分子構造
ペプチド合成
ペプチドは、GL Biochem Ltd.(Shanghai)により合成された。ペプチドの純度は、逆相HPLCによって分析し、マトリックス支援レーザー脱離/イオン化飛行時間型(MALDI-TOF)質量分析法を用いて、純度>99%として特徴を明らかにした。
1.リラグルチド(GLP-1アナログ)-
リラグルチドは、典型的には非経口投与されるペプチド薬である。リラグルチドは、2型糖尿病の治療に承認されている。リラグルチドは、国際公開第98/08871(A1)号の実施例37に開示されている。リラグルチドのアミノ酸配列は、配列番号15に示す。
脂肪酸は、26位でアミノ酸Lysに結合している。34位のアミノ酸Lysは、アミノ酸Argで置換されている。

トランスジェニックアルツハイマー病マウスモデル
動物モデルは、新規薬物の試験を行うために開発されたものである。アルツハイマー病の標準モデルは、ヒトにおいてアルツハイマー病を誘発することが知られているアミロイドの産生のためのヒト突然変異遺伝子を発現するトランスジェニックマウスである。これらのAPP/PS1トランスジェニックマウスは、脳内のアミロイドの凝集、記憶喪失およびシナプス喪失などのアルツハイマー病の症状のいくつかを再現する(Raddeら、2006年)。
動物(6匹/群)は、30mlの氷冷PBSおよび30mlの氷冷4%パラホルムアルデヒドで経心的に灌流して、脳を後固定した。脳を摘出し、組織を凍結保護するために新鮮な30%スクロース含有PBS溶液に入れ、低温槽上で厚さ40μmに切断した。切片は、立体規則に従って選択し、最初の切片は無作為に切り取り、その後5つの切片ごとに切り取った。7~13の切片/脳を分析した。
マウスにおいてパーキンソン様症状を誘発するための標準的なモデルは、化学物質(MPTP)の注射である(Liら、2009)。この化学物質は、ドーパミンを産生する頭脳のニューロンを損なわせるか、または死滅させる。マウスは、運動障害を発症し、脳内のドーパミン作動性ニューロンは、その数および機能が減少する。酵素チロシンヒドロキシラーゼ(TH)は、ドーパミンを合成するのに必要とされる。THの喪失は、ドーパミン産生の喪失を意味する(Harkavyiら、2008年)。
ロータロッドは、時間とともに加速する回転ポールで構成されている。マウスをロッド上に置き、回転を加速することにより運動技能を試験する。回転が増すにつれて、動物は握りを失い、ロッドの下に配置された緩衝材上に落ちる。マウスに、3週間、連続した2日間の各日に45分の試行間間隔で3回の試行を行った。各動物の持久時間を記録し、平均を計算した。事後ボンフェローニ検定による一元配置ANOVAを用いてデータを分析した。
6匹/群の動物を、ドーパミン産生のマーカーであるチロシンヒドロキシラーゼ(TH)の発現について分析した。線条体(ブレグマ11~10.2)およびSNpc(ブレグマ24.80~26.04)からの冠状脳切片(20μm)を、THを認識する抗体を用いて免疫組織化学により分析した。
これらの結果は、アルツハイマー病およびパーキンソン病の動物モデルにおいて、GLP-1/GIPデュアルアゴニストペプチドの両方(DA4およびDA5)が単一のGLP-1およびGIPアナログよりも優れた神経保護を示したことを実証したものである。したがって、本実施例のデータは、ペプチドDA4およびDA5が両方とも、例えばアルツハイマー病および/またはパーキンソン病などの神経変性障害の治療および/または防止における使用に好適であると考えられ得ることを実証している。
実施例2の材料および方法は、記載した以外は実施例1と同じである。実施例2で使用する試薬としては、p-PI3Kp85、PI3Kp85、p-Akt(ser473)、Akt、p-GSK3β(Y216)、およびGSK3β(Abcam、Inc.Cambridge、UK)に対する抗体のみが挙げられる。p-タウに対する抗体(AT8)(Thermo Scientific USA)。マウス抗βアクチン、抗ウサギIgG、抗マウスIgG、CY3抗マウスIgG、RIPAライセート、BCA プロテインアッセイキット、DAPI染色液および抗蛍光減衰剤(Boster(Wuhan、China))。
