潜在型ヘパラナーゼ;

からなる群より選択される少なくとも1つのタンパク質の存在を検出する、ドライアイを有する対象体を特定するための方法を提供する。その後、ドライアイを有する患者は、

本明細書に記載のようにラクリチンに基づく活性を有する組成物は、眼表面に関する疾患、傷害または病態を処置するために開示される。一実施態様によれば、ラクリチンポリペプチド、ラクリチンの生物活性断片、非天然ラクリチンペプチド、またはラクリチンのペプチドミメティック誘導体を含む組成物を提供する。一実施態様において、組成物は、配列番号1、配列番号7、配列番号8、配列番号5および配列番号6からなる群より選択される配列、または1、2、3、4もしくは5つのアミノ酸修飾により配列番号1、配列番号7、配列番号8、配列番号5もしくは配列番号6と異なる配列を含む。一実施態様において、組成物は、配列番号7もしくは配列番号8からなる群より選択される配列、または1、2、3、4もしくは5つのアミノ酸置換により配列番号7もしくは配列番号8と異なる配列を含み、そして更なる実施態様において1、2、3、4または5つのアミノ酸置換は、保存的アミノ酸置換である。

組成物を含むラクリチンを用いる処置に適した患者は、次の状態のうちの1以上を有する患者を含む:
a) 処置前の対象体の涙液が、正常な非ドライアイの涙液と比較して90kDa脱グリカン化SDC-1レベルの低下を含む;
b) 処置前の対象体の涙液が、正常な非ドライアイの涙液と比較して25kDa SDC-1レベルの上昇を含む;
c) 処置前の対象体の涙液が、正常な非ドライアイの涙液と比較して不活性型ラクリチン-Cスプライスバリアントレベルの上昇を含む;
d) 処置前の対象体の涙液が、正常な非ドライアイの涙液と比較して潜在型ヘパラナーゼレベルの低下を含む;
e) 処置前の対象体の涙液が、正常な非ドライアイの涙液と比較して活性化ヘパラナーゼレベルの上昇を含む;および
f) 手術を受けるまでの時間にかかわらず、対象体が、PRK(レーザー屈折矯正角膜切除術)もしくはLASIK(レーザー角膜内切削切除形成術)手術または眼の他の外科手術から回復中であり、PRKまたはLASIK手術を受け、ドライアイを患っている任意の対象体を含む。一実施態様において、過去1日、1カ月、6カ月、1年または数年以内にPRKまたはLASIK手術を受けた対象体は、本明細書に記載の製剤を含むラクリチンを用いる処置から恩恵を受け得る。

例えば、タンパク質またはペプチドの一次配列を変更するが、通常その機能は変更しない保守的アミノ酸の変化がなされ得る。このため、10以上の保守的アミノ酸変化は、典型的に、ペプチド機能に影響を及ぼさない。一実施態様によれば、保守的アミノ酸置換は、次のグループ内での置換を含む。

ドライアイ疾患の同定
手順
細胞培養、コンストラクトおよび抗体
ヒト角膜上皮(HCE-T)細胞をRIKEN BioResource Center(つくば市、日本)から購入し、継代3および15間を使用した。HCE-T細胞を培養し、4 mg/mlインスリン、100μg/ml EGF、500μg/mlコレラ毒素および5μl/ml DMSOを含むDMEM/F-12中で保存した。プライマリーヒト角膜上皮細胞(PCS-700-010)をATCC(Manassas、VA)から購入し、推奨の培地中に広げた。
眼瞼と眼の間でミリメートルグラデーションを有するフィルターウィッキング「シルマー」試験紙の挿入により0.5%プロパラカイン麻酔した眼から涙液を採取し、個々に-70℃で保存した。溶出前に、正常またはドライアイの涙液総量を各試験紙へ引っ張られた涙液のミリメートルから推定した。これは、溶出のためにそれぞれ用いられるPBSの最終量を定義した。貯留された正常の涙液または貯留されたドライアイの涙液を-70℃で使用するまで保存した。
FOXO3転座アッセイについて、α-MEM(5.54 mm グルコース)中カバーガラス上にサブコンフルエンス(約50%)までHCE-T細胞を3回培養し、IFNG(100 units/ml;Roche Applied Science)中で一晩感作し、10 nmラクリチンまたはC-25を含むまたは含まないTNFを加えたα-MEM(50 ng/ml;PeproTech, Rocky Hill, NJ)で1:100希釈した正常またはドライアイの涙液で15分間処理した。細胞を洗浄し、4%パラホルムアルデヒドで固定し、FOXO3(1:200;Millipore, Billerica, MA)、続いてヤギ抗-ウサギ二次抗体の免疫染色し、Zeiss LSM 700顕微鏡で見た。
