Description
慢性炎症性疾患の予防および/または治療のための薬物の製造における試薬の使用であって、
前記試薬は、
(1)炎症因子として細胞外に分泌される異常量(異常な含有量)のIFP35および/またはNMIの活性を阻害する物質、
(2)炎症因子として細胞外に分泌されるIFP35および/またはNMIの量および/または活性を阻害する物質、
(3)IFP35および/またはNMIの炎症因子としての細胞外への分泌を阻害する物質、
(4)IFP35および/またはNMIの量および/または活性の異常な増加を阻害する物質、
(5)IFP35および/またはNMIの細胞外への分泌を阻害する物質、
(6)炎症因子として慢性炎症性疾患を引き起こすIFP35および/またはNMIを阻害する物質、あるいはDAMPsとして慢性炎症性疾患を引き起こすIFP35および/またはNMIを阻害する物質、
(7)IFP35および/またはNMIがインターフェロン、TNF、IL1および/またはIL6である一部の炎症因子の発現および分泌を上方制御することを阻害する物質、
(8)IFP35および/またはNMIがTLR4と相互作用し、TLR4/MD2を介してNF-κBを活性化することを阻害する物質のうちの少なくとも一つである。
前記試薬は、

一方で、本発明は、治療有効量のインターフェロン誘導タンパク質35kD(IFP35)および/またはN-Myc相互作用タンパク質(NMI)の阻害剤と、薬学的に許容される担体とを含む、ウイルスに感染された個体の炎症反応を治療または緩和するための医薬組成物を提供する。
いくつかの実施形態において、前記ウイルスは、コロナウイルス科のウイルス、例えば新型コロナウイルス(COVID-19)、SARSウイルス、MERSウイルスであるか、またはオルトミクソウイルス科のウイルス、例えばインフルエンザウイルス(例えばA型インフルエンザウイルス)である。
以下の実施例は、本発明のいくつかの実施形態を説明するためにのみ提供されており、制限的な目的または性質はない。
マウスのMSモデルを構築し、IFP35抗体で治療する。結果は、図7に示される。IFP35抗体の治療により、マウスのMSの症状は、有意に軽減される。
マウスのNMI遺伝子をノックアウトする。NMIノックアウトマウスのMSの症状を観察する。結果は、図6および図8に示される。NMIをノックアウトした後、マウスのMSの症状は軽減される。
細胞実験によると、LPS、MOGおよびH37Raによって誘導されるマクロファージ(MS動物モデルと同様)と比較して、正常細胞が誘導された後、マクロファージにおけるIFP35/NMI発現量は、有意に上昇し、細胞外に分泌されるため、血清中でIFP35/NMI含有量の増加を検出することができることを示す。同時に、LPSおよびNMIの刺激下でのミクログリアM1型マーカー分子TNF、iNOS、IL1βのmRNAの発現等、他のいくつかの炎症サイトカインの発現および血清での含有量も増加していることが分かり、iNOS、HMGB1、TNF、iNOS、IL1βなどが図1、図2および図3に示される。
BMDM培養:
頚椎脱臼法によりマウスを犠牲死させ、マウスの二つの後肢を取り、70%アルコールに1分間浸し、后骨の筋肉を極力除去し、注射器で二重抗体(ペニシリン)を含むPBSを吸い込み、骨髓腔を洗浄する。400gで10分間遠心分離し、上清を捨て、赤血球溶解液1mLで赤血球を30秒溶解し、次にPBS 10mLを加えて赤血球溶解液を中和する。400gで10分間遠心分離し、10%のFBS(Gibco)、1%のペニシリン、1%のL-グルタミンのDMEM(Gibico)完全培地を加え、最終濃度が20ng/mLであるMCSF(peprotech)を加え、37℃、5%CO2のインキュベーターで培養する。
10%のFBS(Gibco)、1%のペニシリン、1%のL-グルタミンのDMEM(Gibico)の完全培地、37℃、5%CO2のインキュベーターで培養する。
10%のFBS(Gibco)、1%のペニシリン、1%のL-グルタミンの1640(Gibico)の完全培地、37℃、5%CO2のインキュベーターで培養する。
