本発明をさらに詳細に検討する前に、次の用語および手法を最初に定義する:
本発明の抗体および抗原結合ドメインは、RANKL/OPGbpに選択的に結合するが、すなわち、それらは、他の抗原に対するよりも強い結合親和性によってRANKL/OPGbpに優先的に結合する。抗体はヒトRANKL/OPGbpに選択的に結合するが、非ヒトRANKL、たとえばマウスRANKLにも検出できるほど結合する可能性がある。あるいはまた、抗体はもっぱらヒトRANKLまたはRANKに結合するが、それぞれ非ヒトRANKLまたはRANKには検出可能な結合をしないこともある。
第1の態様において、本発明は化合物を提供するが、その化合物は、オステオプロテジェリン結合タンパク質(OPGbp)/NFκB活性化受容体リガンド(RANKL)のNFκB活性化受容体(RANK)との結合のアンタゴニストもしくは阻害剤であって、精巣において(生殖)細胞増殖を刺激するのに使用するため、ならびに/または、哺乳動物において男性不妊の治療および/もしくは予防に使用するため、ならびに/または、哺乳動物において男性受精能を誘導もしくは改善するのに使用するためのものである。
本発明の他の実施形態によれば、上記のように使用するための化合物は、OPGbp/RANKLペプチド、特にヒトOPGbp/RANKLペプチドと免疫反応する化合物である。あるいはまた、抗体はRANKペプチド、特にヒトRANKペプチドと免疫反応することができる。免疫反応性化合物は、具体的には、抗体、抗原結合ドメイン、抗体もしくは抗原結合ドメインの断片、または抗体もしくは抗原結合ドメインの誘導体からなる1群から選択することができる。
i) 配列番号2または配列番号4に記載のアミノ酸配列;
ii) 配列番号3のヌクレオチド1から951に示すヌクレオチド配列を含有するDNA分子によりコードされるアミノ酸配列;
iii) 配列番号11に記載のアミノ酸配列;
iv) i)-iii)に記載の配列のうちいずれか1つの部分配列であって、配列番号11に記載のRANKポリペプチドに結合することができる前記部分配列;ならびに
v) i)-iii)に記載の配列のいずれかに対して少なくとも90%の配列同一性を有する配列;たとえば、i)-iii)に記載の配列のいずれかに対して少なくとも91、92、93、94、95、96、97、98、99または99.5%の配列同一性を有する配列。
i) 配列番号5(ヒトRANKL D-Eドメイン:GFYYLYANICFRHHETSGDLATEYLQLMVYVTKTSIKIP)に記載のアミノ酸配列;および
ii) i)のアミノ酸の部分配列であって、当該部分配列もしくはバリアントが配列番号6(ヒトDEエピトープDLATE)に記載の配列を含んでなる、前記部分配列。
i) 配列番号7(αRANKL-1の重鎖)に記載のアミノ酸;
ii) 配列番号7に記載のアミノ酸配列の部分配列;ならびに
iii) i)およびii)に記載のアミノ酸配列のいずれかに対して少なくとも90%の配列同一性、たとえばi)およびii)に記載の配列のいずれかに対して少なくとも91、92、93、94、95、96、97、98、99、または99.5%の配列同一性、を有するアミノ酸配列。

(実施例)
RANKLシグナル伝達は、骨の恒常性、免疫系、および乳汁分泌に重要である。RANKLは、隣接細胞の膜にある受容体RANKと特異的に結合する細胞外リガンド-ドメインを有する、膜貫通タンパク質である。RANKの結合および活性化は、多くの場合NFκB活性化を介して細胞内シグナル伝達を引き起こし、それは細胞周期に影響を及ぼす。OPGはRANKLの細胞外ドメインに結合してRANKの活性化を妨げるため、RANKLとRANKとの相互作用は、近傍の細胞からのOPG分泌によって調節される。したがって、RANKLの生物学的影響は、RANKL、RANKおよびOPGの存在および存在量に左右される。さらに複雑なことに、RANKLは3つの異なるアイソフォーム1、2、および3として存在する。1つは膜貫通型、また1つは細胞質型であり、もう1つは分泌型である。異なるアイソフォームは必ずしも同じ影響を及ぼすとは限らないと考えられ、細胞質非分泌型は概してRANKL発現の阻害物質である。これは、RANKLの阻害がモデルにおける個別アイソフォームの発現に応じて反対の影響を与える可能性があることを意味する。