涙液は、タンパク質、脂質および代謝物において豊富な半透明膜として血管の無い膜上皮および血管のある結膜に溜まる。眼瞼を保湿するその能力を超えて、涙液は、光の屈折に不可欠である。涙液の役割は、角膜上皮の健康の促進において等しく重要で、薬物または点眼剤によって置き換えられない。涙液が慢性的に不十分であるとき、上皮はストレスを受け、状況をさらに悪化させる炎症性サイトカインを放出する。ドライアイは、全人口の5〜6%に影響を与えており、閉経後の女性および老年者においてそれぞれ6〜9.8%および34%まで増加する。最も一般的な眼の疾患であるが、単一のゴールドスタンダード診断テストはなく、効果的な処置もない。現在の方法としては:a)対象体調査票、b)眼表面損傷のローズベンガルまたはリサミングリーン染色、c)涙液量のシルマー試験紙測定、d)涙液破壊時間、e)涙液蒸発率、f)涙液メニスカス高さまたは半系、g)涙膜インデックスまたはターンオーバー率、h)涙液のオスモル濃度、i)リゾチームまたはラクトフェリンアッセイ、およびj)涙シダ状結晶分析が挙げられ、これらの各々は多数の欠点を有する。
一般的に用いられる「人口涙液」は、ドライアイに関連する症状をこれらの症状の原因に対処することなく一時的に軽減する。抗炎症剤シクロスポリンの眼科用製剤が現在広く使用されている。それおよび他の抗炎症剤が臨床試験されているが、一般的にドライアイ対象体の約15%のみに有益である。炎症の結果に着目するか、または他の臓器系のために開発された薬物を適用するよりむしろ、眼表面の天然の生態および何がドライアイにおいてかけているかを考慮することに利益がある。正常なウサギの眼に局所添加したときに基礎的流涙を促進するラクリチンモノマーという天然の涙液タンパク質の下方制御は、ドライアイ疾患の上流の扇動者であり得る。なぜドライアイにおいてラクリチンモノマーが少ないのか。ラクリチンモノマーは、涙液中の組織トランスグルタミナーゼ(TGM2)により不活性型多量体に架橋される。ヒト涙液からのすべてのラクリチン単量体、多量体および断片を免疫枯渇することによりこれを実証した。免疫枯渇させた涙液中にスパイクした組み換えラクリチンは、37℃で一晩インキュベーション後、二量体、三量体および四量体を形成した。涙液なしの陰性コントロールにおいて、少量の二量体が形成された。架橋は、シンデカン-1を標的とするラクリチン分裂促進ドメイン(アミノ酸100〜109)内にグルタミン106を含む。架橋ラクリチンは、本質的にあまりシンデカン-1と結合せず、活性がより低い(図5;右2つの棒グラフ)。ブロッティングは、正常なヒトの涙は0.6μM TGM2を含み、それ故に単量体ラクリチンの陰性レギュレーターとして作用するように思われることを示唆する。ヒト角膜上皮細胞は、TGM1およびTGM2のmRNAの両方を発現する。両方のmRNA発現は、高浸透圧ストレスで特にTGM1を増加させるが、TGM1は涙液中で検出されなかった。したがってラクリチンは、ドライアイにおいて架橋の促進および非活性化を受け得る。
T涙液ラクリチンのモノマー形態は、眼表面ストレスの緩和に関与する多機能因子である。それはまた、基礎的流涙のアゴニストである。モノマーラクリチンは、細胞表面のシンデカン-1(SDC1)のヘパラナーゼ(HPSE)脱グリカン化形態を標的とする。しかしながら、重合化ラクリチンおよびラクリチン-Cスプライスバリアントは両方、SDC1を標的とできず、それ故に不活性である。我々は、SDC1またはHPSEいずれかがドライアイの涙液においてモノマーラクリチンを置き換えられ得るか、およびSDC1またはHPSEが不適当でありウ得るかを調べた。
25アミノ酸ラクリチンのC末端断片の安定性
