動的連続流酵素リアクター
発明の背景
本発明は、血液凝固反応速度及び体内と非常に近い環境中で活性を燐脂質に依存
しているその他の動的酵素反応の速度をin vitroで測定するための動的
連続流酵素リアクター及び方法に関するものである。
人及び動物の凝固系は、止血の維持及び血栓(血液凝塊)形成にも太き(貢献し
ている。凝固は本質的には連続的段階(cascade)であり、通常は血液及
び他の組織中に不活性な酵素前駆物質、即ちチモーゲンとして存在する各凝固因
子が順次活性化されて蛋白質分解酵素になり、それが系列中の次のチモーゲンを
選択的に襲い、それによってこれが活性酵素に変るのである。プロセスの各段階
で増幅がおこり、最初は小さかった刺激が究極的にはかなりの量のフィブリン凝
塊を生成する。
凝血の連続段階は二つの別々の径路として始まり、それらは最後は一つになる。
一つの径路は血液に固有であるが、もう一つの径路は“外因性”といわれている
。なぜならばその径路は通常は血液中に存在しない凝固因子によって引き金をひ
かれるからである。固有の径路は負傷後の止血に大きな役割を演じる。外因性径
路は種々の病的状態、例えば広汎性内皮損傷、進行癌、内毒素血(endoto
xemia)及び妊娠合併症において活性化され得る。
現在、因子■、ビタミンに一依存血漿凝固因子蛋白質及び組織因子、通常は血球
と結合していない細胞結合蛋白質が相互作用するとき体内で凝固が開始するとい
うことを示す証拠がかなりある。[例えばネマーソン(Nemerson)の“
血液(Bfood)″71巻、1−8ベージ、 198B参照]。この相互作用
の結果、活性化されたコンブレタスが生成し、それは酵素活性をもち、他の二つ
の蛋白質、因子X及び因子■をそれらの酵素的活性型、因子X、及び因子■、に
それぞれ変換することによって凝固を開始する。(一般的には、活性血液凝固因
子の前駆物質、即ちチモーゲンはローマ数字であられし、その活性型は下付き文
字“a”によって示される。例えば因子Xはチモーゲンを、因子X は活性化さ
れた因子を表す。
組織因子は殆どすべての細胞の表面に存在する血液凝固促進(procoagu
lant)蛋白質であり、普通は血液と直接接触することはない。しかし組織因
子は種々の薬物学的仲介物質、例えば腫瘍壊死因子、インターロイキン−1及び
内毒素によって刺激されて内皮細胞及び単球内に誘発される。外因性凝血径路は
、因子■−ビタミンに一依存性セリンプロテアーゼチモーゲンを錯体化し、活性
化する組織因子によって引き金をひかれる。組織因子による因子■の活性化はカ
ルシウムイオンの存在下でおこり、因子■の構造変化に起因すると信じられてい

。因子■チモーゲンから因子■ 活性酵素への変換は、チモーゲンのarg−i
1eペプチド結合を切断し、Gla部分を含む軽鎖と酵素活性部位を含む重鎮と
を有する因子■、を生成することによって実現する。
チモーゲン、因子■が凝固促進活性をもつならば、凝固開始は単純に、通常は因
子■を組織因子から分離している物理的障壁の破壊に続いておこるはずである。
こうして、止血がおこるためには、血液凝固を開始する負傷そのもので十分であ
る。チモーゲンがその誘導酵素に比較して少量の活性をもっことを決定するには
困難を伴う。というのは、活性チモーゲンは、痕跡量の酵素が混入した不活性チ
モーゲンと同じ活性をもつからである。たいていの場合、この問題のアプローチ
は、単に自活性部位(ac目ve 5ite−divected)酵素阻害剤、
例えばジイソプロピルフルオロ燐酸(DFP)、或いは適当なりロロメチルケト
ンでチモーゲンを処理し、それにより混入酵素を阻害するということである。
