明 細 書
ヒトのモノクローン性抗HIV−1−抗体本発明は、免疫学的に特殊なエイズ治
療の分野およびHIV−感染した細胞の生体内での画像の分野に関する。より詳
細には、本発明は、ヒトのモノクローン性抗HI V−1−抗体および該抗体か
ら得られた免疫学的化学薬品、ならびに該免疫学的化学薬品を使用する治療法に
関する。ヒトのモノクローン性抗体(h umAbs)の生産は、感染および他
の疾病の免疫学゛的治療に使用することができる抗体の新しい源を表わすことが
できる。マウス起源の代わりにヒト起源のmAbを使用することは、ヒトにおけ
る抗体応答の誘導を阻止する。マウス抗体のFc部分は、異種の投与された蛋白
質に対して抗体応答を最小にするためにヒトへの注入よりも先に除去しなければ
ならない。しかし、ヒトのモノクローン性抗体は、ヒト個体の相同蛋白質の一部
を表わす、ヒト免疫不全ビールス(HI V)に感染したヒトの血清の場合には
、抗ビールス抗体は、感染後の一定期間に亘って後天性免疫不全症候群(AID
S)の如何なる臨床学的発現もなしに検出することができた。活性免疫応答の前
記状態で高い数の抗原−特異的B細胞は、循環していることが予想された。この
日細胞は、ヒトモノクローン性抗HIV抗体を発生させるための融合成分として
使用された。本明細書中では、つぎのことが記載されるニ
ーHIV抗厚に対するヒトmAbの発生、−HIV感染した細胞の成長の実質的
な阻害、一記載したヒト抗H夏V−1−mAbおよびA鎖毒素と接合した免疫毒
素の調製、
一免疫毒素を用いてのHIV−1感染した細胞の選択的殺害、
一抗体依存性細胞毒性によるHIV−1感染した細胞の選択的殺害、
−ヒトモノクローン性抗HIV−1抗体を用いての細胞感染の選択的阻害、
一ヒト血清中のHIV−1抗原の選択的測定。
発明の詳細な説明
物質および方法
ヒト抗HIVモノクローン性抗体()(u抗HIV−1−mAb)
本発明の好ましい実施態様によれば、使用したhu抗HIV−1−mAbは、ケ
ンネット(Kennet)等著、“Monoclonal antibodie
s and functional cell 1ines;progress
and applictions”、PIenum Press(New Y
ork)、1984に記載された公知のハイブリッド形成方法により得られたI
gG1サブクラスを有する。ハイブリドーマ3D6の試料は、E CA CC%
Porton Dovn、 5alisbury S P 4 0 J G、英
国に寄託した。3D6は、HIVエンベロープ蛋白質を認める抗体を生じる6種
類のハイブリドーマの中のただ1つ寄託したものであった。寄託しt:ハイブリ
ドーマのECAACC寄託番号および寄託データは次の通りであった:8711
0301 ; 1987年11月3日。
融 合
記載した方法で実施した融合(K6hler等、Europ、 Journal
of Immunologys 6 (1976) = 292 )。
PEG1500との細胞融合よりも先に、HIV T血清の正の供与体からの
抹消血液リンパ系細胞は、3回血清遊離細胞培養液で洗浄された。細胞は、融合
よりも3日前にアメリカヤマゴボウの根茎(PWM)5n9と直ちに融合された
か、または該根茎で刺激された。細胞は、HAT感受性融合細胞と5:1の比で
混合された。細胞は、DMSO7,5%(Serva、 F RG)の存在下に
PEG 1500 42%(Ferak、F RG)と融合された。細胞は、
希釈を制限することによってクローン化されかつサブクローン化された。
ハイブリドーマのスクリーニング
19Gおよび19Mイソタイプは、公知方法でペルオキシダーゼ接合した抗ヒト
1gG、19M(重鎮特異的)抗体を用いてサンドイッチェンザイムリンクトイ
ムノソルパントアッセイ(ELISA)によって培養上澄み液から分析した。特
異的抗体のスクリーニングは、10μg/m(lの濃度でHIVを使用すること
によリサンドイッチELISAによって実施された。細胞培養から単離されl;
