Description
IGF−Iは、インスリン様成長因子(Insulin-like growth factor)であり、主に肝臓から分泌され、IGF−I受容体に作用することにより、各種臓器で種々の生理機能を発現する。このことからIGF−Iは、種々の疾患への治療が期待される。IGF−Iは、プロインスリンのアミノ酸配列と比較して、約40%と高い相同性を有することから、インスリン受容体にも結合して、インスリン様の作用を発現することがある(非特許文献1)。また、IGF−I受容体は、インスリン受容体のアミノ酸配列と比較して、約60%と高い相同性を有することから、両受容体はヘテロ二量体を形成することがある(非特許文献1)。なお、インスリンは、インスリン受容体に作用することにより、強力な血糖低下作用を発現することから、血糖降下薬として治療に用いられている。
IGF−I受容体はα鎖及びβ鎖で構成され、L1、CR、L2、Fn1、Fn2及びFn3の6つの細胞外ドメイン、膜貫通ドメイン、及び細胞内ドメインを含む膜貫通タンパク質である(非特許文献2)。IGF−I受容体の細胞内ドメインは、チロシンキナーゼを有する。細胞外ドメインであるCR(cysteine−rich domain)は、IGF−IがIGF−I受容体に結合した時の当該受容体の立体構造の変化に伴う、細胞内のチロシンキナーゼの活性化に関与する。IGF−I受容体は、ホモ二量体複合体(ホモ型)を形成して、IGF−Iが結合すると、受容体キナーゼを活性化することにより信号を送る。また、インスリン受容体とのヘテロ二量体複合体(ヘテロ型)を形成して、インスリン又はIGF−Iが結合すると、受容体キナーゼを活性化することにより信号を送る(非特許文献3、4)。
IGF−Iは、身長や体重増加等の成長促進作用、及び糖代謝促進や血糖低下作用等のインスリン様代謝作用が明らかにされている。ヒト組換えIGF−Iであるメカセルミンは、インスリン受容体異常症の高血糖、高インスリン血症、黒色表皮腫及び多毛といった症状を改善することが認められている。また、成長ホルモン抵抗性低身長症の成長障害を改善することが認められている(非特許文献5)。
IGF−Iの成長促進作用として、IGF−Iはヒト軟骨細胞のDNA合成能を亢進させることが知られている。また、IGF−Iの投与は、下垂体摘除ラットの体重を増加させ、大腿骨骨長を伸長させる(非特許文献5)。
IGF−Iを介した細胞増殖活性の亢進は、IGF−I受容体の持続的な活性化が必要である(非特許文献6)。IGF−I受容体を過剰発現させた動物では筋肉量が増加している(非特許文献7)。また、IGF−I/IGFBP3の持続投与は、大腿骨近位部骨折患者の握力を亢進させ、介助なしでの座位からの立ち上がり能力を改善させる(非特許文献8)。高齢のヒト及びマウスの筋肉中のIGF−I濃度は、若齢と比較して低下することが知られているが(非特許文献9、10)、IGF−Iを筋組織特異的に強制発現させた高齢マウスでは、野生型マウスと比較して、筋肉量が改善した(非特許文献11)。
グレリン受容体の作動薬であるアナモレリンは、廃用性筋萎縮症であるカケキシアの臨床試験において、除脂肪量を増加させた。一方、副作用として、吐き気及び血糖値の上昇が認められる(非特許文献12)。
ミオスタチンは、アクチビン受容体II(ActRII)に作用して、Akt/mTORを阻害する、骨格筋形成の負の制御因子である(非特許文献13〜15)。抗ミオスタチン抗体であるLY2495655は、全人工股関節置換術を実施した患者及び高齢者の筋肉量を増加させる(非特許文献16、17)。
また、抗ActRII抗体であるビマグルマブは、神経筋疾患患者の筋肉量を増加させる(非特許文献18)。しかし、骨格筋の形成を促進させ、治療のために使用できる薬は現在のところ存在しない。
ヒト組換えGH製剤(成長ホルモン製剤)はGH受容体を活性化し、IGF−Iを分泌させ、成長促進効果が認められる。一方、1日1回投与の皮下注射剤のため、服薬コンプライアンス(投与忘れなど)に起因した成長効果の低下が認められている(非特許文献19)。GH製剤の動態を改善し、週1回又は2週に1回投与の持続型GH製剤が開発中である。
しかし、服薬コンプライアンスを改善し、成長促進作用を有する、治療のために使用できる薬は現在のところ存在しない。さらに、GH製剤は、GH受容体活性化に対する感受性が低下した、GH受容体異常、又はGH治療に抵抗性を有する患者に対して、成長効果の低下が認められている(非特許文献20)。
IGF−Iは、GH受容体の下流で作用することから、GH受容体活性化に対する感受性が低下した患者に対しても成長促進作用を有する唯一の治療薬である。しかし、IGF−I製剤は1日2回投与の注射剤であり、服薬コンプライアンスが悪いだけでなく、副作用として低血糖が認められている(非特許文献21)。IGF−Iの服薬コンプライアンス、及び低血糖を改善させ、治療のために使用できる薬剤は現在のところ存在しない。
IGF−Iのインスリン様作用として、血糖低下作用が知られている。IGF−Iは、ラット筋肉由来細胞においてグルコース取込み作用を亢進させる(非特許文献5)。また、IGF−Iの投与は、ラットの血糖値を低下させる(非特許文献5)。
IGF−Iによる血糖低下作用は、臨床上の副作用として低血糖を惹起させることが報告されている(非特許文献21)。さらに、IGF−Iは、ヒトへの投与により低血糖を起こすことから、治療開始時は、低用量から順次適当量を投与し、投与後の血糖値等を含む各種臨床所見の観察が必要となる(非特許文献5)。
IGF−Iは、IGF−I受容体の下流シグナルであるAktのリン酸化の亢進を介して血糖低下作用を発現する。Aktの活性型変異体は、3T3−L1細胞のグルコース取込みを亢進させる(非特許文献22)。一方、Akt2を欠損させたマウスは、血糖値が上昇した(非特許文献23)。また、ラット筋肉由来細胞においてAkt阻害剤は、インスリン刺激によるグルコース取込みを阻害する(非特許文献24)。さらに、IGF−Iは、血糖低下作用に関与するインスリン受容体を活性化させることが知られている。これらのことから、IGF−Iによる血糖低下作用には、Aktの過剰な活性化及びインスリン受容体の活性化が関与することが考えられる。
IGF−Iの血中半減期は、短いため治療では頻回投与が必要となる。実際に、ヒト組換えIGF−Iであるメカセルミンは、血中半減期が約11時間から16時間であり、低身長症の治療では1日1回から2回の投与が必要である(非特許文献5)。
血中のIGF−Iの約70から80%はIGFBP3と結合している。IGF−Iの遊離体が生理活性を示す。IGFBP3との結合は、IGF−Iの血中半減期を約10時間から16時間に維持している(非特許文献1)。
IGF−IとIGFBP3の配合剤であるIPLEXは、血中半減期が約21時間から26時間とIGF−Iと比較して長く、1日1回の投与を可能にした薬剤である(非特許文献23)。しかし、IPLEXは市場から撤退している。
IGF−Iの動態を改善させたPEG化IGF−Iも開発が試みられたが、治療に用いられている薬剤は存在しない(特許文献1)。
IGF−Iは多種の臓器に作用し、その生理機能は多岐にわたることが知られている(非特許文献21)。
IGF−Iは、中枢神経系において、IGF−I受容体の活性化を介して、ミトコンドリアの保護及び抗酸化作用による神経保護作用があることが報告されている(非特許文献26、27)。IGF−Iは、傷害後の神経突起の形成を促進させる(非特許文献28)。
IGF−Iは、成長促進の主要な因子である(非特許文献29、30)。実際に、ヒト組換えIGF−Iであるメカセルミンは、低身長症の治療薬として臨床で用いられている。
