Description
Claims (42)
- 1.ジヒドロカンフェロール(dihydrokaempfemrol;DHK )ヒドロキシル化酵素又はその誘導体もしくは部分をコードするか、又はこれを コードする配列に対して相補的なヌクレオチド配列を含で成る核酸単離体。
- 2.前記核酸がDNA又はcDNAである、請求項1に記載の核酸単離体。
- 3.前記酵素が3′,5′−ヒドロキシラーゼである、請求項1又は2に記載の 核酸単離体。
- 4.前記酵素がペチュニア、バーベナ、ヒエンソウ、ブドウ、アイリス、フリー ジア、アジサイ、シクラメン、ポテト、パンジー又はナス由来である請求項1〜 3のいずれか1項に記載の核酸単離体。
- 5.前記酵素がペチュニア由来である、請求項4に記載の核酸単離体。
- 6.図9又は10に記載されているか又はそれに対して少なくとも40%の類似 性を有するヌクレオチド配列の実質的にすべて又は部分を含んで成る核酸配列を 有する、請求項4又は5に記載の核酸単離体。
- 7.トランスジェニック植物に存在する場合の請求項1〜6のいずれか1項に記 載の核酸単離体。
- 8.前記トランスジェニック植物がパラ、ペチュニア、キク、カーネーション、 ガーベラ、ゼラニウム又はユリズイセンである、請求項7に記載の核酸単離体。
- 9.トランスジェニック植物がバラ又はペチュニアである請求項8記載の核酸単 離体。
- 10.核酸単離体を植物細胞又は組織に移行させることができるベクター分子中 に含まれる、請求項1〜9のいずれか1項に記載の核酸単離体。
- 11.前記移行がアグロバクテリウム(Agrobacterium)との共存 培養を必要とする、請求項10に記載の核酸単離体。
- 12.前記ベクター及び核酸単離体が図11に示すpCGP90である、請求項 10又は11に記載の核酸単離体。
- 13.組換えジヒドロカンフェロール(DHK)ヒドロキシル化酵素又はその誘 導体もしくは部分。
- 14.前記酵素が3′,5′−ヒドロキシラーゼである、請求項13に記載の組 換え酵素。
- 15.前記酵素がペチュニア、バーベナ、ヒエンソウ、ブドウ、アイリス、フリ ージア、アジサイ、シクラメン、ポテト、パンジー及びナス由来である、請求項 13又は14に記載の組換え酵素。
- 16.前記酵素がペチュニア由来である、請求項15に記載の組換え酵素。
- 17.図9又は10に記載の又はそれに対して少なくとも40%の類似性を有す るアミノ酸配列の実質的にすべて又は部分を含んでなるアミノ酸配列を有する、 請求項15又は16に記載の組換え酵素。
- 18.トランスジェニック植物中に存在する場合の請求項13〜17のいずれか 1項に記載の組換え酵素。
- 19.前記トランスジェニック植物がバラ、ペチュニア、キク、カーネーション 又はガーベラである、請求項18に記載の組換え酵素。
- 20.前記トランスジェニック植物がバラ又はペチュニアである、請求項19に 記載の組換え酵素。
- 21.組換えジヒドロカンフェロール(DHK)ヒドロキシル化酵素を発現する ことができるか、又はDHKヒドロキシル化酵素に翻訳できるmRNA分子の全 部もしくは部分に実質的に相補的な核酸配列の転写を指令するトランスジェニッ ク植物の製造方法であって、請求項1又は6に記載の核酸単離体を該核酸単離体 の最終的発現を可能にする条件下に適当な植物の細胞に導入し、該細胞からトラ ンスジェニック植物を再生し、そして該トランスジェニック植物を前記核酸単離 体の発現を可能にするのに十分な時間及び条件下で成長せしめる、ことを含んで 成る方法。
- 22.前記組換え酵素が3′,5′−ヒドロキシラーゼである、請求項21に記 載の方法。
- 23.前記核酸単離体の発現が発生段階により制限される、請求項21又は22 に記載の方法。