ヒト組織サンプル
地方倫理委員会からの承認後、ヘルシンキ宣言にしたがって、コペンハーゲン大学病院(Rigshospitalet)、デンマーク、の男性病クリニックから患者を募集した。TGCTのために実施された精巣摘除術の検体から成人精巣サンプルを得た。腫瘍周囲の組織は、CIS細胞または正常な/低下した精子形成のいずれかを有する精細管を含んでいた。各サンプルを断片に分割し、その断片をRNA抽出のために急速凍結して-80℃で保存するかまたは、ホルマリンもしくはパラホルムアルデヒド中で4℃にて一晩固定した後パラフィンに包埋した。熟達した病理学者が全サンプルを評価し、TGCTのための免疫組織化学(IHC)マーカーを使用して、腫瘍の組織学的サブタイプを確認した。
Leuven Vdr-/-マウスおよび対照を作製し、標準的な齧歯動物飼料(1.0% カルシウム、0.7% リン、Sniff R/M-H)を用いて飼育した。すべての動物は、国立衛生研究所(National Institutes of Health)「実験動物の管理と使用に関する指針」(Guide for the Care and Use of Laboratory Animals)にしたがって取り扱われ、すべての実験はルーヴェン・カトリック大学(KU Leuven)の「動物実験倫理委員会」(Ethical Committee Animal Tests)によって承認された。代表的なマウス群をそれぞれ10または15週齢で屠殺した。マウスを麻酔下で失血させて、ホルモン測定用に血清を採取した。それぞれの精巣を断片に分割し、その断片をRNAもしくはタンパク質抽出のために急速凍結して-80℃で保存するかまたは、パラホルムアルデヒド(PFA)もしくはブアン(Bouin)溶液中で4℃にて一晩固定してからパラフィンに包埋した。
RANKL、OPG、およびRANKについて、免疫組織化学的に全組織を染色した。手短に述べると、免疫組織化学(IHC)染色は、標準的な間接ペルオキシダーゼ法にしたがって実施し、マイヤーヘマトキシリンで対比染色した。すべての実験は、一次抗体なしで染色した陰性対照とともに実施した。連続切片を使用して、RANKL、RANK、およびOPGの同時発現を調べた。抗原回復は、切片を15分間TEGバッファー(5 L中にTris 6.06 g、EGTA 0.95 g、pH 9.0)中でマイクロウェーブ処理することによって達成された。切片を2%過酸化水素中で洗浄して内在性ペルオキシダーゼをブロックした。その後、すべての切片を2%非免疫ヤギ血清もしくはマウス血清(Zymed Histostain kit, San Francisco,CA, USA)と共にTris緩衝食塩水(TBS)中でインキュベートし、交差反応を最小限にした。一次抗体はSanta Cruz Biotechnology Inc., Santa Cruz, CAから購入した。ウェスタンブロッティングによってすべての抗体をテストしたが、最初の実験は、さまざまな抗体希釈度(1:50から1:500)、さまざまなバッファー(TEG、クエン酸、尿素)、およびさまざまな固定液(ホルマリン、エタノール、および、なし)を用いて行った。4℃にて16時間のインキュベーション後、切片を、ビオチン標識ヤギ抗ウサギIgG(Zymed Histostain kit)とともに、またはビオチン標識ロバ抗ヤギIgGとともに(1:400)インキュベートしてから、ペルオキシダーゼ標識ストレプトアビジン複合体(Zymed Histostain kit)を第3段階として使用した。アミノエチルカルバゾール(Zymed Histostain kit)を用いて可視化を行った。インキュベーションステップの間にスライドをTBSで洗浄した。
ヒト
RNAレベルで、本発明者らは、完全な精子形成を有する正常な精細管を含む精巣において、RANKL、RANK、およびOPGの顕著な発現を見いだした(図1)。RANKLのすべてのアイソフォームが精巣において発現された。セルトリ細胞単独においてはただ1つのアイソフォームが発現されたが、それは卵巣で発現されているアイソフォームとは異なっていた。タンパク質レベルでは、成人セルトリ細胞および雄生殖細胞においてRANKLの発現が見いだされた(図3)。発現は細胞質型および膜貫通型であった。
