Description
「放射線療法(irradiation therapy)」なる語は、内部および外部放射線療法、放射免疫療法、並びにX線、ガンマ線、アルファ粒子、ベータ粒子、光子、電子、中性子、放射性同位体、およびその他の形態の電離放射線を含む各種放射線の使用を含む、複数の種類の放射線療法(radiation therapy)を称するものとして当該技術分野で一般に用いられている。本明細書において、「放射線療法(irradiation therapy)」、「放射線療法(radiation therapy)」、「放射線照射(radiation)」および「放射線照射(irradiation)」は、特に断らない限り、これら全てのタイプの放射線療法を包含する。放射線治療器には様々な種類があり、それらはわずかに異なる仕方で動作する。放射線療法セッションの数と期間は、癌の種類と癌が体内のどこにあるかによる。表在性皮膚癌では、数回の短い治療しか必要としないが、体内の深部の癌ではより長期の治療が必要である。
本明細書において「患者」とは、哺乳動物、好ましくはヒトを含む動物をいう。
8-アザグアニンは潜在的な抗腫瘍活性を有するプリン類似体である。8-アザグアニンは、酵素のtRNA-グアニンリボシルトランスフェラーゼ(tRNA-グアニントランスグリコシラーゼ)によって触媒される転移リボ核酸(tRNA)への取込みに対してグアニンと競合することによってtRNAの修飾を妨害する。tRNAの改変されたグアニン修飾は、細胞分化および腫瘍性形質転換に関係している。8-アザグアニンはまた、43Sおよび80S開始因子複合体の生成を抑制し、それによって翻訳の開始を妨害しタンパク質合成を抑制する。この剤は、腫瘍細胞の増殖を抑制し、細胞分化を刺激する。
メブヒドロリン(ナパジシル酸塩)は、鼻アレルギーやアレルギー性皮膚症を含む、ヒスタミン放出によるアレルギー症状の症状緩和に用いられる抗ヒスタミン薬である。
フルルビプロフェンは、変形性関節症(関節の内層の破壊に起因する関節炎)および関節リウマチ(関節の内層の腫脹に起因する関節炎)に起因する痛み、圧痛、腫れ、および硬直を緩和するために使用される。フルルビプロフェンは、NSAIA(非ステロイド系抗炎症薬)と呼ばれる薬のクラスに含まれている。それは、痛み、発熱、炎症を引き起こす物質の生体の生産を止めることによって働く。プロピオン酸類のこの非ステロイド性抗炎症剤は、構造的および薬理学的にフェノプロフェン、イブプロフェン、およびケトプロフェンに関連しており、他のプロトタイプNSAIAと同様の薬理学的作用を持っている。フルルビプロフェンは、抗炎症作用、鎮痛作用、および解熱作用を示す。市販のフルルビプロフェンは、(+)S-および(-)R-鏡像異性体のラセミ混合物である。S-鏡像異性体はほとんどの抗炎症作用を保有しているようだが、両方の鏡像異性体は鎮痛作用を持っている。他のNSAIAと同様に、フルルビプロフェンの抗炎症作用は、プロスタグランジン合成経路におけるアラキドン酸のプロスタグランジンG2(PGG2)への変換およびPGG2からプロスタグランジンH2(PGH2)への変換に関与する酵素であるシクロオキシゲナーゼ(COX)の可逆的阻害を介して起こる。これにより、炎症、痛み、腫れおよび発熱に関与するプロスタグランジンの濃度が効果的に低下する。フルルビプロフェンは非選択的COX阻害剤であり、COX-1および-2の両方の活性を阻害する。フルルビプロフェンはまた、プロスタグランジン阻害活性の点で最も強力なNSAIAの1つである。

ジギトキシンに類似した強心配糖体であるジゴトキシンは、リエントリー機構(reentry mechanisms)による鬱血性心不全と上室性不整脈の治療、および慢性心房細動の治療における心拍数の制御に使用される。
ジギトキシン