IGF−Iは、肝硬変の治療に有用であると考えられている。肝硬変は、肝障害又は慢性肝疾患から病態進展したものであり、肝臓の線維化を伴う疾患である。肝硬変モデル動物において、IGF−Iの投与は、肝臓の線維化を抑制した(非特許文献31)。
IGF−Iは、腎臓の発達、機能にも関与することが知られている。腎臓のメサンギウム細胞において、IGF−Iは、糖毒性による酸化ストレス及びアポトーシスに対して保護作用がある(非特許文献32)。IGF−Iは、腎症の治療薬として期待される。
IGF−Iは、その多彩な生理作用から、種々の疾患の治療薬として期待される。しかし、副作用である血糖低下作用、及び短い半減期による複数回の投与が臨床で用いるための課題である。
抗体製剤は、一般的に半減期が長く、月に1回から2回の投与で有効性を示す。IGF−I受容体アゴニスト抗体は、in vitroでの受容体活性化作用が報告されているが、in vivoにおいてIGF−I受容体に対するアゴニスト活性を示した抗体の報告は無い(非特許文献33〜37)。
抗体3B7及び抗体2D1は、in vitroにおいて細胞のDNA合成を亢進する(非特許文献34)。
抗体11A1、11A4、11A11及び24−57は、in vitroにおいてIGF−I受容体のチロシンのリン酸化を亢進する(非特許文献35)。
抗体16−13、17−69、24−57、24−60、24−31、及び26−3は、in vitroにおいて細胞のDNA合成、及びグルコース取込みの亢進作用を有することが示されており、これらの抗体は血糖低下作用を有する可能性がある(非特許文献36,37)。
IGF−I受容体に結合する抗体はIGF−IとIGF−I受容体の結合を阻害するアンタゴニスト作用を利用して、悪性腫瘍等の治療への応用が試みられている。しかしながら、既存のIGF−I受容体アンタゴニスト抗体は単独治療において高血糖等の副作用が多いだけでなく(非特許文献38)、他の抗癌剤との併用により高血糖の発現率が上昇することから(非特許文献39)、治療への適用は限定的なものになるものと考えられている。
[1]脊椎動物のIGF−I受容体に特異的に結合し、脊椎動物由来細胞の増殖誘導活性を有する、抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
[2]脊椎動物由来細胞の増殖誘導活性が、天然型IGF−Iと同程度以上である、[1]に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
[3]脊椎動物由来細胞の増殖誘導活性のin vitroにおけるEC50値が、天然型IGF−Iに対して1/20以下である、[1]又は[2]に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
[4]培養下の脊椎動物由来細胞に接触させた場合における、当該培養細胞との接触時間に対する当該培養細胞の増殖誘導作用の持続性が、天然型IGF−Iと比べて改善されている、[1]から[3]のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
[5]天然型IGF−Iが配列番号1に記載のアミノ酸配列を有するヒトIGF−Iである、[2]から[4]のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
[6]脊椎動物由来細胞の増殖誘導活性のin vitro におけるEC50値が、0.1nmol/L以下である、[1]から[5]のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
[7]脊椎動物に非経口投与されることによって、当該脊椎動物の筋肉量及び/又は体長の増加を誘導する活性を有する、[1]から[6]のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
[8]脊椎動物へ1週間に1回以下の頻度で投与する、[1]から[7]のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
[9]脊椎動物が、ヒト、又はモルモット、サル、ウサギ、ウシ、ブタ、ウマ、ヒツジ、イヌもしくはニワトリを含む非ヒト動物、さらにはヒトIGF−I受容体を発現させた非ヒト動物である、[1]から[8]のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
[10]脊椎動物由来細胞の増殖を誘導する用量において、分化筋細胞でのグルコース取込みを誘導しないことを特徴とする、[1]から[9]のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
[11]in vitroにおける脊椎動物由来細胞の増殖誘導活性を示すEC50値の100倍以上の用量においても分化筋細胞でのグルコース取込みを誘導しないことを特徴とする、[10]に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
[12]脊椎動物由来細胞がヒト又はヒト以外の哺乳動物に由来する筋芽細胞である、[10]又は[11]に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
[13]脊椎動物に非経口投与され、当該脊椎動物の筋肉量及び/又は体長の増加を誘導する用量において、当該脊椎動物の血糖値を低下させないことを特徴とする、[7]から[12]のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
[14]脊椎動物に非経口投与され、当該脊椎動物の筋肉量及び/又は体長の増加を誘導する有効用量に対して10倍以上の用量においても、当該脊椎動物の血糖値を変動させないことを特徴とする、[13]に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
[15]IGF−I受容体のCRドメインに結合する、[1]〜[14]のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
[16]IGF−I受容体のCRドメインに結合し、IGF−I又はIGF−IIとIGF−I受容体の結合を阻害する、抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
[17]IGF−I受容体のCRドメインの配列におけるProSerGlyPheIleArgAsnX1X2GlnSerMet(X1はGly又はSer、X2はSer又はThr)を含むエピトープ又はその近傍に結合することを特徴とする、[15]又は[16]に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。

IGFとはインスリン様成長因子(Insulin−like Growth Factor)のことをいい、IGF−IとIGF−IIが存在する。IGF−I及びIGF−IIは、後述のIGF−I受容体(インスリン様成長因子―I受容体:Insulin−like Growth Factor−I Receptor)に結合して、細胞内に細胞分裂や代謝のシグナルを入れるアゴニスト活性を有する生体内のリガンドである。IGF−I及びIGF−IIは、IGF−I受容体と構造的に類似性のあるインスリン受容体(INSR)にも弱く交差的に結合することが知られている。本明細書では、生理的機能等がよりよく知られているIGF−Iを主に扱うが、IGF−I受容体とリガンドとの結合を介する作用や疾患等の検討を行う場合にはIGF−IとIGF−IIの両者の作用を含めて記載することがある。