RNAレベルで、本発明者らは、10週齢および15週齢動物の精巣において、RANKL、RANK、およびOPGの顕著な発現を見いだした(図2)。RANKLのすべてのアイソフォームが精巣において発現された。タンパク質レベルでは、2つの異なるマウス系統の成体セルトリ細胞においてRANKLの発現が見いだされた。発現は細胞質型および膜貫通型の混合であった。染色強度は、非常に弱いものから正常までさまざまであったが、RANKL発現はセルトリ細胞の大半において検出可能であった。RANKLはVdr KOマウスにおいてダウンレギュレートされているとみられたことから、RANKLが精巣においてもビタミンDによって制御されていることがわかる。このことから、存在する主要なRANKLアイソフォームはアイソフォーム1であることがわかるが、それは、RANKLアイソフォーム3が骨においてビタミンDで制御されておらず、Vdr消失後にRANKLが10分の1に減少する事実があるためである。
本明細書では、RANKL、RANK、およびOPGが精巣において発現されることを示している。もっぱらセルトリ細胞において発現されるRANKLのアイソフォームがあると思われるが、一方RANKは生殖細胞で発現され、OPGは精細管周囲細胞において発現される。この発現パターンはマウスからヒトまで保存されており、それはRANKLシグナル伝達が精子形成に関与する可能性を示唆する。精細管周囲細胞およびセルトリ細胞からのシグナル伝達が生殖細胞の運命を決定することが知られており、本発明者らの知る限り、これが、精細管周囲細胞、セルトリ細胞、および生殖細胞に関する最初のシグナル伝達である。

ヒト細胞株
生殖細胞の特性をあまり保っていない、EC由来細胞株NTera2、ならびに生殖細胞の特性を保持している、セミノーマ由来TCam-2細胞株を本研究に使用した。細胞は、グルタミン (58.5 mg/ml)、ペニシリン (100 U/ml)およびストレプトマイシン (100 mg/ml) (すべてGibco)を添加したDMEM (NTera2) またはRPMI 1640 (TCam-2)中で標準条件下37℃にて5% CO2雰囲気において増殖させた。増殖への影響を調べるために、RANKL (100 ng/ml)、CSF-1 (100 ng/ml)、デノスマブ(100 ng/ml)を、単独で、または組み合わせて、培地に添加した。溶媒としてDMSOを使用したので、すべての対照サンプルはDMSO処理した。細胞を25-cm2 フラスコに蒔き、37℃にて5分間0.05%トリプシン-EDTAを用いて剥離した。一部を遠心沈降させ、その細胞ペレットを、RNA精製およびRT-PCR用のcDNA合成のために瞬間凍結した。それぞれのmRNAを標的とする特異的プライマーを用いてqRT-PCRを行った。それぞれのプライマーの組み合わせから得られた代表的なバンドを評価のために配列決定した(Eurofins MWG GmbH,、ドイツ)。
生存分析は、レサズリンの酸化型の減少、ならびに同時に起こる蛍光中間体の増加を測定することによって、生細胞の代謝活性を判定する。細胞はビルケルチュルク(Buerker-Tuerk)計算盤(Tiefe 0.100 mm、0.0025 mm2)にて手作業で計数し、5 × 103 細胞/ウェルを96ウェルプレートのウェルに蒔いて一晩インキュベートしてから処理した。同時処理実験のために、1 × 103 細胞/ウェルを96ウェルプレートに蒔き、一晩インキュベートしてから、選択された化合物100 ng/mlを用いて20時間の最初の処理を行った。陰性対照(培地のみ)および陽性対照(標準培地中1 × 103 細胞)は、すべてのプレート上でインキュベートされた。細胞の生存はTOX-8 in vitro毒性アッセイ(Sigma Aldrich)を用いてメーカーの説明書にしたがって測定した。処理の終了時点でアッセイキットのレサズリン色素を培地に添加した後4-6時間インキュベートした。次にプレートをELISAマイクロプレートリーダー(Tecan, Sunrise)で測定した。パーセント生存率を計算するために、未加工の吸光度値は、基準波長およびブランクサンプル(培地のみ)からの差し引き値とした。その結果得られた値を平均し(それぞれの処理についてn=8)、最終的な吸光度値を生成した。未処理細胞を100%生存とみなし、それを用いて処理細胞のパーセント生存率を算出した。各実験は、独立した実験として少なくとも3回繰り返した。
