのトータルゲインを供給する2つの増幅器で増幅した。このゲイ
ンを可変減衰器で調整した。増幅し、そして濾波した後、ホモダイン検波のため
二重平衡ミクサのRFポートに信号を向かわせた。最終的なFM線形(ラインシ
ェイプ)を再び低域で濾波してから、データ収集のための80386パーソナル
コンピユータとインクフェイスで接続されているディンタルオシロスコープ(テ
クトロニクス(Tektronix)型番号 2403A)によって抽出したバ
ーレイ波長計(Burleigh Wavemeter)(型番号 WA20D
L)を用いてレーザの波長を0.01−am−’分解能で測定した。レーザは6
246 cm−’ないし6232cm’の同調範囲を持っていた。我々の成果を
それぞれ’ ” CO2および”C2に関する6237.43cm ’および6
237.16cm−’の2つの接近した吸収ライン上に集めた。これらのライン
はこの適用にとって最適ではなくて、我々はレーザの同調範囲により最適ライン
に達するのが制限された。HITANデータベースによれば、これらの吸収ライ
ン(相対的な天然存在比について補正されている)のラインストレングスは1.
592X10” cmおよび8.794X10−” cmである。ヒトの呼気に
関する実験を遂行する前に、純粋な”co、および目CO2を用いて、これらの
ラインを先ず同定した。我々が正しいラインを監視していることを確認するため
にHITANデータベース中の幾つかのその他のCO2ラインのライン位置と強
さを同定して、測定した。
いずれかの吸収種に由来する吸収横断面(absorption cross−
section)および従ってTTFMS信号は衝突ブロードニング(coll
isional broadening)のために圧力の関数である。これらの
実験で監視されたラインは異なる衝突ブロードニング係数(”Cotについては
0. 074 cm−’a tm−’、そして”CO2については0.076c
m−’atm一つを持っていた。我々は吸収横断面の比δ、すなわち我々が圧力
の関数として監視した12CO2および”co、ラインの何倍かの濃度Nを算出
し、そしてそれが9〜50トルの圧力範囲にわたって約3%だけ変化することを
見出した。実際上の理由から、大抵の測定は50トルよりも低い圧力で遂行した
。比較的低い周波数ではレーザダイオードを変調するのが容易なので、圧力の拡
大(broadening)を最小限に保ち、そして吸収ラインの幅を変調周波
数に匹敵するように保持するのが望ましい。臨床に適用される計測器を作るとい
う点からみても、患者か大きな容量のものを比較的高い圧力で充填しないですむ
場合、より容易になるであろう。我々の場合、小さな風船に0.5リツトルの呼
気試料を詰め込むのは多重光路容器に50トルの全圧まで充填するのに十分以上
であった。
監視された2本のラインはこのように異なる強さを持っていたので、2つの吸収
ラインにわたって走査したとき、CO2増幅器チェインの中のゲインを変える必
要があった。したがって、両方のラインを検出するためには我々の系が必要なダ
イナミックレンジを持っていることを保証しなければならなかった。RFデザイ
ンよび容器内のガス圧力の関数として我々の系の応答度およびダイナミックレン
ジを検量するために幾つかの運転を行って信号の直線性を確認した。このデータ
は我々の系が少なくとも35dbのダイナミックレンジを有すること、および信
号が40ないし50トルまで大体直線に保持されていることを示した。
6A図は50トルの呼気試料と40dbのトータルゲインを用いて6237゜4
3 cm−’で”co、吸収ライン全体にわたって調べた波長走査を示している
。
同様に6B図は同じ50トルの呼気試料から6237.16cm−’で” CO
2吸収から得られた信号を示している。この場合のゲインは70dbであった。
30dbのゲイン差を引き起こすこれらの2つの信号の比は168.3である。
H1TANデータベースに示されたラインストレングスと圧力拡大係数を用いる
と、50トルのこれらのラインに関する吸収横断面の理論的な比は実験上の値よ
りも稍低い166.4になることが計算で出された。この差は増幅器チェーンに
おける小さな非直線性と我々の圧力計の不正確な点に帰することができる。この
比は圧力を徐々に18トルまで低下させると約3%減少する。”co2の走査に
おいては、走査の終わりに向って別の弱い線が顕著になる。我々はこの線が5.