10%のFBS(Gibco)、1%のペニシリン、1%のL-グルタミンのDMEM(Gibico)の完全培地、37℃、5%CO2のインキュベーターで培養する。
MOGおよびH37Raを最終濃度が100ng/mLである上記細胞に加えて8時間刺激する。
マウス組換えタンパク質mNMIを発現させ、インビトロでマウス初代ミクログリアを刺激し、細胞濃度を2×106に調節し、5μg/mLのmNMIで6時間インキュベートし、QPCRでマクロファージの分極マーカーを検出する。
BMDM細胞のトータルRNA抽出は、康為世紀生物公司のRNA抽出キットカタログ番号CW0597 BV2細胞のトータルRNA抽出用Trizol(invitrogen)を使用する。
PrimeScript(登録商標) II 1st Strand cDNA Synthesis Kit カタログ番号6210AおよびSYBR(登録商標) Premix Ex Taq(登録商標)(Tli RNaseH Plus)、ROX plus Q-PCRキットのカタログ番号RR42LRは、すべてTAKARA社製である。
実験で選択されたマウスは、バイタルリバー実験動物技術(Vital River Laboratory Animal Technology)社から購入した、8~12週齢のC57BL/6バックグラウンドの雌マウスである。オリゴデンドロサイトタンパク質MOG35-55およびフロイント完全アジュバントCFAで乳化し、各マウスに0.2mgのMOG35-55および0.2mgのCFAを皮下注射し、百日咳毒素PTXと組み合わせ、各マウスの腹腔内に500ngを注射し、48時間目でPTXを1回注射する。偽処置グループは、MOG35-55の代わりにPBSを投与し、残りは、同量のCFAおよびPTXで処理し、マウスの発症情况および生活状態を毎日観察し、記録する。マウスの発症後に、マウスの発症に対して採点し、採点基準は、次のとおりである。0:臨床症状なし、1:マウスの尾の麻痺、2:マウスの片方の後肢の麻痺または両後肢の衰弱、3:マウスの両後肢の麻痺、4:マウスの両後肢の麻痺かつ前肢が影響を受けること、5:マウスの死にかけることである。採点基準に従って、発症マウスに対して採点して評価した。
灌流および脱血されたマウス脊髄組織100mgを取り、RIPA(Proteinase inhibitor cocktail、Roche)を氷上で30分間加え、12000rpm、4℃で10分間遠心分離し、5X SDS-PAGEローディング緩衝液(loading buffer)で95℃、5分間行う。
C57BL/6 WT、IFI35-/-およびNMI-/-は、チャレンジ実験グループとして300pfuの用量でA/PR8毒株に感染し、同時に、C57BL/6野生型マウスに陰性対照グループとして同量のPBSを接種する。マウスの血清中のIFP35およびNMIのタンパク質レベルがウイルス感染後三日目に変化するかどうかを検出し、同時に、血清中のTNFおよびIL6の含有量を検出する。
マウスにIFP35モノクローナルハイブリドーマ細胞を腹腔内注射し、7~10日後、マウスの腹水を採取し、次のようにプロティンGアグロスビーズ(Protein G Agrose beads)(GE)で腹水の抗体を精製する。
(1)プロティンGアグロスビーズスラリー(slurry)1mLを、タンパク質精製充填カラムに加え、
(2)リン酸塩緩衝液10mLで精製カラムのマトリックスを洗浄し、
(3)腹水1mLをリン酸塩緩衝液5mLに加え、精製カラムに加え、4℃で2時間インキュベートし、
(4)洗浄:20mLのリン酸塩緩衝液でカラムを洗浄し、
(5)溶出:適切な量の0.1Mグリシン(Glycine)(PH=2.5~3.0)緩衝液で抗体を溶出し、ここで溶出チューブに1M PH=10.0のTris-HCl緩衝液を事前に加え、溶出液を中和し、
限外ろ過チューブで抗体を濃縮し、かつ抗体濃度を測定する。
マウスEAEモデルを構築し、IFP35抗体治療グループおよびIgG対照グループに分け、12日目に抗体の投与を開始し、各マウスに毎日100μg(5mg/kg)の抗体を10日間連続的に静脈内注射し、マウスの二つのグループの臨床症状を比較する。
