Description
本発明は、メルティング分析を用いてターゲット核酸配列を定量する方法に関する。
(a)一連の反応を繰り返すサイクルを通じて核酸試料内のターゲット核酸配列を増幅させて、ラベリングモイエティ(labeling moiety)を含む二量体(duplex)を形成させる段階であって;前記二量体は、二本鎖形態の前記増幅されたターゲット核酸配列、前記ターゲット核酸配列とプローブとの間のハイブリダイゼーションによって形成された二量体、または前記ターゲット核酸配列の存在に依存的に形成された二量体を含み;前記二量体の形成は、前記ターゲット核酸配列の増幅に比例して増加し;前記ラベリングモイエティは、前記二量体の結合(association)または解離(dissociation)過程の間に検出可能なシグナルを発生させ;
(b)前記段階(a)の繰り返し過程の間に、前記二量体から検出可能なシグナルが検出される温度範囲でのメルティング分析を既定の(predetermined)サイクルで実施することによって、少なくとも2つの既定のサイクルに対するメルティングピークカーブを得る段階;及び
(c)前記メルティングピークカーブを用いて、前記ターゲット核酸配列を定量する段階。
(a)本発明は、核酸増幅前にメルティングピーク分析を実施する核酸増幅内の少なくとも2つのサイクルをあらかじめ決定し、少なくとも2つの既定のサイクルでメルティングピーク分析を実施し、引き続き、メルティングピークカーブからのデータ値(例えば、存在または不存在、高さ及び面積)を用いてターゲット核酸配列を定量する方法で実施される。
従来技術のうち、ターゲット増幅後、一回のメルティング分析を実施する定量方法は、定量結果がターゲット増幅のサイクルによって変わりうるという深刻な問題を有している。本発明は、ターゲット増幅過程の間に少なくとも2つの既定のサイクルでメルティングピーク分析を実施するために、従来の方法よりもさらに正確かつ意味のある定量結果を提供することができる。
米国特許第8,039,215号に開示された方法の場合、閾値メルティングピーク最大値が出たサイクルを確認するために、ターゲット増幅の各サイクルごとにメルティング分析を実施し、これは、遥かに長い分析時間を招く。本発明は、既定のサイクルでのみメルティングピーク分析を実施するために、さらに迅速かつ便利にターゲット核酸配列を定量することができる。
(b)米国特許第8,039,215号に開示された方法の場合、特定サイクルでメルティングピーク最大値が閾値よりも大きい場合、これら間の差をターゲット核酸配列の定量に利用しない。一方、既定のサイクルでのメルティングピークのあらゆる高さまたは面積値をターゲット核酸配列の定量に利用できる。
(c)本発明は、二量体のTm値を利用するメルティングピーク分析を基盤とするので、一種の標識を利用する場合にも、複数のターゲット核酸配列を同時に検出及び定量することができる。
(d)本発明は、ターゲット核酸配列の相対的定量を可能にする。また、本発明は、既知濃度を有する対照群を用いて絶対的方式でターゲット核酸配列を定量することができる。
段階(a):ラベリングモイエティを含む二量体の形成
本発明によれば、ターゲット核酸配列は、増幅されてラベリングモイエティを含む二量体を形成する。
引き続き、ターゲット核酸配列の増幅の間に、少なくとも2つの既定のサイクルで二量体からのシグナルを一定範囲の温度で検出する(メルティング分析)。シグナル検出は、一定範囲の温度で段階(a)から形成された二量体のメルティングによって発生するシグナル検出、及び一定範囲の温度で段階(a)から形成された二量体のメルティング後、ハイブリダイズによって発生するシグナル検出を含む。
次いで、メルティングピークカーブを用いてターゲット核酸配列の定量を実施する。
本発明者らは、得られたあらゆるメルティングピークを用いてサイクリックメルティングカーブ(cyclic melting curve)分析を行う場合、ターゲット核酸配列の定量が可能であるか否かを確認しようとした。ターゲット核酸配列の定量のために、メルティング分析を含むPTOCE分析を利用した。

本発明者らは、最初のメルティングピークを用いてサイクリックメルティングカーブ分析を行う場合、ターゲット核酸配列の定量が可能であるか否かを確認しようとした。ターゲット核酸配列の定量のために、メルティング分析を含むPTOCE分析を利用した。
Claims (5)
- 次の段階を含むメルティングピークカーブを用いて核酸試料内のターゲット核酸配列を定量する方法であって、:
