機能性RDH12配列番号3をコードするコドン最適化核酸配列を、完全なコザックコンセンサスをNotI部位に組み込んで5’末端に付加し、3’末端にBclI及びBamHI部位(クローニングの制限部位)を付加することにより、天然ヒトRDH12配列を修飾することで生成した。TGA終止コドンをBclI部位に組み込んで最適なエピトープタグ標識を容易にした。また、この実施形態は、上記に示された特定の制限酵素の使用を回避する。
本発明者らは、p618骨格に基づくmycタグを含み、また含まないヒト天然RDH12のcDNAを含有するアデノ随伴ウイルスプロウイルスのシス-プラスミドを作製した(p618で使用された配列に関しては、参照により本明細書に援用される米国特許第9,249,425号を参照のこと)。これらのプロウイルスプラスミドでは、hRDH12のcDNAは構成的CMV.CβAプロモーター(CBAe)、すなわちpAAV.CMVe.天然-hRDH12及びAAV.CBAe.h-天然RDH12.mycにより活性化される。

患者特異的な人工多能性幹細胞(iPSC)における近年の進歩により、疾病の原因を研究するための適切なin vitroモデル系が提供される。RDH12の機能を研究するためのin vitroモデルを開発するため、iPS細胞をヒトRDH12患者から作製し、特性評価を行った。AAV2.CMV.CβA-天然-hRDH12による感染が外因性RDH12の産生をもたらしたことを確認するため、本発明者らは、iPS細胞のアリコートに対して、1×103、1×104、1×105または2×105、3×105ベクターゲノム(vg)/細胞のAAV2CMV.CβA-天然-hRDH12を用いて形質導入した。48時間後、細胞溶解物を収集し、mycタグに特異的な抗体を用いてウエスタンブロッティングにより分析した。形質導入された細胞溶解物において、用量依存性が明らかなヒトRDH12の産生が認められた。図4の結果を参照のこと。
BALB/c由来のRdh12-/-マウスを、Dr.Anne Kasus-Jacobi,University of Oklahomaから入手した。これらの動物は、通常の周期的な光照射の下で飼育した場合、ヒト網膜変性の表現型を示さないが、明るい光に曝露された後に変性を起こす。
rAAV粒子はまた、適切な緩衝化担体中の懸濁液中の1010~1012GCまたはウイルス粒子の網膜下注射による投与後に、ヒト対象の網膜の細胞を形質導入するためにも用いられる。形質導入細胞または網膜におけるコドン最適化hRDH12の発現は、網膜及び視覚機能によって評価される。
Claims (16)
- 機能性ヒトRDH12をコードし配列番号5のヌクレオチド1~948を含む核酸分子。
- 機能性ヒトRDH12をコードし配列番号5のヌクレオチド1~948を含むヌクレオチド配列を含む発現カセット。
- 前記ヌクレオチド配列が、前記発現カセットを含む宿主細胞中で前記ヌクレオド配列によってコードされている機能性ヒトRDH12の発現を誘導する発現制御配列に作動的に関連する、請求項2に記載の発現カセット。
- 前記発現制御配列がロドプシンキナーゼプロモーターを含む、請求項2または3に記載の発現カセット。
- 機能性ヒトRDH12をコードし配列番号5のヌクレオチド1~948を含む発現カセットに隣接するAAV逆方向末端反復配列と、前記発現カセットを含む宿主細胞中で前記コードされた機能性ヒトRDH12の発現を誘導する発現制御配列とを含む、組換えアデノ随伴ウイルス(AAV)発現カセットであって、前記発現制御配列がロドプシンキナーゼプロモーターを含む、前記組換えAAV発現カセット。
- 請求項2~5のいずれか一項に記載の発現カセットを含むベクターまたはプラスミド。