- 24.前記組換え酵素がペチュニア、バーベナ、ヒエンソウ、ブドウ、アイリス 、フリーシア、アジサイ、シクラメン、ポテト、パンジー又はナス由来のもので ある、請求項23に記載の方法。
- 25.前記組換え酵素がペチュニア由来のものである、請求項26に記載の方法 。
- 26.前記核酸単離体又は酵素が図9又は図10に記載されているのと実質的に 同じか又はそれに対して少なくとも40%の類似性を有するヌクレオチド配列又 はアミノ酸配列を有する、請求項23又は24に記載の方法。
- 27.前記トランスジェニック植物がバラ、ペチュニア、キク、カーネーション 、ガーベラ又はタバコである、請求項21〜26のいずれか1項に記載の方法。
- 28.前記トランスジェニック植物がバラ又はペチュニアである、請求項27に 記載の方法。
- 29.ジヒドロカンフェロール(DHK)ヒドロキシル化酵素をコードするか、 又はそれをコードする配列に相補的なヌクレオチド配列を含んで成る安定に導入 された核酸分子を有するトランスジェニック植物。
- 30.前記酵素が3′,5′−ヒドロキシラーゼである、請求項29に記載のト ランスジェニック植物。
- 31.前記酵素がペチュニア、バーベナ、ヒエンソウ、ブドウ、アイリス、フリ ージア、アジサイ、シクラメン、ポテト、パンジー又はナス由来である、請求項 30に記載のトランスジェニック植物。
- 32.前記酵素がペチュニア由来のものである、請求項31に記載のトランスジ ェニック植物。
- 33.前記酵素が図9又は2に記載のものと実質的に同じかあるいはそれに対し て少なくとも40%の類似性を有するアミノ酸配列を有する、請求項31又は3 2に記載のトランスジェニック植物。
- 34.前記植物がバラ、ペチュニア、キク、カーネーション、ガーベラ、アイリ ス、チューリップ、ユリ、リシアンサス(Lisianthus)、フリージア 、ヒエンソウ、リムニウム(Limunium)又はペラルゴニウムである、請 求項29〜32のいずれか1項に記載のトランスジェニック植物。
- 35.前記トランスジェニック植物がバラ又はペチュニアである、請求項34に 記載のトランスジェニック植物。
- 36.植物からのシトクロムP450分子又は類似分子をコードするか又はそれ をコードする配列に対して相補的なヌクレオチド配列を含んで成る核酸分子をク ローニングする方法であって、1又は複数のオリゴヌクレオチドプライマーを用 いて1又は複数のポリメラーゼ連鎖反応により前記植物の細胞からの核酸分子の 適切な調製物からシトクロムP450ヌクレオチド配列又は相補的配列を増幅す ることを含んで成り、前記プライマーが既知のミクロソーム由来シトクロムP4 50分子の1又は複数のコンセンサス配列に由来するヌクレオチド配列を有する ものである、前記方法。
- 37.前記コンセンサス配列がシトクロムP450分子のヘム結合ドメインから のものである、請求項36に記載の方法。
- 38.前記コンセンサス配列がF(G,S)XGXRXCXGであり、ここでX は任意のアミノ酸である、請求項37に記載の方法。
- 39.前記コンセンサス配列がFMPFGAGXRXCLGであり、ここでXが 任意のアミノ酸である、請求項37に記載の方法。
- 40.前記シトクロムP450分子又は類似分子がDHKヒドロキシル化酵素で ある、請求項36〜39のいずれか1項に記載の方法。
- 41.前記DHKヒドロキシル化酵素が3′,5′−ヒドロキシラーゼである、 請求項40に記載の方法。
- 42.前記クローン化された酵素が図9又は10に示すのと実質的に同じかある いはそれに対して少なくとも40%の類似性を有するアミノ酸配列、又はヌクレ オチド配列によりコードされているアミノ酸配列を有する、請求項40又は41 に記載の方法。