NTera2細胞は、TCAM2細胞とは異なるRANKLの1つのアイソフォームを発現する。TCAM2細胞で発現されるアイソフォームは、セルトリ細胞唯一症候群(SCO)のセルトリ細胞特異的アイソフォームに類似しており、これは卵巣で発現されているものとは異なる。しかしながら、このアイソフォームは正常な精巣のセルトリ細胞において発現する可能性があり、正常なセルトリ細胞およびSCOのセルトリ細胞が異なるRANKLアイソフォームを有することを除外することはできない。RANKおよびその下流のシグナル伝達機構は2つの細胞株のいずれでも発現されているが、それは、2つの細胞株ともRANKL処理に反応することを意味する(図1)。OPGはもっぱらNTera2細胞で発現されるが、それは、この細胞がRANKLの天然の阻害剤を有し、暴露の増加に適応して感受性が低下している可能性があることを意味するが、一方TCAM2はOPGを発現しない。
上記のデータは、RANKL経路の調節が生殖細胞腫瘍に由来するがん細胞の細胞生存に影響を及ぼすことを明確に示す。RANKLおよびデノスマブ処理はいずれも、細胞生存に顕著な変化を引き起こす。観察された反応の相違の一部は、二つの細胞における異なるRANKLアイソフォームに起因する可能性がある。OPGの存在も異なっているが、これは明らかに結果に影響を与える。生殖細胞増殖の変化はきわめて早く引き起こされ、RANKLとデノスマブの両方が生殖細胞生存に強力で急速な影響を及ぼすことができるという事実を裏付ける。
序論
RANKLシグナル伝達は、骨の恒常性、免疫系、および乳汁分泌にとって重要である。RANKLは、隣接する細胞の膜にある受容体、RANKに特異的に結合する、細胞外リガンドドメインを有する膜貫通タンパク質である。RANKの結合および活性化は、多くの場合NFκBの活性化を介して細胞内シグナル伝達を誘導するが、それは細胞周期に影響を及ぼす。RANKLとRANKとの相互作用は、近くの細胞からのOPG分泌によって調節されているが、それは、OPGがRANKLの細胞外ドメインに結合してRANKの活性化を妨げるからである。したがって、RANKLの生物学的影響は、RANKL、RANK、およびOPGの存在および存在量に左右される。さらに複雑なことには、RANKLは3つの異なるアイソフォーム1、2、3として存在する。1つは膜貫通型、また1つは細胞質型であり、もう1つは分泌型である。異なるアイソフォームは必ずしも同じ影響を及ぼすとは限らないと考えられ、細胞質の非分泌型は概してRANKL発現の阻害物質である。これは、RANKLの阻害がモデルにおける個別アイソフォームの発現に応じて反対の影響を与える可能性があることを意味する。本発明者らは、RANKがもっぱら生殖細胞において発現されることを見いだしたが、したがって生殖細胞は、RANKLおよびデノスマブ処理に直接的に反応することができる唯一の細胞である。RANKLはセルトリ細胞と生殖細胞の両方で発現されるが、OPGは精細管周囲細胞において発現される。本発明者らは最近、器官培養モデルを確立したが、そのモデルにおいて、小さな組織断片を、最適化された培地の「懸滴」中で培養する。この培養法は、ヒト精巣組織、精巣がん患者の精巣摘除検体から得られたCISおよび精巣がん腫瘍を含む組織に適用することができる。本発明者らは、正常な精巣、CISおよびTGCTを含む組織を最大で2週間までの間、よくない変化を観察することなしに培養することに成功した。この器官培養モデルは、動態的および機能的な方法でRANKLシグナル伝達経路のメカニズムについて研究することを可能にするが、そのモデルは組織の完全性を保つので、生殖細胞が体細胞ニッチ(セルトリおよび精細管周囲細胞)に維持されるという利点を有しており、処理実験の間、体細胞との持続的な相互作用を可能にする。デノスマブは、内在的に生成されたOPGのようにマウスRANKLもしくはTRAILシグナル伝達を阻害しないので、ヒトRANKLシグナル伝達の人工的な特異的阻害剤である。サルにおいて、通常使用されるより10倍高用量のデノスマブは、主立った精巣の組織学的異常も、精子の運動性低下も引き起こさなかったが、受精能検査は実施されなかった。
ヒト組織サンプル採取および調製