最も多い合成ペプチド薬の制限は、それらのプロテアーゼ感度である。HIVプロテアーゼレトロペプシンおよびカテプシンKのみが、PROSPER(プロテアーゼ特異性予測サーバー)分析に従いLACRIPEPを開裂でき、前者は中間において切断し、後者は最後のアラニンを取り除く。レトロペプシンは、LACRIPEPを不活性化するだろう。一方カテプシンKは、影響を有しないだろう。しかしながら、いずれのプロテアーゼも正常なヒトの涙液において見られない。それにもかかわらず、涙液は他のプロテアーゼに富む。したがって、我々は、2、4、6および16時間37℃で正常なヒト涙液中においてLACRIPEPをインキュベートした。免疫ブロッティングのために、我々は、最初に免疫枯渇によりすべての内因性ラクリチンを取り除いた。37℃で2〜16時間ラクリチン枯渇ヒト涙液中にてインキュベーション後の、種々のタンパク質からのプロテアーゼ感受性陽性コントロール「SN pep」およびLACRIPEP(「N-94」)の免疫ブロットを示す図6Aに示されるように、LACRIPEPは少なくとも16時間非常に安定であった。SN pep、Lacripep(「N-94」)、および6つC末端アミノ酸を有しないLacripep(「N-94/C-6」)の質量分析は、37℃で4時間ラクリチン枯渇涙液中にてインキュベーション後の3つのペプチドの比較による安定性を示す(図6B)。驚くべきことに、より小さいラクリチンのC末端断片(N-94/C-6;配列番号5)は、LACRIPEPペプチド(配列番号7)ほど安定でないことが分かった。質量分析は、Lacripep(「N-94」)および6つC末端アミノ酸を有しないLacripep(「N-94/C-6」)が涙液中で同様の安定性を有し、6つC末端アミノ酸を有しないLacripep(「N-94/C-6」)が62℃(図6B)で29日間リン酸緩衝生理食塩水中にて安定であったことを示唆されたが、免疫ブロッティングは、N-94/C-6が37℃で4時間ラクリチン枯渇涙液にてインキュベーション後エピトープを消費するが、Lacripep(「N-94」)ではそうでないことを示す。したがって、天然ラクリチンの最後の6つのアミノ酸は、N-94をN-94/C-6と比較して優れた医薬ペプチドにするラクリチンの生物活性断片の安定性を高めるのに関連がある。
涙ラクリチンの開裂増強断片は殺菌性である
実験手順
涙および涙免疫枯渇正常ヒトの基礎的な涙を回収した。簡単にいうと、0.5%プロパラカインで麻酔した眼からの涙を予め秤量した芯に回収し、-70℃保存のために急速冷凍した。涙を、各試験紙で30μlのPBSに20分浸し、続いて遠心することにより溶出した。免疫枯渇について、10倍希釈した涙を一晩(4℃)または1時間、室温で、「抗N-65 Lac C末端」または免疫前Igで飽和させたタンパク質Aビーズ(0.2 ml、NAb Spin Kit、Peirce/Thermo Scientific)とインキュベートした。N-65は、65 N-末端アミノ酸を欠くラクリチントランケーション変異体である。遠心(5000×gで1分)後の涙フロースルーを、次いで抗細菌活性についてアッセイした。

および共通逆方向プライマーGGTGGTCATATGTATATCTCCTTCTTAAAG(配列番号11)を使用して産生した。全構築物をシークエンシングにより確認した。細菌タンパク質発現および組み換えラクリチンの精製およびラクリチントランケーションを先に記載のように行った(Zhang et al、(2013)J. Biol. Chem. 288, 12090-12101)。簡単にいうと、浄化細胞(ER2566またはBL21-CP)ライセートを、50 mM Tris、0.5 M NaCl(pH 8)で平衡化したキチンカラム(IMPACT-CN System;New England Biolabs Inc., Beverly, MA)に載せ、続いて20カラム容積で洗浄し、50 mM 2-メルカプトエタノールで16時間、室温で同じ緩衝液で溶出し、PBS(4℃)に対して広範な透析を行い、タンパク質定量化を行った。さらにDEAE精製で約9-kDaラクリチンタンパク分解性断片および細菌混入物を除去し、その中でラクリチンをリン酸緩衝液、pH 7.2中の140 mM NaClを用いるフロースルーとして回収した。合成ペプチドN-80/C-25、N-94、N-94/C-6、N-94/C-10、N-94/C-15、N-99、およびN-104をGenscript(Piscataway、NJ)で、アセチル化N末端を用いて合成した。純度は95%であった。ラクリチンC末端N-94、N-99、およびN-104以外の全てのC-末端はアミド化された。N-64/C-31はアミド化もアセチル化もされず、University of Virginia Biomolecular Research Facilityにより合成された。ラクリチン約9-kDa断片の性質を、ウェスタンブロットで探究した。簡単にいうと、DEAE前後のラクリチンをSDS-PAGEにより分離し、次いでそれぞれ0.3% Tween 20含有PBS中1:200または1:400に希釈した抗Pep Lac N-末端および抗N-65 Lac C末端抗体を用いて移動およびブロットした。検出はECLを用いた。