(1)Elisaプレートを取り出し、室温に戻せ、希釈された標準品およびサンプルをウェルあたり100mL、室温下で2時間加える。標準品およびサンプルは、1%BSAのPBST(0.05%のTween-20)で希釈する。
(5)終了:ウェルあたり2Mol/LのH2SO4を50mL加える。
ELISAプレート、450nmのスキャンでOD値を得る。
1.MOGおよびH37Raによる、マウスマクロファージからのNMI放出の誘導。
図2の方法:a、100ng/mLのLPSを使用してミクログリアBV2を誘導し、0、2、4、6、8時間で細胞から上清に分泌されるNMIの量を検出し、b、異なる濃度のLPS(0、10、20、50、100ng/mL)を使用してBV2細胞を8時間刺激し、上清中のNMIの量を検出する。図のエラーバーは、三つの繰り返し実験±s.e.mを表し、有意差は、マッチングしないt検定によって検出され、*P<0.05、**P<0.01、***P<0.001。
図3の方法:精製されたマウスNMI(mNMI)タンパク質(5μg/mL)を使用して、マウスミクログリアを6時間誘導し、LPSまたはNMIの刺激下で、ミクログリアのM1型マーカー分子であるTNF、iNOS、IL1β(a)およびM2型マーカー分子であるArg1、IL10、TGF-β、CD206(b)mRNAの変化を検出し、図のエラーバーは、三つの繰り返し実験±s.e.mを表し、有意差は、マッチングしないt検定によって検出され、*P<0.05、**P<0.01、***P<0.001。
図4によると、EAEモデリング後、異なる時点でのマウスのNMI分泌および発現は、偽処置対照実験グループマウスおよびEAEマウスの発症前期間(7日)、初期発症期間(12日)、発症ピーク期間(20日)、疾患緩和期間(30日)のNMI発現を検出すことを示す。結果によると、マウスにおけるMSの発症の様々な期間でのNMIの発現量がMSの発症と密接に関連していることを示す。
図5によると、EAEモデリング後、異なる時点でのマウス脊髄組織内におけるNMIおよび主な炎症因子iNOS、COX2、HMGB1の発現を検出し、正常なマウスの脊髄ではiNOSがほとんど検出されないことが分かり、MSがピーク期間に入ると、iNOSの発現量が最高レベルに達し、COX2およびHMGB1は、MSの発症とともに増加し、NMIの正常な発現量は低く、MSが発症し始めると発現量が最も高いことを示す。NMIおよび他の炎症因子の発現は、MSの発生とともに増加することができ、NMIは比較的早い時期で上方制御される因子の一つであることを示す。マウスの脳脊髄液は入手が難しいため、ここで検出されたのは脊髄組織である。
図6の方法:a、NMIノックアウトマウス(NMI-/-)および野生型マウス(WT)のMS動物モデルを構築し、それらの臨床症状を比較する。採点基準は、次のとおりである。0:臨床症状なし、1:マウスの尾の麻痺、2:マウスの片方の後肢の麻痺または両後肢の衰弱、3:マウスの両後肢の麻痺、4:マウスの両後肢の麻痺かつ前肢が影響を受け、5:マウスの死にかけることである。採点基準に従って、発症マウスに対して採点して評価した。b、NMI-/-およびWTマウスの脊髄組織のHE病理学的切片。c、NMI-/-およびWTマウスの脊髄組織のLFB病理学的切片、IF実験において、GFAPでアストロサイトを標識し、IBA1でミクログリアを標識し、NMI-/-およびWTマウスの脊髄組織の白質(d)および灰白質(e)内側におけるアストロサイトおよびミクログリアの活性化状況を比較する。
