Description
この式は、廃油出口(212)に対する廃棄物出口(214)の位置を決定する。いくつかの実施形態において、Δhは、約0.5~1.5インチに及び得る。
以下は、本発明の非限定的な実施例である。当該実施例は、本発明を実践したところを明示する目的で提供されており、本発明を限定するようには決して意図されていないことを理解されたい。等価形態または置換形態は、本発明の範囲内である。
実施形態1 液体カバースリップ(LCS)は、0.8の比重を有する。Δh=0.5インチについて、油層の高さであるh0は、式
を使用して算出され、h0=2.5インチ、すなわち63.5mmとなっている。
DABを破壊するための、ベンチマークISH/ISH自動機器の廃棄物におけるAOP活性に影響を与える可能性がある様々なレバーを調べるために、調査を実施した。これらのレバーは、蒸留脱イオン水、反応緩衝液およびベンチマークDAB IHC廃棄物マトリクス中で試験した。これらのレバーは、流水式UV光源試験台デバイス(すなわち、直列UV殺菌水製造装置)、トップダウン非接触照射源(tRED試験台)、および、水/油分離とDAB水性廃棄物UV照射とを組み合わせたtRED試験台内で試験した。
BenchMark Ultra機器をスライド染色装置として使用して、実験データを得た。BenchMark Ultra機器上での典型的な液体カバースリップ(LCS)と水との比はおよそ20/80%であり、LCS(油)は典型的には蒸発損失を軽減するために高温段階に使用される。ランダムアクセス処理に起因して、LCSは、組み合わせ廃棄物流中で任意の時点において存在し得る。浮力(図2)と、任意選択のろ過/合体(図26B)との2つの分離原理が、LCSを水性廃棄物から分離するために行使される。これらの分離原理は両方とも、重力によって駆動される流れのみに基づき、いかなる圧送も必要としない。
Milli-Q(登録商標)脱イオン(「DDI」)水、ベンチマーク反応緩衝液およびベンチマークIHC廃棄物マトリクス中のDAB濃度変化を監視するために、HPLC分析を使用してDAB分解速度を判定した。HPLC分析は、フォトダイオードアレイ(「PDA」)検出を備えたWaters Acquity超高速液体クロマトグラフィ(「UPLC」)機器上で実施した。標準的な逆相クロマトグラフィを、ベンチマーク反応緩衝液および蒸留脱イオン水マトリクスにおけるUPLC分析に使用した。イオン対化を用いた逆相HPLCクロマトグラフィを、ベンチマーク廃棄物におけるDAB分析に使用した。DAB試料はろ過または希釈せずに分析した。
Waters ACQUITY UPLC Bridged Ethylene Hybrid(“BEH”)C18 Column 1x50mm(1.7μ)UPLCカラムを使用した。溶離液は、A:液体クロマトグラフィ質量分析(「LCMS」)の水中0.1%ギ酸、および、B:アセトニトリル中の0.1%ギ酸であった。流速は0.2mL/分、カラム温度は40℃、注入量は8μLであった。PDA検出波長は210~499nmであった。(2.4nm分解能かつ20点/秒)。DABおよび2-ヒドロキシピリジン(2-HOPyr)の予測保持時間はそれぞれ0.35分および0.54分であった。代表的なHPLCスペクトル(274nm)が図11A~図11Cに示されている。勾配プロファイルを表1に示す。

SIELC PrimeSep S column 2.1x100mm(5μ)および2.1x10mm guard column HPLCカラムを使用した。溶離液は、50mMギ酸アンモニウム(pH=4.0)中の40%アセトニトリルであった。流速は0.2mL/分、勾配プロファイルは定組成溶離、カラム温度は30°C、注入量は8μLであった。PDA検出波長は210~499nmであった。(2.4nm分解能かつ20点/秒)。DABおよび2-HOPyrの(ベンチマーク廃棄物中の)予測保持時間はそれぞれ2.56分および1.64分であった。代表的なHPLCスペクトル(310nm)が図12A~図12Cに示されている。
A.流水式UV照射試験台
複数のUV照射流水式システムを使用して、UVによって助長されるDAB酸化を調べた。事前設定のランプワット数およびUV照射チャンバ形状を有する市販の直列UV殺菌水製造装置、すなわち、Aquatop 10W直列UV滅菌器(Aquatop製品番号IL10-UV)を使用して、211ガロン/時までの流速で水を処置した。滅菌器は、独立した電源の支援を受けて流体流速を制御する外部可変速度マイクロポンプギアポンプを特徴とするものであった。能動的混合、ドウェル混合、または混合なしで試料を照射した。混合ポンプを用いずに使用された直列UV滅菌器については、試料採取の前に、一様な試料を作成するために転倒混和を実施した。複数のUV滅菌器UVランプワット数を調べた(10、18および55W)。排出物暴露時間を増大させるために、複数のUV照射源が、直列接続で互いに隣接して設置され得る。