普通チモーゲンはほとんど不活性であるから、これは測定可能の活性の完全な喪
失をおこす。しかしながら因子■チモーゲンはそれ自体DFPによって容易に阻
害され、こうしてこの単純なアプローチをうまく回避する。実際、因子■のDF
Pに対する反応性は非常に大きく、このこと自体、因子■チモーゲンの非常に大
きい活性を示唆している。
ウシ因子■及び■、を用いるDFP阻害研究は、因子■チモーゲンは因子■ 活
性の1%よりわづかに小a
さい活性を含むだけとはいえ、因子■が定性的には因子■、と同じ酵素活性をも
つことを示した。DFPはヒト因子■も阻害することがわかり、その速度は因子
■、の阻害速度の3分の1であった。これはウシ蛋白質を用いたときに認められ
た割合とほぼ同じである。

実験は、組織因子と因子■との物理的錯体化によって凝固が簡単に開始するとい
う意見を支持している。
この考え方を支持するその他の証拠は、ウシ因子■及び■ はほとんど同じ解離
常数をもって組織因子に結合するという考察に由来する。ヒト組織因子のソース
として単球を用いた場合、同じ現象がヒト因子■及び■、で認められた。従って
、蛋白質分解的凝固開始を仮定する必要はなく、これによって蛋白質分解的事象
の無限の退行の問題は回避される。チモーゲンのこの活性度は概して異常で、凝
固系において特異であるように見える。因子■の又は実際、因子■ の活性は、
静かな(Biescenj)凝固系と矛盾しない。なぜならば組織因子が存在し
ないときにはそれは凝固の引き金をひ(ことができないからである。
固有の反応性をもつため、因子■は他のすべての既知の凝固チモーゲンと区別さ
れる。このように因子■チモーゲンは酵素活性をもっているから、チモーゲン及
び活性酵素を区別せずに、−緒に言う場合には因子■(a) という表現を用い
る。
組織因子も補因子の中では独特である。というのは凝固因子V及び■並びに凝固
連続段階におけるその他の補因子とは異なり、成熟組織因子の蛋白質は活性をあ
られすためにはそれ以上の処理を必要としないからである。これらの考察をまと
めると、血液凝固の開始のために組織因子にとって必要なことは、それが因子■
と物理的に錯体化することだけであることが示唆される。
組織因子は、燐脂質と結合した膜結合−糖蛋白質であり通常は循環中に存在しな
い。血管が破壊されると血漿凝固因子である因子■が組織因子と錯体化すること
ができ、それによって触媒的活性種(Ap6cie+)が形成され、それは固有
の径路の成分である因子■(血漿トロンボプラスチン成分)及び外因性、及び固
有凝固径路どちらにも含まれる因子X[スチュアート(Sjuarj)因子]を
活性化し、因子■ 及び因子X。
が生成する。組織因子は、血液凝固をモニター且つ研究する診断薬として重要な
臨床的用途をも有する。
因子■は血漿中に痕跡量存在する(約101M)。因子■が著しく欠乏している
人にひどい出血が見られることは、この蛋白質の生理的重要性を示している。因
子■欠乏は稀であるが、最近のデータは、欠乏患者の約16%に普通は死に通づ
るような脳出血がおこることを示唆している。ラグニ(Ragni )らの“因
子■欠乏(Factor Vll Deficiency )、 Amer、
J。
Hemajol、、 10巻、 79−88ページ(1981) 、他方、因
子■を正常レベルの5%しかもたない患者でもほとんど、又は全く出血症状をお
こさないことがある。しかしながら成る一定の因子■レベルでもかなりの臨床的
変動性がある。
種々の病気、例えば癌及び心臓血管病は血管中の凝血形成の増加と関係がある。
心臓血管病の主な治療の中には抗凝固剤、例えばワルファリン及び関連薬剤の使