図7によると、遺伝子ノックアウトがMSマウスの症状を有意に改善することができるため、本発明の発明者らは、IFP35の中和抗体を使用して、MSマウスの症状を軽減できるかどうかを観察することを示す。a図は、精製された本発明に記載のIFP35中和抗体を使用して、SDS-PAGE検出することを表し、ここで、55kD位置は、抗体重鎖であり、25kD位置は、抗体軽鎖であることが分かる。抗体が十分に精製されていることを示す。b図は、当該精製されたIFP35抗体に100μg(5mg/kg)の抗体を10日間(12~22日)静脈内注射して、MSマウスの症状を観察および採点することを示す、当該抗体は、マウスMSの症状を有意に緩和できることが分かる。この結果によると、IFP35ファミリータンパク質を阻害モノクローナル抗体は、多発性硬化症(MS)の治療に使用できることを示す。マウスMSモデリングは、公認された評価基準を最小氏、採点基準は、次のとおりである。0:臨床症状なし、1:マウスの尾の麻痺、2:マウスの片方の後肢の麻痺または両後肢の衰弱、3:マウスの両後肢の麻痺、4:マウスの両後肢の麻痺かつ前肢が影響を受け、5:マウスの死にかけることである。採点基準に従って、発症マウスに対して採点して評価した。
結果は図8に示される。a図.NMIノックアウトは、脊髄炎症におけるCD45陽性白血球の浸潤を減少させ、b図.NMIノックアウトは、脊髄炎症因子iNOSおよびCOX2の発現を減少させる。
結果は図9に示される。LPS(i.p.、10mg/kg)でLPS誘導性炎症モデルを構築した後、時間とともにNMI(a図)およびPCT(b図)を変化させ、同様に、腸結紮CLPは、マウス炎症モデル16時間後、NMI(c図)およびPCT(d図)の含有量を構築する。この図によると、PCTと比較して、NMIは、炎症の発生を早期(1~2時間)に検出でき、NMIのバックグラウンド発現は、PCTよりも少なく、NMIがPCTよりも優れた臨床リスク指数検出指標として使用できることを示す。
一.抗体のヒト化:
1.実験方法:PCR部位特異的突然変異誘発方法を介してマウス抗体の特定のアミノ酸残基位置のDNA配列に突然変異を導入して、マウス抗体のいくつかのアミノ酸を変化させて、ヒト抗体配列に変換することにより、抗体のヒト化改変を実現する。次に、配列が改変された抗体遺伝子をpCDNA3.1、pCDNA3.4等の対応する抗体発現ベクターにクローニングする。ヒト腎臓細胞株eExpi293FまたはHEK293T細胞に形質転換して発現し、次に、細胞外に分泌される発現抗体を精製する。発現および精製の改変後の抗体と抗原IFP35との結合能を検出する。使用される機器は、BiACoまたはITC等である。Elisa法を使用して抗原に対するヒト化改変後の抗体との結合能を検出することができる。抗原は、ヒトIFP35タンパク質である。
1.実験方法:実験は、一般的な突然変異プラスミド構築方法およびタンパク質抗体発現方法を採用する。まず、PCRランダム突然変異方法により、ヒト化改変後の一つの抗体の組み合わせAE001-H3+L3のCDR領域のDNA配列にランダムに突然変異を導入することにより、ヒト化抗体のCDR配列を改変する。次に、改変後の抗体配列は、pCDNA3.1またはpCDNA3.4等の対応する抗体発現ベクターにクローンされる。ランダム突然変異抗体ライブラリーを構築する。ライブラリープラスミドをヒト腎臓細胞株293TまたはExpi293F細胞に移して発現し、次に細胞外に分泌される発現抗体を精製する。改変後の抗体およびインビトロで精製された抗原IFP35またはNMIまたはIFP35/NMI複合体の発現および精製を使用して、結合力を検出する。使用される機器は、BiACoまたはITC等である。Elisa法を使用して、CDR領域が改変された後、抗原に対する抗体の結合能を検出する。

IFP35のNIDドメイン(アミノ酸124-220のフラグメント)を免疫原として使用して、マウスを免疫し、マウス脾臓からIFP35に対するモノクローナル抗体をスクリーニングして製造し、35NIDmAbとして記録する。
IFP35 NIDの構造ならびに35NIDmAb FabおよびIFP35 NIDの複合体の構造に加えて、本発明の発明者らは、NMI NIDの構造を分析した。NMI NIDの全体的な構造は、IFP35 NIDの構造と非常によく似ている。IFP35 NIDの35NIDmAb Fabの相互作用に関与するアミノ酸と比較して、NMI NIDの対応する位置のアミノ酸は、IFP35 NIDとは異なり(以下の配列の比較図を参照)、対応する残基は、それぞれArg185、Asn186、Lys215、Lys218、Lys219、Glu221、Tyr222、Pro223、Tyr225、Cys230、Arg232、Thr234である。これらのアミノ酸は、NMI NIDと35NIDmAbとの相互作用に影響を与えることができる。しかし、これらのNMIアミノ酸残基は、NMI上の抗体結合エピトープを表しており、スクリーニングして中和抗体を取得するのに役立つ。

