Description
Claims (24)
- a)再利用可能な液体試薬を試薬リザーバからキャッシュリザーバへ吸い上げるステップであって、該キャッシュリザーバは、前記試薬リザーバおよびフローセルの少なくとも1つのチャネルと流体連通している、吸い上げるステップと、
b)前記試薬を前記キャッシュリザーバから前記フローセルの少なくとも1つのチャネル上へ輸送するステップと、
c)前記フローセルのチャネル上の前記試薬の少なくとも30%、40%、50%、60%、70%、80%、90%、または100%を前記キャッシュリザーバへ輸送するステップであって、前記フローセルからの前記液体試薬は、前記フローセルと接触後、前記試薬リザーバへ戻らない、輸送するステップと、
d)ステップb)とステップc)とを繰り返して前記フローセル上の前記液体試薬の再利用を達成するステップと、を含む試薬を再利用する方法。 - 1つ以上の前記キャッシュリザーバは、ポンプと流体連通しており、該ポンプは、前記キャッシュリザーバからフローセルへ液体試薬を移動させ、かつ該フローセルから該キャッシュリザーバへ液体試薬を戻すよう構成され、前記フローセルへの試薬の進入および前記フローセルからの試薬の退出は、前記フローセルの同じポートを通して行われる、請求項1に記載の方法。
- ステップ(c)において前記キャッシュリザーバへ輸送されなかった、前記フローセルからの試薬は、廃棄物リザーバへ輸送される、請求項2に記載の方法。
- ステップb)およびステップc)のうちの一方または両方の輸送するステップは、前記キャッシュリザーバと前記フローセルとを流体接続するバルブを介して行われる、請求項1に記載の方法。
- ステップb)およびステップc)のうちの一方または両方の輸送するステップは、単一方向への流体の流動により行われる、請求項1に記載の方法。
- ステップb)およびステップc)のうちの一方または両方の輸送するステップは、相互流動により行われる、請求項1に記載の方法。
- 前記液体試薬は、塩基配列決定を行うための試薬を含む、請求項1に記載の方法。
- 前記試薬は、ポリメラーゼを含む、請求項7に記載の方法。
- 前記試薬は、ヌクレオチドまたは種々のヌクレオチドの混合物を含む、請求項7に記載の方法。
- 一種または複数種の試薬を前記フローセル上に流した後でステップd)を行う、請求項1に記載の方法。
- ステップc)において前記キャッシュリザーバに戻った試薬は、該キャッシュリザーバ内の類似の試薬と混ざり、それにより、ステップb)とステップc)とを複数回繰り返すことから類似の試薬を含む混合試薬が形成される、請求項1に記載の方法。
- 前記キャッシュリザーバに戻った試薬と新しい試薬との混合は、前記キャッシュリザーバ内では最小限に抑えられる、請求項1に記載の方法。
- 試薬カートリッジからフローセルへ試薬を送達するための流体システムであって、
試薬カートリッジとフローセルの注入口との間で流体連通が達成されるよう構成した複数のチャネルを備える試薬マニホールドと、
前記マニホールドの複数のポートから下方へ伸びる複数の試薬シッパーであって、前記各試薬シッパーは、試薬カートリッジの試薬リザーバへ挿入され、前記試薬リザーバから前記シッパーへ液体試薬が吸い上げられるよう構成した、前記複数の試薬シッパーと、
前記リザーバと前記フローセルの注入口との間の流体連通を媒介するよう構成した少なくとも1つのバルブと、を備えた流体システムを用いて行われる、請求項1に記載の方法。 - 拡散接合された多層試薬マニホールドであって、該マニホールドは少なくとも10個、15個、または20個のポートを備え、該各ポートはシッパーを介して別の試薬リザーバから試薬を吸い上げ、前記マニホールド内の流体チャネルを介してフローセルの1つ以上のチャネルと流体連通するよう構成された、試薬マニホールドを用いて行われる、請求項1に記載の方法。
- (i)光透過性表面を具備するフローセル、
(ii)核酸試料、
(iii)配列決定反応のための試薬を保存する試薬リザーバ、および
(iv)前記試薬リザーバをキャッシュリザーバを介して前記フローセルと流体連通させるとともに、前記試薬を前記フローセルに送達する流体ラインシステム
を備えた流体システムを提供するステップ(a)と、
(i)xおよびy次元の画像平面において広視野画像を検出するように構成した対物レンズを具備するミクロ蛍光計、および
(ii)試料ステージ
を備える検出装置を提供するステップ(b)と、
塩基配列決定作業における流体操作を前記流体システム内で実行し、該塩基配列決定作業における検出操作を前記検出装置において実行するステップ(c)であって、
(i)前記流体ラインシステムによって前記試薬が前記フローセルに送達され、
(ii)前記ミクロ蛍光計によって前記核酸のフィーチャーの広視野画像が検出され、
(iii)前記試薬の再利用のために、少なくとも一部の試薬が前記フローセルから前記キャッシュリザーバに取り出される、ステップ(c)と、を含む塩基配列を決定する方法。 - 前記流体システムは、廃棄物リザーバをさらに備え、前記塩基配列決定作業における流体操作は、前記少なくとも一部の試薬を前記フローセルから前記廃棄物リザーバに取り出すことをさらに含む、請求項15に記載の方法。
- 前記キャッシュリザーバは、ポンプと流体連通しており、該ポンプは、前記キャッシュリザーバから前記フローセルへ試薬を移動させ、かつ該フローセルから該キャッシュリザーバへ試薬を戻すよう構成され、前記フローセルへの試薬の進入および前記フローセルからの試薬の退出は、前記フローセルの同じポートを通して行われる、請求項15に記載の方法。
- ステップ(c)(iii)において前記キャッシュリザーバへ輸送されなかった、前記フローセルからの試薬は、廃棄物リザーバへ輸送される、請求項15に記載の方法。
- ステップ(c)(i)またはステップ(c)(iii)における試薬送達は、前記キャッシュリザーバと前記フローセルとを流体接続するバルブを介して行われる、請求項15に記載の方法。
- ステップ(c)(i)またはステップ(c)(iii)における試薬送達は、単一方向への流体の流動により行われる、請求項15に記載の方法。
- ステップ(c)(i)またはステップ(c)(iii)における試薬送達は、相互流動により行われる、請求項15に記載の方法。
- 前記試薬は、塩基配列決定を行うための試薬である、請求項15に記載の方法。
- 前記試薬は、ポリメラーゼである、請求項22に記載の方法。
- 前記試薬は、ヌクレオチドまたは種々のヌクレオチドの混合物である、請求項22に記載の方法。