(a)一連の反応を繰り返すサイクルを通じて核酸試料内のターゲット核酸配列を増幅させて、標識を含む二量体を形成させる段階であって;前記二量体は、二本鎖形態の前記増幅されたターゲット核酸配列、前記ターゲット核酸配列とプローブとの間のハイブリダイゼーションによって形成された二量体、または前記ターゲット核酸配列の存在に依存的に形成された二量体を含み;前記二量体の形成は、前記ターゲット核酸配列の増幅に比例して増加し;前記標識は、前記二量体の結合または解離過程の間に検出可能なシグナルを発生させ;
(b)前記段階(a)の繰り返し過程の間に、前記二量体から検出可能なシグナルが検出される温度範囲でのメルティング分析を既定のサイクルで実施することによって、2−5つの既定のサイクルに対するメルティングピークカーブを得る段階であって;前記既定のサイクル間の間隔は少なくとも5サイクルであり;及び
(c)前記メルティングピークカーブを用いて、前記ターゲット核酸配列を定量する段階であって;前記段階(c)が、下記(c−1)、(c−2)及び(c−3)のいずれかの工程により行われ、:
(c−1)前記既定のサイクル別に基準数値を付与し、前記メルティングピークカーブが出た既定のサイクルの基準数値を合算することにより定量数値を計算して、初期ターゲット核酸配列の量を定量する工程であって、前記基準数値は、規定のサイクルに昇順または降順で付与されるか、または各サイクルに同じ数値の基準数値が付与される工程、
(c−2)前記既定のサイクル別に基準数値を付与し、最初のメルティングピークカーブが出た既定のサイクルの基準数値を用いて定量数値を計算して、初期ターゲット核酸配列の量を定量する工程であって、前記基準数値は、規定のサイクルに昇順または降順で付与され、ターゲット核酸配列の量が多くなるほど、より早いサイクルでメルティングピークカーブが得られる工程、
(c−3)既定のサイクル別に基準数値を付与し、(i)前記最初のメルティングピークカーブが出た既定のサイクルの基準数値、及び(ii)最初のメルティングピークカーブの高さの最大値または最大面積値を用いて定量数値を計算して、初期ターゲット核酸配列の量を定量する工程であって、前記基準数値は、規定のサイクルに昇順または降順で付与され、ターゲット核酸配列の量が多くなるほど、より早いサイクルでメルティングピークカーブが得られ、最初のメルティングピークカーブが出た既定のサイクルから計算された前記定量数値の最小値が、前記既定のサイクル直後のサイクルで最初のメルティングピークカーブが出る場合に、計算された定量数値の最大値よりも大きいか、同じ数値である工程、
前記ターゲット核酸配列の定量は、他の核酸試料または既知核酸濃度を有する対照群(control)試料を前記段階(a)〜(c)に適用して得た定量数値と比較することで実施される方法。 - 前記二量体の標識は、前記二量体のうち何れか一方の鎖に連結された単一標識、前記二量体のうち何れか一方の鎖にいずれも連結されたレポーター分子及びクエンチャー分子を含む相互作用的二重標識、前記二量体の一方の鎖に連結されたレポーター分子及び他方の鎖に連結されたクエンチャー分子を含む相互作用的二重標識、または前記二量体に挿入されるインターカレーティング染料であることを特徴とする請求項1に記載の方法。
- 前記ターゲット核酸配列の増幅は、温度が変化する条件下で、増幅用プライマーと前記ターゲット核酸配列との間のハイブリダイゼーション、前記プライマーの延長及び延長鎖の解離を含む一連の反応を繰り返すことで達成され、前記サイクルは、前記一連の反応の1回繰り返しを1つの単位として有することを特徴とする請求項1に記載の方法。
- 前記ターゲット核酸配列の増幅は、等温条件下に一連の反応を繰り返すことで達成され、前記サイクルは、一定時間間隔で実施される前記一連の反応の繰り返しを1つの単位として有することを特徴とする請求項1に記載の方法。
- 前記ターゲット核酸配列の増幅は、PCR(polymerase chain reaction)、LCR(ligase chain reaction)、GLCR(gap filling LCR)、Q−beta(Q−beta replicase amplification)、SDA(stand displacement amplification)、3SR(self−sustained sequence replication)、NASBA(nucleic acid sequence−based amplification)、TMA(Transcription−Mediated Amplification)、またはRCA(Rolling Circle Amplification)によって実施されることを特徴とする請求項1に記載の方法。