(1)IFP35 NIDおよびNMI NIDの発現および精製
IFP35 NIDをコードする遺伝子フラグメント(残基124~220)をpGEX-6p-1にクローニングし、大腸菌で発現させる。IFP35 NIDのプラスミドをBL21(DE3)に形質転換する。LB培地に100mg/Lのアンピシリンを加え、37℃で細菌を培養する。OD600が0.8~1.0に達する場合、最終濃度が0.5mMであるイソプロピル(isopropyl)-β-D-チオグラクトシダーゼ(thioglactosidase)を使用して16℃で20時間誘導する。その後、4000rpmで15分間遠心分離して細菌を収集する。溶解緩衝液(20Mm Tris at pH8.0、400mMのNaCl、5%のグリセロール(Glycerol))で細菌を再懸濁し、超音波ディスラプターで細菌を破壊し、その後、16000rpmで30分間高速で遠心分離する。遠心分離された上清液をGSTアフィニティーカラムと1時間結合させた後、GSTアフィニティーカラムを洗浄する。溶出緩衝液(20mMのTris at Ph8.0、150mMのNaCl、30mMのグルタチオン(Glutathione))で組換えタンパク質を溶出し、溶出されたタンパク質をPPase酵素で消化し、イオン交換カラム(高塩緩衝液:20mMのTris at Ph8.0、1MのNaCl、5%のグリセロール、低塩緩衝液:20mMのTris at Ph8.0、100mMのNaCl、5%のグリセロール)およびゲルろ過クロマトグラフィーカラムSuperdex200(緩衝液:20mMのTris at Ph8.0、150mMのNaCl)でさらに精製する。SDS-PAGEタンパク質接着剤およびクマシーブリリアントブルー染色を使用して、組換えタンパク質の純度および完全性を検証する。
文献で報告された方法を使用して、35NIDmAbハイブリッド腫瘍細胞を作成し、35NIDmAb抗体を生成する。35NIDmAbを取得した後、PBSに1mMのEDTAおよび1mMのを加え、この緩衝液を使用してパパイン(papain)を溶解する。精製された35nidmabをパパインで溶解し、100:1(35nidmab:パパイン)の質量比で37℃で6~8時間反応させて、35NIDmAb Fabを取得する。
結晶化プロセスにおいて、35NIDmAb FabおよびIFP35 NIDを1:1の比率で混合し、superdex200を使用して、ゲルろ過クロマトグラフィーによって分離および精製する。結晶スクリーニングは、ハンギングドロップ法を使用し、16℃で0.2μLのタンパク質(10mg/mL)+0.2μLのプール液の混合物でスポットする。35NIDmAb FabおよびIFP35 NIDの結晶は、0.1MのTris(pH8.5)、0.2MのMgCl2、18%のPEG3350を含むプール液で成長する。結晶液に15%のDMSOを加えて低温で結晶を保護し、上海シンクロトロン放射装置のビームライン(beamline)BL17UおよびBL18Uで結晶を回折する。HKL3000で回折データを処理する。
人体(患者または健常者)の血清中のIFP35および/またはNMIの検出方法
(1)サンプル採取:4mLの静脈血を空腹時に採取し、抗凝固剤を含まない使い捨ての真空採血管に入れ、3000r/minで15分間遠心分離し、血清を分離した後、血清サンプルチューブを56℃の水浴に30分間以上入れてウイルスを不活化する。その後、サンプルチューブはテストのために-20℃または-80℃の冷蔵庫に保管され、凍結と解凍の繰り返しを回避する。
(2)サンプルの検出:
酵素免疫測定法(ELISA)を使用して、血清中のIFP35および/またはNMIの濃度を検出し、検出ステップは、次のとおりである。
