Description
核酸を含有する試料をカラムに添加すると、緩衝液、変性剤(塩酸グアニジンなど)、Triton X-100、イソプロパノール及びpH指示薬を含有してよい結合溶液の(カラムのシラノール基に対して)低いpH及び塩濃度によって、核酸が結合する;
カラムを5mM KPO4、pH8.0又は類似物、80%EtOH)で洗浄する;及び
カラムを緩衝液又は水で溶離する。
i)ポリメラーゼと封鎖剤及びdNTPとを混合して、それらの混合物を得る工程、及び
ii)混合物を乾燥させて乾燥試薬組成物を形成する工程
を含む核酸増幅用の乾燥試薬組成物を製造する方法が提供される。
i)核酸を含有する溶液を上記の乾燥試薬組成物とともにインキュベートする工程及び、
ii)核酸を増幅させる工程
を含む、核酸増幅方法が提供される。
i)本明細書上文に記載される方法を用いて核酸を増幅して、増幅核酸を作製する工程、
ii)増幅核酸を検出する工程、及び
iii)所望により増幅核酸を定量化する工程
を含む検出系を用いる、増幅核酸の検出及び/又は定量化方法が提供される。

正常なヒト血液(Tissue Solutions Ltd);

実験結果
qPCRを用いる、セルロースマトリックスからの乾燥した血液スポットからのDNA測定
PCR試薬をシクロデキストリンと合し、表1中の以下の条件下で凍結乾燥した。
血液をスポットしたFTAを、シクロデキストリンを含有するか又は含有しない核酸増幅試薬ケークとともにウェルに添加した。標準物質及び試料を適切なウェルに添加した。プレートを1000rpmで1分間遠心し、密封した。PCRは、MJ Research PTC-200サーモサイクラーで製造業者の取扱説明書に従って実行した。
Claims (9)
- 核酸増幅方法であって、
i)固相支持体に固定化された核酸を、水と、封鎖剤、ポリメラーゼ及びデオキシリボヌクレオチド三リン酸(dNTP)を含む凍結乾燥試薬組成物とともにインキュベートする工程と、
ii)核酸を増幅する工程と
を含んでおり、溶解試薬が固相体支持体に組み込まれており、固相支持体が洗浄されていない、方法。 - 前記封鎖剤が、α-シクロデキストリン、β-シクロデキストリン、γ-シクロデキストリン、6-O-α-D-マルトシル-βシクロデキストリン、ヒドロキシエチル-β-シクロデキストリン、ヒドロキシプロピル-β-シクロデキストリン及び2-ヒドロキシプロピル-β-シクロデキストリン並びにそれらの誘導体からなる群から選択されるシクロデキストリンである、請求項1記載の方法。
- 増幅方法がポリメラーゼ連鎖反応であるか、或いは逆転写ポリメラーゼ連鎖反応又は等温増幅を含む、請求項1又は請求項2記載の方法。
- 固相支持体がセルロース系マトリックスである、請求項1乃至請求項3のいずれか1項記載の方法。
- 固相支持体が予め打ち抜いたディスクの形態又はFTAの予め打ち抜いたディスクの形態である、請求項4記載の方法。
- 増幅が単一の反応器で実行される、請求項1乃至請求項5のいずれか1項記載の方法。
- 検出系を用いて増幅核酸を検出及び/又は定量化する方法であって、
i)請求項1乃至請求項6のいずれか1項記載の方法を用いて核酸を増幅して増幅核酸を生成する工程と、
ii)増幅核酸を検出する工程と
を含む方法。 - iii)増幅核酸を定量化する工程をさらに含む、請求項7記載の方法。
- 検出系がPCR画像形成系である、請求項7又は請求項8記載の方法。