IGF−I受容体はインスリン様成長因子−I受容体(Insulin−like Growth Factor−I Receptor)のことをいう。本明細書において「IGF−I受容体」は、特に断りがない限り、IGF−I受容体タンパク質を意味する。IGF−I受容体はα鎖とβ鎖からなるサブユニットが2つ会合した構造のタンパク質である。配列番号2に示したヒトIGF−I受容体のアミノ酸配列においては、そのアミノ酸配列のうち、31番目から735番目のアミノ酸配列からなる部分がα鎖に相当し、β鎖は740番目以降の配列に相当する。IGF−I受容体のα鎖はIGF−Iの結合部分を有し、β鎖は膜貫通型の構造であり、細胞内へのシグナルを伝える働きをする。IGF−I受容体のα鎖は、L1、CR、L2、FnIII−1及びFnIII−2a/ID/FnIII−2bのドメインに分かれる。配列番号2に示したヒトIGF−I受容体のアミノ酸配列においては、31番目から179番目の部分がL1ドメイン、180番目から328番目の部分がCRドメイン、329番目から491番目の部分がL2ドメイン、492番目から607番目の部分がFnIII−1ドメイン及び608番目から735番目までがFnIII−2a/ID/FnIII−2bドメインに相当する。中でもCR(cysteine−rich domain)ドメインは、IGF−IがIGF−I受容体に結合した時の当該受容体の立体構造の変化に伴う、β鎖の細胞内チロシンキナーゼの活性化に関与する。ヒトのIGF−I受容体のアミノ酸配列は、EMBL−EBIのUniProtKB−アクセッション番号P08069等から参照することが可能であるが、配列表の配列番号2にも示す。
抗体とは、ジスルフィド結合により相互結合された少なくとも2つの重(H)鎖及び2つの軽(L)鎖を含む糖タンパク質である。それぞれの重鎖は、重鎖可変領域(VHと略される)及び重鎖定常領域を含み、重鎖定常領域は、3つのドメイン、CH1、CH2及びCH3を含む。それぞれの軽鎖は、軽鎖可変領域(VLと略される)及び軽鎖定常領域を含む。軽鎖定常領域は、1つのドメイン、CLを含む。軽鎖の定常領域にはλ鎖及びκ鎖と呼ばれる2種類が存在する。重鎖の定常領域にはγ鎖、μ鎖、α鎖、δ鎖及びε鎖が存在し、その重鎖の違いによって、それぞれIgG、IgM、IgA、IgD及びIgEという抗体のアイソタイプが存在する。VH及びVL領域は、さらにフレームワーク領域(FR)と称される、より保存されている4つの領域(FR−1、FR−2、FR−3、FR−4)と、相補性決定領域(CDR)と称される超可変性の3つの領域(CDR−1,CDR−2,CDR−3)に細分される。VHは、アミノ末端からカルボキシ末端へ、FR−1、CDR−1(CDR−H1)、FR−2、CDR−2(CDR−H2)、FR−3、CDR−3(CDR−H3)、FR−4の順番で配列された3つのCDR及び4つのFRを含む。VLはアミノ末端からカルボキシ末端へ、FR−1、CDR−1(CDR−L1)、FR−2、CDR−2(CDR−L2)、FR−3、CDR−3(CDR−L3)、FR−4の順番で配列された3つのCDR及び4つのFRを含む。重鎖及び軽鎖の可変領域は、抗原と相互作用する結合ドメインを含む。
本発明の抗IGF−I受容体抗体は、IGF−I受容体のCRドメインをエピトープとし、IGF−I受容体の一次構造(アミノ酸配列)及び高次構造と高い類似性を有するINSRには結合性を有さない。
本発明の抗IGF−I受容体抗体は、IGF−I受容体のCRドメインに結合することにより、IGF−I受容体が二量体を形成したホモ型の受容体、あるいはIGF−I受容体とINSRが二量体を形成したヘテロ型の受容体を活性化させることが考えられる。
本発明の抗IGF−I受容体抗体は、脊椎動物のIGF−I受容体に特異的に結合し、細胞増殖の誘導活性を有するものであれば、配列は特に限定されない。
しかしながら、各CDR配列として、特定のアミノ酸配列を有することが好ましい。具体的には以下のとおりである。なお、本発明においてアミノ酸配列の「同一性」(identity)とは、一致するアミノ酸残基の割合を意味し、相同性(similarity)とは、一致又は類似するアミノ酸残基の割合を意味する。相同性及び同一性は、例えばBLAST法(NCBIのPBLASTのデフォルト条件)により決定することができる。
1)脂肪族側鎖を有するアミノ酸残基:グリシン、アラニン、バリン、ロイシン、及びイソロイシン残基。
2)脂肪族ヒドロキシル側鎖を有するアミノ酸残基:セリン及びトレオニン残基。
3)アミド含有側鎖を有するアミノ酸残基:アスパラギン及びグルタミン残基。
4)芳香族側鎖を有するアミノ酸残基:フェニルアラニン、チロシン、及びトリプトファン残基。
5)塩基性側鎖を有するアミノ酸残基:リシン、アルギニン、及びヒスチジン残基。
6)酸性側鎖を有するアミノ酸残基:アスパラギン酸及びグルタミン酸残基。
7)硫黄含有側鎖を有するアミノ酸残基:システイン及びメチオニン残基。

本発明の抗IGF−I受容体抗体と、IGF−I受容体に対して競合結合する抗体も、本発明の範囲に含まれる。本発明において「競合結合」とは、複数種のモノクローナル抗体が抗原と共存する際に、一方の抗体の抗原への結合が、他方の抗体の抗原への結合により阻害される現象を意味する。一般的には、一定量(濃度)のモノクローナル抗体に対して、別のモノクローナル抗体の量(濃度)を変えて加えていった場合に、前者の一定量のモノクローナル抗体の抗原への結合量が低下する添加量(濃度)を測定することによって測定可能である。その阻害の程度は、IC50又はKiという値で表すことができる。本発明の抗IGF−I受容体抗体と競合結合するモノクローナル抗体とは、本発明の抗IGF−I受容体抗体、例えばIGF11−16抗体を10nMで用いて抗原抗体結合を検出した際に、IC50が通常1000nM以下、中でも100nM以下、更には10nM以下の抗体をいう。競合結合の測定を行う場合、用いる抗体を酵素、蛍光物質、発光物質、放射性同位元素等で標識を行い、その標識した物質の物理的及び/又は化学的特性に適した測定方法を用いて検出することでその測定を行うことも可能である。
本発明の抗IGF−I受容体抗体は、他の脊椎動物のIGF−I受容体に対して交差反応を有することが好ましい。交差反応とは、その抗体が抗原抗体反応をするIGF−I受容体の動物種(例えばヒト)とは異なる、他の動物種の抗原に対する抗体の結合性を示す。その抗体が抗原抗体反応をするIGF−I受容体の動物種以外である、ヒト、又はモルモット、サル、ウサギ、ウシ、ブタ、ウマ、ヒツジ、イヌもしくはニワトリを含む非ヒト動物のIGF−I受容体と交差反応性を有することが好ましい。実施例7において抗IGF−I受容体抗体であるIGF11−16抗体は、ヒトIGF−I受容体のCRドメインにおけるProSerGlyPheIleArgAsnGlySerGlnSerMetの配列と結合することが示された。サル(カニクイザル)、ウサギ、モルモット、ウシ、ヒツジ、ウマ及びイヌのIGF−I受容体の相同的な部分において、このProSerGlyPheIleArgAsnGlySerGlnSerMetの配列が保存されており、これらの種の間でのIGF−I受容体と結合交差性がある。また、マウス及びラットにおいては、当該相同的な部分のアミノ酸配列がProSerGlyPheIleArgAsnSerThrGlnSerMetとなっており、この部分に結合する抗IGF−I受容体抗体を取得することで、マウスやラット等のIGF−I受容体と結合し、IGF11−16と同様の性状や機能を有する抗体を取得することが可能である。