2.1 試薬、サンプル、標準品の製造:
サンプルおよび試薬を徐々に室温(18~25℃)まで溶解する。標準品を5000pg/mL、2500pg/mL、1250pg/mL、625pg/mL、312pg/mL、156pg/mL、78pg/mLの順に希釈する。標準品の希釈液(0pg/mL)は、空白ウェルとして直接使用される。検出結果の妥当性を確保するために、各検出の対照品は、すぐに使用できるように調整する必要がある。
2.2 検出:
基準ウェル、試験サンプルウェル、空白ウェルをそれぞれ設定する。ELISAプレートを覆い、37℃で2時間インキュベートする。液体を廃棄し、スピンドライし、試験作業溶液Aを追加し、ELISAプレートを覆い、37℃で1時間インキュベートする。ウェル内の液体を廃棄し、スピンドライし、プレートを5回洗浄する。試験作業溶液Bを加え、37℃で1時間インキュベートし、ウェル内の液体を廃棄し、スピンドライし、プレートを5回洗浄する。各ウェルに90μLの基質溶液を加え、ELISAプレートを覆い、暗所で37℃で発色し(反応時間は、15~25分間に制御し、基準ウェルの最初3~4ウェルに明確な青色の勾配がある場合、最後の3~4ウェルの勾配が明らかでない場合、終了することができる)、各ウェルに50μLの停止溶液加えて、反応を停止し、停止溶液の追加順は、基質溶液と同じであり、色が不均一な場合、ELISAプレートを静かに振って、溶液ができるだけ均一になるようにしてください。ELISAプレートの底に水滴がなく、ウェルに気泡がないことを確認した後、マイクロプレートリーダーを使用して各ウェルの光学密度を測定する。
2.3 結果の処理:
標準曲線から血清中のIFP35および/またはNMIの濃度を計算し、SPSS統計ソフトウェアを使用してデータを処理し、平均値±標準差を使用して、サンプルグループおよび対照グループのデータに対するt検定を表し、P<0.05は、差が統計学的に意味を有することを示す。
HEK293T(ATCC、CRL-11268)、A549(ATCC、CRM-CRL-185)およびMDCK(NBL-2)(ATCC CCL-34TM)細胞株をDMEM培地で培養し、10%のウシ胎児血清(Gibco)を追加する。THP1(ATCC、TIB-202TM)およびRAW264.7細胞(ATCCTIB-71TM)をRPMI1640培地(Gibco、C1875500BT)で培養する。9日齢のSPFニワトリ胚は、広東大華農生物技術有限公司から購入した。A型インフルエンザウイルスA/プエルトリコ/8/1934(H1N1)(PR8)毒株をSPFニワトリ胚で増殖させ、ショ糖密度勾配遠心分離で精製した後、MOCK細胞でウイルス力価を測定する。
IFP35モノクローナル抗体(H00003430-M01)は、Abnova会社から購入した。NMI抗体(ab183724)は、abcamから購入した。A型インフルエンザウイルスNS1抗体(sc-130568)は、Santa Cruzから購入した。抗A型インフルエンザウイルス核タンパク質抗体は、abcam(ab128193)から購入した。E6446(二塩酸塩)(HY-12756A)(TLR7/9阻害剤)およびCU-CPT-9b(HY-112051)(TLR8阻害剤)は、Med ChemExpress(MCE、米国)から購入した。TLR3 dsRNA阻害剤(614310)は、Milliporeから購入した。Resatorvid(TAK-242)は、Selleck(米国)から購入した。
C57BL/6(B6)野生型マウスは、広東省医学実験動物センター(GDMLAC)から購入した。公表された文献報告によると、C57BL/6マウスモデルを使用して、NMI-/-またはIFP35-/-純合子ホモ接合型遺伝子ノックアウトマウスを構築する。実験で使用されるすべてのマウスは、8~12週齢であり、性別が一致している。マウスは、SPF環境で飼育され、飼育条件は、中山大学動物福祉使用委員会の基準を満たす。
