Description
保存された(conserved)ポリヌクレオチドの配列相同性または同一性は、標準配列アルゴリズムによって決定され、用いられるプログラムによって確立されたデフォルトのギャップペナルティが一緒に利用されてもよい。実質的に、相同性(homologous)を有するかまたは同一(identical)の配列は、一般的に、配列全体または全長と少なくとも約50%、60%、70%、80%または90%の中間または高い厳しい条件(stringent conditions)でハイブリッドしてもよい。ハイブリッド化はポリヌクレオチドでコドンの代わりに縮退コドンを含有するポリヌクレオチドも考慮される。
92番のチミン(T)がグアニン(G)、シトシン(C)、またはアデニン(A)に置換;
94番のチミン(T)がグアニン(G)、シトシン(C)、またはアデニン(A)に置換;
102番のアデニン(A)がグアニン(G)、シトシン(C)、またはチミン(T)に置換;
103番のアデニン(A)がグアニン(G)、シトシン(C)、またはチミン(T)に置換;
249番のグアニン(G)がチミン(T)、シトシン(C)、またはアデニン(A)に置換;
251番のシトシン(C)がチミン(T)、グアニン(G)、またはアデニン(A)に置換;または、これらの置換の組み合わせを含むものであってもよい。
L-glutamate + L-cysteine + ATP <-> gamma-glutamyl cysteine + ADP + Pi
様々な菌株を含有している麹から菌株を収得し、それを改良してグルタチオン生産能を有する菌株を選別した。
CJ-5菌株のグルタチオン生産能をさらに改善するために、以下の方法で突然変異を誘導した。
前記実施例2-1の結果から、GSH1タンパク質の86番位置がグルタチオン生産に重要であると判断し、GSH1タンパク質の86番目のシステインアミノ酸が他のアミノ酸に置換された変異タンパク質を発現するようにサッカロマイセス・セレビシエ(S. cerevisiae)CEN.PK2-1D及びサッカロマイセス・セレビシエ(S. cerevisiae)CJ-5菌株の変異菌株を製作し、グルタチオン生産量の増加を確認した。
前記CC02-2490菌株のグルタチオン生産能をさらに改善するために、以下の方法で突然変異を誘導した。
プロモーター変異の有無によるグルタチオン生産能を比較するために、実施例2-1で製作したCC02-2490菌株及び実施例2-3で製作したCC02-2544菌株のグルタチオン生産能を測定した。対照群として野生型であるCEN.PK1-D菌株とCJ-5菌株のグルタチオン生産能も測定し、下記表4に示した。
菌株によるプロモーター変異の効果を確認するために、実施例2-3で確認したプロモーター変異を野生型菌株、CJ-5菌株及び実施例2-1で製作したCC02-2490に導入してグルタチオン生産能を評価した。
先の実施例で確認した内容に基づき、置換位置のうち一部のみ変異を導入してもグルタチオン生産能が増加するかを調べるための実験を行った。
先の実施例で確認した内容に基づいて、置換位置のうち一部のみ変異を導入してもグルタチオン生産能が増加するかを調べるための実験を行った。具体的には、-250(C→T)変異(1点変異(1))または-252(G→A)変異(1点変異(2))、及び前記変異の両方をCEN.PK1-D及びCJ-5菌株に導入し、グルタチオン生産能を測定して下記表9及び表10に示した。
先の実施例で確認した内容に基づいて、置換位置のうち一部のみ変異を導入してもグルタチオン生産能が増加するかを調べるための実験を行った。

Claims (13)
- 配列番号1または2のポリヌクレオチド配列において、92番、94番、102番、103番、249番及び251番ヌクレオチドで構成された群から選択されるいずれか1つ以上のヌクレオチドが他のヌクレオチドに置換され、配列番号1または2のポリヌクレオチド配列と90%以上100%未満の配列同一性を有し、プロモーター活性を有し、前記92番ヌクレオチドの置換がシトシンへの置換であり、前記94番ヌクレオチドの置換がシトシンへの置換であり、前記102番ヌクレオチドの置換がシトシンへの置換であり、前記103番ヌクレオチドの置換がチミンへの置換であり、前記249番ヌクレオチドの置換がアデニンへの置換であり、前記251番ヌクレオチドの置換がチミンへの置換である、ポリヌクレオチド。
- 前記プロモーター活性を有するポリヌクレオチドは、249番及び251番ヌクレオチドが他のヌクレオチドに置換されたものである、請求項1に記載のポリヌクレオチド。
- 前記プロモーター活性を有するポリヌクレオチドは、92番、94番、102番及び103番ヌクレオチドが他のヌクレオチドに置換されたものである、請求項1に記載のポリヌクレオチド。
- 前記プロモーター活性を有するポリヌクレオチドは、92番、94番、102番、103番、249番及び251番 ヌクレオチドが他のヌクレオチドに置換されたものである、請求項1に記載のポリヌクレオチド。
- 前記プロモーター活性を有するポリヌクレオチドが、配列番号3~32の中から選択されるいずれか1つのポリヌクレオチド配列で構成されたものである、請求項1に記載のポリヌクレオチド。
- 請求項1~5のいずれか一項に記載のポリヌクレオチドを含む遺伝子発現用組成物。
- 請求項1~5のいずれか一項に記載のポリヌクレオチド及び目的タンパク質をコードする遺伝子を含むベクター。
- 前記目的タンパク質が、グルタミン酸システインリガーゼ(Glutamate-cysteine ligase)活性を有するポリペプチドである、請求項7に記載のベクター。
- 請求項1~5のいずれか一項に記載のポリヌクレオチド、前記ポリヌクレオチド及び目的タンパク質をコードする遺伝子を含むポリヌクレオチドまたはそれを含むベクターを含む、サッカロマイセス属微生物。
- 前記目的タンパク質が、グルタミン酸システインリガーゼ(Glutamate-cysteine ligase )活性を有するポリペプチドである、請求項9に記載のサッカロマイセス属微生物。
- 請求項9に記載の微生物を培地で培養する段階を含む、グルタチオン製造方法。
- 前記培養された微生物、前記微生物の乾燥物、前記微生物の抽出物、前記微生物の培養物、及び前記微生物の破砕物から選択された1つ以上からグルタチオンを回収する段階をさらに含む、請求項11に記載のグルタチオン製造方法 。
- 配列番号1または2のポリヌクレオチド配列において、92番、94番、102番、103番、249番及び251番ヌクレオチドで構成された群から選択されるいずれか1つ以上のヌクレオチドが他のヌクレオチドで置換され、配列番号1または2のポリヌクレオチド配列と90%以上100%未満の配列同一性を有し、前記92番ヌクレオチドの置換がシトシンへの置換であり、前記94番ヌクレオチドの置換がシトシンへの置換であり、前記102番ヌクレオチドの置換がシトシンへの置換であり、前記103番ヌクレオチドの置換がチミンへの置換であり、前記249番ヌクレオチドの置換がアデニンへの置換であり、前記251番ヌクレオチドの置換がチミンへの置換である、ポリヌクレオチドのプロモーターとしての使用。