ビールスは、平らな底面のマイクロ滴定板中に塗布され、未結合の物質は洗浄除
去される。培養上澄み液は、4℃で一晩中インキユベートされた。未結合の物質
を洗浄除去しt;後、特異的抗体は、ペルオキシダーゼで標識化された抗ヒト−
!9−重鎖特異的抗体によって検出された。
イムノプロッティング
自然のバイロンから得られたHIVエンベローフ蛋白質を使用することによるイ
ムノプロット:精製されたウィルスは、変質され、5DS2.5%および2−メ
ルカプトエタノール5.0%で90℃で5分間還元され、かつ10%のポリアク
リルアミドスラブゲルに使用された6分離後、蛋白質バンドは、ニトロセルロー
スシート(Schleicber & 5chQl1%F RG )に電子伝達
されt;。このシートをドライミルク5%で遮断した後、ストリップが切断され
、かつl:2の希釈した培養上澄み液SmQ中に浸漬された。結合した抗体は、
)(RPで標識されI;抗ヒトIgG抗体と反応されかつNiCl20.1%を

クローン化された物質から得られたHIVエンベロープ蛋白質を使用することに
よるイムノプロット:5DS−ゲル電子泳動法は、レンムリ (Laemmli
) (Nature227、l 970 ; 680−585)の方法に従って
実施された。それぞれ遺伝的に設計されたエシェリキア コリ E 、COl

ら誘導された、電気泳動的に分離されたエンベロープまたはコア蛋白質は、ニト
ロセルロースにエレクトロプロットされた。牛血清アルブミン3%(PBSに溶
解したBSA)で遮断した後に、ストリップはPBS緩衝液中で希釈された試料
(トリトン(Triton) X −1000,1%、BSA 1%、ゼラチ
ン 0.5%)中に一晩中浸漬され、3回洗浄され、かつペルオキシダーゼで標
識化されたヤギ抗ヒトγ−鎖と一緒にインキュベートされた。
商業的に入手可能なプロットストリップを使用することによるイムノプロット:
デュポン社Du Pantのロフト番号7044128からのプロットストリッ
プは、当社の推薦により使用されIこ。
商業的に入手可能なEIA
商業的に入手可能なEIAは、モノクローン性抗体の特異性を検査するために使
用された。キャッチーエリサ(catch −elisa)および競合的EIA
の双方とも使用されに、ラブシステムズ(Labsystems;(Finla
nd))、オルガノン(Organon;(Belgium))、パスツール(
Pasteur;(Fr、ance))、バイオクロム(Biochrom;(
FRG))、アボット(Abott;(USA))からの検定法およびオルガノ
ン(Organon;(Delgium))からの競合的EIAが使用された。
ペプチドのマツピング
異なるペプチドは、抗原結合部位の評価のために評価された。ペプチドAおよび
Bは、刊行物の記載により合成された。CおよびDは、バイオクロム(llli

多細胞系列の108個の細胞は、ニトロセルロース膜上にスポットされた。ハイ
ブリッド形成試料は、ヒトDNAに対して特異的でありかつゲノム中で約300
000回繰り返されているクローン化されt:A]u配列を含有する32p−標
識化されたプラスミド ブルール(Blur) 8であッf二(Sch+eid
他、5cience216.1982 ; 1065)。−晩に亘る予備ハイブ
リッド形成およびハイブリッド形成の後、膜は3回100mQのSSC中で2回
、0.1%のSDS中で1回室温で5分間洗浄され、かつ2回200m12のS
SC中で1回、0.1%のSDS中で1回60℃で30分間洗浄されIこ。
免疫蛍光法
固定)(IV−1感染した細胞(H9細胞)は、hu抗HIV−mAbと一緒に
高温保持された。mAbの結合は、FIFT標識された抗ヒト!gGで未結合の
mAbを洗浄除去した後に1秒間のインキュベーションによって証明された。
3B6抗体および3A6抗体の精製
精製は、公知方法で実施されt;。澄明にされI;培養上澄み液は、セファデッ
クス(Sephadex)G −25カラム(Pharmacia)上でのゲル
濾過を使用することにより脱塩され、かつさらにCM−セファロースファストフ