すべてのベクターは、シーケンシングによって検証され、一過性発現のためにExFect2000トランスフェクション試薬(Vazyme、T202-1)で細胞をトランスフェクトする。簡単に説明すると、ウイルス感染の24時間前に、293T細胞に各プラスミドを一過性にトランスフェクトし、次に5MOI PR8ウイルスを6ウェルプレートの細胞に接種する。細胞を1時間インキュベートした後、PBSで1回洗浄してから、FBSを含まない正常なDMEM培地と交換する。次に、上清液を収集して、ウイルス力価を測定し、異なる時点でのサンプルを収集し、Westernブロット電気泳動によって分析する。
TRIzol LS試薬(米国)を使用して、細胞または肺組織からトータルRNAを抽出する。次に、特異的プライマーU12A:AGCAAAAGAGGレトロウイルスゲノムRNA(1μg)を使用し、PrimeScript II第一の鎖cDNA合成キット(Takara)を使用して逆転写システムを構成する。ウイルスmRNAは、最初にmRNA分離キット(中国葉森生物技術有限公司)で精製し、次にオリゴ(oligo)(dT)18をプライマーとして逆転写反応する。cDNA産物をテンプレートとして使用し、SYBR Green Mix(Applied Biosystems)を使用してqPCR増幅を行う。Quant Studio5(Applied Biosystems、Thermo Fisher Scientific)を使用して、定量的PCR反応を実行する。標準曲線法を使用してデータを分析する。
マウスmIFP35(E10460m)ELISAキットは、EIAab社から購入した。ヒトIFP35 ELISAキット(OKEH02088)は、AVIVA SYSTEM BIOLOGYから購入した。ヒトhNMI(CSB-EL015 893HU)、マウスmNMI(CSB-EL015893MO)ELISAキットは、すべてCUSABIO社から購入した。取扱説明書に従ってELISA実験を行う。簡単な説明は次のとおりである。100μLの多重希釈の標準品、検出サンプル、陰性および空白対照をそれぞれ対応する検出ELISAプレートに追加し、サンプルごとに二つの複製(二重)を設定する。メンブレンを覆った後、37℃で2時間インキュベートする。メンブレンをはがし、ウェル中の液体を捨て、紙タオルの上にバックオフするように逆様にして置き、軽く叩いて残りの液体を捨てる。各ウェルに100μLのビオチン標識(1×)の検出用抗体を加え、メンブレンを覆い、37℃で1時間インキュベートする。メンブレンをはがし、ウェル中の液体を捨て、紙タオルの上にバックオフするように逆様にして置き、軽く叩いて残りの液体を捨てる。1×洗浄緩衝液でプレートを3回洗浄する。各ウェルに100μLのHRP標識の検出二次抗体を加えた後、メンブレンを覆い、37℃で1時間インキュベートする。メンブレンをはがし、ウェル中の液体を捨て、紙タオルの上にバックオフするように逆様にして置き、軽く叩いて残りの液体を捨てる。1×洗浄緩衝液でプレートを5回洗浄する。各ウェルに90μlのTMB基質を加え、暗所で37℃で15~30分間インキュベートする。各ウェルに50μlの停止溶液を加え、ELISAプレートリーダーでOD450吸光度値を読み取る。
すべてのマウス肺組織を4%のパラホルムアルデヒドで固定し、パラフィンに包埋し、4μmのスライスに切断する。次に、ヘマトキシリンおよびエオシン(H&E)ですべての切片を染色し、顕微鏡下で組織の損傷、壊死および炎症性細胞の浸潤状況を確認し、様々な領域をランダムに選択して写真を撮る。
インフルエンザウイルスの研究での患者の血液および正常なボランティア対照サンプルは、中山大学医学部、中山大学第6付属病院、中山大学第1付属病院、広州医科大学第1附属病院、広東省の婦人や子供などからのものである。血清を分離した後、-80℃で保存する。血清サンプルの検出は、すべての参加者のインフォームドコンセントを得た上で関連病院の人体実験論理委員会によって承認された(201701093)。
中華人民共和国国務院が承認した「実験動物管理規則」に従って動物実験を行う。当該動物実験は、中山大学動物福祉使用委員会(IACUC)によって承認され、ライセンス番号は、SYXK2016-0112である。インフルエンザウイルスに関連するすべてのの操作は、第3レベルのバイオセーフティラボで完了した。
