Description
項1. 以下の(1)~(3)の少なくとも1種のポリペプチドからなるプロテアーゼを有効成分とする、腸内菌叢改善剤。
(1)配列番号1に示すアミノ酸配列からなるポリペプチド。
(2)配列番号1に示すアミノ酸配列において、1個又は数個のアミノ酸が置換、付加、挿入又は欠失されてなり、且つ、配列番号1に示すアミノ酸配列からなるポリペプチドと同等のプロテアーゼ活性を有するポリペプチド。
(3)配列番号1に示すアミノ酸配列に対する配列同一性が80%以上であり、且つ、配列番号1に示すアミノ酸配列からなるポリペプチドと同等のプロテアーゼ活性を有するポリペプチド。
項2. 項1に記載の腸内菌叢改善剤を含む、腸内菌叢改善用の内服用医薬品。
項3. 項1に記載の腸内菌叢改善剤を含む、腸内菌叢改善用の食品用添加剤。
項4. 項1に記載の腸内菌叢改善剤を含む、腸内菌叢改善用の飲食品。
項5. 以下の(1)~(3)の少なくとも1種のポリペプチドからなるプロテアーゼを腸内菌叢の改善が必要とされる者に経口適用する、腸内菌叢の改善方法。
(1)配列番号1に示すアミノ酸配列からなるポリペプチド。
(2)配列番号1に示すアミノ酸配列において、1個又は数個のアミノ酸が置換、付加、挿入又は欠失されてなり、且つ、配列番号1に示すアミノ酸配列からなるポリペプチドと同等のプロテアーゼ活性を有するポリペプチド。
(3)配列番号1に示すアミノ酸配列に対する配列同一性が80%以上であり、且つ、配列番号1に示すアミノ酸配列からなるポリペプチドと同等のプロテアーゼ活性を有するポリペプチド。
項6. 腸内菌叢の改善処置に使用される、以下の(1)~(3)の少なくとも1種のポリペプチドからなるプロテアーゼ。
(1)配列番号1に示すアミノ酸配列からなるポリペプチド。
(2)配列番号1に示すアミノ酸配列において、1個又は数個のアミノ酸が置換、付加、挿入又は欠失されてなり、且つ、配列番号1に示すアミノ酸配列からなるポリペプチドと同等のプロテアーゼ活性を有するポリペプチド。
(3)配列番号1に示すアミノ酸配列に対する配列同一性が80%以上であり、且つ、配列番号1に示すアミノ酸配列からなるポリペプチドと同等のプロテアーゼ活性を有するポリペプチド。
項7. 以下の(1)~(3)の少なくとも1種のポリペプチドからなるプロテアーゼの、腸内菌叢改善剤の製造のための使用。
(1)配列番号1に示すアミノ酸配列からなるポリペプチド。
(2)配列番号1に示すアミノ酸配列において、1個又は数個のアミノ酸が置換、付加、挿入又は欠失されてなり、且つ、配列番号1に示すアミノ酸配列からなるポリペプチドと同等のプロテアーゼ活性を有するポリペプチド。
(3)配列番号1に示すアミノ酸配列に対する配列同一性が80%以上であり、且つ、配列番号1に示すアミノ酸配列からなるポリペプチドと同等のプロテアーゼ活性を有するポリペプチド。
本発明の腸内菌叢改善剤は、特定のプロテアーゼを有効成分とすることを特徴とする。以下、本発明の腸内菌叢改善剤について詳述する。
本発明の腸内菌叢改善剤は、以下の(1)~(3)の少なくとも1種のポリペプチドからなるプロテアーゼを有効成分として使用する。このように、特定のプロテアーゼを選択し、これを使用することにより、腸内の乳酸菌やビフィズス菌等の善玉菌を増加させ、腸内菌叢を効果的に改善することが可能になる。
(1)配列番号1に示すアミノ酸配列からなるポリペプチド。
(2)配列番号1に示すアミノ酸配列において、1個又は数個のアミノ酸が置換、付加、挿入又は欠失されてなり、且つ、配列番号1に示すアミノ酸配列からなるポリペプチドと同等のプロテアーゼ活性を有するポリペプチド。
(3)配列番号1に示すアミノ酸配列に対する配列同一性が80%以上であり、且つ、配列番号1に示すアミノ酸配列からなるポリペプチドと同等のプロテアーゼ活性を有するポリペプチド。
(プロテアーゼの活性の測定法)
まず、0.6重量%カゼイン溶液(pH3.0)5mLを試験管にとり37℃に保温する。続いて、プロテアーゼ活性の測定対象となるサンプルを含む酵素溶液(溶媒は水を使用)1mLを加えて37℃で正確に10分間おいた後、0.44mol/Lトリクロロ酢酸溶液5mLを加え、反応を停止させる。37℃で30分間おいたのち、ろ紙ろ過して得られたろ液2mLを別の試験管にとり、0.55mol/L炭酸ナトリウム5mL、3倍希釈フォリン試液1mLを順に加える。37℃で30分間おいた後に吸収波長660nmにおける吸光度を測定する。ブランク操作は、トリクロロ酢酸溶液添加後に酵素溶液を加える。また別途10~40μg/mLのチロジン溶液を用いて、先述のろ液に対する操作と同様の操作をすることによりチロジン検量線を作成する。本条件下、1分間にチロジン1μgに相当するフォリン試液呈色物質の増加をもたらす酵素量を1Uとする。算出には以下式を用いる。
本発明の腸内菌叢改善剤の適用量については、該剤が使用される製品の種類、用途、期待される効果、適用形態等に応じて適宜設定すればよいが、例えば、成人1日当たり前記プロテアーゼの摂取又は投与量として、0.1~3,000mg、好ましくは1~2000mg、より好ましくは1~1000mg、より一層好ましくは2~500mg、さらに好ましくは2~150mg、最も好ましくは5~100mgが挙げられる。