ローカラム(Sepharose fast flow column)(Ph
armacia)上でクロマトグラフィー処理された。CM−セファロースファ
スト70−カラム(Sepharose fast flowcolumn)の
溶出液は、限外濾過によって濃縮され、かつフェニル−スーパーロース(Phe
nyl −Superose)(Pharmacia)を使用することにより再
クロマトグラフィー処理された。クロマトグラフィ一工程は、ファーマシア社
Phar+macia companyの推菖により実施された。
フェニル−スーパーロース(Phenyl −5uperose)カラムに溶液
を負荷する前に粗製mAb溶液は、2モルの硫酸アンモニウム溶液によって希釈
された。
3A6tllは、CM−セファロース7アスト70−(Sepharose f
ast flow )および蛋白質−Aスーパーロース(5uperose)を
使用することにより陽イオン交換クロマトグラフィーによって精製された。
リシン−A鎖毒素の製造
リシンは、ヒマ(Ricinus Co+e+eunis)から公知方法によっ
て抽出される。リシンは、粉砕された全体のヒマ(Ricinus Commu
nis)から抽出されるか、或イハl: −F 加工の副生成物であるヒマのケ
ーキから抽出される。ヒマのケーキは、3回40 (v/v)%−60(v/v
)%の石油エーテル5容量で抽出することによって脱脂される。
次に、空気乾燥された物質は、燐酸塩緩衝食塩水(PBS;NaC(10,15
モル、燐酸塩 0.01モル、pH7,2)中で一晩中抽出される。抽出液は、
ナイロンガーゼを通して濾過することによって澄明にされ、引続き1500gで
1時間遠心分離される。澄明な上澄み液は、4℃で飽和硫酸アンモニウムで沈澱
される。硫酸アンモニウム60%の最終的な濃度で、沈澱物は捕集され、かつ遠
心分離によって収集され(1500g、1時間)、PBSの最小量中に再溶解さ
れ、かつ抽出液が硫酸アンモニウムを含有しなくなるまでPBSに対して透析さ
れる。
アフィニティークロマトグラフィー
澄明にされかつ透析された毒素抽出液は、レクチンアフィニティークロマトグラ
フィーによって後精製される。ゲル(Sepharose 4 B s Pha
rmacia社)は、1モルのプロピオン酸で少なくとも4週間室温で前処理さ
れ、レクチンの結合能は増大される。
カラムクロマトグラフィーは、10未満の温度で処理され、レクチン結合を最適
化する。
澄明にされかつ透析された抽出液は、PBS平衡にされt;酸で前処理されたセ
ファロース(5epharose)4B−カラムに使用される。試料の使用後、
カラムはUV吸収が基本線に戻るまでPBSで洗浄される。毒素は、他のレクチ
ンと一緒にPBS中の100ミリモルのガラクトースで溶離される。更に、この
混合物は、セファクリル(Sephacryl)S 200 HRでゲルクロ
マトグラフィーによって分離される。試料の容量は、全床量の3〜4%を越えて
はならない。毒素は、他のレクチンから前記条件下で完全に分解される。アフィ
ニティーカラムから回収された毒素は、セファクリル(Sephacryl)S
20 OHRカラムでの使用前に限外濾過(ミリ孔PTGC膜)によってlo
mg/m4に濃縮される。毒素ピークは捕集され、かつフィルター滅菌された。
滅菌毒素溶液は、A鎖とB鎖とに切断される前に一30℃で凍結されて貯蔵され
た。
A鎖およびB鎖へのりシンの切断
アフィニティー精製されかつ濃縮された毒素は、5%のメルカプトエタノール溶
液によってA鎖とBlとに切断される。この毒素は、β−メルカプトエタノール
(最終的濃度5%)およびガラクトース(最終的濃度0.5モル)で完成された
0、1モルのトリス(Tr 1s)−HCQのp H8,5の緩衝液中5mg/
mQの濃度で室温で一晩中インキユベートされ、引続き37℃で2〜3時間イン
キュベートされる。この毒素は、インキュページ夏ン緩衝液で平衡にされたセフ
ァデックス(Sephadex) G 25カラムでのゲルクロマトグラフィー