定量的データは、平均値±SDで示し、ペアリングしない学生のt検定を使用して、グループ間の差が統計学的意味を有するかどうかを判断する。ログランク検定を使用して、異なる治療を受けたマウスの生存率に有意差があるかどうかを分析する。P値が0.05未満である場合、統計学的意味を有すると見なされ、P値が0.001未満である場合、有意差があると見なされ、P>0.05、*P<0.05、**P<0.01、***P<0.001。
本発明の発明者らは、指定された病院によって新型コロナウイルス肺炎について収集された新型コロナウイルス感染症の5人の重症/重篤の患者の血漿サンプルを検出し、すべての患者のIFP35およびNMIレベルが有意に上昇したことを発見した。同様に、より多くの(数十の)患者血清サンプルを使用してさらに分析すると、同じ結果が得られる。さらに注目すべきは、患者の臨床症状と予後を合わせて見ると、IFP35およびNMIのレベルには高い相関関係があり、即ち、IFP35およびNMIのレベルが高いほど、患者の臨床症状は重症になり、予後は悪化する(本研究でIFP35およびNMIのレベルが最も高い患者は、最終的に死亡する)。図15は、5人の患者の血清IFP35およびNMIの結果を示し、一部のサンプル中のIFP35の含有量は、500~700pg/mLのレベルに達し、これは、本発明の発明者らによって以前に発見された敗血症患者の致死レベル(約700pg/mL)に近いレベルに達し、これは、IFP35および/またはNMIが、特に重症/重篤の患者において、新型コロナウイルス感染の炎症反応に関連していることを示すため、COVID-19に感染された患者の血液検出マーカーとして使用して、医療スタッフが患者の炎症性疾患の重症度および予後を判断するのに役立てることができる。
本発明の発明者らの予備研究(早時研究)は、LPS刺激がマクロファージにNMIを発現および放出させることを誘導できることを示すため、本研究では、陽性対照として使用し、PBSを陰性対照として使用する。
C57BL/6マウスモデルでNMI-/-ノックアウトマウスを構築する。一つのグループのC57BL/6野生型またはNMI-/-ノックアウトマウスをそれぞれ2×106pfuのPR8ウイルスで試験し、感染されたマウスの90%を殺すことができる。すべてのマウスを毎日監視して、生きているかどうか、体重が減っているかどうか、および病気の臨床症状があるかどうかを判断する(例えば、無気力、脱毛、しわの寄った毛皮、腰下ろしの姿勢、急速な浅い呼吸、聞こえるラ音)。各マウスの毎日の臨床スコアは、0(無症状)~5(死にかけ)等である。図19のA図およびB図に示されるように、NNMI-/-マウスは、軽度の臨床症状を示し、体重が減少する(%)。対照的に、野生型マウスの臨床スコアははるかに高く、体重は、30%近く減少する。H&E染色の結果(図19のC図):PR8に感染された野生型マウスの肺組織が損傷され、炎症性滲出液およびうっ血が肺泡空間および肺泡間隔を満たす。NMI-/-マウスの肺組織は、PBSに感染された対象マウスと同様に、明らかな病変がなく、基本的に無傷のままである。実験が終わる際に、野生型マウスと比較して、NNMI-/-マウスは、致死的なPR8感染(LD90)から大きい程度で保護される(図19のD図)。
IFP35の外因性阻害が同様の保護効果を達成できるかどうかを検証するために、薬物としてIFP35中和抗体を使用して、ウイルス感染の1日前に12匹のC57BL/6野生型マウスを治療し、同時に、陰性対照として同量のマウスIgGを摂取して、C57BL/6野生型マウスの別のグループを治療し、各マウスに200μgの抗体を5日間連続して静脈内注射し、次に、すべてのマウスは、0日目にLD90のPR8ウイルスを接種する。各マウスに対して体重を測定し、しわの寄った毛皮、無気力、忍び寄り、腰下ろし、息切れ、聞こえるラ音などを監視し、毎日臨床症状に対して採点し、2週間持続する。実験プロトコルの概略図は、図21のA図を参照する。