本発明の腸内菌叢改善剤は、前記プロテアーゼの作用によって、腸内でビフィズス菌及び乳酸菌等の善玉菌の菌数を増加させ、善玉菌が優勢な菌叢を形成させることができるので、腸内環境の健全化、健全な腸内環境の保持等の目的で使用される。特に、本発明の腸内菌叢改善剤は、腸内でビフィドバクテリウム属細菌及びラクトバシスル属細菌の菌数を増加させる作用に優れているので、ビフィドバクテリウム属細菌及びラクトバシスル属細菌の腸内菌数増加剤として使用することもできる。
本発明の腸内菌叢改善剤は、経口摂取又は経口投与により経口適用される。本発明の腸内菌叢改善剤は、経口摂取又は経口投与され、腸内に到達すると、腸内菌叢を改善する作用を発揮するので、飲食品、内服用医薬品、飼料、ペットフード等の各種製品に配合して使用することができる。
前述するように、前記プロテアーゼは、腸内菌叢を改善する作用がある。従って、本発明は、更に、前記プロテアーゼを腸内菌叢の改善が必要とされる者に経口適用する、腸内菌叢の改善方法;腸内菌叢の改善処置に使用される、前記プロテアーゼ;並びに、前記プロテアーゼの腸内菌叢改善剤の製造のための使用を提供する。これらの発明における具体的態様については、前記「1.腸内菌叢改善剤」の欄に記載の通りである。
1.プロテアーゼの調製
配列番号1に示すアミノ酸配列のポリペプチドからなるプロテアーゼは、生産菌である微生物を固体培養法で培養して得られた麹抽出液から調製した。具体的には、小麦ふすまからなる固体培養培地にて、アスペルギルス・オリゼを25±5℃にて約3日間培養した。更に、培養後の麹を水に浸漬させることでポリペプチドを抽出し、ケイソウ土ろ過により麹を除いて、ポリペプチド溶液を得た。次いで、ポリペプチド溶液を限外ろ過膜にて濃縮した後に、脱塩によりポリペプチド粗精製液を得た。ポリペプチド粗精製液をメンブランフィルターろ過にて除菌した後、噴霧乾燥機にて噴霧乾燥することで粉末状の粗精製ポリペプチドを得た。更に、この粗精製ポリペプチドを、順にイオン交換カラムクロマトグラフィー、疎水カラムクロマトグラフィー、ゲルろ過クロマトグラフィー、脱塩カラムクロマトグラフィー、及び凍結乾燥の精製工程に供し、精製ポリプチド粉末を得た。また、得られた精製ポリペプチドをSDS-ポリアクリルアミドゲル電気泳動及びCBB染色に供し、確認されたポリペプチドのバンドについて、酵素処理した後にLC-MS/MS解析し、配列番号1に示すアミノ酸配列からなるポリペプチドであることを確認した。
SDラット(オス、3週齢、株式会社広島実験動物研究所より入手)を用いて、配列番号1に示すアミノ酸配列のポリペプチドからなるプロテアーゼの摂取が腸内菌叢に及ぼす影響を調べた。実験は、対照群1及び2と実施群1及び2の4群に分け、対照群1は8匹、対照群2は4匹、実施群1及び2は4匹のラットを用いた。それぞれの群について表1に示す飼料を14日間自由に摂取させた。
得られた結果を表3に示す。対照群1及び2に比べて実施群1及び2の方が、腸内のビフィズス菌及び乳酸菌の菌数が共に高値であった。即ち、配列番号1に示すアミノ酸配列のプロテアーゼの摂取により、ビフィズス菌及び乳酸菌の増殖が促進され、腸内菌叢が改善されることが確認された。また、単なるペプチドの投与ではなく、酵素活性のあるプロテアーゼを摂取することで、腸内菌叢が改善されることが確認された。
Claims (5)
- 以下の(1)~(3)の少なくとも1種のポリペプチドからなるプロテアーゼを有効成分とする、腸内菌叢改善剤。
(1)配列番号1に示すアミノ酸配列からなるポリペプチド。
(2)配列番号1に示すアミノ酸配列において、1個又は数個のアミノ酸が置換、付加、挿入又は欠失されてなり、且つ、配列番号1に示すアミノ酸配列からなるポリペプチドと同等のプロテアーゼ活性を有するポリペプチド。
(3)配列番号1に示すアミノ酸配列に対する配列同一性が80%以上であり、且つ、配列番号1に示すアミノ酸配列からなるポリペプチドと同等のプロテアーゼ活性を有するポリペプチド。 - 請求項1に記載の腸内菌叢改善剤を含む、腸内菌叢改善用の内服用医薬品。
- 請求項1に記載の腸内菌叢改善剤を含む、腸内菌叢改善用の食品用添加剤。
- 請求項1に記載の腸内菌叢改善剤を含む、腸内菌叢改善用の飲食品。
- 以下の(1)~(3)の少なくとも1種のポリペプチドからなるプロテアーゼの、腸内菌叢改善剤の製造のための使用。
(1)配列番号1に示すアミノ酸配列からなるポリペプチド。
(2)配列番号1に示すアミノ酸配列において、1個又は数個のアミノ酸が置換、付加、挿入又は欠失されてなり、且つ、配列番号1に示すアミノ酸配列からなるポリペプチドと同等のプロテアーゼ活性を有するポリペプチド。
(3)配列番号1に示すアミノ酸配列に対する配列同一性が80%以上であり、且つ、配列番号1に示すアミノ酸配列からなるポリペプチドと同等のプロテアーゼ活性を有するポリペプチド。