涙液中のラクリチン殺菌活性-涙は眼の表面を環境病原体から保護し、分泌促進性マイトジェンラクリチンに富み、これは、基底および反射で涙を流す間、眼の上を流れる。ラクリチンはタンパク分解性に処理されたC末端がヒト汗の殺菌活性に貢献するダームシジンと21%同一である。我々は、ラクリチンまたはラクリチン断片が殺菌活性を有するか否かの決定を探索した。半希釈した基礎的な涙は大腸菌増殖を完全に遮断し、大腸菌は、緑膿菌同様発展途上国における細菌結膜炎の顕著な貢献因子である。我々は、ラクリチンおよびC末端ラクリチン断片の両者を免疫枯渇するために固定化抗N-65 Lac C末端抗体(ab C末端)または免疫前Ig(偽枯渇)を通した涙で試験した。両者を、大腸菌および緑膿菌の用量依存的攻撃のために10倍希釈した。偽枯渇涙は大腸菌および緑膿菌コロニーを、涙液量依存的方法で抑制した。これは、リン酸緩衝液陰性コントロールとして無効であったC末端抗体-免疫枯渇させた涙と対照的であった。
ラクリチンが殺菌性であるか否かを探索する理論的根拠は、タンパク分解性処理されたC末端がそのヒト汗における殺菌活性に貢献するダームシジンとの21%同一性であり、涙に存在することである。驚くべきことに、ダームシジン一次配列相同性はラクリチン活性の源ではなかった。ダームシジンの殺菌性SSL-25ペプチドと、合成ペプチドとしては不活性型であった相同ラクリチン領域の間で、同一性はわずか40.7%である。むしろ、合わせて、N-65処理細胞における膜不透過性SYTOX Greenの急速侵入により明らかなように、細菌膜接触および挿入に適した、N-末端両親媒性α螺旋および疎水性C末端コイルドコイル尾部からなるハイブリッドドメインである、7%ダームシジン同一性を有するラクリチンN-104断片がコア活性を具体化した。驚くべきことに、C末端25アミノ酸断片KQFIENGSEFAQKLLKKFSLLKPWA(配列番号7;アミノ酸95-119;N-94)は十分活性であり、一方、6末端アミノ酸(例えばKQFIENGSEFAQKLLKKFS;配列番号5)の除去は、ペプチドの殺菌活性を実質的に低下させることが判明した。
眼科用製品の局所適用が患者のコンプライアンスのために最もポピュラーで忍容性の良好な投与経路のままであるが、点眼剤のバイオアベイラビリティは、まばたき、基準および反射涙液分泌、ならびに鼻涙の排流により激しく妨害される。局所処置の治療指標を高める1つの解決法は、薬物担体としてポリマーナノ粒子の適用による。
材料および装置
TB DRY(登録商標)Powder Growth MediaをMO BIO Laboratories, Inc.(Carlsbad、CA)から購入した。NHS-rhodアミンをThermo Fisher Scientific(Rockford、IL)から購入した。SV40-Adeno ベクタートランスフォーム角膜細胞(RCB 2280、HCE-T)を理研細胞バンク(日本)から購入した。ウシ下垂体抽出物(BPE)および事前認定(prequalified)ヒト組み換え上皮細胞増殖因子1-53(EGF)を加えたケラチン生成細胞-SFM培地をGibco Invitrogen(Life Technologies、NY)から購入した。細胞浸透性のカルシウム指示薬Fluo-4 AMをLife Technologies(NY)から購入した。0.5 mmバー(burr)を有するAlgerbrush IIをThe Alger Company, Inc.(TX)から購入した。自社飼育の12週齢雌性非肥満糖尿病性(NOD)(Taconic Farms、Germantown/NY、USA)マウスを用いてインビボ実験を行った。