図9A~図9Cに示すように、液体がUV照射源(「UVランプ光」としても参照される)に直に接触しないトップダウンUV照射システムを調査するための試験台が準備された。この試験台では、試料暴露時間を制御するために下方の金属トレー内の所定の経路を通じて液体が蛇行している間に、2つの18Wランプを使用して、上方から流体マトリクスを照射した。UV光源は、アレイの電力を増大させるために互い(例えばUVライトアレイなど)に対して並列接続で隣接して設置され得る。代替的に、UV光源は、暴露時間を増大させるために直列に設置されてもよい。進行経路を有しない第2の底面照射プラスチック容器を準備した。UV光は液体媒体を通るときに吸収され、ランプから最も遠い水性層が受けるUV照射はより少なくなる。この試験台を使用して、液層厚および廃棄物マトリクス不純物(すなわち、LCS、DAB酸化生成物など)がDAB UV分解速度に及ぼす影響を調べた。
コストが低く、放出波長が離散的であることから、水銀灯を一次UV光源として使用した。より連続的な放出スペクトルを有するランプとしては、キセノンアークランプ、重水素アークランプ、水銀-キセノンアークランプ、メタルハライドアークランプ、および、タングステン-ハロゲン白熱灯が挙げられる。UV LED、UVレーザなどのような他のUV光源が利用可能であり、同様の影響をもたらし得る。直列UV滅菌器のAquatop 10W UVランプ光源を、蛍光検出器によって光源の放出スペクトルを読み取ることによって特性化した。蛍光検出器に対して光を放出するための小孔を含有するスズ箔でランプを包み込んだ。関連するUV光波長およびそれらの波長の光子エネルギーのリストを、下記において表2に見ることができる。253nmにおけるUVC光が、より高いエネルギーをもたらすため、DAB酸化のほとんどを実施することができる。

A.試料混合の影響
マイクロポンプユニットを使用した様々な機械的混合によって、Aquatop 10W直列UV滅菌器(Aquatop製品番号IL10-UV)において最初のDAB UV酸化実験を実施した。蒸留脱イオン水中のDAB試料(68.1mg/L)を、3%過酸化水素をラジカル開始剤として用いて(およそ225mLのDAB試料あたり1mL)処置した。試料採取の前に一様な試料を作成するために、一定の混合または迅速なドウェル混合のいずれかで、UV光によって試料を照射した(10分)。反応混合物を、10分間を通じて2分ごとに、UPLCによって分析した。図13Aに示すように、DAB酸化速度は機械的混合を通じて加速された。混合は、DABを酸化する均等過酸化水素UV酸化生成物(HO)の拡散を補助した。
様々な過酸化水素ラジカル開始剤濃度を用いて、上記のようにAquatop 10W直列UV滅菌器においてDAB UV酸化実験を実施した。蒸留脱イオン水中のDAB試料(68.1mg/L)を、3%過酸化水素(およそ225mLのDAB試料あたり1mL)の存在下または非存在下で照射した。試料はUV光によって(60~90分)照射した。試料採取の前にマトリクスを一様に混合するために、エアポケットを使用してAquatop直列滅菌器を転倒させた(2回)。反応混合物を、10分ごとにUPLCによって分析した。
様々な試料マトリクス(すなわち、蒸留脱イオン水対ベンチマークIHC廃棄物)を用いて、上記のようにAquatop 10W直列UV滅菌器においてDAB UV酸化実験を実施した。マトリクス中のDAB試料(蒸留脱イオン水中の68.1mg/L DABおよびベンチマークIHC廃棄物中の136.2mg/L)を、3%過酸化水素の非存在下で照射した。TSA増幅IHC廃棄物を含有する新鮮なベンチマークUltra OptiView DABを、水性層が重力によってLCSの大部分から分離されるこの実験に使用した。ベンチマークIHC廃棄物は、依然としていくらかのLCSを含有していた。このLCSは、ベンチマークIHC廃棄物界面活性剤によって微細に分散される。両方の試料をUV光によって75分にわたって照射した。試料採取の前にマトリクスを一様に混合するために、エアポケットを使用してAquatop直列滅菌器を転倒させた(2回)。反応混合物を、10分ごとにUPLCによって分析した。