用も含まれる。それらはビタミンに一依存性凝血因子(例:因子■、■、 IX
、 X)の合成を阻害する。この治療が種々の血栓塞栓症、肺動脈塞栓症、心筋
梗塞(心臓発作)の発生率を低下させることを示す研究が多数である。しかしな
がらワルファリン治療は、時には致命的な出血をかなり高い発生率でおこす。
ワルファリン型抗凝固剤の投与量をモニターする標準的方法は、クイックone
−stageプロトロンビン時間を用いるこ方法である。静止的条件下で行われ
るこの試験では、患者血漿の試料を37℃にあたためる。それから組織トロンボ
プラスチン(粗組織因子)の懸濁液をカルシウムイオンと共に血漿試料に加え、
凝固時間を測定する。正常な凝固時間は12+/−0,5秒である。この抗凝固
剤の治療的範囲は、凝固時間を正常値の1.2乃至1.5倍の範囲にする薬剤血
中濃度である。このように狭い範囲のため、臨床検査では正確さが必要であるが
、この正確さには達しないことが多い。これらの不正確さの原因は抗凝固剤のい
くつかの出血性副作用にあると考えられている。
因子■は同様な方法で測ることができる。
患者血漿の希釈液を正常血漿に加え、one−stageプロトロンビン時間試
験を行う。試験試料中の因子■の量は試験試料の凝固時間を正常血漿の希釈液で
得られるそれと比較することによって推定される。実際、現在では凝固系のすべ
ての試験が種々の血漿の凝固時間の測定(常に静止条件下の試験管中)に基づい
ている。しかしながらin vivoにおける血液凝固は動いている流れの中で
おこるのが常であるから、静止系においては流れが酵素反応に与える影響を正し
く評価することはできない。
今では、種々の血液凝固チモーゲンをカルシウムイオンと共に、内面を平らな燐
脂質の二重層膜で被覆した管状ハウジング部分を一定の流速で通すことによって
、特異的血液凝固酵素反応が動的方法でより特異的に行われ得ることが判明した
。任意に、そしてより好ましくは、その平らな膜が精製組織因子を含んでいる。
ハウジングを通過する試薬は、因子■又は因子■ のどちらか(それは組織因子
と結合して酵素的活性種を形成する)を、組織因子−因子■錯化合物の基質とな
る因子■又は因子Xと共に含む。因子■ 又は因子X の産生速度は、これらの
因子のために適した試験法によって容易に分析される。動的反応は、現在の静的
方法によるよりも、活性因子の産生をより特異的に分析することができる。
更に、例えば患者からの血漿試料をこのような燐脂質膜被覆デバイスを一定流速
で且つ本発明のその他の条件下で通過させ、ることにより、血漿試料と組織因子
含有−燐脂質膜との接触によって生成、する特異的酵素生成物の測定が可能とな
り、特異的凝固因子欠乏に関する貴重な情報が得られ、先行方法によるよりも鋭
敏に患者の正しい抗凝固パラメーターをモニターすることができる。
発明の酵素リアクターは、血液凝固反応以外の燐脂質依存性酵素反応を行い、分
析するためにも用いられる。このような反応は、試薬溶液中の種々の不活性の酵
素反応成分が燐脂質二重層膜−被覆−管状ハウジングを通過して流れることを含
む。試薬溶液中の不活性酵素成分はハウジングの内面の燐脂質膜と相互作用して
酵素的活性になり、反応産物を溶出液中で分析することができる。
発明の概要
本発明によると、体内に見出されるものと非常に近い環境において血液凝固反応
速度及びその他の燐脂質依存性酵素反応の速度をin vNroで測定できる動
的連続流酵素リアクターが提供される。本発明によると、種々の凝固因子の活性
化速度、特に因子Xから因子X への活性化及び因子■から因子■ への活性化

(両方共因子■又は因子■ を介する)の速度を測定する方法、並びにトロンビ