75IXIO−”のラインストレングスを有する6237.06cm−’におけ
る12Cl l ()l 8 ()によるものと決定した。これらの走査に関す
る信号対雑音比は十分に100を超え、これは”CO2/”CO2比について1
%よりも大きい測定精度を与える。ヒトの呼気に関する”Cot / 12CO
2比の天然の変動は1%の水準にあるので、この精度は医療上の診断に利用する
のに十分である。我々の系の感度は純粋な12CO2、純粋な”co□の両方と
呼気試料を用いて検量した。我々の装置は一般にIX]、O−’はどの小さな吸
収を測定することができた。RAM、エタロンフリンジまたレーザ過多の雑音を
引き去る努力は払われなかった。感度は雑音引き去り機構(noise 5ub
traction scheme)を使用することによって、またファラデーア
イソレータを用いてレーザ中に戻される光フィードバックの効果を減することに
よって多分改善できるであろう。
これらのラインは0. 27 cm−lシか離れていなかったけれども、2本の
ラインのために実質的に異なるゲインが必要であったという事実は特定の増幅器
ゲインを用いる単一の走査測定を不可能にした。別々のレーザダイオードを用い
て各ラインを監視することはできるけれども、両方のラインに対して単一のレー
ザを使用するのが好ましい。これは強度がより近く一致し、そして波長の間隔が
より狭くなっている一組のラインを要求する。これらの要求を満たす吸収の幾つ
かの組がある。例えば、6228.6938cm−’の”co、ラインおよび6
228.4370cm−’の”CO2ラインは監視するのに遥かに優れたライン
である。
これらの50トルにおける吸収横断面の比はl004であるので、RFエレクト
ロニクスにとっては遥かに小さいダイナミックレンジが適している。我々のレー
ザの同調性(t、unabilitY)は制限されているため、我々はこれらの
吸収を利用できなかったが、様々なレーザダイオードは適切な精度をもってこれ
らのラインを同時に監視することができた。
携帯用計測器
図7は本発明を具体化している計測器150の透視図である。この計測器はレー
ザイルミネーションサブシステム15、容器12Rおよび12s、および検出器
20Rおよび20S、およびエレクトロニクスおよび駆動およびモジュレーショ
ンサーキットリー30を結合しているオプティックスモジュール160を含む主
要ハウジング155を包含している。マイクロコンピュータ95は好ましくはケ
ーブルを経て主要ハウジングに連結しているラップトツブ(laptop)装置
として提供されている。
結論
要約すると、本発明は幾つかの医療試験を成し遂げるための比較的安価で、かつ
安全な技術を提供することが理解できる。室温のレーザを使用すると、大きくて
費用がかかり、かつ低温を必要とする装置の必要性が回避され、また実行される
このような試験を成し遂げるための装置をコンパクトな携帯用計測器の形にする
ことができる。
上に述べたことは特定の実施態様の完全な説明であるととも、それに代わる構成
、変更および等価のものを使用することができる。例えば、前に論じた特定の吸
収線は1. 6μmの波長範囲にあって、これは現在市販されている室温レーザ
ダイオードで測定することができる。しかし、研究書違は2μmないし3μmの
範囲で作動する室温装置を一般に運転しており、そしてこのような装置が適して
いると考えられる。このような装置を利用することができる一対の吸収ラインは
4870.436cm−’のl l CO、吸収ラインと4870.4917c
m−’の1′CO□吸収ラインであろう。さらに、この波長領域における運転は
一酸化炭素およびメタンのようなその他の化合物の同位体積までこの技術を拡張
することを可能にするであろう。
それ故、これまでの説明および図解は特許請求の範囲によって定義される本発明
の範囲を限定するものと解すべきではない。
Claims (7)
- 1.分析されるガス試料を保有するための試料容器;第一および第二の同位体種 の既知の比を有するガスの一定容量を保有するための対照容器; 1μmないし3μmの範囲にある第一および第二の波長の光を前記試料容器およ び前記対照容器の両方に通過させるためのレーザダイオードイルミネータ;そし て 前記第一の波長は前記第一の同位体を含む分子の吸収ラインにあり;前記第二の 波長は前記第二の同位体を含む分子の吸収ラインにあり;前記第一および第二の 各波長の光を個々のレジームに従って変調するための手段; 前記第一および第二の各波長の光を前記個々のレジームに従って復調して、前記 試料容器および対照容器の各々の中で前記第一および第二の各波長の吸収に対応 するそれぞれの信号を提供するための手段;および前記それぞれの信号を互いに 関連させて前記試料容器中の同位体存在比の表示を提供するための手段; を含む、分子状ガス試料中の第一および第二の同位体種の存在に関する情報を測 定する装置。
- 2.分析されるガス試料を保有するための試料容器;第一および第二の同位体種 の既知の比を有するガスの一定容量を保有するための対照容器; 第一および第二の波長の光を前記試料容器および前記対照容器の両方に通過させ るためのレーザダイオードイルミネータであって、ほぼ0〜50℃の範囲にある 作動温度を有する前記レーザダイオードイルミネータ;そして前記第一の波長は 前記第一の同位体を含む分子の吸収ラインにあり;前記第二の波長は前記第二の 同位体を含む分子の吸収ラインにあり;前記第一および第二の各波長の光を個々 のレジームに従って変調するための手段; 前記第一および第二の各波長の光を前記個々のレジームに従って復調して、前記 試料容器および対照容器の各々の中で前記第一および第二の各波長の吸収に対応 するそれぞれの信号を提供するための手段;および前記それぞれの信号を互いに 関連させて前記試料容器の中の同位体存在比の表示を提供するための手段; を含む、分子状ガス試料中の第一および第二の同位体種の存在に関する情報を測 定する装置。
- 3.前記イルミネータが 単一のレーザダイオード;および 前記レーザダイオードの少なくとも1つの作動特性を変化させて前記第一および 第二の波長を包含する範囲にわたってレーザ波長を変化させるための手段;を含 む請求の範囲1または2の装置。
- 4.前記イルミネータが 前記第一および第二の波長でそれぞれ作動する第一および第二のレーザダイオー ド を含む請求の範囲1または2の装置。
- 5.前記変調するための手段が、与えられた光周波数の光を第一および第二のR F変調周波数において変調して、前記第一および第二の変調周波数のインテグラ ルマルチプルにより光周波数から変位したマルチプル側波帯を生じさせるための 手段を含み;そして 前記復調させるための手段が、前記第一の変調周波数と前記第二の変調周波数と の差の特定のインテグラルマルチプルにおいて信号を検出するための手段を含む 、請求の範囲1または2の装置。
- 6.周波数の差の前記特定のインテグラルマルチプルが、その差自体である請求 の範囲5の装置。