は脂溶性強心配糖体であり、細胞膜ナトリウムカリウムATPアーゼを阻害し、細胞内ナトリウムおよびカルシウムレベルの増加と細胞内カリウムレベルの減少をもたらす。研究では、細胞内カルシウムの増加は細胞死に先行し、細胞内カリウムの減少はカスパーゼ活性化とDNA断片化を増加させ、アポトーシスと癌細胞増殖の阻害を引き起こす。
様々なジギタリス種から分離されるラナトシドCは、鬱血性心不全と不整脈の治療に使用される。
ドキソルビシン塩酸塩は、抗腫瘍活性を持つアントラサイクリン系抗生物質であるドキソルビシンの塩酸塩です。細菌Streptomyces peucetius var. caesiusから単離されるドキソルビシンは、ダウノルビシンのヒドロキシル化同族体である。ドキソルビシンはDNAヘリックスの塩基対間に挿入され、それによってDNA複製を防ぎ、最終的にタンパク質合成を阻害する。さらに、ドキソルビシンは、トポイソメラーゼIIを阻害し、その結果、DNA複製中に切断酵素とDNAが結合した複合体の増加と安定化をもたらし、その後、二本鎖切断後のヌクレオチド鎖の連結を防ぐ。ドキソルビシンは酸素フリーラジカルも形成し、細胞膜脂質の脂質過酸化に続く細胞毒性をもたらす。酸素フリーラジカルの形成はまた、アントラサイクリン系抗生物質の毒性、すなわち心臓および皮膚の血管への影響にも寄与する。

(a)候補化合物を腫瘍細胞培養物に加え、
(b)該腫瘍細胞培養物を1~20グレイ(Gray)の線量で処置し、
(c)該細胞培養物で起きた放射線照射媒体細胞捕食を測定する
工程を含み、
ここで、対照レベルと比較して放射線照射媒体細胞捕食を促進する化合物は放射線療法治療を強化することができる。
化学物質、細胞株および培養条件:
特に断らない限り、化学物質はSigma-Aldrichから購入した。抗生物質、培地、細胞培養のための添加剤はLife Technologiesから得た。ベンジルオキシカルボニル-Val-Ala-Asp(OMe)-フルオロメチルケトン(Z-VAD-fmk)はBachemから、組換えマウスTNF-アルファはR&D systemsから得た。ヒト結腸癌HCT116細胞はMcCoy's 5A培地で培養し、マウス線維肉腫細胞株L929はDulbecco変法イーグル培地で培養した。全ての培地に10%加熱不活化ウシ胎仔血清(FBS)、10mM HEPES緩衝液、2mM L-グルタミン、10U/mLペニシリンナトリウムおよび10μg/mL硫酸ストレプトマイシンを添加した。
細胞を6-ウエルプレート、12-ウエルプレートまたは25cm2フラスコに播種し、ガンマ線照射器IBL-637(Cs137、1Gy/分、gamma CIS-BioInternational, IBA, Saclay, France)で呈示の線量で放射線照射した。
培養培地を除去後、HCT116細胞を、10μMの5-クロロメチルフルオレセインジアセタート(CMFDA、緑色蛍光)または5-(および-6)-(((4-クロロメチル)ベンゾイル)アミノ)テトラメチルローダミン(CMTMR、赤色蛍光)(Molecular Probes-Life Technologie)を含有する前もって温めた培地で37℃にて45分間インキュベートした。その後、HCT116細胞を前もって温めた培地で2回濯ぎ、37℃にて1時間インキュベートした。染色した細胞を呈示のごとく処理し、細胞死プロファイリング分析のため培養した。