本発明のある態様における抗IGF−I受容体抗体は、脊椎動物由来細胞の増殖誘導活性を有する。IGF−I受容体アゴニスト抗体の存在はすでに知られていたものの、初代培養細胞、その中でも筋芽細胞において細胞の増殖誘導活性を示した抗体の報告は無かった。さらに、in vitroにおいてIGF−IのEC50値よりも低用量で細胞の増殖誘導活性を有する抗体の報告もなかった。本発明における脊椎動物由来細胞とは、好ましくは哺乳類、鳥類、爬虫類、両生類又は魚類に由来する細胞であり、より好ましくは哺乳類又は鳥類に由来する細胞であり、さらにより好ましくは、ヒト、サル、ウサギ、モルモット、ウシ、ブタ、ヒツジ、ウマ又はイヌに由来する細胞である。これらの種に由来する細胞で、IGF−I受容体を発現し、当該IGF−I受容体に本発明の抗IGF−I受容体抗体が交差反応する細胞であれば、本発明の抗IGF−I受容体抗体によって細胞増殖が誘導されうる。また、本発明の抗IGF−I受容体抗体と交差反応性がある種のIGF−I受容体を発現されるように改変された細胞や動物、又は当該改変動物に由来する細胞も、本発明における脊椎動物由来細胞に含まれる。
を有することが示されたのは、本発明の抗IGF−I受容体抗体が初めてである。
本発明のある態様における抗IGF−I受容体抗体は、脊椎動物由来の分化筋細胞での細胞内へのグルコースの取込み作用及び/又は脊椎動物での血糖値に影響を及ぼさない特徴を有する。IGF−IはIGF−I受容体へのアゴニスト作用の一部として細胞へのグルコースの取込みの上昇や血糖値の降下作用を起こすことが知られているが、IGF―I受容体アゴニスト抗体として機能する本発明での抗IGF−I受容体抗体では細胞での増殖誘導活性のin vitroにおけるEC50値の100倍以上の用量においても分化筋細胞でのグルコース取込みを誘導せず、動物に非経口的に投与した場合に筋肉量の増加を誘導する有効用量のさらに10倍以上でも血糖値を変動させないという性状を示すことは予想外の効果である。また、IGF―I受容体アンタゴニスト抗体としての性状としても、脊椎動物由来細胞の分化筋細胞でのグルコース取込み及び/又は脊椎動物での血糖値に影響を及ぼさないという特徴は、従来のIGF−I受容体アンタゴニスト抗体をヒトの治療で用いる際の未充足の課題であった高血糖等を回避するうえで有利な効果である。本発明における脊椎動物由来細胞とは、好ましくは哺乳類、鳥類、爬虫類、両生類又は魚類に由来する細胞であり、より好ましくは哺乳類又は鳥類に由来する細胞であり、さらにより好ましくは、ヒト、サル、ウサギ、モルモット、ウシ、ブタ、ヒツジ、ウマ又はイヌに由来する細胞である。また、本発明の抗IGF−I受容体抗体と交差反応性を有する脊椎動物種由来のIGF−I受容体あるいは結合性を有するように変異を導入したIGF−I受容体を発現されるように改変された細胞や動物、又は当該改変動物に由来する細胞も、本発明における脊椎動物由来細胞に含まれる。
本発明における抗体は、当業者に周知の技法を用いて得ることができる。本発明における抗体は、ポリクローナル抗体、又はモノクローナル抗体(ミルスタイン(Milstein)等、ネイチャー(Nature)(England)1983年10月6日発行、305巻5934号、p.537〜540)であることができる。例えば、ポリクローナル抗体は配列番号2のIGF−I受容体のペプチドを抗原として哺乳動物に感作し、当該動物の血清等から回収することができる。また、ペプチドを抗原として用いる場合には、BSAやKLH等のキャリアタンパク質やポリリジン等に結合させた形態での抗原を用いることができる。また抗原として用いるペプチドの具体的例としては、配列番号2の部分配列であるProSerGlyPheIleArgAsnGlySerGlnSerMetを用いることができるが、これに限定されるものではない。当該発明におけるモノクローナル抗体は、抗原を感作した哺乳動物から免疫細胞を取り出して骨髄腫細胞等と細胞融合させることにより得られたハイブリドーマをクローニングして、その培養物から回収することができる。そのようなモノクローナル抗体の取得方法としては、実施例1に記載されており、それによって得られたモノクローナル抗体としては配列番号9のVHアミノ酸配列、配列番号10のVLアミノ酸配列を有するモノクローナル抗体(IGF11−16)が挙げられるが、それらに限定されるものではない。
本発明の抗IGF−I受容体抗体は、IGF−Iに関連した状態又はIGF−I受容体への作用に起因する疾患の治療薬又は予防薬として利用可能である。具体的には、IGF−Iに関連した状態又はIGF−I受容体アゴニスト抗体での治療又は予防の対象となる疾患としては、廃用性筋萎縮、低身長症、肝硬変、肝線維化、糖尿病性腎症、慢性腎不全、ラロン症、老化、子宮内胎児発育遅延(IUGR)、心血管保護、糖尿病、インスリン抵抗性、メタボリックシンドローム、骨粗しょう症、嚢胞性線維症、筋強直性ジストロフィー、エイズ筋減弱症、HIVに伴う脂肪再分布症候群、クローン病、ウェルナー症候群、X連鎖性複合免疫不全症、難聴、神経性無食欲症及び未熟児網膜症、ターナー症候群、プラダー・ウィリー症候群、シルバー・ラッセル症候群、特発性低身長、肥満、多発性硬化症、潰瘍性大腸炎、低筋肉量、心筋虚血、低骨密度、IGF−I受容体アンタゴニスト抗体での治療又は予防の対象となる疾患としては、神経芽細胞腫、横紋筋肉腫、骨肉腫、小児がん、先端巨大症、卵巣がん、膵臓がん、良性前立腺肥大症、乳がん、前立腺がん、骨がん、肺がん、結腸直腸がん、頚部がん、滑膜肉腫、膀胱がん、胃がん、ウィルムス腫瘍、転移性カルチノイド及び血管作動性腸管ペプチド分泌腫瘍に関連する下痢、ビポーマ、ウェルナー−モリソン症候群、ベックウィズ−ヴィーデマン症候群、腎臓がん、腎細胞がん、移行上皮がん、ユーイング肉腫、白血病、急性リンパ芽球性白血病、脳腫瘍、膠芽腫、非膠芽腫性脳腫瘍、髄膜腫、下垂体腺腫、前庭神経鞘腫、未分化神経外胚葉性腫瘍、髄芽腫、星状細胞腫、乏突起膠腫、脳室上衣腫、脈絡叢乳頭腫、巨人症、乾癬、アテローム性動脈硬化症、血管の平滑筋再狭窄、不適切な微小血管増殖、糖尿病性網膜症、グレーヴズ病、全身性エリテマトーデス、慢性甲状腺炎、重症筋無力症、自己免疫性甲状腺炎、及びベーチェット病が挙げられる。特に、本発明の抗IGF−I受容体抗体は廃用性筋萎縮及び/又は低身長症の治療薬又は予防薬としての使用が好ましい。また、本発明の抗IGF−I受容体抗体は投与によって血糖値の変動を生じさせない点において優れている。