ン産生速度の測定法も提供第1図は本発明の一実施態様に従う、因子■ 及び組
織因子による因子Xの活性化を示す。

発明の詳細な説明
本発明は、平らな燐脂質の二重層から成り、任意に、且つより好ましくは酵素又
は酵素補因子を含む脂質膜で内面を被覆した管状ハウジングから成る動的連続流
酵素リアクターに関するものである。管状ハウジングは、特異的液体試薬をその
中に供給する手段に連結可能の一端をもち、第二の開口はそこからの溶出液を集
め分析する手段に連結可能である。管状ハウジングは、両端が開いている、又は
開放可能の毛細管であることが好ましい。
種々の精製凝固因子又はチモーゲン、又はそれらに代る血漿をカルシウムと共に
ポンプで酵素リアクターを通過させ、燐脂質膜中の酵素又は補因子と反応させる
。これらの因子がリアクターに入る時の流速を調節し、リアクターを出る溶出液
中の生成物濃度を測定することによって、一連の血液凝固段階の種々のチモーゲ
ンから活性生成物への活性化率(速度)を推定し、生成物形成に対する流れの影
響を測定することができる。その上、活性化種が定常的産生レベルに達するまで
の時間も計算できる。
発明の好ましい実施態様によると、燐脂質二重層は、血液凝固因子の活性化を開
始する酵素補因子である精製組織因子を含む。リアクターを通過する凝固因子は
因子■又は因子■ 、並びに因子■又は因子Xである。
凝固因子はカルシウムイオンと共に燐脂質膜−被覆−毛細管を通過し、管の内面
の組織因子含有−燐脂質二重層膜上を流れる。膜中の組織因子が流動物質中の因
子■又は因子■ に接触するとき毛細管の内側に酵素的活性なコンブレタスが形
成される。この酵素的活性なコンブレタスは、これはこれで流動物質中の因子■
を因子■ に、又は因子Xを因子X に活性化する。

リアクター人口における因子■又は因子X及び因子■又は因子■ の流速及び濃
度を調節し、リアクターからの溶出液中の因子■ 又は因子X の濃度をモニタ
a
−することによって、因子■が因子■、に又は因子Xが因子X に活性化される
速度が計算され、それに対する流れの影響が測定できる。その上因子■又は■。
の種々の濃度の場合、因子■ 又は因子X、の定常的産生に達するまでの時間も
計算できる。
因子■は因子■ に置き代えることができ、その反対も可能である。なぜならば
既述のように、チモーゲンの活性は活性酵素の約1%に過ぎないとはいえ、チモ
ーゲン因子■は定性的にはその活性酵素誘導体である因子■ と同じ血液凝固促

1−8ページ(198B)。こうして因子■は前処理して因子■、にすることな
く、原材料としてこの酵素リアクターに用いられる。
本発明のその他の実施態様によると、精製因子■又は因子■ と、精製凝固因子
である因子V、因子■。
因子■及び因子Xとの混合物を、プロトロンビンと共に膜被覆−毛細管に導入す
る。それからトロンビン産生−9因子■ 産生−9又は因子X、産生速度を測定
し、種々濃度の各凝固因子のトロンビン−1因子IX +、又は因子X 産生
に対する影響をモニターすa
ることができる。
本発明のもう一つの実施態様によると、血漿を一定の流速で燐脂質膜−被覆酵素
リアクターに導入し、燐脂質膜中の組織因子との相互作用によって凝固因子の活
性化を開始する。それから選択した因子、例えばIX、X 又はプロトロンビ
ンなどの産生を測定することができる。この実施態様は、プロトロンビン時間の
静的測定によって得られるよりも特異的に凝固形成を評価することができ、血液
凝固不全患者又は抗凝固剤治療を受けている人の評価に適用できる。
連続流酵素リアクターの毛細管の大きさは様々で、内径は約0.10乃至1.1
0mm、長さは約1乃至15cmである。低い壁剪断速度(約205ec−”)