非処理のHCT116細胞をCMFDA(緑色の蛍光、CMFDA+)またはCMTMR(赤色の蛍光、CMTMR+)で標識し、処理したHCT116細胞をCMFDA(緑色の蛍光、CMFDA+)で標識した。以下の細胞混合を行った:非処理のCMTMR+ HCT116細胞を非処理のCMFDA+ HCT116細胞と、または非処理のCMTMR+ HCT116細胞を処理したCMFDA+ HCT116細胞と混合した。ついで、ROCKの薬理学的阻害剤、Y27632(30μM)、パン-カスパーゼ阻害剤(pan-caspase inhibitor)、Z-VAD-fmk(ZVAD、100μM)、カスパーゼ-1の阻害剤、Ac-YVAD-cmk(YVAD、100μM)、ネクロプトーシス阻害剤、ネクロスタチン-1(Necrostatin-1)(NEC1、30μM)、オートファジーを阻害する液胞型プロトンATPアーゼ(vacuolar type H(+)-ATPase)(V-ATPase)阻害剤、バフィロマイシン(Bafilomycin)A1(BafA1、50nM)、抗有糸分裂活性を有するCdksの阻害剤、ロスコビチン(Roscovitine)(Rosco、10μM)の存在下または不在下、24時間共培養した。24時間共培養後、剥離細胞および接着細胞の両者を回収し、温めた完全培地中、37℃で1時間、Hoechst 33345(10μg/mL)で染色した。ホスファチジルセリン(PS)暴露および原形質膜の透過性を検出するため、標識したHCT116細胞をビオチン-アネキシンV(BD Pharmingen;製造業者による推奨)、0.5μg BV786-ストレプトアビジン(BD Biosciences)および3μM DRAQ7(BioStatus)とともに連続的に25℃で15分間インキュベートした。PBS溶液で洗浄後、イメージングフローサイトメーターFlowSightR(AmnisR, EMD Milliporeの一部)を用いて直ちに試料を分析した。データはINSPIREソフトウェアを用いて20×倍率で得た。励起のため、405nm、488nm、および561nmのレーザーを用いた。それぞれ、420-480nm、745-800nm、642-745nm、480-560nm、595-642nmおよび430-505nmのチャネルを用い、明視野、アネキシンV-BV786、DRAQ7、CMFDA、CMTMRおよびHoechst 33345染色を検出した。試料当たり細胞の少なくとも1000の事象を分析した。異なるチャネルにわたって蛍光シグナルを正規化するため、追加の単一標識した対照を調製した。得られたデータをIDEAS分析ソフトウェア(v6.1; Merck-Millipore)を用いて分析した。ゲーティング戦略は以下の通りであった。焦点を合わせた細胞(focused cells)についてグラジエントRMS特性(Gradient RMS feature)を用いて細胞をゲーティングした。単一細胞についてアスペクト比と面積の特徴(aspect ratio and area features)を用いて細胞をゲーティングした。捕食検出のため、二重陽性CMFDA+およびCMTMR+染色で細胞をゲーティングした。
PS暴露、原形質膜透過性および細胞周期の進行を検出するため、共培養後、細胞を特定の蛍光プローブ(FITC-結合アネキシンV、ヨウ化プロピジウム、およびHoechst 33342)で順次標識し、フローサイトメトリーにより分析した。剥離細胞および接着細胞の両者を回収し、温めた完全培地中、37℃で1時間、Hoechst 33345(10μg/mL)で染色した。PBSで洗浄後、HCT116細胞を、製造業者の指示に従ってフルオレセインイソチオシアネート(FITC)-結合アネキシンV(BD Biosciences)およびヨウ化プロピジウム(PI, 1μg/mL)(Sigma)を添加した1X結合緩衝液に懸濁した。ついで、LSRIIフローサイトメーター(Becton Dickinson)およびFlowJoソフトウェアv10を用いて試料を分析した。共焦点蛍光顕微鏡のため、3,7%パラホルムアルデヒド-PBS中で15分間共培養後にHCT116細胞を固定し、1μg/mLのHoechst 33342(Invitrogen)で15分間対比染色した。ついで、Apochromat 63×1.3NAおよび63× 1.15NA油浸レンズを備えた共焦点SPE顕微鏡により細胞を分析した。Leica Aplication Suite (LAS)ソフトウェアを用いた(Leica Microsystems)。