によって出発緩衝液(PBS)からインキュベーション緩衝液(Tris緩衝液
)に移される。
インキュベージ3ン後、試料は、0.1モルのトリス(Tris)/HCQ緩衝
液、p H8,5で平衡にされたDEAE−セフ 70−スフテスト70−(5
epharose fastflow)に使用された。このカラムは、全ての未
結合物質が溶出されるまで0.1モルのトリス(Tris)/HCQ緩衝液、p
H8,5で洗浄される。未結合の物質、本質的には純粋なA鎖、は、捕集され
、かつアシアロ−7エツインーセフアロース(asia’1o−feLuin−
Sepharose)4B カラムを通過し、汚染毒素は除去される。アジアO
−7x フィン−セファロース(asialo−fetuin−5epharo
se) 4 Bは、ファルマシア社(Pharmacia)の推奨によって製造
された。未結合の物質は、捕集され、フィルター滅菌され、かつ4℃で貯蔵され
る。
DEAE−セファロース7アストフロ(5epharose fast flo
w)は、ガラクトース0.1モルおよびNaCθ1モルを含有量る0、1モルの
トリス(Tris) −HCρ緩衝液、pH8,5で再生される。
停電点の測定
hu 抗HIV−1−mAbの等電点は、記載した方法によって測定された。
ファルマライト(Pharmalyte)はキャリヤー両性電解質として使用さ
れた。全体の方法は、ファルマシア社(Pharmacia)の推奨によって実
施された(小冊子: l5oelectric focusiB、Pharma
cia fine chemicals)。
二次的種類および軽鎖の測定
軽鎖およびサブクラスは、ELISAによって測定されt;。アルカリ金属ホス
ファターゼで標識化された特異的な抗ヒトに鎖抗体またはペルオキシダーゼによ
って標識化された特異的な抗ヒトGl、G2、G3およびG4抗体が使用されI
;。
リシンA111[の品質制御
品質制御試験は、精製リシン−Aill上で実施される。還元条件下での勾配5
DS−ポリアクリルアミドゲル電気泳動法により、如何なる汚染物質も存在して
いないことが判明する。それぞれ33キロダルトンで1つのみのバンドおよび3

A鎖毒素とモノクローン性抗体との接合精製されI;モノクローン性抗体5mg
(PBS中1〜2m+?/mff)は、ジメチルホルムアミド中1mg/mQ
で溶解されたS P D P (Phar+oacia)の10倍のモル過剰量
と、室温で30分間反応された。FDP置換された抗体は、セファデックス(S
ephadex) G −25を使用することによりゲル濾過によって脱塩され
た。
280nmで連続的に測定された蛋白質ピークは、捕集され、かつ氷上に置かれ
I;。リシンA鎖5mgは、DTT5ミリモルを用いて室温で1時間還元され、
かつセファデックス(Sephadex) G −25のカラム上で脱塩された
。カラムはPBSで平衡にされた。蛋白質ピークは、捕集され、かつ冷たいFD
P置換された抗体と直ちに混合された。この混合物は、4℃で1時間振盪され、
次いで塩化ナトリウム2モルを含有する0、01モルの燐、酸ナトリウム緩衝液
に対して4℃で透析濾過された。未接合のA鎖のバルクは、セファクリル(Se
phacryl)S 200 HRでゲルクロマトグラフィー処理することに
よって免疫毒素から分離された。
カラムは、塩化ナトリウム3モルを含有する0、02モルの燐酸ナトリウム中で
平衡にされた。
免疫毒素を含有した第1のピークは、プールされ、かつ抗ヒトIyG−セファロ
ース(5epharose) 4 B上でアフィニティ精製された。免疫毒素は
、3.5モルの塩化マグネシウムを用いてアフィニティ力ラムから溶出され、か
つ専ら塩化ナトリウムに対して透析され、次いでPBSに対して透析された。
免疫毒素(0,2−2me/ mQ)は、PTGC膜を使用して限外濾過するこ
とによって試験に必要とされる最終的濃度に濃縮された。最終的な調製物は、滅
菌濾過され、かつ−20℃でアリコート中に貯蔵された。免疫毒素は、還元条件
