Claims (1)
- 【特許請求の範囲】 1.約195から800ヌクレオチド塩基対を含む単離されたDNA分子であっ て、該DNA分子がPlA1決定基を含むGPIIIaのアミノ酸残基配列をコ ードする単離されたDNA分子。 2.配列番号:2、配列番号:6、配列番号:10、配列番号:12、配列番号 :14、配列番号:16、または配列番号:18のヌクレオチド塩基配列を有す る、請求の範囲第1項の単離されたDNA分子。 3.約195から800ヌクレオチド塩基対を含む単離されたDNA分子であっ て、該DNA分子がPlA2決定基を含むGPIIIaのアミノ酸残基配列をコ ードする単離されたDNA分子。 4配列番号:4、配列番号:8、配列番号:20、配列番号:22、配列番号: 24、配列番号:26、または配列番号:28のヌクレオチド塩基配列を有する 、請求の範囲第3項の単離されたDNA分子。 5.外来DNAセグメントに機能しうるように連結された発現ベクターを含む組 換え分子であって、当該外来DNAセグメントが、配列番号:1に示した約残基 1から約残基65の血小板GPIIIaのPlA1またはPlA2決定基のアミ ノ酸残基配列をコードし、当該組換え分子が、それで宿主細胞を形質転換したと きに、天然の血小板GPIIIaPlA1またはPlA2決定基の何れかを発現 する組換え分子。 6.前記外来DNAセグメントが、配列番号:2、4、6、8、10、12、1 4、16、18、20、22、24、26および28から成る辞から選ばれる、 請求の範囲第5項に記載の組換え分子。 7.前記発現ベクターがλgt22である、請求の範囲第5項に記載の組換え分 子。 8.天然の血小板GPIIIaPlA1またはPlA2決定基を含むポリペプチ ドの製造方法であって、該方法が、以下の工程:配列番号:1に示した約残基1 から約残基65のアミノ酸残基をコードする外来DNAセグメントに機能しうる ように連結された発現ベクターを含む組換え分子で宿主細胞を形質転換する工程 ;さらに、該形質転換宿主細胞を前記ポリペプチドが発現するに十分な時間培養 条件下で維持す工程 を含むポリペプチド製造方法。 9.前記宿主細胞が大腸菌である、請求の範囲第8項に記載の方法。 10.前記ベクターがλgt22である、請求の範囲第9項に記載の方法。 11.前記DNAセグメントが配列番号:2、4、6、8、10、12、14、 16、18、20、22、24、26および28を有する群から選ばれる、請求 の範囲第10項に記載の方法。 12.前記ポリペプチドが融合ポリペプチドとして発現される、請求の範囲第8 項に記載の方法。 13.さらに前記発現ポリペプチドを回収する工程を含む、請求の範囲第8項に 記載の方法。 14.配列番号:1のアミノ酸残基を含む血小板GPIIIaの約65から約2 66アミノ酸残基を含む単離されたポリペプチドであって、前記ポリペプチドが 血小板GPIIIaPlA1,またはPlA2決定基を示す単離されたポリペプ チド。 15.前記ポリペプチドが、配列番号:3、5、7、9、11、15、17、1 9、21、23、25、27および29から成る群から選ばれるアミノ酸残基を 有する、請求の範囲第14項に記載の単離されたポリペプチド。 16.第二のポリペプチドに機能しうるように連結された第一のポリペプチドを 含む、天然の血小板GPIIIaPlA1またはPlA2決定基を免疫学的に模 倣した融合ポリペプチドであって、 前記第一のポリペプチドは、前記第二のポリペプチドにペプチド結合した免疫学 的に不活性なポリペプチドまたは蛋白のアミノ酸残基配列を示し;前記第二のポ リペプチドは、血小板GPIIIaの約65から約266アミノ酸残基を含み、 配列番号:1のアミノ酸残基を含み、さらに、血小板GPIIIaPlA1また はPlA2決定基を示す融合ポリペプチド。 17.前記第一のポリペプチドがβ−ガラクトシダーゼのアミノ酸配列を有する 、請求の範囲第16項に記載の融合ポリペプチド。 18.前記第二のポリペプチドが配列番号:3、5、7、9、11、13、15 、17、19、21、23、25、27および29から成る群から選ばれる、請 求の範囲第17項に記載の融合ポリペプチド。 19.天然のGPIIIaPlA1またはPlA2決定基と免疫的に反応するヒ トの抗体の存在についてヒト抗体含有身体サンプルを分析する方法であって、以 下の工程: (a)前記体サンプルを以下の抗原: (i)GPIIIaの配列を有し、天然のPlA1またはPlA2決定基を示し 、さらに配列番号:1のアミノ酸残基配列を含む約65から266アミノ酸残基 を含む単離されたポリペプチド、または(ii)GPIIIaの約65から26 6アミノ酸残基のアミノ酸残基配列を含み、天然のPlA1またはPlA2決定 基を示し、さらに、配列番号:1のアミノ酸残基配列を含む融合蛋白;と混合す ることによって水性免疫反応混合物を形成する工程;(b)前記水性免疫反応混 合物を、存在している抗体が前記抗原と免疫的に反応して免疫反応生成物を形成 するために十分な時間、生物学的な分析条件下で維持する工程;さらに (c)前記形成した免疫反応生成物の存在を検出し、それによって前記抗体の存 在を検出する工程; を含む方法。 