Description
a)7〜10の範囲にあるpHを有し、かつカルシウムの水溶液、リン酸塩(phosphate)の水溶液およびクエン酸イオンの溶液を含有する混合物を20℃〜40℃の範囲の温度で30秒間〜10分間の範囲の時間にわたって維持する工程と、
b)工程a)の得られた溶液から未反応のイオンを除去し、それによって1つ以上のリン酸カルシウムのナノ粒子(CaP−NP)の懸濁液を得る工程と、
c)工程b)の懸濁液から1つ以上のリン酸カルシウムのナノ粒子(CaP−NP)の生成物を回収する工程とを含む。
− 「治療用/診断用化合物(複数可)」は、目標の器官に対して放出するための1種以上の医療用化合物、例えば、核酸、ペプチド、合成化合物、または診断プローブを意味する。
− 150〜231nmの範囲にある「Z平均」は、動的光散乱(Dynamic Light Scattering:DLS)によって測定された平均流体力学的径を意味する。
− 「−27.0mV〜−41.0mVの範囲にあるζ電位を有する負に帯電したCaP−NP」とは、電気泳動光散乱(Electrophoretic Light Scattering:ELS)によって測定されたζ電位を有するCaP−NPを意味する。
− 「分裂因子(SF)」は、ウェイナー エス.(Weiner S.)およびバール−ヨセフ オー.(Bar−Yosef O.)(1990年)、「States of preservation of bones from prehistoric sites in the Near East: a survey」、Journal of Archaeological Science、第17巻、p.187−196に従ってNPのFT−IRスペクトルから計算される結晶化度を測るものである。
a)7〜10の範囲にあるpHを有し、かつカルシウムの水溶液、リン酸塩の水溶液およびクエン酸イオンの溶液を含有する混合物を20℃〜40℃の範囲の温度で30秒間〜10分間の範囲の時間にわたって維持する工程と、
b)工程a)の得られた溶液から未反応のイオンを除去し、それによって1つ以上のリン酸カルシウムのナノ粒子(CaP−NP)の懸濁液を得る工程と、
c)工程b)の懸濁液から1つ以上のリン酸カルシウムのナノ粒子(CaP−NP)の生成物を回収する工程とを含む。
実施例1A. 本発明のリン酸カルシウムナノ粒子(CaP−NP)の調製。
12.5体積のCaCl2(10〜50mM)およびNa3(C6H5O7)(40〜200mM)の溶液、1体積のNaOH(0.1〜0.5M)の溶液、および12.5体積のNa2HPO4(12〜60mM)の溶液を含有する溶液を調製し、次いで37℃の水浴中に5分間入れた。
工程a)におけるクエン酸イオンの存在の重要性を評価するために、CaP−NPの寸法およびコロイド安定性に対するクエン酸塩の効果をDLSによって予め評価した。
結晶化の間に未反応のイオンを除去するために、実施例1Aにおいて示したように透析を用いた。
実施例1Aから得られたNPの細胞内移行を評価するために、NPをイソチオシアン酸フルオレセイン(FITC)で標識した。
マイクロRNAの封入を伴うCaP−NPの調製は、以下に記載するようにマイクロRNA化合物の含有によって行われる実施例1Aに記載したような調製プロトコルに詳細に従って実施した。マイクロRNA miR−133に対応するマイクロRNAは、合成ヌクレオチド配列(ドイツ国IBAによって合成)である。
CaP−NP−miR粒子の特性分析を下記の表3に示す。
CaP−NP−miRに封入されたmiR−133の正確な量を評価するために、CaP−NP−miRから抽出された全核酸に対して定量的PCR(qPCR)測定を実施した。実施例3に記載したような、合成中に用いられる異なる濃度(2μg、25μgおよび50μg)のmiR−133を用いたCaP−NP−miRの3つの調製物から、500μLのCaP−NP−miRの溶液をPurezol試薬(プロメガ(Promega))によるRNAの抽出に用いた。次いでCaP−NP−miRの各調製物について抽出した合計40ngのRNAをuniversal cDNA Synthesis II kit(エキシコン(Exiqon))を用いて逆転写した。次に、逆転写反応の1/40を後続のmiR−133に特異的なqPCRに用いた。前記qPCRは、Vila(商標)7 Real−Time PCR System(アプライド バイオシステム(Applied Biosystem))上でSYBR(登録商標) Select Master Mix(インビトロジェン(Invitrogen))を用いて、3回繰り返して実施した。次に、既知量のmiR−133の合成オリゴから導出されるcDNAの連続希釈法を用いた絶対的定量化法を用いて、miR−133の正確な量を求めた(40フェムトモルの出発点から1:10希釈)。CaP−NPに結合したmiR−133の量を、線形標準曲線において導出されるCtをトレースすることにより推定した。CaP−NPに結合したmiR−133aの量を表4に示す。結果は、反応中に用いられるマイクロRNAの50%超がCaP−NP内に封入されたことを示している。