本発明のある態様における抗IGF−I受容体抗体は、ヒト以外の動物での畜産用途又は獣医学的用途に用いることができる。本発明の抗IGF−I受容体抗体を畜産用途又は獣医学的用途で用いることができる動物としては、好ましくはヒト以外の哺乳類、鳥類、爬虫類、両生類又は魚類に属する動物であり、より好ましくはヒト以外の哺乳類又は鳥類に属する動物であり、さらにより好ましくは、サル、ウサギ、モルモット、ウシ、ブタ、ヒツジ、ウマ又はイヌから選択される動物である。現在、ウシ成長ホルモンやブタ成長ホルモンがウシでの乳汁産生量の増大や子ブタの成長促進のために使われているが、これらの作用は成長ホルモンによって発現が誘導されるIGF−Iの作用によるものと考えられている(h. JiangとX. Ge、Journal of Animal Science、Vol.92、pp21−29、2014)。従って、本発明の抗IGF−I受容体抗体は、そのアゴニスト作用を利用することで、同様に動物の乳汁産生の亢進や、胎児や出生後の動物の成長促進等に用いることができる。その他、本発明の抗IGF−I受容体抗体を用いることができる用途の例としては、動物における筋肉量の増量又は脂肪重量に対する筋肉重量比の増大、摂取飼料の体組織への効率的変換、繁殖効率増大や種保存のための生殖能力の増強、動物における外傷や消耗性疾患での消耗的症状の治療等が挙げられるが、これらに限定されるわけではない。また、本発明の抗IGF−I受容体抗体は、別の態様であるアンタゴニスト作用を利用することで、動物での悪性腫瘍の治療、繁殖頻度の制御、個体の成長の制御やその他の用途にも用いることができる。本発明における抗IGF−I受容体抗体を用いる場合、投与する動物に合わせて抗体のフレームワークや定常領域のアミノ酸配列を改変して免疫原性を低下させる等、当業者であれば適宜その構造を改変したうえで用いることも可能である。
脊椎動物由来細胞をin vitroにおいて維持、増殖及び/又は分化させるための細胞培養技術において、IGF−I又はその誘導体が多く用いられており、細胞培養用の試薬として市販されている。IGF−Iは安定性の問題等から長期の培養においては時間が経つにつれて効果が減弱する可能性があり、安定的な細胞培養を行うためには適宜濃度を調節する等の対応が必要になる。また、IGF−Iは細胞へのグルコースの取込みを誘導することから、細胞内グルコース濃度の上昇によって細胞の代謝や特性の変化が誘導されたり、培地中のグルコース濃度が減少して培養環境が変化する可能性がある。本発明の抗IGF−I受容体抗体はIGF−Iと比較して安定性が高く、細胞に接触してからの細胞増殖を誘導する時間が長く、IGF−Iより低濃度で細胞増殖誘導活性を示し、かつ、細胞内へのグルコース取込みを誘導しないという特徴を持つ。本発明の抗IGF−I受容体抗体は、細胞培養においてその培地に適量を添加して使用されるほか、培養を行う容器の固相に吸着又は固定して用いることが可能で、それによって使用量を削減したり、固相に付着する細胞に対して有効に細胞増殖誘導を行うことができる。本発明における脊椎動物由来細胞とは、好ましくは哺乳類、鳥類、爬虫類、両生類又は魚類に由来する細胞であり、より好ましくは哺乳類又は鳥類に由来する細胞であり、さらにより好ましくは、ヒト、サル、ウサギ、モルモット、ウシ、ブタ、ヒツジ、ウマ又はイヌに由来する細胞である。また、当該細胞としては、初代培養細胞、株化細胞又はそれらの細胞の形質転換体細胞、遺伝子組み換え動物に由来する細胞等を用いることが可能である。さらに、本発明の抗IGF−I受容体抗体を用いた培養の対象としては、脊椎動物又はその遺伝子改変動物に由来する臓器や組織等も含まれる。本発明の抗IGF−I受容体抗体は、細胞を用いた物質生産のための培養や、細胞自体を用いる細胞医療・再生医療での培養工程で用いることができる。
マウスモノクローナル抗体は、Kohlerら(Nature 256:495−497、1975)のハイブリドーマ法により作製し得る。IGF−I受容体アゴニスト抗体は、マウスにヒトIGF−I受容体を発現させた細胞を用いて免疫して、標準的なハイブリドーマ技術を使用して作製した。全ての動物実験は、施設の規則に従って実施した。マウスから採取した脾臓由来の細胞とマウスミエローマ細胞株(P3U1)との融合による標準的な方法を使用して実施した。ヒポキサンチン、アミノプテリン、及びチミジンを含有する培地を使用してハイブリドーマを選択した。ハイブリドーマの培養液を用いて、IGF−I受容体を発現させた細胞を用いたCell ELISAによる結合性評価、及びPathHunter(登録商標)によるIGF−I受容体の細胞内チロシンキナーゼの活性化の評価を実施し、陽性ハイブリドーマ含有ウェルを選択した。このウェルに含まれるハイブリドーマを限界希釈法によってシングルクローン化した。このシングルクローン化した陽性ハイブリドーマを無血清培養して、培養液中からプロテインAカラム(Ab−Capcher、プロテノバ)を使用してモノクローナル抗体を精製した。このモノクローナル抗体を使用して、ヒト筋芽細胞増殖活性評価によりIGF−I受容体アゴニスト抗体であるIGF11−16を見出した。
IGF−I受容体アゴニスト抗体の抗体アイソタイプを決定するために、抗体のアイソタイプに特異的な抗体を用いて、ELISAを実施した。PBSにて2000倍希釈した抗マウスIgG抗体(TAGO、6150)を、50μL/ウェルで96ウェルプレート(Nunc、MaxiSorp)に添加し、4℃で一晩静置した。96ウェルプレートを3%BSA/PBSに置換したものをELISAに使用した。IGF−I受容体アゴニスト抗体を、抗マウスIgG抗体を固定化させた96ウェルプレートに30μL/ウェルで添加し、室温にて1.5時間反応させた。洗浄液にて洗浄した後、マウスIgGの各種アイソタイプに特異的に反応する抗体、抗マウスIgG1抗体−ALPコンジュゲート(SBA、1070−04)、抗マウスIgG2a抗体−ALPコンジュゲート(SBA、1080−04)、抗マウスIgG2b抗体−ALPコンジュゲート(SBA、1090−04)及び抗マウスIgG3抗体−ALPコンジュゲート(SBA、1100−04)、を30μL/ウェルで添加し、室温にて1時間反応させた。基質(PNPP)を100μL/ウェルで添加し、室温にて45分間反応させ、吸光度405−550nmを算出した。吸光度405−550nmの値を結合活性として評価した。
IGF11−16は、抗マウスIgG1抗体に反応性を示すことから、抗体のアイソタイプはIgG1であった。
IGF−I受容体アゴニスト抗体の軽鎖及び重鎖の遺伝子配列を決定するために、SMARTer(登録商標) RACE法を実施した。抗体を産生するハイブリドーマ由来のRNAから開始及び終始コドンを含む抗体の重鎖及び軽鎖の遺伝子の断片をSMARTer(登録商標) RACE法により取得し、その塩基配列を決定した。ハイブリドーマ由来のTotal RNAを鋳型としてSMARTer(登録商標) RACE 5’/3’ Kit(634859、Clontech)を用いて、1st strand cDNAを合成した後、PCR反応によりcDNAを増幅させた。そのcDNAを鋳型として、キットに付属のユニバーサル配列に対するプライマーと、抗体の重鎖及び軽鎖にそれぞれ特異的なプライマーを用いてPCR反応を行った。マウス抗体の軽鎖(kappa)はAccession番号BC080787、マウス抗体のIgG1はLT160966を参照してプライマーを設計した。マウス抗体の軽鎖に対するプライマーの塩基配列は、ggtgaagttgatgtcttgtgagtggを設計し、マウス抗体の重鎖に対するプライマーの塩基配列は、gctcttctcagtatggtggttgtgcを設計し、それぞれを実験に用いた。得られたPCR産物は5‘RACE PCR産物としてTAクローニングに用いた。
ヒト(配列番号2、NP_000866)、モルモット(配列番号11、XP_003475316)、カニクイザル(配列番号12、NP_001248281)、ウサギ(配列番号13、XP_017193273)、ラット(配列番号14、NP_494694)及びマウス(配列番号15、NP_034643)のIGF−I受容体に対するIGF−I受容体アゴニスト抗体の結合活性を検討するために、各種IGF−I受容体を発現させた細胞を用いてCell ELISAを実施した。
インスリン受容体に対するIGF−I受容体アゴニスト抗体の結合活性を検討するために、ヒトインスリン受容体を発現させた細胞を用いてCell ELISAを実施した。