では生成物の形成は、等式(L/Q)2/3により、毛細管の大きさに比例する
。この式においてLは管の長合、Qはリアクターを通る流速で、この式は拡散調
節反応を示している。高剪断速度(1005ec−1以上)では、拡散はあまり
重要でなく、酵素動力学が主になると考えられる。
毛細管は、先ずそれを沸騰洗剤溶液、例えばスパークリーンTM(Sparkl
een”)に浸し、その後それを超音波洛中で蒸溜脱イオン水ですすぐことによ
って準備される。それから120℃で乾かし、組織因子を含む脂質小胞の懸濁液
を満たす。それから毛細管に0.14MNaCfと1mg/mlウシ血清アルブ
ミン(BSA)を含む0.OIMN−2−ヒドロキシエチルピペラジン−N−2
−エタンスルフォン酸(HEPES)緩衝溶液(H′CAでP Hを7.5に調
節)をどっと流す。
充填毛細管は最後は緩衝液中に室温で保存し、部材が空気にさらされないように
する。
精製組織因子を含む脂質小胞懸濁液は、好ましくはバッチ(Bacb)らの“再
構成された燐脂質小胞中の組織因子に結合する因子■:ホスファチジルセリンに

(られる。この方法は、透析可能の非イオン性洗剤オクチルグルコジッドの大過
剰の存在下で組織因子を燐脂質小胞に挿入する段階を含む。透析により洗剤を除
去すると、大きい燐脂質小胞内に精製組織因子が自然に挿入される。小胞はホス
ファチジルセリン又はその他の酸性燐脂質と、ホスファチジルコリンとの混合物
からつくられる。0乃至40%ホスファチジルセリン(p s)と60乃至10
0%ホスファチジルコリン(PC)との混合物を約1乃至10IIIo1組織因
子対100.OOOmol燐脂質の比で組織因子と結合させるのが好ましい。
精製組織因子は既知の方法でウシ脳又はヒト脳又は胎盤からつ(られ(例えばス
パイサー(Spicer)ら、Proc、 Najl、 Acad、 Sci
、 84巻、 5148−5152ページ。
(1987)或いは1987年6月12日提出のマネーソンらの米国特許出願第
062.166号に提供される組換えDNAクローニング法によってつくられる
。
第2図及び第4図に関しては、酵素リアクターを用いる場合、毛細管(1)を貯
蔵用緩衝液から取出し、カルシウムイオン溶液と、因子■又は因子■、と、因子
V。
因子■、因子■、因子X及びプロトロンビンから成る精製凝固因子の中の一つ以
上とを含む注射器(3)に連結する。これらの精製因子は公知の蛋白質分離法に
よって得るか、組換えDNAクローニング法によってつくられる。ポンプ(4)
を用いて溶液を一定速度で毛細管(1)を通し、種々の凝固因子を組織因子含有
一平面燐脂質膜と反応させる。活性化された酵素生成物を含む溶出物質を毛細管
(1)の出口でコレクタ中に集め分析する。
それから選択した酵素の産生速度を測定する。因子■ 又は因子X の産生速度
を測定する一つの方法は、a
出発材料としてトリチウム標識化因子■又は因子Xを用い、その後毛細管溶出液
中に生成した酸性溶性トリチウムの量を測定することである。その他に標準ラジ
オアッセー又は蛍光(蛍光発生)アッセー法を用いて本発明のりアクタ−中にあ
られれる酵素反応の生成物を分析することができる。蛍光アッセーを用いる場合
、種々の化学的成分の相互反応の後に、毛細管溶出液の蛍光を時間の関数として
測定する。
所与の酵素の産生速度を測定するもう一つの方法は、酵素生成物に特異的な色素
原基質を酵素リアクターの溶出液に加え、それからその流れを連続流光度計を通
過させるというものである。例えば第4図に示されるように因子Xaの産生速度
を測定しようとする場合、色素原基質52222@(6)(ロッテンバーグ(L
oHenberg) ら、Meth、 Enxymol、 80巻、
