本発明のポリペプチドUFV150558、UFV150850、UFV160655、UFV160656、UFV160664及びUFV160665)をコードするDNAフラグメントを合成し(Genscript)、pcDNA2004プラスミド(社内で改変した強化CMVプロモーターを有するpcDNA3ベクター)にクローニングした。調製した発現プラスミドの293fectin(商標)トランスフェクション試薬(Invitrogen)を用いた一過性トランスフェクションにより、Freestyle(商標)培地中で培養したHEK293F細胞中でポリペプチドを産生した。ExpiFectamine(商標)トランスフェクション試薬(Gibco、ThermoFisher Scientific)を用いた一過性トランスフェクションにより、ExpiCHO(商標)発現培地中のExpi-CHO細胞培養物においてポリペプチドを産生した。Expi-CHO細胞の培養のために、ExpiFectamine CHOエンハンサー及びExpiCHOフィード(Gibco、ThermoFisher Scientific)を、トランスフェクションの1日後に添加した。分泌されたポリペプチドを含有する培養上清を、7日目~11日目(ExpiCHO細胞について)の間に遠心分離によって回収し、続いて0.2μmボトルトップフィルター(Corning)で濾過した。
回収した培養上清中の発現ポリペプチドのレベルを、OCTETプラットフォームを使用するバイオレイヤー干渉法によって評価した。要するに、ビオチン化mAb CR9114をストレプトアビジン(SA)バイオセンサー(Pall ForteBio)に固定し、その後、明確な精製相同ポリペプチドの希釈系列の結合シフトを評価することによって標準曲線を確立した。続いて、本発明のポリペプチド(動態緩衝液中に希釈された5~15μg/mL)を含む、予め希釈された回収培養上清の結合シフトを測定し、確立した標準曲線を使用して、ポリペプチドの濃度を計算した。
ポリペプチド発現レベル及び三量体含量を、トランスフェクション後9日目に、3つの独立した70mL ExpiCHOトランスフェクションについて決定した。結果を図5に示す。親デザイン/構築物とも呼ばれる、前述の構築物5367(配列番号16)(46位から始まって306位のアミノ酸までのアミノ酸のHA1ドメインにおける欠失を含み、HA1ドメインにおける欠失部分を置換する4Gリンカーを含む)と比較して、H1N1 A/Brisbane/07/59ベースのポリペプチドUFV160655(47位から始まって306位のアミノ酸までのアミノ酸のHA1ドメインにおける欠失を含み、HA1ドメインにおける欠失部分を置換する4Gリンカーを含まず、点変異Y392P、R404Q、E434Q、及びS442Aを含む)(配列番号103)は、発現レベルが明らかに増加し(最大40倍)、約500mg/L培養上清に達した(図5A)。
精製
ポリペプチドを2段階プロトコルによって精製した。最初に、回収し清澄化した培養上清を、Porosビーズ(ThermoFisher Scientificから入手)に固定化したHA特異的単一ドメイン抗体からなるアフィニティーレジン(Capture Select)を充填したHiScale 16/20カラム(GE Healthcare)にロードした。このレジンは、H1株由来のヘマグルチニン蛋白質に対する特異性が高い。三量体の単離を向上させるため、カラムに意図的に約15%過剰ロードした。50mMトリス、0.5M NaCl、pH7.4中での結合及び平衡化に続いて、ポリペプチドを、0.1Mトリス、2M MgCl2、40%プロピレングリコール、pH7.4への段階勾配を適用することによって溶出した。UVシグナル(A280)に基づいて、溶出画分をプールし、Millex-GV 0.22μmフィルター膜(Merck Millipore)を通して濾過した。続いて、精度を高める目的で、すなわち、最小量の多量体及び単量体タンパク質を除去するために、収集した溶出ピークを、泳動用緩衝液(20mMトリス、150mM NaCl、pH7.8)中で平衡化したSuperdex 200pg 26/60カラム(GE Healthcare)に適用した。三量体画分をプールし、高速液体クロマトグラフィー(HPLC)Infinity 1260シリーズセットアップ(Agilent)における分析SEC-MALSによって純度を評価した。精製された各ポリペプチドの40μgをTSKgel G3000SWxlカラム(Sigma-Aldrich)に流し(1mL/分)、溶出した物質のモル質量を、miniDAWN Treos多角度光散乱検出器及びOptilab T-rex示差屈折率検出器(Wyatt Technology)によって、溶出した物質のモル質量を測定した。データをAstra6ソフトウェアパッケージで解析し、分子量計算は屈折率シグナルから導いた。