IGF−I受容体アゴニスト抗体のIGF−I受容体に対するエピトープを同定するために、IGF−I受容体の各種ドメインを、IGF−I受容体と類似構造を有するインスリン受容体のドメインと置換した変異体に対するIGF−I受容体アゴニスト抗体の結合性を測定した。
(置換体1)ヒトインスリン受容体のL1ドメインからL2ドメインまでを、ヒトIGF−I受容体のL1ドメインからL2ドメインに置換した置換体、hIGFIR[L1−L2]/hINSR。
(置換体2)ヒトIGF−I受容体のL1ドメインからL2ドメインまでを、ヒトインスリン受容体のL1ドメインからL2ドメインに置換した置換体、hINSR[L1−L2]/hIGFIR。
(置換体3)ヒトIGF−I受容体のL1ドメインを、ヒトインスリン受容体のL1ドメインに置換した置換体、hINSR[L1]/hIGFIR。
(置換体4)ヒトIGF−I受容体のL2ドメインを、ヒトインスリン受容体のL2ドメインに置換した置換体、hINSR[L2]/hIGFIR。
IGF11−16のエピトープであるCRドメインから、さらに詳細なエピトープを同定するために、IGF11−16のIGF−I受容体に対する結合性の種差から結合配列を推定した。図1に、それぞれの種におけるIGF−I受容体のCRドメインのアミノ酸配列を示す。
(CRドメインの置換体1)ヒトIGF−I受容体(NP_000866、配列番号2)のアミノ酸配列、245番目及び247番目のアスパラギン酸及びアラニンを、それぞれアスパラギン及びトレオニンに置換させた。
(CRドメインの置換体2)ヒトIGF−I受容体(NP_000866、配列番号2)のアミノ酸配列、294番目のグルタミン酸を、アスパラギン酸に置換させた。
(CRドメインの置換体3)ヒトIGF−I受容体(NP_000866、配列番号2)のアミノ酸配列、315番目及び316番目のグリシン及びセリンを、それぞれセリン及びトレオニンに置換させた。
薬剤のIGF−I受容体に対する結合特性(結合速度及び解離速度)を検討するために、表面プラズモン共鳴(SPR)法により測定した。
IGF−I受容体アゴニスト抗体のIGF−I受容体に対する活性化作用を検出するために、PathHunter(登録商標)IGF1R Functional Assay(DiscoverX)を用いてIGF−I受容体の下流シグナルの活性化を測定した。
IGF−I受容体アゴニスト抗体のヒト筋芽細胞に対する増殖活性を検討するため、ヒト筋芽細胞に薬剤を添加して、4日後の細胞内のATP量を測定した。
モルモット筋芽細胞(Cell Applications)を、SkBM−2(Lonza、CC−3246)に1%BSAを含む培地を使用して、96ウェルプレート(Collagen type I coated)に、0.1mL/ウェル(4×103cells/ウェル)で播種し、37℃、5%CO2の条件でインキュベートした。細胞播種の翌日に各種薬剤を25μL/ウェルで添加し、37℃、5%CO2の条件で4日間インキュベートした。細胞増殖の指標として細胞内のATP量を、CellTiter−Glo(登録商標)Luminescent Cell Viability Assay(Promega)を使用して測定した。4日間インキュベートした96ウェルプレートを、培養液が50μL/ウェルとなるように上清を除き、30分以上室温にて静置した。CellTiter−Glo(登録商標)試薬を50μL/ウェルで添加して10分以上反応させた後にルミノメーター(Tristar、ベルトールド)にて発光シグナルを測定した。
IGF11−16とIGF−Iの作用持続性を比較するために、IGF11−16又はIGF−Iを添加した18時間後に培地を交換して、IGF11−16及びIGF−Iを除去した条件でヒト筋芽細胞の増殖活性を測定した。
IGF11−16のグルコース取込み作用を検討するために、ヒト分化筋細胞を用いて、放射標識された3H−2−Deoxy Glucoseの取込み量を測定して、IGF−Iの作用と比較した。
IGF−I受容体アゴニスト抗体のin vivoでの薬効を確認するために、モルモットにIGF11−16を単回投与して、2週間後の筋肉量を測定して、IGF−Iを持続投与した時の作用と比較した。筋肉量増加作用とは、モルモットの筋肉重量をコントロール群と比べて5%以上増加させることとする。
IGF−I受容体アゴニスト抗体のin vivoでの血糖低下作用の有無を確認するために、モルモットにIGF11−16を単回投与して、継時的に血糖値を測定して、IGF−Iの単回投与時の血糖低下作用と比較した。血糖低下作用とは、血糖値を50mg/dL以下に低下させる、又は低血糖症状を起こす作用とする。
IGF−I受容体アゴニスト抗体のin vivoでの骨に対する薬効を確認するために、IGF−Iを持続投与、及び成長ホルモン(GH)を1日1回反復投与した時の作用と比較した。下垂体を摘出されたモルモットにIGF11−16を単回投与して、2週間後の脛骨の長さ、及び成長板軟骨の厚さを、成長促進効果の指標として測定した。IGF11−16(0.3mg/kg及び1mg/kg)を、下垂体摘出モルモットに単回皮下投与した。比較対照としてヒト組換えIGF−I(メカセルミン)を、浸透圧ポンプ(アルゼット)を使用して皮下に埋め込み、1mg/kg/日となるように持続投与した。また、別の比較対照としてヒト組換えGH(ジェノトロピン(登録商標))を、1mg/kgの用量で、1日1回、反復皮下投与した。薬剤投与の2週間後、モルモットを麻酔下で放血致死させ、脛骨近位部の成長板軟骨の厚さ、及び脛骨の長さを測定した。結果を図9及び図10に示す。
IGF11−16の0.3mg/kgの単回投与群の成長促進効果は、ヒト組換えIGF−Iを1mg/kg/日で持続投与した群(IGF−I)と同程度であった。また、IGF11−16の1mg/kgの単回投与群の成長促進効果は、ヒト組換えGHを1mg/kg/日で反復投与した群(GH)と同程度であった。このことから、IGF11−16は、単回投与により、IGF−Iの持続投与、及びGHの1日1回の反復投与と同等の薬効を有することが示された。臨床でのヒト組換えIGF−I(メカセルミン)及びヒト組換えGH(ジェノトロピン(登録商標))の用法用量は、それぞれ1日1回から2回の皮下注射、及び週6回から7回の皮下注射である。一方、in vivoにおいてIGF11−16は2週に1回の投与でIGF−Iの持続投与、及びGHの1日1回反復投与と同等の有効性を示すことから、IGF−I及びGHと比較して持続性に優れることが示された。
IGF−Iの血中動態
モルモットを12時間絶食させ、ヒト組換えIGF−Iを、0.3、1、3及び10mg/kgで皮下投与した。モルモットは、投与24時間後まで絶食させた。覚醒状態のモルモットを、投与前(0時間)、投与1、2、4、8、10及び24時間後に採血して、血漿中のヒトIGF−I濃度をELISA(DG100、R&D)により測定した。結果を図11に示す。
モルモットを12時間絶食させ、IGF−I受容体アゴニスト抗体を、0.3、1、3、10、30及び100mg/kgで皮下投与した。モルモットは、投与24時間後まで絶食させ、24時間後に再給餌した。覚醒状態のモルモットを、投与前(0時間)、投与2、4、8、10、24、48及び72時間後に採血して、血漿中のIGF11−16濃度をELISAにより測定した。結果を図12に示す。
血漿中のIGF11−16濃度は投与用量に依存して上昇し、投与48時間以降も血漿中のIGF11−16濃度は投与24時間後と比較して約50%以上を維持していた。IGF11−16の血中動態はIGF−Iと比較して持続性に優れていることが示された。