および非還元条件の双方の下で5DS−PAGEによって分析された。免疫毒素
の置換は、ラジオイムノアッセイによって測定された。
抗体1モルあたりA鎖l〜2モルの平均的置換が観察されt;。

hu抗HI V −m A bの一連の希釈液(PBS中でB5Al%およびT
riton X −100)は、4℃で一晩中インキユベートされた精製HIV
エンベロープ蛋白質で被覆された平らな底面を有するマイクロ滴定容器(+m1
crotiter veils)中で分散させた。未結合の物質を洗浄除去した
後、結合した接合体は、1.4−7エニレンジアミンとの反応によって測定され
た。展開された色は、492nmで測定された。較正曲線は、光学密度対希釈度
をプロットしたものであった。半飽和の希釈は、この較正曲線から計算された。
半量大飽和に相当する希釈液でのHu抗HIV−mAbは、AIDSを患ってい
る患者からの血清と混合されたか、または常法のスクリーニング法(ELISA
)および確認試験(ウェスタンプロット)によって血清陽性であることが測定さ
れた発端者からの血清と混合された。接合体(半最大希釈)と、発端者からの血
清との混合物は、HIVエンベロープ蛋白質で被覆されたマイクロ滴定容器中で
インキュベートされ、洗浄され、かつ染色されj;。
前記試料の光学密度は、半量大飽和時に抗HIV−mAbと比較される。半量大
飽和よりも低い値は、ヒト血清と、ヒトモノクローン性抗−HIV抗体との間の
競合を示す。
HIV−1ピリオンの感染性に対する中和活性hu抗HIV−1−mAbの中和
活性のための検定法は、公知方法により実施された(J、 VirolOgy、
61.2024−2028、(1987) ;Nature 316.72−
74.1985 ; Biotechnology 5 940〜946.19
87)。12日百計50%の接種された培養基中で感染を惹起させるための量の
20倍に等しいHI V−1用量(20xT I CD 20)は、H9FI
I胞系統を感染させるt;めに使用された。細胞は、ビールスの接種後3日目、
6日目、9日目および12日後に捕集され、かつ感染の百分率は、免疫蛍光法に
よって測定されI;。100%の感染は、6日目ないし9日目の間に観察された
。また、中和効果は、モノクーロン性抗体の存在下に種々の希釈率で試験された
。
3D6抗体の中和活性は、ビールスの接種直後に抗体の1回量の添加(最終的濃
度IPg/m(+)によって評価された。感染速度は、感染された細胞の百分率
として表わされj二。
細胞毒性試験
細胞毒性試験に使用した試験細胞系統は、HIV−1感染したH9細胞であった
。HI V−1感染したH9細胞は、HIV−1ビールスの感染性に対する中和
活性下で記載しt;ように得られt;。感染されていないH9細胞は、陰性の対
照として使用された。ビールスの接種後6〜9日後に収集されI;細胞は、媒体
1mQ中に懸濁された。hu mAb毒素接合体の種々の希釈液は、感染され
I;細胞および陰性の対照に添加された。37℃で24時間のインキュベージ1
ン後に、細胞はPBSで洗浄され、35S−メチオニンで補われt;メチオニン
遊離媒体が添加された。細胞は37℃で2時間インキュベートされ、次に媒体は
遠心分離によって除去され、細胞は、ガラス繊維フィルターに移され、かつメチ
オノン1mg/mffを含有するトリクロル酢i!10%で3回洗浄された。こ
の細胞は、フィルター上で空気乾燥され、次いでシンンチレーシ2ン液体中に移
される。放射能は、シンチレーションカウンター中で計数された。細胞毒性は、
対照未処理蛋白質合成の50%(TCIDso)を生じた接合体の組織培養抑制
量と、して表わされた。
実験結果
種々の実験には、本発明を詳説しかつ次の実施態様に利用するための実施例とし
て役立つことが示されている。
ヒトの起源、生化学的性質、免疫化学的特性、試験管内および生体内での挙動は
、代表的な結果によって証明されている。

第6図:355−メチオニン配合によって測定された蛋白質合成の抑制率は、細
胞毒性に対する測定値と見なされる。分子量を180000ダルトンと仮定する