- AAVカプシドと、機能性RDH12をコードし配列番号5の配列番号1~948を含むヌクレオチド配列であって、対象の光受容体細胞において前記RDH12の発現を誘導する調節配列の制御下にある前記ヌクレオチド配列とを含む、組換えアデノ随伴ウイルス(AAV)ベクターであって、任意に、前記AAVカプシドが、AAV8カプシドもしくはそのバリアント、AAV7カプシドもしくはそのバリアント、AAV5カプシドもしくはそのバリアント、またはAAV2カプシドもしくはそのバリアントである、前記組換えAAVベクター。
- 前記rAAVが自己相補的AAVである、請求項7に記載の組換えAAVベクター。
- AAV8カプシドおよび発現カセットを含む組換えAAVベクターであって、前記発現カセットが、
a)5'AAVITR;
b)ロドプシンキナーゼプロモーター;
c)機能性ヒトRDH12をコードし配列番号5のヌクレオチド1~948を含むヌクレオチド配列;および、
d)3'AAVITR
を含む、前記組換えAAVベクター。 - 請求項2~4のいずれか一項に記載の発現カセット、請求項6に記載のベクターまたはプラスミド、または請求項7~9のいずれか1に記載の組換えAAVベクターを含む、宿主細胞。
- 眼疾病の治療に有用な核酸分子、及び対象の眼への送達に適した担体または賦形剤を含む組成物であって、前記核酸分子は、機能性ヒトRDH12をコードし配列番号5のヌクレオチド1~948を含むヌクレオチド配列、及び眼細胞中で前記機能性ヒトRDH12の発現を誘導する調節配列を含む、前記組成物。
- 眼への送達に適した薬学的に許容される担体と、AAVカプシドタンパク質と、機能性ヒトRDH12コードし配列番号5のヌクレオチド1~948を含むヌクレオチド配列であって対象の光受容体細胞中で前記機能性ヒトRDH12の発現を誘導する調節配列の制御下にあるヌクレオチド配列を含む核酸分子、を含むウイルスベクターとを含む、
組成物。 - 哺乳動物対象のRDH12媒介性疾患の治療に使用するための、請求項12に記載の組成物。
- 哺乳動物対象の眼疾病または失明病の進行を治療するか、遅延または停止させるか、或いは前記対象の少なくとも部分的な視力を回復するのに使用するための、請求項12又は13に記載の組成物であって、前記疾病がRDH12をコードする遺伝子の欠損または変異から生じ、任意に、前記組成物が網膜下または硝子体内に投与される、前記組成物。
- 対象のLCA13またはRP53に関連する視力喪失の予防、進行の阻止または改善をするのに使用するための組成物であって、前記組成物が、AAVカプシドと、機能性ヒトRDH12コードし配列番号5のヌクレオチド1~948を含むヌクレオチド配列であって前記対象の光受容体細胞中で前記機能性ヒトRDH12を発現する調節配列の制御下にあるヌクレオチド配列を含む核酸分子とを含む、組換えアデノウイルス随伴(AAV)ベクターを含み、及び薬学的に許容される担体を含む、前記組成物。
- それを必要とする対象のLCA13またはRP53を治療または予防するのに使用するための組成物であって、
前記組成物が、機能性ヒトRDH12コードし配列番号5のヌクレオチド1~948を含むヌクレオチド配列であって、光受容体細胞中で前記ヌクレオチド配列によってコードされる前記機能性ヒトRDH12を発現する調節配列の制御下にある前記ヌクレオチド配列、を含む核酸分子を保有する組換えウイルス、及び薬学的に許容される担体を含み、
前記使用が、
(a)LCA13もしくはRP53に罹患しているか、または発症する危険性がある対象を特定し、
(b)遺伝子型解析を行ってRDH12遺伝子の変異を特定し、
(c)非侵襲性網膜イメージング及び機能的研究を実施して保持された光受容体の領域を特定し、および、
(d)前記組成物を有効濃度で前記対象に投与すること、を含んでなる、
前記組成物。