実施例1Aで得られたCaP−NPを生体適合性および毒性について、心臓の細胞系統HL−1を増大する投与量(0〜500ug/mL)のCaP−NPに曝露して、インビトロで試験した。第1工程として、死細胞のみを選択的にマークすることができる染料であるトリパンブルーによる除外アッセイによって、細胞毒性の評価を実施した。図6Aに示すように、HL−1系統は試験した量の投与に耐性を有したが、高投与量(>125ug/mL)のみにおいて細胞の死亡率の増大が観察された。続いて、CaP−NPの投与後に誘発される任意のアポトーシス反応の評価にカスパーゼ−3−7アッセイを用いた。図6Bに示すように、アポトーシス活性に関する有意差は、≧250ug/mLの高濃度のCaP−NPにおいてのみ観察された。従って、双方のアッセイは、CaP−NPを心筋に対する目的の化合物の内部移行のための可能な担体として選択した。
CaP−NPの細胞内移行を評価するために、HL−1細胞を20μg/mLの濃度の実施例2のCaP−NP−FITCに曝露し、投与後24時間に共焦点顕微鏡分析を実施した。図6Cに示すように、別々の細胞内小胞区画におけるCaP−NP−FITCの明確な内部移行が得られた。この内部移行が実際に活性エンドサイトーシス機構によるものであったかを判定するために、原形質膜の陥入およびエンドサイトーシスに関わるタンパク質であるクラスリンまたはダイナミンに特異的な阻害剤を用いた。従って、HL−1細胞をこれらの阻害剤で前処置し、次いで増大する濃度のCaP−NPに曝露した。図6Aに示すように、クラスリン/ダイナミン媒介エンドサイトーシス阻害は、すべての投与量においてCaP−NPによって誘発される(トリパンブルーによる除外アッセイにおける)細胞毒性を著しく低減し、よって、CaP−NPの細胞吸収の阻害を反映する。
心筋細胞のような興奮性細胞上でのカルシウムに基づいたナノ粒子の使用における主な問題の1つは、電気生理学的特性に対する潜在的影響である。従って、2つの細胞タイプ(HL−1および成体マウス心室心筋細胞)の生物物理学的特性について検討した。最初に、20μg/mLの実施例1AのCaP−NPの短期投与後および長期投与後のHL−1細胞における活動電位(AP)の特性を分析した。具体的には、投与後24時間に、HL−1を長期の実験(CaP−NPと共に24時間のインキュベーション)に用い、一方、マウス心筋細胞は短期の実験(CaP−NPと共に4時間のインキュベーション)に用いた。長期条件および短期条件の双方において、全細胞構成におけるパッチクランプ法によって実施した電気生理学的実験は、休止細胞の生物物理学的特性に関して、および閾値を超える電気刺激(図7〜図11)後に得られる活動電位特性について、処置した試料および対照試料の双方の間において有意差を示さなかった。よって、これらの細胞の電気的表現型を決定し、それらに興奮性の典型的な特性を与えるパラメータ、例えば、膜電位(Vm)、膜容量(Cm)、膜抵抗(Rm)、活動電位閾値(AP)、AP幅(APA)、AP(dV/dTmax)の最大上昇率およびAP(APD)の持続時間などは、上記の濃度のCaP−NPの投与後に、有意に変更されなかったことが示された。また、典型的な心臓の細胞AP特性に基づいて検討した前記2つの細胞タイプに通常存在する主なイオン電流(カリウム電流、ナトリウム電流およびカルシウム電流)の、またパッチクランプ法による、分析は、対照と、短期条件および長期条件の双方においてCaP−NPによって処置したものとの間において有意差を示さなかった。
miR−133(実施例3で使用)を模倣する合成二本鎖オリゴ(synthetic duplex oligo)を内部に封入したCaP−NPの細胞内移行を評価した。従って、本発明の生成物をmiR−133にコンジュゲートされたCaP−NP(CaP−NP−miR)として、実施例3に従って合成miR−133の溶液を用いて調製した。筋肉に特異的なマイクロRNAであるmiR133は、陰性のβ−アドレナリン作動性受容体調節薬であることが知られていた。
実施例1Aにおいて生成したCaP−NPが、心臓の活動に影響を与えることなく、成功裡に心筋に到達したかを評価するために、インビボの試験を成体ラットにおいて行なった。ロッシ(Rossi)ら、AJP、2008年の文献に記載されているように麻酔した動物に、気管経路によって生理食塩水そのもの(CTL)、または動物の体重1kg当たり3mgの濃度でCaP−NPを含有する生理食塩水を投与した。処置後4時間に、前記動物を電気生理学的分析に供した。心外膜表面に配置した電極装置から得られた電位図を図15に示す。図14および下記の表5に示すように、電位図はP、PQ、QRS、QT、ERP、RRに関して2つの実験群の間に定性的差異を示さなかった。