実験前に、動物は実験室で30分間の順応を行った。オープンフィールドは、中央領域として中央に4つの正方形を画定し、その残りを周辺領域として画定して、16の同じサイズの正方形に分割した。各マウスは、5分間、中央から装置(40cm×40cm×40cm)に個別に入れ、合計距離、ならびに中央での時間の割合を、Smart 3.0ソフトウェアシステム(Panlab、Spain)により同時に記録した。各試験の前に、装置を70%エタノールで洗浄した。
空間作業記憶は、Y字型迷路における自発的交替挙動を記録することにより査定した。迷路は、長さ30cm、高さ7cm、幅15cmの3本のアームで作られ、そのアームは正三角形の中央領域に集合させた。各マウスを中央の三角領域に配置し、8分間のセッション中に迷路を自由に移動させた。全アームへの侵入およびアームの順序は、Smart 3.0ソフトウェアシステムによって記録した。交替は、重なり合う三組の3つのアームへの連続した侵入として定義した。交替率は、実際の交替数と可能な交替数の比(アーム侵入総数から2を引いた値として定義される)に100を掛けた値として計算した。
古典的モリス水迷路(MWM)試験を用いて、マウスの空間基準記憶を調べた。水迷路は、主にステンレス鋼プールおよび逃避のためにプール内にプラットフォームを備えていた。水迷路の壁には、いくつかのランドマークを固定させた。プールは、22±2℃の水道水で満たした。第1の段階は、獲得試行段階であり、5日の訓練日(1~5日目)、および毎日行う4回の試行で構成されていた。プール(直径120cm、高さ50cm)を4つの象限に分け、水面下1cmに隠れた逃避プラットフォーム(直径12cm)を第1象限の中央に配置した。プールの円周に沿って等間隔に配置された4点は、開始位置として使用し、この位置は、1日4回の試行において無作為に選択した。マウスがプラットフォームに上がるか、または時間が1分に達すると、記録を停止した。逃避潜時は、Ethovision 3.0ソフトウェアシステム(Noldus Information Technology、the Netherlands)によって記録した。第2の段階は、プローブ試行段階であった。獲得作業を終えた翌日(6日目)に、空間記憶を査定するためにプローブ試験を実施した。プラットフォームをプールから取り出し、動物を1分間自由に泳がせた。標的象限(第1象限)における遊泳時間の割合およびプラットフォームを通過する回数を記録した。逆転MWM試験を用いて、マウスの再学習能力および認知の柔軟性を観察した。古典的なMWM試験の後、プラットフォームを反転象限(第3象限)に移動させ、マウスを上記と同様の4日間の獲得試行および1日間のプローブ試行のために水中に入れた。第3の段階は、可視可能なプラットフォーム試験であった。プラットフォームを水面より1cm上に上げ、マウスを無作為に選択した2つの象限に入れた。標的までの水泳時間を記録した。
マウスをウレタン(Sigma、UK、1.5g/kg、i.p.)で麻酔し、L-LTPを記録するために脳定位固定装置(RWD Life Science、China)に入れた。頭皮を切り取り、頭蓋骨を露出させ、頭蓋骨の一方の側に小さい穴を開けた(ブレグマの後側2.0mmおよび正中線の外側1.5mm)。
インビボでの海馬のL-LTPを記録後、マウスを、腹腔内注射により抱水クロラール(0.007ミリリットル/g)で麻酔し、組織を脱水のために30%スクロース溶液に移した後、脳組織を24時間パラホルムアルデヒドで固定した。脳組織を採取し、-80℃で凍結し、次いで厚さ30μmにスライスし、凍結切片をPBS中に入れ、正常ヤギ血清(Solarbio、Beijing、China)でブロックし、続いて一次抗体、二次抗体、およびDAPIを添加した。リン酸化タウタンパク質は、共焦点レーザー走査型顕微鏡で観察した。