ヘルシンキ宣言にしたがって、地方倫理委員会からの承認後、コペンハーゲン大学病院(デンマーク)の発達生殖研究部門(Department of Growth and Reproduction)の男性病クリニックから患者を募集した(認可番号H-1-2012-007)。すべての参加者は、精巣がん治療のための精巣摘除術に先だって、説明を受けて同意を与えた。TGCTのために実施された精巣摘除術の検体から成人精巣サンプルを得た。外科的切除後ただちに精巣摘除検体を病理研究部門に送り、腫瘍と肉眼的に正常な部分に分けた。組織の大部分は診断分析のために割り当てられ、残りを研究用とした。研究用に割り当てられたサンプル部分をただちに培地(下記を参照されたい)に入れ、実験室に運んだ。外科的切除から2時間以内に、検体を〜1mm3片にカットし(平均的な精細管は直径150μmである)、DMEM F12、ペニシリン(100U/ml-1)、ストレプトマイシン(100mg/ml-1)、インスリン、トランスフェリン、およびセリン(× 1)を10%ウシ胎仔血清(FBS)または0.1%ウシ血清アルブミン(BSA)のいずれか一方とともに含有する培地での懸滴培養の中に入れた。培養条件最適化の初期段階において、DMEM、RPMI-1640、およびMEMαを含めていくつかの異なる基本培地をテストしたが、生存もしくは組織形態に明白な相違は見られなかった(データ示さず)。すべての細胞培地および追加成分は、BSA (Sigma-Aldrich, Broendby, Denmark)を除いてGibco (Naerum, Denmark)から購入した。培養を始めるために、30μLの培地の液滴をペトリ皿(NUNC細胞培養ペトリディッシュ)の蓋の上に準備した。個々の組織片をそれぞれの液滴中に置いた後、ディッシュを注意深く裏返し、液滴をそのままに維持して組織が浮遊するようにした。脱水乾燥を防ぐためにディッシュの底にDPBS (10ml)を添加した。組織は、5% CO2 中34℃にて最大14日間培養し、培地は48時間ごとに交換した。実験準備には、次に行う:固定、RNA精製、および生存アッセイ用にそれぞれ同じ起源の組織片から切り取った少なくとも3回分の組織片を含めた。培養期間が終了したら、サンプルを、4%パラホルムアルデヒド(PFA)固定液中に集めた後(室温にて30分、4℃にて一晩)組織学的分析および免疫組織化学法のためにパラフィン包埋し、RNA精製およびリアルタイムPCR分析のためにRNAlater(Ambion, Austin, TX, USA)に入れ、または生存分析に備えた。
組織片が生きた細胞を含んでいるかを評価するために、生存分析を使用し、TOX-8 in vitro毒性アッセイ(Sigma Aldrich)を用いてメーカーの説明書にしたがって全代謝活性を測定した。手短に述べると、組織片を90μL培地および10μLレサズリン色素(Sigma-Aldrich)とともに96ウェルディッシュに移し、すべてのプレートに含められた陰性実験対照(培地+レサズリン色素、組織なし)とともに34℃にて5時間インキュベートした。そこで、レサズリン(青)の酸化型の減少、および同時に起こる蛍光中間体(赤)の増加を、メーカーの説明書(Sigma-Aldrich)にしたがってELISAマイクロプレートリーダー(Tecan Sunrise, Maennedorf, Switzerland)で測定した。
簡単に述べると、パラフィン切片を脱パラフィンして再水和した。抗原回復は、切片を15分間、回復バッファー中でマイクロウェーブ処理することによって達成された。次に切片をTris緩衝食塩水(TBS)で希釈した2%非免疫ヤギ血清(Zymed Histostain kit, San Francisco,CA, USA)または0.5%粉乳と共にインキュベートし、交差反応を最小限にした。一次抗体、希釈剤、および回復バッファー。4℃にて16時間および室温で1時間のインキュベーション後、切片を、ビオチン標識ヤギ抗ウサギIgG(Zymed Histostain kit)とともに、またはビオチン標識ヤギ抗マウスIgGとともに(1:400)インキュベートしてから、ペルオキシダーゼ標識ストレプトアビジン複合体(Zymed Histostain kit)を第3段階として使用した。アミノエチルカルバゾール(AEC)(Zymed Histostain kit)を用いて可視化を行い、深紅色を生じた。