治療用途の可能性をさらに調査するために、実施例3においてマイクロRNAについて記載したような模倣ペプチド(MP)の含有によって行われる実施例1Aにおけるように、CaP−NPを生成した。MPは、新規なクラスの陽性変力物質のうちに入る短い9aaペプチド(米国ジーンスクリプト(Genescript)により合成)である。従来にない機構によって作用すること(すなわち、カルシウム依存性の収縮期収縮をもたらすトリガ要素であり、かつチャネルゲート特性を変化させることがない、電圧依存性L型カルシウムチャネルの細胞表面密度の正常化)により、MPは、LTCC密度、従って心筋収縮能がダウンレギュレートされる心機能障害(すなわち糖尿病性心筋症、DM)の状態において心拍の力を回復する。DMを誘発するために、膵臓のインシュリンを生成するβ細胞に有害な化合物であるストレプトゾトチン(STZ)をマウスに注射した。興味深いことに、MP−CaP−NPによるDMマウスの10日間の吸入治療により、心機能は完全に回復し、一方、MP単体またはスクランブル−CaP−NP(CaP−NP−HA)を投与した場合には効果が得られなかった(図16A)。加えて、同一の治療されたマウスから分離した心筋細胞の機能分析により、MP−CaP−NPがカルシウム電流(Ica)を回復することが明らかとなった(図16B)。
Claims (17)
- −41.0mV〜−27.0mVの範囲にあるζ電位を有する負の表面電荷を備えた1つ以上のリン酸カルシウムのナノ粒子(CaP−NP)を含有する生成物の調製方法であって、該方法は、
a)7〜10の範囲にあるpHを有し、かつカルシウムの水溶液、リン酸塩の水溶液、およびクエン酸イオンの溶液を含有する混合物を20℃〜40℃の範囲の温度で30秒間〜10分間の範囲の時間にわたって維持する工程と、
b)工程a)の得られた溶液から未反応のイオンを除去し、それによって1つ以上のリン酸カルシウムのナノ粒子(CaP−NP)の懸濁液を得る工程と、
c)工程b)の懸濁液から1つ以上のリン酸カルシウムのナノ粒子(CaP−NP)の生成物を回収する工程とを含む、方法。 - 工程a)の混合物中に、1種以上の治療用/診断用化合物の水溶液も存在する、請求項1に記載の方法。
- 工程c)から回収された前記1つ以上のリン酸カルシウムのナノ粒子(CaP−NP)は、1種以上の治療用/診断用化合物によって表面機能化されている、請求項1に記載の方法。
- 工程a)の混合物中のカルシウムの水溶液は、20〜200mMの範囲のモル濃度を有する塩化カルシウムの溶液である、請求項1乃至3のいずれか一項に記載の方法。
- 工程a)の混合物中のリン酸塩の水溶液は、24〜240mMの範囲のモル濃度を有するNa2HPO4の溶液である、請求項1乃至4のいずれか一項に記載の方法。
- 工程a)の前記温度は35〜40℃の範囲にある、請求項1乃至5のいずれか一項に記載の方法。
- 工程a)の混合物の維持時間は約5分間である、請求項1乃至6のいずれか一項に記載の方法。
- 前記クエン酸イオンの溶液は、40〜800mMの範囲のモル濃度を有するクエン酸ナトリウムの水溶液である、請求項1乃至7のいずれか一項に記載の方法。
- 工程a)の混合物は約10のpHを有する、請求項1乃至8のいずれか一項に記載の方法。
- 未反応のイオンを除去する工程b)は、透析膜によって実施される、請求項1乃至9のいずれか一項に記載の方法。
- 前記透析膜は、3500ダルトンのカットオフを有するセルロース膜である、請求項10に記載の方法。
- 透析膜によって実施される除去工程b)は、5〜24時間にわたって行われる、請求項11に記載の方法。
- 前記リン酸カルシウムのナノ粒子(CaP−NP)の回収工程c)は、凍結乾燥によって実施される、請求項1乃至12のいずれか一項に記載の方法。
- 1つ以上のリン酸カルシウムのナノ粒子(CaP−NP)を含有する生成物であって、前記CaP−NPは、−41.0mV〜−27.0mVの範囲にあるζ電位を有する負の表面電荷と、多くとも1.76の分裂因子(SF)とを有するとともに、核酸、ペプチド、合成化合物および診断プローブからなる群から選択される1種以上の表面機能化された治療用/診断用化合物を含有する、生成物。
- 1つ以上のリン酸カルシウムのナノ粒子(CaP−NP)を含有する生成物であって、前記CaP−NPは、1種以上の治療用/診断用化合物を封入し、−41.0mV〜−27.0mVの範囲にあるζ電位と、150〜231nmの範囲にある平均流体力学的径(average mean hydrodynamic diameter)(Z平均として測定)とを有する負の表面電荷を有するとともに、核酸、ペプチド、合成化合物および診断プローブからなる群から選択される1種以上の表面機能化された治療用/診断用化合物を含有する、生成物。
- 請求項14又は15に記載の生成物の、循環器疾患の治療用薬剤を製造するための使用。
- 前記治療は、吸入投与、経腸投与、非経口投与、静脈内投与、腹腔内投与、経口投与、舌下投与、噴霧投与、直腸内投与、眼内投与、局所投与および経皮的投与のうちから選択される投与経路を通じて実施される、請求項16に記載の使用。