ELP(SI)をコードする遺伝子をpET25b(+)ベクター内での再帰的方向性ライゲーションによって合成した。EditSeq(DNAStar Lasergene、WI)において最良のE. coliコドンを用いて、ヒトラクリチンをコードし、分泌シグナルペプチドを有しない配列をデザインした。BseRI部位での挿入を介してラクリチン配列とELP tagの間にトロンビン切断部位をデザインした。pIDTSmart-KANベクター内で5'末端および3'末端にてNdeIおよびBamHI制限消化部位と隣接しているラクリチン遺伝子をIntegrated DNA Technologiesから購入した。

LSI 融合タンパク質の位相図は、DU800 UV-Vis 分光光度計(Beckman Coulter、Brea、CA)を用いて、溶液温度の関数として350 nmでの光学密度変化によって特徴付けられた。Ttは、最大一次導関数の点で定義された。DynaPro-LSR Plate Reader(Wyatt Technology、Santa Barbara、CA)も使用し、動的光散乱法(DLS)を用いてナノ粒子の自己アッセンブリーを測定した。溶液を5から50℃に加熱した場合の光散乱データを一定の温度間隔(1℃)で回収した。レイリー球体モデルを使用して結果を分析し、二乗和値に基づくキュムラントアルゴリズムに適合させた。臨界ミセル温度(CMT)は、Rhが平均的モノマーRhよりも有意に大きい最低温度であると定義された。
100 kVのFEI Tecnai 12 TWIN 顕微鏡(Hillsboro、OR)にてTEMイメージングを行った。簡単に言うと、まず、炭素フィルム(CF400-Cu、Election Microscopy Sciences、Hatfield、PA)を有する銅グリッド上に100 uM 溶液(5 uL)を沈着させた。過剰の溶液を濾紙で除去した後、該検体を2%酢酸ウラニルでネガティブ染色し、次に、30秒後に過剰の酢酸ウラニルを除去した。次に、該検体を、イメージングで使用する前に少なくとも3時間、室温下にて乾燥させた。
ウシ下垂体抽出物(BPE、50 mg/ml)および上皮細胞増殖因子(EGF、5 ng/ml)を含有するケラチン生成細胞-SFM培地(KSFM、Life Technologies、Rockville、MD)中にてSV40不死化HCE-T細胞(理研細胞バンク、日本)を増殖した。35 mm カバーグラス底のディッシュ中にてCa2+イメージング・スクラッチ・アップテイクアッセイに細胞継代4-6を用いた。刺激後の応答性を最適化するために、実験前に24時間、EGFおよびBPE不含培地で細胞を飢餓させた。
HCE-TをCa2+およびMg2+不含リン酸緩衝生理食塩水(PBS)で2回すすいぎ、2.5 mM カルシウムプローブFluo-4AM(Invitrogen Life technologies、NY)を含有する新鮮KSFM培地中にて37℃で20分間インキュベートした。次に、細胞をNaCl リンゲルバッファー(145 mM NaCl、5mM KCl、1mM CaCl2、1mM KH2PO4、1 mM MgCl2、10mM グルコース、および10mM HEPES、モル浸透圧濃度300、pH 7.4)で2回すすぎ、同バッファー中に室温で30分間保持した。Ca2+不含培地について、1 mM Ca2+ を0.5 mM EGTAと交換した。細胞を488 nmで照射し、それらの発光を、Zeiss LSM 510 Meta 共焦点顕微鏡システムを使用して510 nmで3.15秒ごとにモニターした。目的のフィールドは24〜45 細胞を含有しており、画像解析ソフトウェアを用いて各領域の蛍光強度変化を算出した。Fluo-4AMの蛍光強度の変化を使用してCa2+動態を評価した。該データは、各時点での蛍光強度(Ft)から最初の時点での蛍光強度(F0)の変化率として表した:(Ft - F0)/F0 X 100%。
スクラッチアッセイのために、コンフルエントHCE-T単層をp200ピペットチップで一直線に削り取ってひっかき傷を作った。細胞をBPEまたはEGFを含まないKSFM培地ですすいでデブリを除去し、次に、BPE(50 mg/ml)およびEGF(5 ng/ml)を含有する新鮮KSFM培地、LSI、または増殖因子を含まない培地(処理なし)と一緒にインキュベートした。Zeiss LSM 510 Meta 共焦点顕微鏡システムを用いて、処理開始時と24時間後の創傷の位相コントラスト画像を撮った。