全細胞タンパク質を溶解緩衝液(1%NP40、20mmol/L HEPES、10mmol/L KCl、1mmol/L EDTA、10%グリセロール、プロテアーゼおよびホスファターゼインヒビタータブレットを含む)で抽出した。タンパク質抽出物(30μg)を4-12%NuPAGERNovexR Bis-Trisゲル(Life Technologies)にかけ、4℃でImmobilonポリフッ化ビニル(PVDF)膜(Thermo Scientific)に移した。ブロッキング後、膜を4℃で一夜、カスパーゼ-3(#9662)、開裂したカスパーゼ-3(Asp175)(#9661)、ミオシン軽鎖2(MLC2)(#3672)、ホスホ-MLC2(Ser19)(#3675)、LC3A/B(#4108)、p-(S)-CDKs Substrate (#9477)(Cell Signaling Technologyから得た)に特異的な一次抗体とインキュベートした。GAPDH(#MAB374)はMilliporeから購入した。ついで、西洋わさびペルオキシダーゼ結合ヤギ抗マウスまたは抗ウサギ(Southern Biotechnology)抗体を1時間インキュベートし、ImageQuant LAS 4000ソフトウェア支援イメージャー(GE Healthcare)を用いてSuperSignal West PicoR試薬(Thermo Fisher Scientific)またはECLTM Prime Western Blotting Detection System (GE Healthcare)で暴露した。
各実験を少なくとも3回繰り返して比較できる結果を得た。特に断らない限り、数字は一つの代表的な実験からの定量データを示す(平均±SEM、n=3)。データをPrism v.5.03(GraphPad Software, La Jolla, CA, USA)により解析した。統計的有意性は両側スチューデントt検定により評価した。全ての実験において、p値<0.05は統計的に有意であるとした。
マルチスペクトルイメージングフローサイトメトリーを用いた細胞死プロファイリングは非細胞自立性細胞死の様式および細胞自立性細胞死の様式の同時検出を可能にする:
細胞がNCAプロセスにより死ぬ能力は、細胞死プロセスを考慮する本発明者らの方法論的アプローチを概念的な視点から再考するよう本発明者らを導いた。実際、考慮すべき形態学的および/または生化学的パラメーターの選択並びに細胞死を検出するのに用いる技術アプローチは、予想される結果を前もって事前に定義しており、細胞捕食やエントーシスなどの新たな致死プロセスの特定を可能にしないかまたは非常にまれに特定する。放射線療法の分野もまたこの問題に直面している。実際、別の研究で、放射線照射がアポトーシス、オートファジー細胞死、ネクローシスまたは有糸分裂死などの多くの致死的プロセスを引き起こすことが明らかになっている[35,36]。最近、別の研究で、同じ細胞型に同じ線量を照射するとアポトーシスを引き起こす可能性があることが示されたが[37]、有糸分裂死も引き起こした[38]。以前の研究では、放射線照射後に非常に急速に観察されるアポトーシスの開始および有糸分裂死は、長期的に観察されるクローン形成生存率(clonogenic survival)と相関しないことが明らかにされた[35]。これらの研究は、放射線照射された細胞の除去に関与する未知の致死プロセスの存在を強調した。さらに、腫瘍増殖の制御および転移細胞の排除における細胞捕食およびエントーシスの影響を明らかにする出版物の増加は、NCADのこのプロセスの開始を追跡することを促している。
