Description
本願は、2014年9月17日出願の米国暫定特許出願62/051,915(その内容は、参照によって全体が本明細書に組み込まれる。)に対する優先権を主張するものである。

発明の分野
本発明は、相乗的に効果的な量で、(i)(x)少なくとも一つの植物成長刺激タンパク質またはペプチド;および(y)融合タンパク質をバチルス細胞の外膜に局在させる標的配列を含む融合タンパク質を発現する組換え外膜産生性バチルス細胞;および(ii)昆虫、ダニ、線虫および/または植物病原体に対する活性を向上させる能力を示す本明細書に開示の特定の殺菌剤から選択される少なくとも一つの殺菌剤を含む組成物に関する。さらに本発明は、この組成物の使用ならびに植物成長を促進し、植物の健康を増進し、および/または植物および植物部分の全体的損傷を低減する方法に関するものである。
融合タンパク質は、バチルス・セレウス(Bacillus cereus)科構成員の外膜への融合タンパク質を標的とする標的配列、外膜タンパク質、または外膜タンパク質断片ならびに(a)植物成長刺激タンパク質またはペプチド;(b)病原体から植物を保護するタンパク質もしくはペプチド;(c)植物のストレス抵抗性を促進するタンパク質またはペプチド;(d)植物結合性タンパク質またはペプチド;または(e)植物免疫系エンハンサータンパク質またはペプチドを含む。バチルス・セレウス(Bacillus cereus)科構成員細菌で発現される場合、これらの融合タンパク質は、胞子の外膜層を標的とするものであり、そのタンパク質またはペプチドが胞子の外側に露出するような物理的配置となっている。

前記融合タンパク質は、標的配列、外膜タンパク質、または外膜タンパク質断片および少なくとも植物結合性タンパク質もしくはペプチドを含むことができる。その植物結合性タンパク質もしくはペプチドは、植物のいずれかの部分(例えば、植物根または植物の地上部分、例えば葉、茎、花、または果実)または植物体に特異的もしくは非特異的に結合する能力を有するタンパク質またはペプチドであることができる。従って、例えば、前記植物結合性タンパク質もしくはペプチドは、根結合性タンパク質もしくはペプチド、または葉結合性タンパク質もしくはペプチドであることができる。
本明細書に記載の融合タンパク質は、組換え外膜産生性バチルス細胞によって発現することができる。融合タンパク質は、上記で記載の融合タンパク質のいずれかであることができる。
多くのバチルス・セレウス(Bacillus cereus)科構成員株が。固有の有益な特質を有する。例えば、一部の株は、植物成長促進効果を有する。本明細書に記載の融合タンパク質のいずれも、そのような株で発現させることができる。
本明細書に記載の組換えバチルス・セレウス(Bacillus cereus)科構成員のいずれにおいても、前記融合タンパク質は、標的配列、外膜タンパク質、または融合タンパク質の外膜タンパク質断片由来のプロモーターの制御下に発現することができる。例えば、融合タンパク質がB.アントラシス(B. anthracis)スターン(Sterne)BclA(例えば、配列番号1のアミノ酸20から35、配列番号1のアミノ酸1から35、配列番号1、または配列番号60)由来の標的配列を含み、または融合タンパク質が全長BclA(配列番号2)また全長BclAの断片(例えば、配列番号59)を含む場合、融合タンパク質はB.アントラシス(B. anthracis)スターン(Sterne)(例えば、配列番号85のプロモーター)のゲノム中のBclA遺伝子と通常関連しているプロモーターの制御下に発現することができる。
概して、「殺菌性」とは、物質が真菌の死亡率を高め、増殖速度を阻害する能力を意味する。
本発明によれば、前記組成物は、相乗的に有効な量で、a)(i)植物成長刺激性化合物の産生もしくは活性化に関与する酵素;細菌、真菌もしくは植物の栄養源を分解もしくは改変する酵素;および病原体から植物を保護するタンパク質もしくはペプチドからなる群から選択される少なくとも一つの植物成長刺激タンパク質もしくはペプチド;および(ii)融合タンパク質をバチルス細胞の外膜に局在化させる標的配列を含む融合タンパク質を発現する組換え外膜産生性バチルス細胞;およびb)本明細書に開示の少なくとも一つの特定の殺菌剤を含む。
本発明の1態様は、増量剤、溶媒、自発性促進剤、担体、乳化剤、分散剤、凍結保護剤、増粘剤およびアジュバントからなる群から選択される少なくとも一つの補助剤をさらに含む上記の組成物を提供することにある。それらの組成物は、製剤と称される。

実施例1:複数の有効成分の組み合わせの効力についての式
有効成分組み合わせの活性が、個別に施用した場合の有効成分の活性の合計を超える場合には、有効成分の相乗効果が存在する。2種類の有効成分の所定の組み合わせについての予想される活性は、下記のように計算することができる(Colby, S. R., ″Calculating Synergistic and Antagonistic Responses of Herbicide Combinations″,Weeds 1967, 15, 20−22参照)。

トリフロキシストロビンおよびエンドグルカナーゼを発現する組換えバチルス・ツリンギエンシス(Bacillus thuringiensis)細胞(「BEE」)の発酵産物の組み合わせの効力を分析するため、実験を行った。25から28℃および湿度50%の植物成長室のライトラック上、小さい3インチ正方形ポット中の合成培地および砂の無菌混合物で、トウモロコシ種子を約14日間にわたり成長させた。各ポットでは、2個の種子を植えた。植え付け時には、各ポットの成長培地を、下記に記載の処理液に浸した。14日後、植物を、植物バイオマスについて測定した。下記の表において、UTCは未処理対照を指す。「計算値」は、上記コルビー式を用いて計算される予想効果値を指し、「効力」は観察された実際の効果を指す。
20℃および湿度70%の温室中、ローム性砂でトウモロコシ種子を約11日間成長させる。処理時点から約11日後、若芽を土から上で切り取り、新鮮重量を求める。