- 配列番号2に記載のアミノ酸配列を有する天然型ヒトIGF−I受容体に特異的に結合し、配列番号2に記載のアミノ酸配列における315番目及び316番目に相当するアミノ酸残基を含む結合部位に結合すると共に、ヒト由来細胞の増殖誘導活性を有する、抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
- ヒト由来細胞の増殖誘導活性が、配列番号1に記載のアミノ酸配列を有する天然型ヒトIGF−Iと同程度以上である、請求項1に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
- ヒト由来細胞の増殖誘導活性のin vitroにおけるEC50値が、配列番号1に記載のアミノ酸配列を有する天然型ヒトIGF−Iに対して1/20以下である、請求項1又は2に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
- 培養下のヒト由来細胞に接触させた場合における、当該培養細胞との接触時間に対する当該培養細胞の増殖誘導作用の持続性が、配列番号1に記載のアミノ酸配列を有する天然型ヒトIGF−Iと比べて改善されている、請求項1から3のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。

- 脊椎動物の筋肉量及び/又は体長の増加を誘導する用量において、当該脊椎動物の血糖値を低下させないことを特徴とする、請求項6から10のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
- 脊椎動物の筋肉量及び/又は体長の増加を誘導する有効用量に対して10倍以上の用量においても、当該脊椎動物の血糖値を変動させないことを特徴とする、請求項11に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
- IGF−I受容体のCRドメインの配列におけるProSerGlyPheIleArgAsnGlySerGlnSerMetを含むエピトープに結合する、請求項1から12のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
- サル、ウサギ、モルモット、ウシ、ブタ、ヒツジ、ウマ、イヌ、ニワトリ、マウス、又はラットのIGF−I受容体にも特異的に結合し、各動物細胞のIGF−I受容体のCRドメインの配列におけるProSerGlyPheIleArgAsnX 1 X 2 GlnSerMet(X 1 はGly又はSer、X 2 はSer又はThr)を含むエピトープに結合すると共に、各動物細胞の増殖誘導活性を有する、請求項1から13のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
- ヒト、又はモルモット、サル、ウサギ、ウシ、ブタ、ウマ、ヒツジ、イヌ、ニワトリ、ラット、又はマウスを含む非ヒト動物のIGF−I受容体と交差反応性を有する、請求項1から14のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
- 当該抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体の抗原抗体反応は、解離平衡定数(KD)が1×10-8M以下の親和性強度を有することを特徴とする、請求項1から15のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
- 当該抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体が、1)〜4)のうち少なくとも一つの特徴を有する、請求項13から16のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
1)脊椎動物由来細胞の増殖誘導活性を有する。
2)脊椎動物の筋肉量及び/又は体長の増加を誘導する活性を有する。
3)脊椎動物由来細胞の増殖を誘導する用量において、分化筋細胞でのグルコース取込みを誘導しない。
4)脊椎動物の筋肉量及び/又は体長の増加を誘導する用量において、当該脊椎動物の血糖値を変動させない。 - 抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体が、1)〜4)の少なくとも一つの特徴を有する、請求項13から17のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
1)IGF−Iによる脊椎動物由来細胞の増殖を阻害する。
2)脊椎動物におけるIGF−Iに起因する細胞増殖性疾患における細胞増殖を抑制する。
3)IGF−Iによる脊椎動物由来細胞の増殖を阻害する用量において、分化筋細胞でのグルコース取込みに影響しない。
4)脊椎動物におけるIGF−Iに起因する細胞増殖性疾患における細胞増殖を抑制する用量において、当該脊椎動物の血糖値を変動させない。 - 抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体が、Fab、scFv、Diabodyもしくは二重特異性抗体、又はそれらの誘導体である、請求項1〜18のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
- 重鎖可変領域のCDR−1(CDR−H1)配列として配列番号3又は配列番号3のいずれか1か所のアミノ酸残基が置換、欠失、もしくは挿入されたアミノ酸配列、
重鎖可変領域のCDR−2(CDR−H2)配列として配列番号4又は配列番号4のいずれか1か所又は2か所のアミノ酸残基が置換、欠失、もしくは挿入されたアミノ酸配列、
重鎖可変領域のCDR−3(CDR−H3)配列として配列番号5又は配列番号5のいずれか1か所又は2か所のアミノ酸残基が置換、欠失、もしくは挿入されたアミノ酸配列、
軽鎖可変領域のCDR−1(CDR−L1)配列として配列番号6又は配列番号6のいずれか1か所又は2か所のアミノ酸残基が置換、欠失、もしくは挿入されたアミノ酸配列、
軽鎖可変領域のCDR−2(CDR−L2)配列として配列番号7又は配列番号7のいずれか1か所のアミノ酸残基が置換、欠失、もしくは挿入されたアミノ酸配列、及び
軽鎖可変領域のCDR−3(CDR−L3)配列として配列番号8又は配列番号8のいずれか1か所又は2か所のアミノ酸残基が置換、欠失、もしくは挿入されたアミノ酸配列、
を含むアミノ酸配列からなる請求項1から19のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。 - 免疫グロブリンのフレームワーク配列をさらに含む、請求項20に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
- 免疫グロブリンのフレームワーク配列が、ヒト、又はモルモット、サル、ウサギ、ウシ、ブタ、ウマ、ヒツジ、イヌ、ニワトリ、マウスもしくはラットを含む非ヒト動物の免疫グロブリンの各クラスにおけるフレームワーク配列である、請求項21に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
- 重鎖可変領域として配列番号9又は配列番号9と90%以上の相同性を有するアミノ酸配列、及び軽鎖可変領域として配列番号10又は配列番号10と90%以上の相同性を有するアミノ酸配列を含むアミノ酸配列からなる請求項1から22のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