ことに基づいて、免疫毒素のモル濃度は計算された。免疫毒素(3D6)−リシ
ン−A鎖のTCIDsoはlOnm未満である。

モノクローン性抗体のクローンおよびヒト起源の発生種々のクローンは、抹消血
液リンパ系細胞および融合細胞系統の融合によって得られた。11種類の11イ
ブリドーマの特異性は、第1表に示されている。

このハイブリドーマ系統(3D6)によって産生されf:h u mA bv
cHI V −1は、本発明を説明するための例として使用されている。3D6
抗体のイムノプロットは、第1a図、第1b図および第C図に示されている。
3A6、l 9H2,23H7,37G12および42B9抗体のイムノプロッ
トは、第1d図に示されている。
モノクローン性抗体のヒト起源は、32P−標識化プラスミドロブルール(Bl
ur) 8のハイブリッド形成によって示されている。ハイブリドーマ細胞のD
NAを用いた場合のオートラジオグラフは、第2図に示されている。また、抗ヒ
ト19Gおよびヒト141M抗血清との反応および二次的種類を測定するための
より特異的な血清は、モノクローン性抗体がヒト起源であることを検定する。
抗体の生化学的性質および免疫化学的特性ハイブリドーマN o 、3 D 6
によって産生されたモノクローン性抗体のイソタイプは、Gである。ELISA
によって測定されたサブクラスはGlである。ファルマシア(Pharmac
ia)固定系統システムを使用しての等電焦点合わせによって測定された等電点
け、pH8,6の範囲内にある。
また、結果の総括は第1表に示されている。
免疫蛍光法
固定HIV感染されt;細胞は、3D6抗体と一緒にインキュベージコンされた
。マーカー接合体抗ヒト!gc−prTcによって惹起されI;特異的蛍光法は
、第3図に示されている。
モノクローン性抗体および関連せる免疫毒素の試験管内での挙動
中和活性
3D6抗体lpg/mI2までの存在下でH9細胞をH!■ビールス細胞の20
倍のTCIDsoによって感染させることはできなかった。中和活性は第4図に
示されている。
20倍のTCID5.を用いてビールス接種してから9日後に3D6抗体0.1
.ug/mffは、H9細胞中のHIV感染を阻止することができた。3A6抗
体の中和活性は、第5図に示されている:6日遅れて観察することができた。感
染速度は免疫蛍光法によってそくていされt;。
細胞毒性
リシンA鎖と共役的に結合したモノクローン性抗体(3D6)は、抗体の細胞毒
性を証明するために使用された。細胞毒性は、355−メチオニン配合によって
測定された。物質および方法について記載したように得られた、3D6抗体とり
シンA鎖との間の接合体から構成されている免疫毒素は、特異的にHI V−1
感染したH9細胞を殺害した。355−メチオニン配合率は、対照の百分率で表
わされている(第6図)。
hu 抗HIV−mAbと、血清陽性者およびエイズまたはブリエイズを患っ
ている患者から自然に生じる抗体との比較
請求の範囲に記載に記載されt:モノクローン性抗体と、HIV感染後にヒトに
おいて自然に生じる抗体との比較のために、競合的EIA(酵素免疫検定法)が
選択された。3D6モノクロ一ン性抗体はペルオキシダーゼと接合され、半量大
飽和の希釈が測定された。
半量大飽和の希釈時に3D6抗体は、エイズの臨床的発現のない血清陽性者から
の異なる血清と混合され、かつエイズまたはプリエイズを患っている患者からの
血清と1:lで混合された。血液中での3D6抗体と、自然に生じる抗体との競
合は、半量大飽和の%で表わされている。
3D6抗体による半量大飽和は、100%と見なされている。第■表には、3D
6−抗体ペルオキシダーゼ接合体および異なる血清を使用することによる競合的
EIAからの値が示されている。

ペプチドCおよびDを3D6抗体と反応させた。同様の競合的配列は、yp12
0の場合にも見い出されI;。これは、5p41とgp120との反応の結果で
ある。
商業的に入手可能なEIA
商業的に入手可能なEIAの結果は、第8図に示されている。全てのEIAの場