アミノ末端の共有結合的修飾を介して、SIおよびLSI ナノ粒子をNHS-rhodアミン(Thermo Fisher Scientific Inc、Rockford、IL)とコンジュゲートした。コンジュゲーションを100 mM ホウ酸バッファー(pH 8.0)中にて4℃で2時間(LSI)または一晩(SI)行い、次に、PD10カラム(GE Healthcare、Piscataway、NJ)にて脱塩して、遊離色素を除去した。簡単に言うと、細胞をBPEおよびEGFを含まない新鮮培地ですすいだ後、該ディッシュに10 mM rhodアミン標識タンパク質を加えた。37℃で種々の時間インキュベーとした後、該細胞をすすぎ、Zeiss LSM 510 Meta 共焦点顕微鏡システムを用いて画像を得た。
簡単に言うと、12週齢雌性NODマウスをキシラキシン/ケタミン(60-70 mg + 5 mg/kg)の腹腔内注射で麻酔し、加熱パッドの上に置いた。眼洗液(OCuSOFT、Inc.、TX)で眼表面を浄化した後、Algerbrush II(The Alger Company、Inc.、TX)を用いて右眼の角膜上皮を基底膜まで取り除いた;左眼は、反対側の対照として無傷のままであった。マウスを、12時間間隔で24時間、BPE(50 mg/ml)および EGF(5 ng/ml)を含有するKSFM培地、100 mM LSIまたは100 mM SIを2回投与して(5 ml)治療したか、または処置しなかった。眼表面を0.6 mg/ml フルオレセイン(Akorn、IL)5 mlで染色した後、12時間後および24時間後にMoticam 2300カメラを用いて擦過傷の画像を得た。
前実験を少なくとも3回反復した。Ca2+媒介蛍光の最大蛍光強度変化を、対応のないt検定を用いて解析した。一方向ANOVAを用い、次に、Tukeyのpost hoc検定を用いてスクラッチ創傷治癒定量化を解析した。二元配置分散分析を用い、次に、Bonferroni post検定を用いてHCE-Tアップテイクを解析し、Kruskal-WallisノンパラメトリックANOVAを用いてマウスの角膜上皮の擦過傷からの回復を解析した。Mann-Whitney U検定を用いて、処置12時間後のLSIとLS96との角膜創傷治癒比較を解析した。0.05未満のp値は統計学的に有意であるとみなした。

LSIのインビトロ分裂促進活性に後押しされて、本発明者らは、さらに、外因性LSIがHCE-Tに進入可能であるかを研究した。かくして、該細胞を種々の時点でNHS-rhodアミン標識LSIおよびSIナノ粒子と一緒にインキュベートした。ラクリチン媒介アップテイクと一致して、LSIは、時間に依存して、HCE-Tへの細胞アップテイクを受けた。有意な細胞進入は、インキュベーションの10分後に見られ、1時間後に、LSIナノ粒子は核周囲領域内に蓄積した。定量化後、LSIは、SIナノ粒子よりも有意に高い細胞質蛍光を示した(p<0.0001)。種々のサイズ、形状および変化を有するナノ物質は、医用画像処理、組織標的化、および細胞アップテイクに広く用いられている。さらに最近、下流シグナル伝達を調節するためにより効果的に膜受容体を架橋するためのナノ粒子の使用は、特に抗体媒介受容体架橋において、大きな注目を集めている。
本発明者らは、局所点眼剤によるLSIナノ粒子インビボ効力の研究を進めた。この研究において、本発明者らは、LSIナノ粒子の創傷治癒効果を評価するために、雌性NODマウスの角膜上皮性擦過傷モデルを開発した。非肥満糖尿病性(NOD)マウスは、ヒトの創傷治癒不良のための動物モデルとして頻繁に使用される。細胞増殖の低下、筋線維芽細胞表現型の発病の遅延、プロコラーゲンI mRNA 発現の減少、およびアポトーシス細胞死の制御異常がNODグループにおいて見られた。NODマウスモデルは、LSIナノ粒子のインビボ活性の評価のために選択された。簡単に言うと、Algerbrush IIを用いて角膜縁領域にダメージを与えずに動物の右眼に直径約2 mmの円形の擦過傷を作った。画像処理の直後に、5 mlの100 mM LSIナノ粒子、SIナノ粒子または対照EGF + BPEを眼表面に局所投与し、この処置を創傷開始の12時間後に1回繰り返した。