341−361ページ、 1981)を溶出液に加えてから光度計を通過させる
。酵素リアクター中で生成する因子X の濃度は405、nmにおける吸光度の
変化によって測定される。
或いは、例えばトロンビン産生速度を測定しようとする場合は、酵素リアクター
溶出液が光度計に入る前に色素原基質52238”(7)(ロッテンバーグら、
Mefh、 Enxymol、 30巻、 341−361ページ、 19
81)をそれに加える。1種類以上の酵素産生速度を同時に測定しようとする場
合、酵素リアクターの出口の流れを分け、各流れに異なる色素原基質を加え別々
に吸光度を測定する。比例ポンプ(8)を用いて流れを混合段階(9)にまで送
り、その徒刑々の流れは分光光度計(10)及び(11)に送られる。
1種類以上の酵素生成物について測定した濃度レベルを用いて、これら生成物の
各々で定常的産生レベルに達するまでの時間を計算することができる。分析の目
的で、所与の生成物の定常的産生レベルの1/2に達する時間(T17゜)を測
定してもよい。これはデータの比較をより容易にする。今や本発明の連続流酵素
リアクターを用いて得られるこの新しいパラメーター、即ち定常状態の産生に達
するまでの時間は、血液凝固メカニズムの分析を高める。これに対し、従来の静
的凝固アッセイは定常状態的条件に関する情報を与えることができず、定常状態
に達するための時間に与える流速の影響を評価することもできない。
本発明の酵素リアクターは、最低3000 5ec−”の壁体剪断速度まで安定
である。これは人体の血管系の最大平均壁体剪断速度、即ち約2000 5ec
−”乃至5000 5ec−1に匹敵する。
以下の非制限的実施例は本発明をより詳しく説明するだめのものである。
実施例1
米国特許出願第062.166号に記載の組換えDNAクローニング法によって
つくられるか或いはスパイサーらがP[OC,Na11. Acad、 S
ci、 84巻、 5184−5152 。
1987に記載したようにウシ脳又はヒト脳又は胎盤粉末から公知の蛋白質分離
法によって得られる精製組織因子を含む燐脂質小胞は次の方法で作成される。
CHCで、中でPS及びPCをO:100(PS:pc)から40 : 60
(PS : PC)までのモル比で一つにし、N2気流下、次いで真空中で2時
間乾燥し、硼珪素ガラス管壁に薄いフィルムを形成する。それからO,LM
NaCJ及び0.05Mトリス、PH7,5(TBS)中に溶かした15倍過剰
モルのオクチルグルコジッド(200mM)を加え、混合物を時々攪拌しながら
完全に透明になるまで室温でインキ0.1%トライトン”X−100を含むTB
S中の精製組織因子を燐脂質−オクチルグルコシラド標本に加え、トライトン@
X−100濃度が<0.02%。
組織因子:燐脂質:オクチルグルコシラドのモル比が1乃至10:100.00
0:1.500,000゜である最終溶液を形成した。
最終物質中の蛋白質及び燐脂質を正確に定量するために、痕跡量の[”C] P
C及び3H−組織因子を加えた。3H組カウント対14Cカウントの比は約10
乃至100対1であった(組織因子濃度に依って)。液体シンチレーション計算
法のために一部をとり、残りを室温で3X1リツトルのTBSに対して72乃至
96時間透析した。その後その物質をセファロース0CL−2Bカラム(1,5
X55cm)でTBS中で室温でゲル濾過した。3H−組織因子及び14C−燐
脂質の回収を液体シンチレーション計数法で測定した。
生成懸濁液は2mMPS/PC脂質小胞と20乃至200nM組織因子とから成
っていた。
標準ガラス毛細管を準備するために、先ずそれらを1gスパークリーン”/ 5