合に、Abott ELISAを使用することを除いてプラスの結果が得られた
。オルガノン(Organon)(2)検定物質は、競合的EIAであった。

Claims (1)
- 【特許請求の範囲】 1.ヒトモノクローン性抗体において、a)選択的にHIV−1のエンベロープ 蛋白質に結合し、 b)選択的にHIV−1感染した細胞に結合していることを特徴とする、ヒトモ ノクローン性抗体。 2.ヒトモノクローン性抗体において、a)選択的にHIV−1のコア蛋白質に 結合し、b)選択的にHIV−1感染した細胞に結合していることを特徴とする 、ヒトモノクローン性抗体。 3.ヒトモノクローン性抗体において、a)選択的にHIV−1(HTLV I II/LAV)のエンベロープ蛋白質に結合し、 b)選択的にHIV−1(HTLV III/LAV)感染した細胞に結合し、 c)リシンA鎖に結合した場合には、10ナノモル未満のHIV−1感染したH −9細胞に対してTCID50%を示し、 d)GまたはMインクイプを示し、 e)HIV−1パイロンに対して中和活性を示すことを特徴とする、ヒトモノク ローン性抗体。 4.ヒトモノクローン性抗体において、a)選択的にHIV−1のコア蛋白質に 結合し、b)選択的にHIV−1(HTLV III/LAV)感染した細胞に 結合し、 c)選択的にp24またはp55またはp656に結合し、 d)リシンA鎖に結合した場合には、10ナノモル未満のHIV−1感染したH −9細胞に対してTCID50を示し、 e)GまたはMインクイプを示し、 f)HIV−1バイロンに対して中和活性を示すことを特徴とする、ヒトモノク ローン性抗体。 5.ハイプリドーマによって得られた次の抗体:a)3D6 e)54E7 b)3D9 f)81C7 c)24G3 d)25C2 2 g)該基の員と官能的に等価であるモノクローン性抗体の1つである、請求項1 記載のヒトモノクローン性抗体。 6.ハイブリドーマによって得られた次の抗体:a)3A6 b)19H2 c)23H7 d)37G12 e)42B9 g)該基の員と官能的に等価であるモノクローン性抗体の1つである、請求項1 または4に記載のヒトモノクローン性抗体。 7.HIV−1のエンベロープ蛋白質 a)gp 160、 b)gp 120および c)gP 41および d)HIV−1感染した細胞 に結合している、請求項1記載のモノクローン性抗体。 8.HIV−1のコア蛋白質 a)P 24、 b)P 55 c)P 65 d)HIV−1感染した細胞 に結合している、請求項2記載のモノクローン性抗体。 9.マウス×ヒトハイブリドーマにおいて、請求項1記載のヒトモノクローン性 抗体およびハイブリドーマの子孫を生じることを特徴とする、マウス×ヒトハイ ブリドーマ。 10.マウス×ヒトハイブリドーマにおいて、請求項1または3に記載のヒトモ ノクローン性抗体およびハイブリドーマの子孫を生じることを特徴とする、マウ ス×ヒトハイブリドーマ。 11.マウス×ヒトハイブリドーマにおいて、請求項2記載のヒトモノクローン 性抗体およびハイブリドーマの子孫を生じることを特徴とする、マウス×ヒトハ イブリドーマ。 12.マウス×ヒトハイブリドーマにおいて、請求項2または4に記載のヒトモ ノクローン性抗体およびハイブリドーマの子孫を生じることを特徴とする、マウ ス×ヒトハイブリドーマ。 13.免疫毒素において、請求項1または3に記載の抗体および細胞毒性部分の 接合体であることを特徴とする、免疫毒素。 14.免疫毒素の細胞毒性部分がリシンのA鎖である、請求項13記載の免疫毒 素。 15.免疫毒素の細胞毒性部分がアブリンのA鎖である、請求項13記載の免疫 毒素。 16.免疫毒素の細胞毒性部分がジフテリア毒素のA鎖である、請求項13記載 の免疫毒素。 17.免疫毒素において、請求項7記載の抗体および細胞毒性部分の接合体であ ることを特徴とする、免疫毒素。 18.細胞毒性カクテルにおいて、請求項14および請求項15に記載の免疫毒 素からなることを特徴とする、細胞毒性カクテル 19.