Description
〔1〕 交流電流を用いて発生させたプラズマと、水蒸気を含む含水気体との反応物である活性酸素種含有気体を照射する活性酸素種照射ユニットを含んでなる殺菌装置であって、50〜300℃に加熱した前記含水気体を供給する水蒸気供給ユニットを更に含み、ここで、前記活性酸素種含有気体が飽和水蒸気量以上の水分を含むことを特徴とする、殺菌装置。
〔2〕 交流電流を用いて発生させたプラズマと、水蒸気を含む50〜300℃に加熱した含水気体との反応物である、飽和水蒸気量以上の水分を含む活性酸素種含有気体を照射することを含むことを特徴とする、殺菌方法。
本発明の殺菌装置における、含水気体の供給温度の検討を行った。
芽胞菌Bacillus atrophaeusの菌液を用いて、各種濃度(2×105〜2×107CFU/mL濃度範囲において3水準)の菌液を調製した。得られた菌液を図2に示すように、樹脂キャップ(素材ポリエチレン)1個あたり1μL×9spotで菌付けを行った(各濃度n=5)。なお、菌付けした樹脂キャップは24時間滅菌シャーレ内に静置して乾燥したものを用いた。
図1に示す本発明の殺菌装置を用いて、活性酸素種含有気体を菌付けした樹脂キャップの上方30mmの距離から1個あたり0.5秒間を2回(合計1秒間)照射し、照射後のキャップは滅菌シャーレに回収した。なお、本発明の殺菌装置の使用条件は次の通りであった。
(殺菌装置の使用条件)
交流電流のジェネレータユニット1:周波数14kHz、電圧300V、電流11A
変圧器ユニット2:昇圧後の電圧20kV
コントロールパネル3:空気供給量6L/min、酸素供給量3L/min(以上、ノズルユニット4へ)、空気供給量3L/min(蒸発器ユニット6へ)
ノズルユニット4:活性酸素種含有気体照射温度は表1に示す通り、照射スピード50000mm/sec
冷却ユニット5:冷却水5℃
蒸発器ユニット6:電熱線300℃、含水気体供給量6.7L/min(プラズマ噴流/含水気体供給量(体積比)=9/6.7)
ヒーター6a:配管温度は表1に示す通り
ポンプユニット7:水供給量は表1に示す通り
照射台8:表面温度25℃
活性酸素種含有気体の照射を行った樹脂キャップを滅菌シャーレから取出し、5mLのTSA液体培地(BD Falcon社製)を注入し、微生物の増殖に好適な35℃で3日間培養した。培養後、微生物増殖により培地が濁った滅菌シャーレ個数を陽性としてボトルキャップ数としてカウントし、最確数法(MPN法)により殺菌活性値LRV(Log Reduction Value)を算出した。結果を表1に示す。なお、殺菌活性を示す「D」値とは、キャップ1個当たりの菌数を常用対数で表し(LOG値)、処理前の菌数(LOG値)から処理後の菌数(LOG値)を減算した値のことであり、数が大きい程殺菌活性が高く、4.5D以上であれば食品容器の殺菌処理として問題ないことを示す。

Claims (5)
- 交流電流を用いて発生させたプラズマと、水蒸気を含む含水気体との反応物である活性酸素種含有気体を照射する活性酸素種照射ユニットを含んでなる殺菌装置であって、100〜300℃に加熱した前記含水気体を供給する水蒸気供給ユニットを更に含み、ここで、前記活性酸素種含有気体が飽和水蒸気量以上の水分を含み、前記活性酸素種含有気体の温度が55〜80℃である、殺菌装置。
- 更に、殺菌対象物を50℃以下にて載置する照射台を含んでなる、請求項1記載の殺菌装置。
- 交流電流を用いて発生させたプラズマと、水蒸気を含む100〜300℃に加熱した含水気体との反応物である、飽和水蒸気量以上の水分を含む活性酸素種含有気体を照射する、殺菌方法であって、前記活性酸素種含有気体の温度が55〜80℃である、殺菌方法。
- 殺菌対象物と活性酸素種含有気体の温度差が10℃以上である、請求項3記載の殺菌方法。
- 請求項1又は2に記載の殺菌装置を用いる、請求項3又は4記載の殺菌方法。