- ヒト、又はモルモット、サル、ウサギ、ウシ、ブタ、ウマ、ヒツジ、イヌ、ニワトリ、マウスもしくはラットを含む非ヒト動物の免疫グロブリンの各クラスにおける定常領域をさらに含む、請求項1から23のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体。
- 請求項1から24のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体をコードするポリヌクレオチド配列からなる核酸分子。
- 請求項25に記載の核酸分子を少なくとも一つ含むクローニングベクター又は発現ベクター。
- 宿主細胞に請求項26に記載のベクターが導入された組換え体細胞。
- 請求項27に記載の組換え体細胞を培養し、前記組換え体細胞から産生される当該抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体を精製する工程を含む、請求項1から24のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体の製造方法。
- 請求項1から24のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体、請求項25に記載の核酸分子、請求項26に記載のベクター、あるいは請求項27に記載の組換え体細胞を含む医薬組成物。
- 請求項1から24のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体、請求項25に記載の核酸分子、請求項26に記載のベクター、あるいは請求項27に記載の組換え体細胞以外の活性成分をさらに含む、請求項29に記載の医薬組成物。
- 活性成分が成長ホルモン又はそのアナログ、インスリン又はそのアナログ、IGF−II又はそのアナログ、抗ミオスタチン抗体、ミオスタチンアンタゴニスト、抗アクチビンIIB型受容体抗体、アクチビンIIB受容体アンタゴニスト、可溶性アクチビンIIB型受容体又はそのアナログ、グレリン又はそのアナログ、フォリスタチン又はそのアナログ、ベータ2アゴニスト、及び選択的アンドロゲン受容体モジュレーターから1つ以上選択される、請求項30に記載の医薬組成物。
- 活性成分がコルチコステロイド、制吐薬、オンダンセトロン塩酸、グラニセトロン塩酸、メトロクロプラミド(metroclopramide)、ドンペリドン、ハロペリドール、シクリジン、ロラゼパム、プロクロルペラジン、デキサメタゾン、レボメプロマジン、トロピセトロン、癌ワクチン、GM−CSF阻害薬、GM−CSF DNAワクチン、細胞に基づくワクチン、樹状細胞ワクチン、組換えウイルスワクチン、熱ショックタンパク質(HSP)ワクチン、同種腫瘍ワクチン、自己腫瘍ワクチン、鎮痛薬、イブプロフェン、ナプロキセン、トリサリチル酸コリンマグネシウム、オキシコドン塩酸、抗血管形成薬、抗血栓薬、抗PD−1抗体、ニボルマブ、ペムブロリズマブ、抗PD−L1抗体、アテゾリズマブ、抗CTLA4抗体、イピリムマブ、抗CD20抗体、リツキシマブ、抗HER2抗体、トラスツズマブ、抗CCR4抗体、モガムリズマブ、抗VEGF抗体、ベバシズマブ、抗VEGF受容体抗体、可溶性VEGF受容体断片、抗TWEAK抗体、抗TWEAK受容体抗体、可溶性TWEAK受容体断片、AMG 706、AMG 386、抗増殖薬、ファルネシルタンパク質トランスフェラーゼ阻害薬、αvβ3阻害薬、αvβ5阻害薬、p53阻害薬、Kit受容体阻害薬、ret受容体阻害薬、PDGFR阻害薬、成長ホルモン分泌阻害薬、アンジオポエチン阻害薬、腫瘍浸潤マクロファージ阻害薬、c−fms阻害薬、抗c−fms抗体、CSF−1阻害薬、抗CSF−1抗体、可溶性c−fms断片、ペグビソマント、ゲムシタビン、パニツムマブ、イリノテカン、及びSN−38からなる群より選択される成分を含む、請求項30又は31に記載の医薬組成物。
- 請求項1〜24に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体、請求項25に記載の核酸分子、請求項26に記載のベクター、及び請求項27に記載の組換え体細胞の内、いずれか一つ以上を含む、IGF−Iに関連した状態の治療又は予防に用いられる医薬。
- IGF−Iに関連した状態が、廃用性筋萎縮、低身長症、糖尿病性腎症、慢性腎不全、ラロン症、肝硬変、肝線維化、老化、子宮内胎児発育遅延(IUGR)、神経疾患、脳卒中、脊髄損傷、心血管保護、糖尿病、インスリン抵抗性、メタボリックシンドローム、骨粗しょう症、嚢胞性線維症、創傷治癒、筋強直性ジストロフィー、エイズ筋減弱症、HIVに伴う脂肪再分布症候群、火傷、クローン病、ウェルナー症候群、X連鎖性複合免疫不全症、難聴、神経性無食欲症及び未熟児網膜症、ターナー症候群、プラダー・ウィリー症候群、シルバー・ラッセル症候群、特発性低身長、肥満、多発性硬化症、線維筋痛症、潰瘍性大腸炎、低筋肉量、心筋虚血及び低骨密度から選択される、請求項33に記載の医薬。
- 非ヒト動物に投与される動物用医薬である、請求項33又は34に記載の医薬。
- 当該動物用医薬が、筋肉量及び/又は体長の増大、成長の促進、乳汁産生量の増大、繁殖の促進、又は老化の予防の目的で投与される、請求項35に記載の医薬。
- 非ヒト動物が、モルモット、サル、ウサギ、ウシ、ブタ、ウマ、ヒツジ、イヌ、又はニワトリである、請求項35又は36に記載の動物用医薬。
- IGF−I又はIGF−IIのIGF−I受容体への作用に起因する疾患の治療又は予防に用いられる、請求項33〜37のいずれか一項に記載の医薬。
- IGF−I又はIGF−IIのIGF−I受容体への作用に起因する疾患が、肝臓がん、神経芽細胞腫、横紋筋肉腫、骨肉腫、小児がん、先端巨大症、卵巣がん、膵臓がん、良性前立腺肥大症、乳がん、前立腺がん、骨がん、肺がん、結腸直腸がん、頚部がん、滑膜肉腫、膀胱がん、胃がん、ウィルムス腫瘍、転移性カルチノイド及び血管作動性腸管ペプチド分泌腫瘍に関連する下痢、ビポーマ、ウェルナー−モリソン症候群、ベックウィズ−ヴィーデマン症候群、腎臓がん、腎細胞がん、移行上皮がん、ユーイング肉腫、白血病、急性リンパ芽球性白血病、脳腫瘍、膠芽腫、非膠芽腫性脳腫瘍、髄膜腫、下垂体腺腫、前庭神経鞘腫、未分化神経外胚葉性腫瘍、髄芽腫、星状細胞腫、乏突起膠腫、脳室上衣腫、脈絡叢乳頭腫、巨人症、乾癬、アテローム性動脈硬化症、血管の平滑筋再狭窄、不適切な微小血管増殖、糖尿病性網膜症、グレーヴズ病、多発性硬化症、全身性エリテマトーデス、慢性甲状腺炎、重症筋無力症、自己免疫性甲状腺炎ならびにベーチェット病からなる群より選択される疾患である、請求項38に記載の医薬。
- 脊椎動物由来の細胞をin vitroで培養する方法であって、培養の過程で当該脊椎動物由来の細胞を、請求項1から24のいずれか一項に記載の抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体、請求項25に記載の核酸分子、請求項26に記載のベクター、及び請求項27に記載の組換え体細胞のいずれか一つ以上と接触される工程を含む、培養方法。
- 当該接触の工程が、脊椎動物由来の細胞の増殖促進又は分化誘導の目的で行われる、請求項40に記載の培養方法。
- 抗IGF−I受容体抗体もしくはその断片、又はそれらの誘導体が固相に吸着又は固定されている、請求項40又は41に記載の培養方法。