請求項1記載のヒトモノクローン性抗体の予防量を投与することによるヒ トホスト中の細胞のHIV−1感染を阻止する方法。 20.請求項1または2に記載の少なくとも2つの異なるヒト抗体のカクテルを 投与することによるヒトホスト中の細胞のHIV−1感染を阻止する方法。 21.HIV感染した細胞を治療する方法において、請求項1記載の抗体の治療 量をヒトHIV感染した細胞ホストに投与することを特徴とする、HIV感染し た細胞を治療する方法。 22.HIV感染した細胞を治療する方法において、請求項14、15、16お よび17に記載の免疫毒素の治療量をヒトHIV感染した細胞ホストに投与する ことを特徴とする、HIV感染した細胞を治療する方法。 23.HIV感染した細胞を治療する方法において、請求項18記載の細胞毒性 カクテルの治療量をヒトHIV感染した細胞ホストに投与することを特徴とする 、HIV感染した細胞を治療する方法。 24.HIV−1感染した細胞を治療する方法において、ホスト体重1キログラ ムあたり請求項14、15、16および17に記載の免疫毒素約0.01〜20 ミリグラムをヒトHIV感染した細胞ホストに投与することを特徴とする、HI V−1感染した細胞を治療する方法。 25.請求項17記載の免疫毒素をHIv−1感染した細胞ホストに投与するこ とによるヒトHIV−1感染した細胞を治療する方法において、全部の免疫毒素 の全体量がホスト体重1キログラムあたり約0.0l〜約20ミリグラムの範囲 内にあることを特徴とする、HIV−1感染した細胞を治療する方法。 26.細胞毒素において、請求項2または4に記載の抗体および細胞毒性部分の 接合体であることを特徴とする、細胞毒素。 27.細胞毒素の細胞毒性部分がリシンのA鎖である、請求項26記載の免疫毒 素。 28.細胞毒素の細胞毒性部分がアブリンのA鎖である、請求項26記載の免疫 毒素。 29.細胞毒素の細胞毒性部分がジフテリア毒素のA鎖である、請求項26記載 の免疫毒素。 30.細胞毒素において、請求項8に記載の抗体および細胞毒性部分の接合体で あることを特徴とする、細胞毒素。 31.請求項27および28に記載の免疫毒素からなることを特徴とする、細胞 毒性カクテル。 32.請求項2記載のヒトモノクローン性抗体の予防量を投与することによるヒ トホスト中の細胞のHIV−1感染を阻止する方法。 33.請求項2または4に記載の少なくとも2つの異なるヒト抗体のカクテルを 投与することによるヒトホスト中の細胞のHIV−1感染を阻止する方法。 34.HIV感染した細胞を治療する方法において、請求項2記載の抗体の治療 量をヒトHIv感染した細胞ホストに投与することを特徴とする、HIV感染し た細胞を治療する方法 35.HIV感染した細胞を治療する方法において、請求項27、28、29お よび30に記載の免疫毒素の治療量をヒトHIV感染した細胞ホストに投与する ことを特徴とする、HIV感染した細胞を治療する方法 36.HIV感染した細胞を治療する方法において、請求項31に記載の細胞毒 性カクテルの治療量をヒトHIV感染した細胞ホストに投与することを特徴とす る、HIV感染した細胞を治療する方法37.ヒトHIV−1感来した細胞を治 療する方法において、ホスト体重1キログラムあたり請求項27、28、29お よび30に記載の細胞毒素約0.01〜20ミリグラムをヒトHIV感染した細 胞ホストに投与することを特徴とする、ヒトHIV−1感染した細胞を治療する 方法。 38.請求項31記載の細胞毒素をHIV−1感染した細胞ホストに投与するこ とによるヒトHIV−1感染した細胞を治療する方法において、全部の免疫毒素 の全用量がホスト体重1キログラムあたり約0.0l〜約20ミリグラムの範囲 内にあることを特徴とする、ヒトHIV−1感染した細胞を治療する方法。 39.請求項1記載の抗体の少なくとも1つを有するヒト血液または組織培養液 体中のHIV−1感染した細胞を特異的に検出する方法。 40.請求項2記載の抗体の少なくとも1つを有するヒト血液または組織培養液 体中のHIV−1感染した細胞を特異的に検出する方法。