第2の精製工程(サイズ排除クロマトグラフィー)の溶出プロファイルは、大量の非三量体(凝集体及び単量体)ポリペプチド5367がアフィニティーカラムのプールされた溶出画分中に存在することを示した。対照的に、UFV160656のクロマトグラムは少量の凝集体を示しただけで、存在する主要な種は三量体ポリペプチドであった(図6A及びB)。
特性評価
精製ポリペプチドのフォールディング及び温度安定性を、(i)Fabフラグメントの存在下でのSEC-MALS分析、(ii)酵素結合免疫吸着測定(ELISA)、及び(iii)示差走査蛍光測定(DSF)によって評価した。
アミノ酸のみに基づく三量体ポリペプチドの理論的分子量は約90kDaであるが、各タンパク質は5個のN結合グリコシル化モチーフ(NxT/S)を含むので、哺乳動物発現系において産生される場合、分子量はより高くなる。SEC-MALS分析によって決定された分子量は96~106kDaの範囲にあると計算されたが、これはこれらのタンパク質が予想通りかなりグリコシル化されていることを示している(表1)。
デザイン
上記の本発明のインフルエンザヘマグルチニン(HA)ステムポリペプチドを、47位から306位のアミノ酸までのアミノ酸を含むHA1ドメインの一部を欠失させることによって、完全長HAから生成した。連結配列は導入しなかった。親デザインでは、欠失は46~306のアミノ酸からなり、欠失後の2つのHA1末端を、人工的な「GGGG-リンカー」によって連結した(図8)。
表5及び6に列挙したポリペプチドをコードするDNAフラグメントを合成し(Genscript)、pcDNA2004プラスミド(社内で改変した強化CMVプロモーターを有するpcDNA3ベクター)にクローニングした。スクリーニング目的のためのC末端FLAGリンカー-Hisタグを含むポリペプチドを、真核細胞株Expi293F細胞においてマイクロスケール(200μL)で産生した。要するに、細胞を、Opti-MEM(Gibco)中、2.5E+06生細胞(vc)/mLの細胞密度で、96ウェルマイクロプレートフォーマット(Greiner)に播種した。ExpiFectamine293トランスフェクションキット(Gibco)を用いて細胞を一過性にトランスフェクトし、37℃、250rpm、8%CO2、及び湿度75%で3日間インキュベートした。培養上清を、白色96ウェルフィルタープレート(0.22μm PVDF膜)を使用して遠心分離(400×gで10分間)により回収し、凝集体及び細胞残屑を除去した。
全体的には、ポリペプチドは参照タンパク質(UFV160360)と同様のレベルで発現され、代替切断位置(すなわち、代替ヘッドドメイン欠失)を有するデザインと、HA1末端をヘッドドメインに由来するリンカーで連結したデザインとの間で有意差は観察されなかった(図9A)。同様に、bnAb CR9114の結合について有意差は観察されなかった(図9B)。
インフルエンザHA0タンパク質(HA1及びHA2における)の切断がその活性に必要であり、宿主の膜とウイルスの膜との融合を引き起こすことにより、ウイルスゲノムの標的細胞への侵入を促進する。上述した本発明のポリペプチドは全て、切断部位ノックアウト変異R329Qを持って発現され、インビトロ及び/又はインビボでの産生中の分子の推定切断が防止された。
表7に列挙したポリペプチドをコードするDNAフラグメントを、実施例5に記載のように合成した。
一塩基性(R)又は多塩基性切断部位(RRRKK)が挿入されたポリペプチド、すなわち、それぞれUFV160302及びUFV160301では、発現レベルに対する効果は観察されなかった(図10)。両ポリペプチドは、類似したレベルで発現され、329位のアミノ酸のQへの変異によってプロテアーゼ切断に抵抗性のある参照ポリペプチドUFV150850と比較して類似したレベルの三量体含量を示した。
デザイン
