Description
(a)以下の重鎖CDR(complementarity determining region)アミノ酸配列を含む重鎖可変領域;配列表の配列番号1のCDRH1、配列表の配列番号2のCDRH2、及び配列表の配列番号3のCDRH3;そして
(b)以下の軽鎖CDRアミノ酸配列を含む軽鎖可変領域;配列表の配列番号4のCDRL1、配列表の配列番号5のCDRL2、及び配列表の配列番号6のCDRL3。
(a)以下の重鎖CDRアミノ酸配列を含む重鎖可変領域;配列表の配列番号7のCDRH1、配列表の配列番号8のCDRH2、及び配列表の配列番号9のCDRH3;そして
(b)以下の軽鎖CDRアミノ酸配列を含む軽鎖可変領域;配列表の配列番号10のCDRL1、配列表の配列番号11のCDRL2、及び配列表の配列番号12のCDRL3。
(a)以下の重鎖CDRアミノ酸配列を含む重鎖可変領域;配列表の配列番号13のCDRH1、配列表の配列番号14のCDRH2、及び配列表の配列番号15のCDRH3;そして
(b)以下の軽鎖CDRアミノ酸配列を含む軽鎖可変領域;配列表の配列番号16のCDRL1、配列表の配列番号17のCDRL2、及び配列表の配列番号18のCDRL3。
(a)本発明はヒトSema3A及びマウスSema3Aに対して交差結合能を有する抗体を提供する。
(b)本発明の抗体はSema3A発現の高い膠芽細胞腫、膵臓癌、肝臓癌などの多様な癌腫でSema3Aを抑制する抗体治療剤に使用可能である。
(c)Sema3Aは糖尿病性網膜症、自己免疫関節炎、神経障害性疼痛、骨多孔症の治療ターゲットと思われているので、本発明の抗体または抗原結合断片は抗癌治療剤の他にも関連疾患に対する治療剤に使われることができる。
(d)本発明の抗体は高い抗Sema3A結合及びこれに従うSema3A機能抑制によって多様な癌腫由来癌細胞の成長を抑制し、Sema3Aの下位信号伝達物質のうち、ERKリン酸化を抑制して癌細胞の移動を抑制することによって、癌の予防及び治療に非常に有効である。
イソロイシン(+4.5);バリン(+4.2);ロイシン(+3.8);フェニルアラニン(+2.8);システイン/シスタイン(+2.5);メチオニン(+1.9);アラニン(+1.8);グリシン(−0.4);スレオニン(−0.7);セリン(−0.8);トリプトファン(−0.9);チロシン(−1.3);プロリン(−1.6);ヒスチジン(−3.2);グルタミン酸(−3.5);グルタミン(−3.5);アスパラギン酸(−3.5);アスパラギン(−3.5);リシン(−3.9);及びアルギニン(−4.5)。
アルギニン(+3.0);リシン(+3.0);アスパラギン酸(+3.0±1);グルタミン酸(+3.0±1);セリン(+0.3);アスパラギン(+0.2);グルタミン(+0.2);グリシン(0);スレオニン(−0.4);プロリン(−0.5±1);アラニン(−0.5);ヒスチジン(−0.5);システイン(−1.0);メチオニン(−1.3);バリン(−1.5);ロイシン(−1.8);イソロイシン(−1.8);チロシン(−2.3);フェニルアラニン(−2.5);トリプトファン(−3.4)。
既存に製作された合成scFv抗体断片ファージライブラリー(Yang et al., Mol. Cells. 27:225− 235, 2009)を用いてヒトSema3Aに結合するscFv抗体断片をファージディスプレイスクリーニングを通じて同定した。ファージディスプレイスクリーニング過程は、図1と同一である。
4回目のパニングで回収されたファージ粒子は宿主細胞ER2537に感染を通じて培養培地でコロニーと確認された。このコロニーを取って200μl SB/アンピシリン培養培地が加えられて96ウェルプレートに接種後、37℃、2−3時間培養した。
ファージミドの基本構成は図6で確認できるが、前記の過程で使われた宿主細胞ER2537の場合、phage pIIIの前に位置した転写抑制コドン(amber codon(UAG))を抑制するので、scFv単独発現が不可能である。