UVランプワット数を様々に変えて、直列UV殺菌水製造装置内でDAB UV酸化実験を実施した。TSA増幅廃棄物を含有するベンチマークUltra OptiView DAB中のDAB試料(136.2mg/L)を、3%過酸化水素(およそ1.5Lの廃棄物あたり20mL)を用いて照射した。試料採取の前にマトリクスを一様に混合するために、エアポケットを使用してAquatop直列滅菌器を転倒させた(2回)。反応混合物を、10分ごとにUPLCによって分析した。DAB UV酸化速度は、UVランプワット数に比例して増大した(図18)。55Wにおける30分のUV照射の後、DABモノマーは、ベンチマークIHC廃棄物中でDABの検出限界まで低減した。ベンチマークIHC廃棄物中で、DABモノマーがDABの検出限界未満に低減するには、18Wにおいておよそ90分のUV照射が必要であった(およそ1/3のUVランプワット数では3倍長い)。
マトリックス層厚の影響
試料液がUV光源に直に接触しない36WトップダウンUV照射試験台を使用して、DAB UV照射中のマトリクス層厚の許容性を調査した(図9A~図9C)。UV光は液体媒体を通るときに吸収され、ランプから最も遠い水性層が受けるUV照射はより少なくなる。反応緩衝液マトリクス中のDAB試料は、およそ20%(v/v)のLCSを含有する予測廃棄物濃度において調製した。OptiView DAB IHC検出キットDAB色原体、検出過酸化物および阻害剤過酸化物試薬を、ベンチマーク反応緩衝液マトリクスに添加した(250mLあたり143μLの各試薬)。36WトップダウンUV照射試験台底部を、特定のマトリクス層厚(すなわち、10、20または30mm厚)で充填した。DAB試料を、混合せずにUV照射によって処置し、試料を2分~10分ごと、次いで5分ごと、その後、30分を通じて除去した。マトリクス試料を逆相UPLCによって分析した。
試料液がUV光源に直に接触しない36WトップダウンUV照射試験台を使用して、トップダウン構成におけるDAB UV照射中にLCSがいかに許容され得るかを調査した。反応緩衝液マトリクス中のDAB試料は、およそ20%(v/v)のLCSを含有するまたは含有しない予測廃棄物濃度において調製した。OptiView DAB IHC検出キットDAB色原体、検出過酸化物および阻害剤過酸化物試薬を、反応緩衝液マトリクスに添加した(250mLあたり143μLの各試薬)。これらの反応緩衝液/LCS混合マトリクスを、LCSを混合しない状態、20回転倒することによって混合して初期気泡LCSが形成された状態、および、激しく混合して初期不透明LCSエマルションを形成した状態で試験した。
UVラジカル開始剤によるDAB分解
様々な過酸化水素濃度を用いて、上記のようにAquatop 10W直列UV滅菌器においてラジカル開始剤スクリーニング実験を実施した。ベンチマーク反応緩衝液中のDAB試料(68.1mg/L)を、ラジカル開始剤の存在下または非存在下で照射した。ラジカル開始剤モル濃度は、過酸化水素によって以前に使用したモル濃度(250mL試料あたりおよそ0.882mmol)に維持した。ラジカル開始剤をDAB試料に添加し、UV光によって30分にわたって照射した。試料採取の前にマトリクスを一様に混合するために、エアポケットを使用してAquatop直列滅菌器を転倒させた(2回)。反応混合物を、10分間を通じて2分ごとに、次いで、その後5分ごとに、UPLCによって分析した。

超音波分解は、液体中で音響キャビテーションを形成することができ、気泡の形成、成長および爆縮崩壊ならびに熱生成を引き起こす。超音波分解を使用して、熱ラジカル開始剤を熱的に活性化させ、DAB分解を誘発した。MisonixSonicator 3000超音波液体プロセッサを使用して、熱ラジカル開始剤スクリーニング実験を実施した。ソニケータ電圧変換器およびホーンをマイクロチッププローブに接続した。Misonix Sonicator 3000電力設定を7.5に設定し、30分にわたって30秒ごとにオンおよびオフをサイクルするようにプログラムした。RTDベンチマーク反応緩衝液中のDAB試料(68.1mg/L)を、熱ラジカル開始剤の存在下または非存在下で超音波分解した。ラジカル開始剤モル濃度は、過酸化水素によって以前に使用したモル濃度(25mL試料あたりおよそ0.0882mmol)に維持した。超音波分解によって、試料は効率的に混合した。反応混合物を、30分間を通じて5分ごとに、UPLCによって分析した。
DABを酸化するためのベンチマークIHC廃棄物におけるラジカルジェネレータを使用したAOP活性に影響を与える可能性がある様々なレバーを調べるために、調査を実施した。これらのレバーは、蒸留脱イオン水、ベンチマーク反応緩衝液およびベンチマークDAB IHC廃棄物マトリクス中で試験した。これらのレバーは、ラジカルジェネレータがUV照射源を利用するシステムにおいて試験した。試験台は、流水式UV試験台デバイス、トップダウン非接触UV照射試験台、および、DAB水性IHC廃棄排出物の水/油分離とUV照射とを組み合わせる試験台を含む。

Claims (15)
- 診断システム(10)を出る廃棄物流の処置のための受動的重力駆動処置システム(200)であって、前記システム(200)は、