マウスの海馬を解剖し、組織タンパク質抽出試薬中で均質化し、タンパク質濃度をBCA Protein Assay Kitを用いて測定した。各サンプルから合計50μgのタンパク質を使用した。サンプルタンパク質は、12%SDS-ポリアクリルアミドゲル上で分離させた。その後、タンパク質をPVDF膜に移し、非特異的結合を5%BSAでブロックした。一次抗体を用いて膜を4℃で一晩インキュベートし、続いて二次抗体を2時間インキュベートした。標的ストリップ(p-PI3K、t-PI3K、p-AKT、t-AKT、p-GSK3β、およびt-GSK3β)の光学密度は、ゲル画像処理システムを用いて分析した。
この研究に存在する結果は、APP/PS1トランスジェニックADマウスにおける認知機能および病理学的特徴に対するGLP-1/GIPデュアルアゴニストDA5ペプチドの保護作用を確認したものである。DA5ペプチドの神経保護は、海馬シナプス可塑性およびPI3K/AKT/GSK3βシグナル伝達経路の改善と関連している可能性がある。上記の結果は、DA5ペプチドが神経変性障害の治療に有用であり得、AD患者、特にT2DMまたは高血糖症の患者に特に有益であり得ることを示唆している。

- (i)24位のアミノ酸、または(ii)39位もしくは40位のアミノ酸に共有結合している親水性部分を含む、請求項4に記載のペプチド。
- 前記親水性部分が水溶性ポリマーであり、任意により前記水溶性ポリマーがポリエチレングリコール部分であり、かつ任意により約20,000ダルトン~約60,000ダルトンの分子量を有するポリエチレングリコール部分である、請求項4または5に記載のペプチド。
- 前記ペプチドが親油性置換基とコンジュゲートしている、請求項1~6のいずれか一項に記載のペプチド。
- 前記親油性置換基がアシル基を含み、任意により脂肪酸分子であり、さらに、任意により、前記脂肪酸分子が、C-8オクタノイル基、C-10デカノイル基、C-12ラウロイル基、C-14ミリストイル基、C-16パルミトイル基、C-18ステアロイル基、およびC-20アシル基から選択される、請求項7に記載のペプチド。
- 前記親油性置換基が、前記ペプチドのカルボキシル末端でアミノ酸に接着している、請求項7または8に記載のペプチド。
- 前記親油性置換基をアミノ酸残基にコンジュゲートするスペーサーを含む、請求項7~9のいずれか一項に記載のペプチド。
- 前記スペーサーは、天然または非天然アミノ酸由来の残基であり、前記スペーサーは、Gly、Pro、Ala、Val、Leu、Ile、Met、Cys、Phe、Tyr、Trp、His、Lys、Arg、Gln、Asn、α-Glu、γ-Glu、ε-Lys、Asp、Ser、Thr、Gaba、Aib、β-アミノヘキソニル、7-アミノヘプタノイル、8-アミノオクタノイル、9-アミノノナノイル、10-アミノデカノイル、または8-アミノ-3,6-ジオキサオクタノイルの残基を含む、請求項10に記載のペプチド。
- 前記スペーサーがγ-Gluであるか、前記スペーサーがジペプチドであり、任意により前記スペーサーが2個の負に電荷したアミノ酸を含み、さらに任意により、前記スペーサーがγ-Glu-γ-Gluである、請求項11に記載のペプチド。
- 神経障害の治療および/または予防に使用するためのものである、請求項1~12のいずれか一項に記載のペプチド。
- 前記ペプチドは、患者のある領域におけるβ-アミロイドタンパク質プラークの沈着によって引き起こされるか、またはそれに関連する神経障害の前記治療における使用のためのものである、請求項13に記載のペプチド。
- a)シナプス伝達の長期増強の機能障害、または
b)炎症
によって引き起こされるか、もしくはそれに関連する神経障害の前記治療および/もしくは予防に使用するためのものである、請求項13に記載のペプチド。 - 認知機能に影響を及ぼす神経障害、例えば認知症、脳卒中、統合失調症および/または双極性障害の前記治療および/または予防に使用するためのものであって、任意により、前記障害は脳卒中に関連する脳虚血である、請求項13に記載のペプチド。