抗癌治療に関連する細胞プロセスおよび生化学プロセスをよりよく特徴付けるために実施された徹底的な生物学的および薬理学的研究にも拘らず、放射線療法(臨床で最も頻繁に用いられる抗癌治療の一つである)の治療効果の致死メカニズムは未だ知られていない。電離放射線に反応して検出された致死プロセス(アポトーシスや有糸分裂破局など)は、治療効率に直接関係しておらず[35]、未だ知られていないさらなる細胞死の様式が放射線療法の治療効果に寄与しているかもしれないことが示唆された。この文脈で放射線照射した癌細胞の細胞死プロファイリングが決定された。上記方法により、CMFDA-標識細胞を4グレイで放射線照射しまたはせず、24時間後にCMTMR-標識細胞と1:1比で混合し、呈示した各阻害剤の存在下で培養した(補足図1A-1E)。ついで、アネキシンV-BV786、DRAQ7およびHoechst33342染色を用いて、各細胞パートナーのPS暴露、原形質膜完全性およびDNA含量をそれぞれ決定した。隣接細胞集団(CMTMR+細胞、図2A,2Bおよび2D)および非処理対照細胞集団(図2A,2Dおよび2E)でアポトーシス細胞死およびネクローシス細胞死(necrotic cell deaths)の有意の増加が観察されなかったにも拘らず(アネキシンV+DRAQ7-およびDRAQ7+細胞の分析により明らかなように)、全細胞集団(CMFDA+およびCMTMR+細胞集団を考慮することにより明らかなように;図2A-2C)および放射線照射した細胞集団(CMFDA+細胞;図2Bおよび2E)においてこれら両タイプの細胞死の有意な増加が24時間の共培養後に検出されたのであり、本発明の方法が放射線照射しない細胞および放射線照射した細胞の両者の細胞死の様式の検出を可能にすることが示された。また、パン-カスパーゼ阻害剤であるZ-VAD-fmkおよび薬理学的サイクリン依存性キナーゼ阻害剤であるロスコビチン(Rosco)が、放射線照射したCMFDA+細胞の原形質膜の外葉上のPS暴露を阻害し(図2E)、以前に文献に記載されたように[45,46]、放射線照射したCMFDA+細胞は死ぬのにカスパーゼ活性化と有糸分裂の進行の両方を必要とすることが確認された。さらに、バフィロマイシンA1(BafA1)によるオートファジーフラックス(autophagic flux)の障害は、全細胞集団(CMFDA+およびCMTMR+細胞;図2C)、放射線照射していないCMTMR+細胞(図2D)および放射線照射したCMFDA+細胞(図2E)での死につつある細胞(アネキシンV+DRAQ7-およびDRAQ7+細胞)の頻度を増大させ、それゆえ、オートファジーが、放射線照射していない細胞および放射線照射した細胞の両者を死から救済する、電離放射線照射によって引き起こされた生存のための細胞メカニズム(survival cellular mechanism)であることが明らかとなった。さらに、処理した細胞および非処理細胞の細胞周期での進行の同時分析は、細胞死の誘発がG1停止から放射線照射した細胞が逃れてSおよびG2/M期での蓄積に導くことと関係があることを示した(図2F、2Gおよび2J)。全細胞集団またはCMTMR+細胞集団では細胞周期に変化は検出されず、細胞周期の変化は放射線照射した細胞でのみ検出されたことが明確に示された(図2F、2Gおよび2I)。これらの結果は古典的なフローサイトメトリー分析によっても確認されたが(補足図2A-2B)、電離放射線照射後に放射線照射した癌細胞および放射線照射していない癌細胞の両者がカスパーゼ-1依存性細胞死を受けることを示した。これらを総合すると、これらの結果は直接細胞致死およびバイスタンダー効果により癌細胞を同時に排除する抗癌剤の能力を強調した。