00 ml蒸溜、脱イオン水の沸騰洗剤溶液に30分間浸し、それらを超音波洛
中で蒸溜、脱イオン水で5分間づつ3回、合計15分間すすいだ。それから毛細
管を120℃で30分間乾かし、組織因子を含むP S/P C脂質小胞の調製
懸濁液を充填した。10分後管にHEPES/アルブミン緩衝液(0,OIM
HEPES、0.14NaCj2゜1mg/m1BsA、HC矛を用いてPHを
7.5に調節する)を流し込み、その緩衝液に浸して保存し、脂質膜と空気との
接触を阻止する。
実施例2
実施例1に期した方法によって作成し、被覆した毛細管(1,D、=0.56m
m、L=75mm)を因子■、の種々の溶液、10100n!Jチウム標識因子
X及び5HM・CaCJ□を含む注射器に連結した。精密ポンプを用いて、溶液
を27.1μJ2/minの一定の流速で毛細管を通す。管を出る流れについて
生成した酸溶性トリチウムの量を測定することによって、因子X、の産生及び濃
度を分析した。定常状態における結果を1表に示す。

X の濃度は定常状態では因子■2濃度に無関係である。しかしながら第1図は
、上記画濃度の因子■ に対して、時間の経過と共に生成する因子X、の量を説
明している。因子X、が定常状態に達するまでの時間は因子■ の濃度とは逆関
係に変化することが判った。その他に、因子■を同じ条件下で用いても同様な結
果を与えた。即ち、因子■の濃度が減少するにつれて、定常状態に達するまでの
時間は増加した。因子■の濃度0. 1HMでは、因子X、の定常状態濃度は1
0.6HMであった。
因子X8の産生が定常状態に行く過程は除々であったから、因子X、のT17゜
を計算した。第1図では、T1/2は因子■、0.5−101M濃度から得られ
、約4分であることがわかる。これらの因子■ 濃度は人体の正常な因子■濃度
の5乃至100%に相当する一方、体内の因子■濃度が5%以下の場合はひどい
出血がおころのが普通である。第1図に示されるように、因子■、濃度が0.1
1M(正常の1%)及び0.0751M(正常の0.75%)に低下するニつれ
て、T172は著しく増加する。この実施例で発生した壁体剪断速度はヒト静脈
系のそれ、例えば約20乃至40 sec””に近かった。
実施例3
毛細管の大きさ及び流速が異なる以外は、実施例2と同じ実験条件を実施した。
毛細血管の内径0.33=及び長さ125mmの場合の結果を2表に示す。

200 2.40計算値は856sec−1であった。これは小動脈で得られ
る平均壁体剪断速度と、微小循環で得られるそれとの中間である。400μ、&
/minでは、得られた壁体剪断速度は17125ec−”であり、微小循環の
速度に近い。このように、酵素リアクターは生理的範囲に匹敵する剪断速度に対
して明らかに安定的で、あった。よって、凝固系の種々の異常の影響を、静脈系
から微小循環までに互る流れ条件下で評価することができる。
内径的0.10mm乃至1.10胴、長さ約1.0乃至15anの範囲の毛細管
が容認されることがわかった。
管直径の変化は産生ずる生成物の量を変えるが発明の基礎的メカニズムを変える
ことはないことがわかった。
上の実施例は、本発明の連続流酵素リアクターを用いて得ることができるデータ
の型を示す。これは公知の静的方法を用いては得られないデータの型である。
上記は発明の範囲を制限するものではない。なぜならばここに請求する酵素リア
クターを用いてトロンビン生成速度、全血漿中の凝固因子の産生及びその他種々
の燐脂質依存性酵素反応を測定することができる。

Claims (24)