上記の本発明のポリペプチドにおいて、CヘリックスのN末端(分子の上部、図1Cを参照)、特にHA2ドメインの405位のアミノ酸から始まり419位のアミノ酸までのアミノ酸配列を、分子の三量化傾向を改善するために、配列番号113のGCN4三量化ドメインによって置換した。この例では、コイルドコイル界面の最適化がCヘリックスのヘプタッド反復配列の最適化によって首尾よく探索された。表8は、ポリペプチドUFV160090における代替の三量体化配列(配列番号56)を示す。CヘリックスのN末端領域における変異は、明るい灰色で強調されている。UFV160097の三量体化配列(配列番号58)は、実施例1に記載のポリペプチドと同一である。CヘリックスのN末端部分のヘプタッド反復配列の野生型HAとの相違部分を灰色で強調した。
表8に列挙したポリペプチドをコードするDNAフラグメントを、実施例5に記載のように合成した。
ポリペプチド発現レベル、三量体含量、及びCR9114結合についての回収した培養上清のAlphaLISA評価は、Cヘリックス三量体界面(すなわち、上記のポリペプチド中に存在するようなGCN4様反復以外)の代替の最適化が許容されることを示した。タンパク質発現レベルの改善が観察されたが(約2倍)、三量体含量は減少が観察された(約2倍)。CR9114の結合は影響を受けなかった(図11)。
デザイン
ヘマグルチニンはウイルス粒子の表面に位置し、タンパク質のC末端部分がウイルス膜に埋め込まれた膜タンパク質である。本発明のポリペプチドの可溶性バージョンでは、膜貫通ドメイン(TM)の開始時のトランケーションによって膜貫通ドメインが欠失された。さらに、代替のトランケーション位置も評価した(表9及び10)。
表9及び表10に列挙したポリペプチドをコードするDNAフラグメントを、実施例5に記載のように合成した。
代替C末端トランケーションの最小の効果が、ポリペプチドの発現レベルで観察された。UFV150565及びUFV150574を除く全てのバリアントが、回収した培養上清のウェスタンブロット分析において、三量体高さに明瞭なバンドを示した(図12A)。
デザイン
本発明のポリペプチドは、共有結合三量体タンパク質として培養上清から精製される。前述したポリペプチドにおいて、共有結合はBループ(397位)及びCヘリックス(405位)における2つのシステイン残基の導入によって確立され、これらは隣接する単量体のシステイン残基と対合することによってジスルフィド架橋を形成する(単量体間ジスルフィド架橋)。この実施例では、これらのプロトマー間ジスルフィド架橋の2つの代替位置を探索した(表11)。
ポリペプチドをコードするDNAフラグメントを上記のように合成した。回収した培養上清の全ての評価を、実施例7に記載のようにAlphaLISAによって行った。
ポリペプチド発現レベル、三量体含量、及びCR9114結合についての回収された培養上清のAlphaLISAによる評価は、代替のプロトマー間ジスルフィド架橋が参照ポリペプチドUFV160090と同等であるか又はより良好であることを示した(図13)。これらのデータは、398位及び405位(UFV160093)、並びに396位及び408位(UFV160088)におけるシステイン残基の導入が、397位及び405位(UFV160090)に導入されたシステインによって形成されるプロトマー間ジスルフィド架橋の代替物を提供することを示した。
この実施例では、AlOH3アジュバント添加UFV160664の用量範囲の保護効果(追跡期間終了時の生存率に基づく)を、偽免疫(PBS)動物及び固定用量のUFV4900(探索的)と比較評価した。
AlOH3アジュバント添加UFV160664は、H1N1 A/Brisbane/59/07 FL HA ELISA力価が試験した全ての用量でPBS群と比較して有意に増加した(P<0.001;マン・ホイットニーのU検定、最高用量から開始するステップワイズ法、及び多重比較のためのボンフェローニの調整を含む)ので、免疫原性であることが示された。30μgUFV160664投与免疫動物の力価は、30μgUFV4900群に匹敵した(図15)。
本発明によれば、AlOH3アジュバント添加UFV160664は免疫原性であり、致死的H1N1 A/Brisbane/59/07マウスチャレンジモデルにおいて保護を提供することが示された。免疫原性及び保護効果は、AlOH3アジュバント添加UFV4900に匹敵する。
この実施例では、2%Adjuplexアジュバント添加UFV160664の用量範囲の保護効果(追跡期間終了時の生存率に基づく)を、偽免疫(PBS)動物及び固定用量のUFV4900(探索的)と比較評価した。