並行して、同じ共培養において、放射線照射したCMFDA+細胞が隣接細胞を貪食または侵入する能力、細胞捕食関連細胞死(細胞捕食、エンペリトーシスまたはファゴプトーシスなど)またはセルインセル侵入により引き起こされた細胞死(エントーシスなど)の誘発に必要な2つの細胞プロセスを決定した。マルチスペクトルイメージングフローサイトメトリー分析は、ガンマ照射したCMFDA+細胞が隣接細胞の貪食を引き起こすことを明らかにした(ガンマ照射したCMFDA+細胞による「標的」CMTMR+細胞の内在化により明らかなように;図3Aおよび3B)。このプロセスはパン-カスパーゼ阻害剤であるZ-VAD-fmkにより影響を受けなかったが、ROCK1の阻害剤(Y27632)によって抑制され(図3B)、それゆえ、このことは、検出されたセルインセル内在化がアポトーシス細胞のファゴサイトーシス取込み(phagocytic uptake of apoptotic cells)とは区別されるものであり、内在化が起こるにはROCK1活性が必要であることを明らかにした。ガンマ照射した細胞の捕食活性はまた、共焦点顕微鏡によっても確認された(図3D-3F)。興味深いことに、細胞死プロファイリング分析はまた、生きたCMTMR+細胞によるアポトーシスCMFDA+細胞のファゴサイトーシス(バフィロマイシンA1処理によるアポトーシス誘発に引き続いて起こる)から生きた細胞の貪食を区別することを可能にした(図3B)。ついで、貪食されたCMTMR+細胞および放射線照射した貪食するCMFDA+細胞の細胞運命を評価した。貪食されたCMTMR+細胞の殆ど全てが細胞分解の形跡を示すことが観察された(マルチスペクトルイメージングフローサイトメトリー(図3A)および共焦点顕微鏡(図3Dおよび3E)で検出された内在化された細胞のDNA含量の損失により明らかなように)。このプロセスは、パン-カスパーゼ阻害剤であるZ-VAD-fmkおよびカスパーゼ-1の阻害剤であるYVAD-fmkの存在下では有意に低下し、貪食された細胞のIR-媒体死が起こるにはカスパーゼが必要であり、カスパーゼ-1依存性アポトーシス(ピロトーシスとしても知られる)によって実行されることが明らかになった。さらに、捕食細胞の90%はPSを暴露しないかまたは原形質膜の完全性の喪失を示すことが明らかになったが(補足図2C)、このことはIR-媒体細胞貪食の後、内在化された細胞が捕食細胞の生存能力を変更することなく沈殿して死に至ることを強調していた。これらを総合すると、これらの結果は電離放射線照射が直接細胞致死、バイスタンダー致死効果および細胞捕食関連細胞死の組合せ効果によって癌細胞を同時に排除することを示していた。これらの結果は、致死プロセスの際に非細胞自立性細胞死と細胞自立性細胞死との誘発に同時に対処する緊急の必要性を強調している。
ついで、IR-媒体細胞捕食を誘発することのできる化合物を同定するため、化合物のライブラリーのスクリーニングを開発した。それゆえ、8グレイの放射線量で処理しておいたHCT116細胞をオレンジ色のCMTMR細胞追跡剤(cell tracker)または緑色のCMFDA細胞追跡剤のいずれかで染色し、10μMの化合物の存在下で培養した。24時間培養後、細胞を核染色剤(nuclear dye)で染色し(5μg/mlのHoechst 33342;37℃で1時間)、FlowSightR Imaging Flow Cytometerを用いて細胞捕食を分析した。これら各化合物をZ-スコア[47]に従って分類し、それぞれ、13および11の候補化合物を同定した(図4A,4B,4Fおよび4G)。ついで、同定した化合物を、フローサイトメトリー法(処理した細胞のミトコンドリア内膜の脱分極および原形質膜の透過性を同時にモニターすることにより細胞毒性の化合物を排除するため)を共焦点顕微鏡法(候補化合物によるIR-誘発細胞捕食の変調を確認するため)と組み合わせることにより確認した(図4C-Eおよび4H-J)。3つの独立した実験の完了後、効果のない化合物(フェンベンダゾールやカルビマゾールなど)または高い細胞毒性を示した化合物(RN-1-183など)を排除した。最後に、細胞捕食を引き起こすIRの能力を高めることのできる16の化合物が同定された(図4Eおよび4J;表1&2)。