コバルトブルー光の下でフルオレセイン染色を用いて0時、12時間目、および24時間目に創傷の画像を得た。最初の創傷治癒比較研究は、処置グループごとに、反対側の対照として左眼を無傷のままにした4匹のマウスを含んだ。実験後、ImageJを使用して創傷治癒画像を解析した。平均フルオレセイン強度、創傷面積、合計フルオレセイン(合計 = 平均フルオレセイン強度×創傷面積)、初期値のフルオレセインパーセンテージ、初期値の創傷面積パーセンテージ(PctArea)、および初期値の合計フルオレセインパーセンテージを、ブラインド評価者(blind reviewer)によって、Kruskal-Wallisノンパラメトリック検定を用いて12時間目および24時間目にグループ間で比べて決定した。処置によって有意な炎症または他のいかなる有害作用も観察されなかった。特に、12時間目および24時間目のいずれの時点でのLSIも、SI(p = 0.001)、EGF + BPE(p = 0.001)、および非処置グループ(p = 0.001)と比べて、初期創傷面積(PctArea)のパーセンテージを有意に減少させており、LSIが角膜上皮の回復を促進するのに最良の製剤であることを示唆している。フルオレセイン画像処理結果を裏付けるために、本発明者らは、さらに、組織学分析のために24時間後に角膜上皮を処理した。簡単に言うと、角膜を固化し、異常を横切って薄片にし、ヘマトキシリンおよびエオシンによって染色した。
ボーマン膜(BM);角膜実質(ST);デスメ膜(DM);内皮(EN)を評価した。意外なことに、LSI処置グループの角膜上皮は、滑らかな再構成表面をもち、炎症のない完全な回復を示した。フルオレセイン試験によって、SIグループにおける24時間目の染色に対する一部抵抗性が明らかになったが、再生角膜上皮は、分化を完了しなかった。分裂促進的ラクリチンタンパク質は、タンパク質ポリマーナノ粒子上で修飾された場合に活性のままであることを示したので、本発明者らは、次に、ELP媒介粒子アッセンブリーがこの結果を達成するために必要であるかを研究した。インビボでのELPアッセンブリーの有意さに取り組むために、LSIナノ粒子の効力は、LS96と称される熱的に反応しないラクリチン融合タンパク質と直接比較することができる。LSIおよびLS96はどちらも、ラクリチン配列、およびそれに続くELP含有96総五量体反復を含有する;しかしながら、ELP S96は、生理学的温度を超えるまで相分離しない。光学密度測定は、実際に、LS96がリン酸緩衝生理食塩水中にて観察できる相転移を全く示さないことを明らかにした。加えて、DLSは、LSIが37℃でLS96よりも非常に大きな水力学的半径を有することを確認した。
角膜創傷治癒プロセスを促進するために、候補生物薬剤であるマイトジェンラクリチンを眼表面に送達する手段として多価ELPナノ粒子を使用した。このラクリチンELP融合であるLSIは、非修飾SIナノ粒子と類似する熱応答性自己アッセンブリー特性を示し、生理学的に適切な温度にてそのコロナでアクセスできるラクリチンを提供する。LSIナノ粒子は、カルシウム依存性細胞シグナル伝達の引き金を引き、細胞に内部移行し、ヒト角膜上皮性細胞株(HCE-T)の単層におけるスクラッチクロージャーを促進する。角膜上皮の除去後にNODマウスの眼表面に局所的投与した場合、LSIナノ粒子は、SIおよび未処置グループと比べて速い創傷治癒を促進した。最も重要なことに、LSIナノ粒子は、熱依存性アッセンブリーを受けていないLS96と称される対照ラクリチンELP融合と比べて速い角膜上皮再生を生じる。全体的にみて、この研究は、ナノ粒子足場としてのELPの、タンパク質治療薬を眼表面に有効に送達し、擦過傷を修復する潜在能力に光を当てる。
Claims (12)
- 対象体がドライアイを有するかどうかを判定するための方法であって、該対象体から得られた涙液検体中の、
90kDa 脱グリカン化SDC-1;および
25kDa SDC-1;
からなる群より選択される少なくとも1つのタンパク質の存在を検出することを含み、
ここで、
正常な眼からの涙液中に存在するレベルと比較して90kDa 脱グリカン化SDC-1レベルの減少;および/または
25kDa SDC-1の検出;