2%Adjuplexアジュバント添加UFV160664は、PBS群と比較して、0.03μgを除く全ての用量で有意な保護を提供した(P≦0.003;フィッシャーの直接確率検定、構築物に対するボンフェローニ補正、及び段階的検定、最高用量から開始するステップワイズ試験)ことが示された。30μgのUFV160664群の生存率(100%)は、30μgのUFV4900群(100%)と同一であった(図17、上のパネル)。
本発明によれば、2%Adjuplexアジュバント添加UFV160664は、致死的H1N1 A/Puerto Rico/8/34マウスチャレンジモデルにおいて保護を提供することが示された。保護効果は2%Adjuplexアジュバント添加UFV4900に匹敵する。
この実施例では、UFV160664の2回投与によるインビボでの免疫原性及び保護効果(追跡終了時の肺ウイルス量に基づく)を、アジュバント単独免疫動物、及びH1N1 A/Netherlands/602/09ナイーブフェレットチャレンジモデルにおける標準治療の季節性インフルエンザワクチンと比較評価した。
5%Adjuplexアジュバント添加UFV160664は両用量で、アジュバント単独群の力価と比較して有意に高いH1 A/California/07/09HA特異的抗体力価を誘導した(P<0.001;事後t検定、多重比較のためのボンフェローニ調整、及び最高用量で開始するステップワイズ試験を含む打ち切りANOVA)が、SoCは誘導しなかったことが示された(図18)。5%Adjuplexアジュバント添加UFV160664は両用量で、アジュバント単独群の力価と比較して有意に高いH1 A/California/07/09HA特異的抗体力価を誘導した(P<0.001;事後t検定、多重比較のためのボンフェローニ調整、及び最高用量で開始するステップワイズ試験を含む打ち切りANOVA)が、SoCは誘導しなかった(図18)。
本発明によれば、両用量の5%Adjuplexアジュバント添加UFV160664が免疫原性であり、且つ50μg用量はSoCワクチン参照群に匹敵する保護を提供することが示された。
この実施例では、UFV160664の2つの用量のインビボでの免疫原性及び保護効果(追跡終了時の肺ウイルス量に基づく)を、アジュバント単独免疫動物、及び異亜型のH5N1 A/Indonesia/05/05ナイーブフェレットチャレンジモデルにおいて、チャレンジ株に相同のH5 FL HA(探索的)で免疫した陽性対照群と比較評価した。
5%Adjuplexアジュバント添加UFV160664の両用量で免疫した動物及び陽性対照群は、追跡期間を生存したが、アジュバント単独群の生存率は25%であったことが示された(図21)。追跡期間中の累積体重減少は、5%Adjuplexをアジュバントとして添加した50μgのUFV160664で免疫した6匹の動物のうち4匹で、アジュバント単独群と比較して減少した。陽性対照群は50μgUFV160664群の4匹と体重減少が同等であり、アジュバント単独群と比較すると体重の減少は有意に少なかった(P<0.001;事後t検定、多重比較のためのボンフェローニ調整、及び最高用量で開始するステップワイズ試験を含むANOVA)(図22)。
本発明によれば、UFV160664は5μg及び50μgの両用量で、死亡の発生が防がれたことが示された。さらに、UFV160664の50μgの用量では、体重減少が減り、且つ肺及び咽喉のウイルス量が有意に減少し、陽性対照群に匹敵した。
この実施例では、本発明のポリペプチド(特にポリペプチドUFV171590)を発現する核酸を含有するアデノベクター26(Ad26)の用量範囲の体液性及び細胞性免疫原性を評価した。比較のために、対照マウスを、空のアデノベクター、固定用量の2%Adjuplexアジュバント添加UFV160664タンパク質、又はUFV171590プライム、アジュバント添加UFV160664ブーストの異種免疫レジメンで免疫した。
本発明のポリペプチドを発現する核酸を含有するアデノベクターは全ての用量で、空のベクターによる免疫と比較して、有意なH1 A/California FL HA ELISA結合力価を誘導したことが示された(108vp、109vp及び1010vp:p<0.001、尤度比検定に基づくトービット回帰モデル)。(図25)。さらに、空のベクターと比較して、有意なHAステム特異的抗体力価(CR9114競合アッセイにより測定)が、109vp及び1010vpのUFV171590によって誘導された(p<0.001;尤度比検定に基づくトービット回帰モデル)(図26)。アジュバント添加UFV160664によるプライムブースト、及びUFV171590プライム、アジュバント添加UFV160664ブーストはいずれも、有意なH1 A/California/07/09 FL HA結合力価(図25)及びHAステム特異的抗体力価を誘導した(p<0.001 尤度比検定に基づくトービット回帰モデル)(図26)。