- 1.燐脂質依存性酵素反応を行い、分析するための動的連続流酵素リアクターで あって、内面を平らな燐脂質二重層膜で被覆した管状部材から成り、そのハウジ ングは第一の開口が液体流試薬をそこに供給するための手段と連結可能で、第二 の開口がそこからの溶出液を集め、分析するための手段と連結可能である動的連 続流酵素リアクター。
- 2.ハウジングが毛細管である請求の範囲1記載の酵素リアクター。
- 3.ハウジングが、内径約0.1乃至1.1mm、長さ約1.0乃至15cmを 有する請求の範囲1記載の酵素リアクター。
- 4.燐脂質膜が、酵素と酵素補因子とから成る群から選択される最低1つの付加 的成分を含んで成る請求の範囲1記載の酵素リアクター。
- 5.補因子が組織因子である請求の範囲4記載の酵素リアクター。
- 6.燐脂質膜が中性及び酸性燐脂質を含んで成る請求の範囲1記載の酵素リアク ター。
- 7.燐脂質膜がホスファチジルコリン及びホスファチジルセリンを含んで成る請 求の範囲1記載の酵素リアクター。
- 8.燐脂質膜が60乃至100%ホスファチジルコリンと0乃至40%ホスファ チジルセリンとの混合物から成る請求の範囲1記載の酵素リアクター。
- 9.燐脂質膜が70%ホスファチジルコリンと30%ホスファチジルセリンとの 混合物から成る請求の範囲1記載の酵素リアクター。
- 10.燐脂質膜が組織因子を、100,000mole燐脂質に対し約1乃至1 0mole組織因子の割合で含む請求の範囲5記載の酵素リアクター。
- 11.(a)酵素反応を行うための試薬を定められた流速で、内面を平らな燐脂 質二重層膜で被覆した管状部材から成る動的連続的酵素流リアクターの入口に供 給し、試薬及び膜成分は、別々では酵素反応の遂行に対して不活性であるが一緒 になると酵素的活性系を形成し、 (b)試薬を定められた一定流速で酵素リアクターに送り込み、試薬と燐脂質膜 との接触によって媒介される酵素反応が酵素リアクター中でおこり、(c)溶出 液中の酵素反応産物を酵素リアクターの出口から集め、 (d)酵素反応中に生成する生成物の量を適当な試験法によって測定する、 諸段階から成る動的酵素反応を連続的に行い測定する方法。
- 12.燐脂質膜がその他に、酵素と酵素補因子から成る群から選択される最低一 つの付加的成分を含んで成る請求の範囲11記載の方法。
- 13.補因子が組織因子である請求の範囲12記載の方法。
- 14.試薬が血液凝固因子とカルシウムイオンを含んで成る請求の範囲13記載 の方法。
- 15.血液凝固因子が、因子VII及び因子VIIaから成る群から選択される 血液凝固因子を、因子IX,因子X,因子V,因子VIII,プロトロンビン及 び全血から成る群がら選択される最低一つの因子と共に含んで成る請求の範囲1 4記載の方法。
- 16.血液凝固因子が因子VII及び因子IXである請求の範囲14記載の方法 。
- 17.血液凝固因子が因子VIIa及び因子IXである請求の範囲14記載の方 法。
- 18.血液凝固因子が因子VII及び因子Xである請求の範囲14記載の方法。
- 19.血液凝固因子が因子VIIa及び因子Xである請求の範囲14記載の方法 。
- 20.血液凝固因子が因子VII,因子X,因子V及びプロトロンビンである請 求の範囲14記載の方法。

- 22.生成物量を測定する試験法が、ラジオアッセイ,色素発生試験,蛍光発生 試験から成る群から選択される請求の範囲11記載の方法。
- 23.更に、生成した産物量を測定する前に反応産物に特異的な色素発生又は蛍 光発生基質を溶出液に加えることを含んで成る請求の範囲11記載の方法。
- 24.更に、酵素リアクター中で生成する一つ以上の生成物の量を測定できるよ うに、酵素リアクターからの生成物溶出液を分割することを含んで成る請求の範 囲11記載の方法。