Sema3A蛋白質に対する結合能をELISAで確認した後、実際の細胞が分泌するSema3Aに対する抗癌能力を検証するために細胞増殖分析法と細胞移動分析法を用いた。
抗−Sema3A抗体断片をIgG形態に形態転換のためにSema3A scFvの重鎖配列と軽鎖配列の遺伝子をExpi 293F発現システム(life technologies)を用いて形質注入した。
先に抗Sema3A scFvの癌細胞移動抑制を確認したように、IgG形態に転換された3種のSema3A抗体(A08、C10、F11)の癌細胞移動抑制能を再検証した。Sema3Aを過分泌するU87−MG、131、83細胞と2μg/mlの抗Sema3A抗体を用いて細胞移動法を遂行した。細胞移動法は図11から図13のような方法により遂行し、先に記述されている。
Sema3Aが膠芽細胞腫細胞成長に関与するのかを知るために、組換えヒトSema3Aを131、83細胞に処理した後、細胞成長変化を調べた。Eduを用いた細胞増殖アッセイ(proliferation assay)を進行した結果、各々20%、15%の細胞成長が増加することを確認した(図22)。
In vivoで抗−Sema3A F11の抗癌効能を確認するためにSema3Aを過分泌する膠芽細胞腫131細胞を用いた異種移植モデルを作製した。
Claims (11)
- 以下を含むヒトSema3Aに対する抗体またはその抗原結合断片:
(a)配列表の配列番号19のアミノ酸配列を含む重鎖可変領域;そして
(b)配列表の配列番号20のアミノ酸配列を含む軽鎖可変領域。 - 以下を含むヒトSema3Aに対する抗体またはその抗原結合断片:
(a)配列表の配列番号21のアミノ酸配列を含む重鎖可変領域;そして
(b)配列表の配列番号22のアミノ酸配列を含む軽鎖可変領域。 - 以下を含むヒトSema3Aに対する抗体またはその抗原結合断片:
(a)配列表の配列番号23のアミノ酸配列を含む重鎖可変領域;そして
(b)配列表の配列番号24のアミノ酸配列を含む軽鎖可変領域。 - 配列表の配列番号19のアミノ酸配列を含むヒトSema3A及びマウスSema3Aに交差結合する抗体の重鎖可変領域をコードする核酸分子と、
配列表の配列番号20のアミノ酸配列を含むヒトSema3A及びマウスSema3Aに交差結合する抗体の軽鎖可変領域をコードする核酸分子とを含む、組換えベクター。 - 配列表の配列番号21のアミノ酸配列を含むヒトSema3A及びマウスSema3Aに交差結合する抗体の重鎖可変領域をコードする核酸分子と、
配列表の配列番号22のアミノ酸配列を含むヒトSema3A及びマウスSema3Aに交差結合する抗体の軽鎖可変領域をコードする核酸分子とを含む、組換えベクター。 - 配列表の配列番号23のアミノ酸配列を含むヒトSema3A及びマウスSema3Aに交差結合する抗体の重鎖可変領域をコードする核酸分子と、
配列表の配列番号24のアミノ酸配列を含むヒトSema3A及びマウスSema3Aに交差結合する抗体の軽鎖可変領域をコードする核酸分子とを含む、組換えベクター。 - 請求項4から6のいずれかに記載の組換えベクターで形質転換された、宿主細胞。
- (a)請求項1から3のいずれかに記載のヒトSema3Aに対する抗体またはその抗原結合断片の薬剤学的有効量;及び(b)薬剤学的に許容される担体を含む癌の予防及び治療用薬剤学的組成物。
- 前記癌は、乳癌、大腸癌、肺癌、胃癌、肝臓癌、血液癌、骨癌、膵臓癌、皮膚癌、脳癌、子宮癌、鼻咽頭癌、喉頭癌、結腸癌、卵巣癌、直腸癌、膣癌、小腸癌、内分泌癌、甲状腺癌、副甲状腺癌、尿管癌、尿道癌、前立腺癌、気管支癌、膀胱癌、腎臓癌、または骨髄癌であることを特徴とする、請求項8に記載の薬剤学的組成物。
- 癌の予防又は治療用医薬品の製造における請求項1から3のいずれかに記載のヒトSema3Aに対する抗体またはその抗原結合断片の使用。
- 前記癌は、乳癌、大腸癌、肺癌、胃癌、肝臓癌、血液癌、骨癌、膵臓癌、皮膚癌、脳癌、子宮癌、鼻咽頭癌、喉頭癌、結腸癌、卵巣癌、直腸癌、膣癌、小腸癌、内分泌癌、甲状腺癌、副甲状腺癌、尿管癌、尿道癌、前立腺癌、気管支癌、膀胱癌、腎臓癌、または骨髄癌であることを特徴とする、請求項10に記載の使用。
