本願は、2014年10月16日に出願された米国仮出願第62/064,994号、及び2014年10月17日に出願された米国仮出願第62/065,017号の利益を主張し、それらの全体を参照として本明細書にそれぞれ援用する。
本明細書とともに、コンピュータ可読形式の配列表を電子提出によって提出する。前記配列表は、参照としてその全体を援用するとともに、これを2015年10月13日に作成したファイル名P34223WO00_SEQ_PCT.txt、及び327,235バイトのサイズ(MS‐Windows(登録商標)オペレーティングシステムで測定)のファイルに格納する。
配列番号1は、Cry1Da1タンパク質をコードするヌクレオチド配列である。
本明細書において、他の公知の鱗翅類殺虫タンパク質に比べて、鱗翅目種に対して驚くほど高レベルの毒性活性及び広範な殺虫スペクトルを示す改変殺虫タンパク質を提供する。これらの改変殺虫タンパク質は、様々なアミノ酸改変の鋳型として役立つ殺虫性骨格タンパク質に由来する。そのような殺虫性骨格タンパク質の例としては、Cry1Da1及びTIC844(Cry1Da1相同体)が挙げられるが、必ずしもこれらに限定するものではない。TIC844は、Cry1Da1コアトキシン(すなわち、ドメインI、II及びIII)を含む一方で、Cry1Ab3プロトキシンドメインを利用することにより、Bacillus thuringiensis(Bt)において良好な発現を確保している。野生型のCry1Da1プロトキシンドメインを利用した場合、Cry1Da1はBt中でほとんど発現しない。しかしながら、本願で示すように、野生型のプロトキシンドメインを除去し、Cry1Da1コアトキシンコードセグメントとCry1Ab3プロトキシンドメインコードセグメントとをインフレームで融合させると、Btの結晶非産生株においてCry1Da1コアトキシンの発現は顕著に改善する。注目すべき点として、骨格タンパク質TIC844(配列番号14)及びCry1Da1(配列番号2)は、改変殺虫タンパク質において認められるような商業的に有用な鱗翅類阻害スペクトル及び向上した鱗翅類阻害活性を示さない。
改変試験タンパク質の設計及び昆虫バイオアッセイ試験のための試料調製
本実施例は、骨格タンパク質中の関連するアミノ酸残基を同定し、これを改変して、改変試験タンパク質を作製し、得られた改変試験タンパク質のクローニング及び発現を行う方法を示す。
鱗翅目害虫に対する食餌バイオアッセイにおける改変試験タンパク質の試験
本実施例は、実施例1に記載した改変の成果として作製した改変試験タンパク質の試験について示す。
CEW活性の向上を示す改変試験タンパク質
本実施例は、複数回の試験ラウンドにおいて、骨格TIC844またはCry1Da1タンパク質の活性と比較した場合に、いくつかの改変試験タンパク質について、鱗翅類阻害スペクトルの拡張及び/または鱗翅類阻害活性の向上もしくは増大が認められたことを示す。
植物での発現のための改変殺虫タンパク質及び骨格タンパク質をコードする遺伝子の合成
本実施例は、植物での発現のための改変殺虫タンパク質及び骨格タンパク質をコードするポリヌクレオチドの合成を示す。
植物内での改変殺虫タンパク質発現用の発現カセット
本実施例は、植物内で使用するために設計した骨格及び改変殺虫タンパク質をコードするポリヌクレオチド配列を含む発現カセットの構築について述べる。
骨格または改変殺虫タンパク質発現カセットを組み込んだ形質転換ベクター
本実施例は、骨格または改変殺虫タンパク質の植物組織への組み込みについて述べる。
安定に形質転換したトウモロコシにおける遺伝子改変殺虫タンパク質の鱗翅類活性
本実施例は、改変殺虫タンパク質をトウモロコシ植物内で発現させ、それぞれの害虫への食餌として提供した場合に、鱗翅目害虫に対して改変殺虫タンパク質が示す阻害活性について述べる。
安定に形質転換したワタでの改変殺虫タンパク質の鱗翅類活性
本実施例は、改変殺虫タンパク質をワタ植物内で発現させ、それぞれの害虫への食餌として提供した場合に、鱗翅目害虫に対して改変殺虫タンパク質が示す阻害活性について述べる。
安定に形質転換したダイズにおける改変殺虫タンパク質の鱗翅類活性
本実施例は、改変殺虫タンパク質をダイズ植物において発現させ、それぞれの害虫への食餌として提供した場合に、鱗翅目害虫に対して改変殺虫タンパク質が示す阻害活性について述べる。
Claims (13)
- 配列番号44または配列番号40に記載のアミノ酸配列を有する改変殺虫タンパク質であって、前記改変殺虫タンパク質が、鱗翅目昆虫種に対して阻害活性を示す、改変殺虫タンパク質。
- 前記鱗翅目が、Spodoptera及びHelicoverpaからなる群から選択される、請求項1に記載の改変殺虫タンパク質。
- 前記鱗翅目が、Helicoverpa zea及びSpodoptera frugiperdaからなる群から選択される、請求項1に記載の改変殺虫タンパク質。
- 改変殺虫タンパク質をコードするポリヌクレオチドであって、前記ポリヌクレオチドが、異種プロモーターに作動可能に連結し、前記改変殺虫タンパク質が、配列番号44または配列番号40に記載のアミノ酸配列を含む、前記ポリヌクレオチド。
- 改変殺虫タンパク質をコードするポリヌクレオチドであって、前記ポリヌクレオチドが:
a.配列番号43または配列番号39に記載の;または、
b.配列番号44または配列番号40に記載のアミノ酸配列を含む改変殺虫タンパク質をコードする
ヌクレオチド配列を含む、前記ポリヌクレオチド。 - 配列番号43または配列番号39に記載のポリヌクレオチドを含む宿主細胞であって、細菌宿主細胞または植物宿主細胞からなる群から選択される、前記宿主細胞。
- 前記細菌宿主細胞が、Agrobacterium、Rhizobium、Bacillus、Brevibacillus、Escherichia、Pseudomonas、Klebsiella、及びErwiniaからなる群から選択される、請求項6に記載の宿主細胞。
- 配列番号44または配列番号40に記載のアミノ酸配列を有する改変殺虫タンパク質を含有する昆虫阻害性組成物。
- 昆虫阻害有効量の:
a.配列番号44または配列番号40に記載のアミノ酸配列を含む改変殺虫タンパク質;または、
b.配列番号43または配列番号39に記載のポリヌクレオチドによってコードされる改変殺虫タンパク質
を保有する種子。 - 前記鱗翅目害虫を、請求項1に記載の阻害量の改変殺虫タンパク質に接触させることを含む、鱗翅目害虫の防除方法。
- 請求項1に記載の改変殺虫タンパク質を保有するトランスジェニック植物細胞、植物または植物部分。
- 請求項11に記載のトランスジェニック植物細胞、植物または植物部分に前記害虫を曝露することを含む、鱗翅目害虫の防除方法であり、前記植物細胞、植物または植物部分が、鱗翅目阻害量の改変殺虫タンパク質を発現する、前記防除方法。
- 請求項5に記載のポリヌクレオチドを保有する種子の産生方法であって、前記方法が、
a.請求項5に記載のポリヌクレオチドを保有する少なくとも1種の種子を植え付け;
b.前記種子から植物を生長させ;及び、
c.前記植物から種子を収穫することを含み、
前記収穫した種子が、請求項5に記載のポリヌクレオチドを保有する、前記産生方法。
