Description

Claims (14)
- 間隙領域によって分離された凹部を含むパターン化された基材であって、
結合領域を含む、基材と、
前記凹部の各々に付着したシークエンシング表面化学物質であって、
官能化コーティング層と、
該官能化コーティング層にグラフトされたプライマーと、
を含む、シークエンシング表面化学物質と、
架橋ポリアクリルアミド、アガロースゲル、および架橋ポリエチレングリコールからなる群より選択される非グラフト化ヒドロゲルと、
前記結合領域で、前記パターン化された基材に取り付けられた蓋と、
該蓋と前記パターン化された基材との間に画定される流路と、
を含む、フローセル。 - 前記非グラフト化ヒドロゲルが、前記間隙領域の少なくともいくつかの上にもある、請求項1に記載のフローセル。
- 前記官能化コーティング層が、ポリ(N-(5-アジドアセトアミジルペンチル)アクリルアミド-co-アクリルアミド)である、請求項1または2に記載のフローセル。
- フローセルを製造する方法であって、
パターン化されたフローセル基材の凹部に官能化コーティング層を適用する工程であって、凹部が間隙領域によって分離されている工程であって、前記パターン化されたフローセル基材が結合領域を含む、工程と、
プライマーを前記官能化コーティング層にグラフトして、前記凹部にグラフトされた官能化コーティング層を形成する工程と、
少なくとも前記グラフトされた官能化コーティング層上に非グラフト化ヒドロゲルを適用する工程であって、該非グラフト化ヒドロゲルが、架橋ポリアクリルアミド、アガロースゲル、および架橋ポリエチレングリコールからなる群より選択される、工程と、
前記結合領域で、前記パターン化されたフローセル基材に蓋を結合し、これにより、該蓋と前記パターン化されたフローセル基材との間に流路を画定する、工程と、
を含む、方法。 - 前記非グラフト化ヒドロゲルが、前記グラフトされた官能化コーティング層上および前記間隙領域のいくつかの上に配置される、請求項4に記載の方法。
- 請求項4または5に記載の方法であって、前記官能化コーティング層を適用する前に、前記パターン化されたフローセル基材の表面を処理して、前記表面に官能基を付着させて、処理された凹部および処理された間隙領域を形成することをさらに含む、方法。
- 前記凹部に前記官能化コーティング層を適用する工程は、
前記処理された凹部および処理された間隙領域上に官能化コーティング層を適用する工程と、
前記処理された間隙領域から官能化コーティング層を研磨する工程と、
を含む、請求項6に記載の方法。 - 前記非グラフト化ヒドロゲルを適用することが、約0.001%~約0.1%(質量対体積)のヒドロゲル材料を含む水性混合物を適用することを含む、請求項4~7のいずれか1項に記載の方法。
- 前記パターン化されたフローセル基材の結合領域が、それに結合されたスペーサー層を有し、前記非グラフト化ヒドロゲルが適用された後、前記パターン化されたフローセル基材への前記蓋の結合が、前記スペーサー層に前記蓋を結合することを含む、請求項4から8のいずれか1項に記載の方法。
- 前記ヒドロゲルを適用することが、前記グラフトされた官能化コーティング層上にヒドロゲルを選択的に堆積させることを含む、請求項4および6~9のいずれか1項に記載の方法。
- フローセルを製造する方法であって、
凹部が画定された流路を含むパターン化された基材の表面にシランまたはシラン誘導体を付着させ、凹部が間隙領域によって分離され、それによってシラン化凹部およびシラン化間隙領域が形成される工程であって、前記パターン化された基材が結合領域を含む、工程と、
前記シラン化凹部およびシラン化間隙領域に、官能化コーティング層を適用する工程と、
前記シラン化間隙領域からの官能化コーティング層を研磨する工程と、
前記シラン化凹部内の官能化コーティング層にプライマーをグラフトして凹部にグラフトされた官能化コーティング層を形成し、凹部のグラフトされた官能化コーティング層上に非グラフト化ヒドロゲルを適用する工程であって、該非グラフト化ヒドロゲルが、架橋ポリアクリルアミド、アガロースゲル、および架橋ポリエチレングリコールからなる群より選択される、工程と、
前記結合領域で、前記パターン化された基材に蓋を結合し、これにより、該蓋と前記パターン化された基材との間に流路を画定する、工程と、
を含む、方法。 - 前記非グラフト化ヒドロゲルを適用する工程が、約0.001%から約0.1%(質量対体積)までのヒドロゲル材料を含む水性混合物を適用することを含む、請求項11に記載の方法。
- 前記官能化コーティング層が研磨された後、前記プライマーがグラフトされる前に、前記間隙領域の少なくともいくつかに蓋を接合することをさらに含む、請求項11または12に記載の方法。