本発明のポリペプチド(UFV171590)を発現するアデノベクター26は、相同免疫レジメン、又はアジュバント添加UFV160664ブーストと組み合わせた場合に、マウスモデルにおいて、H1 A/California/07/09 FL HAに対する有意な体液性応答及び細胞性応答を誘導することが示された。アジュバント添加UFV160664もまた、有意な体液性免疫応答を誘導したが、UFV171590によるプライムがなければ、検出可能なT細胞応答を誘導しなかった。
哺乳動物細胞におけるタンパク質の発現
本発明のさらなるポリペプチド(すなわち、異なるHA骨格に基づく、図28Aを参照)をコードするDNAフラグメントを合成し(Genscript)、pcDNA2004プラスミド(強化CMVプロモーターを有する社内改変pcDNA3ベクター)中にクローニングした。ExpiFectamine(商標)(Gibco、ThermoFisher Scientific)を用いた一過性トランスフェクションにより、ExpiCHO(商標)発現培地中のExpi-CHO細胞培養物においてポリペプチドを産生した。Expi-CHO細胞培養物に、ExpiFectamine CHOエンハンサー及びExpiCHOフィード(Gibco、ThermoFisher Scientific)を、トランスフェクションの1日後に添加した。7日目に遠心分離し、続いて0.2μm濾過によって、分泌されたポリペプチドを含む培養上清を回収した。
回収した培養上清中の発現ポリペプチドのレベルを、OCTETプラットフォームを使用してバイオレイヤー干渉法により評価した。要するに、ビオチン化mAb CR9114をストレプトアビジン(SA)バイオセンサー(Pall ForteBio)に固定し、その後、明確な精製相同ポリペプチドの希釈系列の結合シフトを評価することによって標準曲線を確立した。続いて、予め希釈したポリペプチドを含有する回収培養上清(カイネティクス緩衝液で約5~15μg/mLに希釈)の結合シフトを測定し、確立した標準曲線を用いて濃度を計算した。
35mLのExpiCHOトランスフェクションの分析は、Hisタグ付きポリペプチドが発現されることを示す(図28A)。発現レベルは42mg/L(骨格H5 A/Vietnam/1203/04)から375mg/L(骨格H1 A/California/07/09)まで変化し、全てのポリペプチドが良好に発現することを示す。さらに、SECプロファイル(図28B)は、発現された各ポリペプチドで、有意な三量体(T)及び単量体(M)画分が検出可能であることを示す。利用した骨格株に応じて、相対的な三量体及び単量体の含量の差が観察された。ポリペプチドの正しいフォールディングをさらに評価するために、関連する抗体(CR9114)及びマルチドメイン(MD3606)の結合をAlphaLISAによって評価した。全てのポリペプチドで、CR9114及びMD3606の両方に対する特異的結合シグナルが観察された(図28C)。発現、SECプロファイル及び結合データは、本発明による変異(例えば、A/California/07/09株に基づくUFV160664の変異)が他のグループ1の骨格に転移可能であることを示す。したがって、ポリペプチドUFV180496、UFV180497、UFV190498、UFV180499、UFV180500及びUFV180501は全て正しくフォールディングされ、三量体であり、培養上清中に分泌された。

Claims (26)
- HA1ドメイン及びHA2ドメインを含むグループ1インフルエンザAヘマグルチニン(HA)ステムポリペプチドであって、
(i)前記HA1ドメインのヘッド領域の欠失;
(ii)前記HA2ドメインの三量体化領域の改変;
(iii)310位に対応するアミノ酸位置のシステイン及び422位に対応するアミノ酸位置のシステイン;
(iv)405位に対応するアミノ酸位置のシステインと組み合わせて397位に対応するアミノ酸位置のシステイン、又は408位に対応するアミノ酸位置のシステインと組み合わせて396位に対応するアミノ酸位置のシステイン、又は405位に対応するアミノ酸位置のシステインと組み合わせて399位に対応するアミノ酸位置のシステイン
を含むアミノ酸配列を含み、