免疫原性細胞死(ICD)誘導物質がIRCCEとして同定されている(ジゴキシン、ジギトキシン、ラナトシドCまたはドキソルビシン塩酸塩など[3,4,48-51])ことを考慮して、放射線療法後にこれら化合物が抗腫瘍免疫を引き起こす能力を評価した。まず、癌腫の2つのマウスモデル(直腸CT26癌腫および線維肉腫MCA205)に対するIRCCEの免疫学的効果を調べるため、前臨床法を開発した。最初に、免疫適格マウスを用い、抗腫瘍ワクチン接種アッセイによりIRCCE+IRの組合せが特異的な抗腫瘍免疫応答を引き起こす能力を評価した。以前に刊行された研究によれば[52]、免疫原性細胞死(ICD)に陥った癌細胞の免疫適格マウスへの注入は、腫瘍抗原に特異的な防御免疫応答を引き起こすに違いない。それゆえ、3x105 MCA205細胞をまず10μMのSG6163Fで24時間処理した。ついで、処理した細胞を200μlのPBS中で8週齢の雌C57BL/6マウスの下腹部に皮下接種した。1週間後、3x104の非処理対照細胞をマウスの反対側腹部に接種した。腫瘍を一般的なキャリパーを使用して毎週評価した。体重が20~25%を超える腫瘍を持つ動物は安楽死させた。IRまたはSG6163Fのみで処理されたマウスは、インビトロで細胞死の有意な増加を誘発できないことが観察された(図5A)。実際には、それらは非処理細胞の接種後に防御応答を示さず、使用した放射線量およびSG6163Fの濃度が抗癌免疫応答を刺激するのに十分ではないことがわかった(図5B)。これら結果は、防御応答は癌細胞の70%がホスファチジルセリンを原形質膜の外葉上に暴露するまで癌細胞が細胞障害性癌治療にインビトロで暴露されたときに初めて検出されることを示す以前に刊行された結果と一致した[48]。驚くべきことに、MCA205細胞の8グレイのIRと10μMのSG6163Fとの組合せ処理が、注射した5匹のマウスのうち3匹で防御応答を誘発したことが観察された(図5B)。これら結果から初めて、処理および放射線照射した細胞の死の有意の増加なしにIRCCE+IRの組合せがIR-媒体抗癌免疫応答を誘発することが明らかになり(図5A)、細胞捕食または細胞捕食関連シグナル伝達経路が腫瘍免疫原性の誘発に寄与しているかもしれないことが示唆された。ついで、10μMのSG6163F(図5Eおよび5F)、VP331(図5Iおよび5J)、ミナプリン二塩酸塩(図5Mおよび5N)またはLOPA87(図5Qおよび5R)の存在下、3x106のCT26細胞を8グレイにて24時間放射線照射した。ついで、以前に記載されたように、細胞を200μlのPBS中で8週齢の雌BALB/cマウスの下腹部に皮下接種した。1週間後、5x105の非処理対照細胞をマウスの反対側腹部に接種し、腫瘍を一般的なキャリパーを使用して毎週評価した。以前に記載されたように、体重が20~25%を超える腫瘍を持つ動物は安楽死させた。各条件の死につつある細胞のパーセントをDiOC(6)3/IP染色で決定することにより評価した(図5E、5I、5Mおよび5Q)。最後に、これら化合物が放射線照射および処理した癌細胞の注射の7日後に注射された癌細胞の増殖を抑制する能力を評価した。IRおよび化合物で処理した癌細胞の注射後に防御応答を示すマウスの頻度の有意の増加が観察された(図5H、5L、5Pおよび5T)。
本発明は、次の態様を含む。
1.対象における放射線療法治療を強化することのできる化合物を同定するインビトロ方法であって、該方法は下記工程:
(a)候補化合物を腫瘍細胞培養物に加え、
(b)該腫瘍細胞培養物を1~20グレイの線量で処置し、
(c)該細胞培養物で起きた放射線照射媒体細胞捕食を測定する