20.前記決定基が天然のGPIIIaP1A1決定基で、前記抗原が、配列番 号:3、配列番号:7、配列番号:11、配列番号:13、配列番号:15、配 列番号:17、配列番号:19およびβ−ガラクトシダーゼのカルボキシ末端に 機能しうるように連結された配列番号:11から成る詳から選ばれる、請求の範 囲第19項に記載の方法。 21.前記決定基が天然のGPIIIaPlA1決定基で、前記抗原が、配列番 号:5、配列番号:9、配列番号:21、配列番号:23、配列番号:25、配 列番号:27、配列番号:29およびβ−ガラクトシダーゼのカルボキシ末端に 機能しうるように連結された配列番号:21から成る群から選ばれる、請求の範 囲第19項に記載の方法。 22.検出が以下の工程: (i)前記免疫反応生成物を、ヒト抗体と免疫的に反応する第二の抗体と混合し て第二の混合物を形成する工程、この場合、前記第二の抗体は機能しうるように マーカーに連結されている; (ii)前記第二の混合物を、前記第二の抗体が該ヒト抗体と免疫的に反応して 第二の免疫反応生成物を形成するに十分な時間、生物学的分析条件下で維持する 工程:さらに、 (iii)前記第二の免疫反応生成物に固定されたマーカーの量を決定し、それ によって天然のGPIIIaPlA1またはPlA2決定基と免疫的に反応する ヒト抗体の存在を決定する工程; を含む、訴求の範囲第19項の方法。 23.前記マーカーが、放射性、酵素、ビオチン、蛍光、および化学ルミネッセ ンス標註から成る群から選ばれる、請求の範囲第22項の方法。 24.前記抗原が、ヒト体液サンプルと抗原とを混合する前に固体支持体に固定 される、請求の範囲第19項の方法。 25.以下の工程: (a)水性媒体中で供与者からの血小板を、GPIIIaPlA1およびPlA 2決定基の一方または他方と免疫的に反応するが両方とは反応しない抗体と混合 して、水性免疫反応混合物を形成する工程;(b)該水性免疫反応混合物を、前 記抗体が免疫的に反応して免疫反応生成物を形成するために十分な時間、生物学 的反応条件下で維持する工程;(c)前記免疫反応生成物の存在を検出し、それ によって当該GPIIIaPlA1またはPlA2決定基の一方または他方の存 在および血小板の型を検出する工程: を含む血小板の型分け方法。 26.前記抗体がGPIIIaPlA1決定基と免疫的に反応し、GPIIIa PlA2決定基とは免疫的に反応しない、請求の範囲第25項に記載の方法。 27.前記抗体がGPIIIaPlA2決定基と免疫的に反応し、GPIIIa PlA1決定基とは免疫的に反応しない、請求の範囲第25項に記載の方法。 28.前記抗体が単クローン抗体である、請求の範囲第25項に記載の方法。 29.以下の要素: GPIIIaPlA1またはPlA2決定基の一方と免疫的に反応するが、両方 の決定基とは免疫的に反応しない抗体を、少なくとも1回の分析に十分な量で含 む容器;および 前記抗体と血小板との免疫反応を検出するための標識:を含む血小板GPIII aPlAの型分けのためのキット形の診断分析系。 30.当該標識が当該抗体に連結されている、請求の範囲第29項に記載の系。 31.さらに、前記血小板GPIIIaPlA1またはPlA2決定基の他方と 免疫的に反応するが、両方の決定基とは免疫的に反応しない抗体を、少なくとも 1回の分析に十分な量で含む第二の容器を含む、請求の範囲第29項に記載の系 。 32.以下の要素: (a)以下の抗原を少なくとも1回の分析を実施するために十分な量で含む容器 ; (i)GPIIIaの配列を有する単離ポリペプチドで、さらに天然のPlA1 またはPlA2決定基を表し、配列番号:1のアミノ酸残基配列を含む約65か ら266アミノ酸残基をその中に含んでいる単離されたポリペプチド、または (ii)GPIIIaの約65から266アミノ酸残基のアミノ酸残基配列を含 み、天然のPlA1またはPlA2決定基を表し、さらに、配列番号:1のアミ ノ酸残基配列を含む融合ポリペプチド、(b)前記ヒト抗体と前記抗原との免疫 反応を検出するための手段;を含む血小板GPIIIaPlA1またはPlA2 決定基に対するヒト抗体の存在について分析するためのキット形の診断分析系。 33.前記抗原が固形支持体に固定されて存在する、請求の範囲第32項に記載 の系。 34.前記抗原が、配列番号:3、5、7、9、11、13、15、17、19 、21、23、25、27、29、およびβ−ガラクトシダーゼのカルボキシ末 端に機能しうるように連結されている配列番号:11又は21から成る群から選 ばれるアミノ酸残基配列を有する、請求の範囲第32項に記載の系。 35.前記ヒト抗体の免疫反応を検出するための前記手段が、別の種からの標識 結合坑ヒト抗体である、訴求の範囲第32項に記載の系。