が、対象体がドライアイを有すると判定するための指標である、該方法。 - 該対象体から得られた涙液検体中の90kDa 脱グリカン化SDC-1および25kDa SDC-1の濃度を測定し、正常な眼からの涙液中に存在するレベルと比較して25kDa SDC-1レベルの増加と一緒になった90kDa 脱グリカン化SDC-1レベルの減少が、対象体がドライアイを有すると判定するための指標である、請求項1に記載の方法。
- 該対象体から得られた涙液検体を25kDa SDC-1の存在について検査し、25kDa SDC-1の検出が、対象体がドライアイを有すると判定するための指標である、請求項1に記載の方法。
- 該対象体から得られた涙液検体を不活性型ラクリチン-Cスプライスバリアントおよび/または潜在型ヘパラナーゼの存在について検査することを更に含み、
不活性型ラクリチン-Cスプライスバリアントの検出および/または正常な眼からの涙液中に存在するレベルと比較して潜在型ヘパラナーゼレベルの減少または活性型ヘパラナーゼの増加が、対象体がドライアイを有すると判定するための指標である、請求項1〜3のいずれか一項に記載の方法。 - 該対象体から得られた涙液検体を25kDa SDC-1および不活性型ラクリチン-Cスプライスバリアントの存在について検査し、25kDa SDC-1および不活性型ラクリチン-Cスプライスバリアントの検出が、対象体がドライアイを有すると判定するための指標である、請求項1に記載の方法。
- 対象体におけるドライアイを処置するための組成物であって、
該組成物はラクリチンポリペプチドを含み、
該対象体は、該対象体から得られた涙液検体中に、
90kDa 脱グリカン化SDC-1;および
25kDa SDC-1;
からなる群より選択される少なくとも1つのタンパク質が存在することを検出することによって、ドライアイを患う患者と特定され、
ここで、
正常な眼からの涙液中に存在するレベルと比較して90kDa 脱グリカン化SDC-1レベルの減少;および/または
25kDa SDC-1の検出;
が、対象体がドライアイを有する患者と特定するための指標であり、
対象体の眼表面への局所投与のために製剤化された組成物。 - 該ラクリチンポリペプチドが、配列番号1に記載の配列を有するポリペプチド、または
KQFIENGSEFAQKLLKKFS(配列番号5);
KQFIENGSEFAQKLLKKFSLLKPWA(配列番号7);
KQFIENGSEFANKLLKKFS(配列番号6);および
KQFIENGSEFANKLLKKFSLLKPWA(配列番号8)、
からなる群より選択される配列を有するラクリチンまたは1もしくは2つのアミノ酸置換により配列番号5、配列番号6、配列番号7もしくは配列番号8と異なる配列を有する配列番号5、配列番号6、配列番号7もしくは配列番号8の誘導体の生物活性断片である、請求項6に記載の組成物。 - 該ラクリチンポリペプチドが、配列KQFIENGSEFAQKLLKKFSLLKPWA(配列番号7)からなる、請求項6に記載の組成物。
- 該ラクリチンポリペプチドが、配列番号5に記載の配列からなり、そのN末端がアセチル化され、およびそのC末端がアミド化されている、請求項6に記載の組成物。
- 予め定められた標準と比較して、減少したレベルの90kDa 脱グリカン化SDC-1が該涙液検体中に存在する場合および/または該涙液検体中の25kDa SDC-1の存在が検出された場合に、該涙液検体を採取した対象体の眼表面と、ラクリチンポリペプチドを含有する医薬組成物とを接触させることを含む、請求項1〜5のいずれか一項に記載の方法。
- 該ラクリチンポリペプチドが、配列番号1に記載の配列を有するポリペプチド、または
KQFIENGSEFAQKLLKKFS(配列番号5);
KQFIENGSEFAQKLLKKFSLLKPWA(配列番号7);
KQFIENGSEFANKLLKKFS(配列番号6);および
KQFIENGSEFANKLLKKFSLLKPWA(配列番号8)、
からなる群より選択される配列を有するラクリチンまたは1もしくは2つのアミノ酸置換により配列番号5、配列番号6、配列番号7もしくは配列番号8と異なる配列を有する配列番号5、配列番号6、配列番号7もしくは配列番号8の誘導体の生物活性断片である、請求項10に記載の方法。 - 該ラクリチンポリペプチドが、配列番号5に記載の配列からなり、そのN末端がアセチル化され、およびそのC末端がアミド化されている、請求項10に記載の方法。