を含み、
ここで、対照レベルと比較して放射線照射媒体細胞捕食を促進する化合物は放射線療法治療を強化することができる、方法。
2.癌細胞におけるIR-媒体細胞捕食を促進するための、表1および2に示す化合物およびその類似体のインビトロ使用。
3.癌を罹患する患者におけるIR-媒体細胞捕食の促進剤として使用するための表1および2に開示されている化合物またはその類似体または該化合物または類似体を含む医薬組成物。
4.放射線療法治療を受けるまたは受けた対象における腫瘍免疫原性を促進するために使用するためのIR-媒体細胞捕食の促進剤。
5.放射線療法治療を受けるまたは受けた対象における有意な防御的抗癌免疫応答を誘発するのに使用するためのIR-媒体細胞捕食の促進剤。
6.それを必要とする対象における放射線療法治療を強化するのに使用するためのIR-媒体細胞捕食の促進剤。
7.それを必要とする対象において放射線療法と組み合わせて癌を治療するのに使用するためのIR-媒体細胞捕食の促進剤。
8.表1および2に開示されている化合物またはその類似体であり、好ましくは、メブヒロリン1,5-ナフタレンジスルホン酸塩、フルルビプロフェン、ミナプリン二塩酸塩、ミリセチン、ジゴキシン、ジギトキシン、ラナトシド、LOPA87、VP331、RN-1-026、SG6163F、VP450およびVP43よりなる群から選ばれる、項4~7に記載の促進剤。

11.放射線療法治療の前に、好ましくは放射線療法治療の24時間前に投与する、項4~10に記載の促進剤。

13.放射線療法治療と同時に投与する、項4~10に記載の促進剤。
14.それを必要とする対象において癌を予防または治療するのに使用するためのミナプリン二塩酸塩。

Claims (9)
- 対象における放射線療法治療を強化することのできる化合物を同定するインビトロ方法であって、該方法は下記工程:
(a)候補化合物を腫瘍細胞培養物に加え、
(b)該腫瘍細胞培養物を1~20グレイの線量で処置し、
(c)該細胞培養物で起きた放射線照射媒体細胞捕食を測定する
を含み、
ここで、対照レベルと比較して放射線照射媒体細胞捕食を促進する化合物は放射線療法治療を強化することができる、方法。 - インビトロで癌細胞におけるIR-媒体細胞捕食を促進するための剤であって、ミナプリン二塩酸塩、メブヒドロリン1,5-ナフタレンジスルホン酸塩、RN-1-001、VP43、VP450、SG6163F、RN-1-026、VP331およびLOPA87よりなる群から選ばれる化合物を含む、剤。
- 癌を罹患する患者におけるIR-媒体細胞捕食を促進するための医薬組成物であって、ミナプリン二塩酸塩、メブヒドロリン1,5-ナフタレンジスルホン酸塩、RN-1-001、VP43、VP450、SG6163F、RN-1-026、VP331およびLOPA87よりなる群から選ばれる化合物を含む、医薬組成物。
- 化合物が、メブヒドロリン1,5-ナフタレンジスルホン酸塩、ミナプリン二塩酸塩、LOPA87、VP331、RN-1-026、SG6163F、VP450およびVP43よりなる群から選ばれる、請求項3に記載の医薬組成物。
- 化合物が、ミナプリン二塩酸塩、LOPA87、VP331およびSG6163Fよりなる群から選ばれる、請求項3または4に記載の医薬組成物。
- 該患者が、脳癌(例えば、神経膠腫)、胃癌、頭頸部癌、膵臓癌、非小細胞肺癌、小細胞肺癌、前立腺癌、結腸癌、非ホジキンリンパ腫、肉腫、精巣癌、急性非リンパ球性白血病または乳癌に罹患している、請求項3~5のいずれか1項に記載の医薬組成物。
- 放射線療法治療の前に投与する、請求項3~6のいずれか1項に記載の医薬組成物。
- 放射線療法治療の後に投与する、請求項3~7のいずれか1項に記載の医薬組成物。
- 放射線療法治療と同時に投与する、請求項3~8のいずれか1項に記載の医薬組成物。