ここで、前記HA1ドメインにおける欠失は、少なくとも47位のアミノ酸に対応するアミノ酸から306位のアミノ酸に対応するアミノ酸までのアミノ酸配列を含み、前記三量体化領域の改変はCヘリックスにおける異種三量体化ドメインの導入を含み、前記Cヘリックスは405位のアミノ酸に対応するアミノ酸から434位のアミノ酸に対応するアミノ酸までのアミノ酸配列を含み、392位のアミノ酸に対応するアミノ酸はP、R又はY、好ましくは、P又はRであり、434位のアミノ酸に対応するアミノ酸はQであり、前記アミノ酸の位置の番号付けは配列番号15のH3番号付けに基づいている、HAステムポリペプチド。 - 前記異種三量体化ドメインはGCN4配列である、請求項1に記載のポリペプチド。
- 前記異種三量体化ドメインは、
CMKQIEDKIEEIESK(配列番号24)、
CIKQIEDKIEEIESK(配列番号25)、
CMEALEKKVDDIEKK(配列番号26)、
CIEALEKKVDDIEKK(配列番号27)、
RMECLEKKVDDIEKK(配列番号28)及び
RIECLEKKVDDIEKK(配列番号29)
からなる群から選択されるアミノ酸配列を含む、請求項1又は2に記載のポリペプチド。 - アミノ酸392に対応するアミノ酸はY、P又はRであり、434位のアミノ酸に対応するアミノ酸はQであり、且つ442位に対応する位置のアミノ酸はAである、請求項1~3のいずれか一項に記載のポリペプチド。
- 405位に対応する位置のシステインと組み合わせて397位に対応する位置にシステインを含む、請求項1~4のいずれか一項に記載のポリペプチド。
- - 395位に対応する位置のアミノ酸はIであり;
- 399位に対応する位置のアミノ酸はY若しくはC、好ましくはYであり;
- 400位に対応する位置のアミノ酸はPであり;
- 401位に対応する位置のアミノ酸はKであり;
- 402位に対応する位置のアミノ酸はSであり;且つ/又は
- 404位に対応する位置のアミノ酸はR若しくはQである、
請求項1~5のいずれか一項に記載のポリペプチド。 - 323位のアミノ酸に対応するアミノ酸はKであり、且つ/又は326位のアミノ酸に対応するアミノ酸はKである、請求項1~6のいずれか一項に記載のポリペプチド。
- 339位のアミノ酸に対応するアミノ酸はTである、請求項1~7のいずれか一項に記載のポリペプチド。
- 前記ポリペプチドはプロテアーゼ切断部位を含まない、請求項1~8のいずれか一項に記載のポリペプチド。
- 329位のアミノ酸はアルギニン(R)でなく、好ましくは329位のアミノ酸はグルタミン(Q)である、請求項9に記載のポリペプチド。
- 前記ポリペプチドは天然の切断部位又は多塩基性の切断部位を含む、請求項1~8のいずれか一項に記載のポリペプチド。

- 少なくとも、516位のアミノ酸に対応するアミノ酸で始まる前記HA2ドメインのC末端部分が欠失されている、請求項1~14のいずれか一項に記載のポリペプチド。
- 前記HA1ドメインにおける前記欠失は1~10個のアミノ酸からなる連結配列によって置換されている、請求項1~15のいずれか一項に記載のポリペプチド。
- 少なくとも配列番号103、104、109又は110の1~231のアミノ酸を含むアミノ酸配列を含む、請求項1~10及び13~15のいずれか一項に記載のポリペプチド。
- 配列番号103、104、109及び110からなる群から選択されるアミノ酸配列を含む、請求項1~10及び13~15のいずれか一項に記載のポリペプチド。
- 請求項1~18のいずれか一項に記載のポリペプチドをコードする核酸。
- 配列番号138~145から選択される核酸配列を含む、請求項19に記載の核酸。
- 配列番号142、配列番号143、配列番号144及び配列番号145からなる群から選択される核酸配列を含む、請求項19又は20に記載の核酸。
- 請求項1~18のいずれか一項に記載のグループ1 HAステムポリペプチドをコードする核酸分子を含むベクター。
- 前記ベクターは組換えアデノウイルスベクターである、請求項22に記載のベクター。
- 請求項1~18のいずれか一項に記載のポリペプチド及び/又は請求項19~21のいずれか一項に記載の核酸及び/又は請求項22又は23に記載のベクター、並びに薬学的に許容される担体を含む医薬組成物。
- インフルエンザウイルスに対する免疫応答の誘導に使用するための、請求項1~18のいずれか一項に記載のポリペプチド、請求項19~21のいずれか一項に記載の核酸、及び/又は請求項22又は23に記載のベクター及び/又は請求項24に記載の医薬組成物。