インキュベーションステップの間にスライドをTBSで洗浄した。陰性対照用に連続切片を処理したが、一次抗体を希釈バッファーのみで置き換えた。対照スライドはいずれも染色を示さなかった。マイヤーヘマトキシリンを用いて対比染色した。連続切片を用いて、PLAP(CISおよびセミノーマ細胞で検出されるシグナル)、KI-67、BrdU取り込み、切断されたカスパーゼ、KIT、およびAP2γの発現を調べた。2人の研究者が独自にすべての染色を評価した。すべての切片はNikon Microphot-FXA顕微鏡(東京、日本)で手動で検査した後、NanoZoomer 2.0 HT (Hamamatsu Photonics, Herrsching am Ammersee, Germany)でスキャンし、ソフトウェアNDPview version 1.2.36 (Hamamatsu Photonics)を用いて解析した。より定量的に免疫組織化学(IHC)染色を評価するために、KI-67-、BrdU-、KIT-、およびAP2γ-染色細胞のパーセンテージを細胞計数によって求めた。増殖マーカーKI-67およびBrdUは、正常な生殖細胞および悪性生殖細胞でのみ発現され、KI-67で薄く染色されたセルトリ細胞はごくまれにしか見られなかった。完全な精子形成を伴う正常な精巣サンプルにおいて、本発明者らは、精原細胞および精母細胞だけを計数した;CISを含有するサンプルにおいて、本発明者らは染色されたCIS細胞の割合も求めた。すべてを数量化するために(KI-67、BrdU、KIT、およびAP2γ)、各患者から得られた、処理あたり3つの組織断片のそれぞれにおいて、最低3.5 mm2の組織を2人の別の研究者が評価した。少なくとも3人の異なる患者から得られた組織片を、溶媒、RANKL、およびデノスマブ処理のために含めた。陽性細胞および陰性細胞をいずれも計数して、染色細胞のパーセンテージを計算した。
培養期間の終了直前にBrdU取り込みアッセイを用いて、増殖する生殖細胞の存在を判定した。BrdU標識試薬(Invitrogen, Camarillo, CA, USA)を培地で1:100に希釈し、組織片はBrdU含有培地で3時間懸濁培養として準備された。次に組織片をPBSで5分間2回洗浄した後、上記のように固定してパラフィン包埋した。BrdUは、免疫組織化学切片において記載したようにBrdU抗体を用いて、免疫組織化学法によって可視化し、陽性染色細胞は増殖していると見なした。
完全な精子形成を含む領域を有する、正常な精巣組織と呼ばれる組織(NT)、およびCIS細胞を含有する精細管を有する精巣組織の全体的な形態は、懸滴組織培養において最大14日間まで維持された。NT組織のヘマトキシリン・エオシン(HE)による染色は、すべての生殖細胞型の存在を実証し、CISマーカーPLAPによる染色は、CIS細胞が存在するかどうかを確認した。7-10日間の培養後、分化の進んだ細胞、特に分裂終了精子細胞、の減少が、一部の患者の組織で観察されたが、これは一貫性のある知見ではなかった。CIS細胞を有する精細管を含む組織において、HEおよびPLAPによる染色は、精細管がおもにCIS細胞(PLAPに陽性)およびセルトリ細胞を含有すること、ならびに組織の形態が最大14日までの培養の間、保存されることを示した。
理論にとらわれず、上記データは、RANKL経路の調節が生殖細胞の生存および増殖に影響を及ぼすことを明確に示している。RANKLおよびデノスマブ処理はいずれも、細胞生存に顕著な変化を引き起こす。観察された反応の相違点の一部は、精巣中に存在するRANKLアイソフォームの違いに起因する可能性がある。生殖細胞増殖の変化は、予想される通りきわめて早く引き起こされ、RANKLとデノスマブの両方が生殖細胞の生存に強力で迅速な影響を及ぼすことができるという事実を裏付ける。
マウスは、薬物治療の薬力学および薬物動態作用を研究するためのモデルとみなされる。残念ながら、デノスマブはマウスRANKLをブロックすることができないので、本発明者らはデノスマブの影響を調べることができなかった。代わりにヒトOPGを使用したが、これは効果的にマウスRANKLの作用をブロックし、すでに実施例4で示したようにデノスマブと同様に作用することができる。
動物
C57BL/6Jマウス、15匹の雄、7-8週齢、Taconic Europe A/Sより。マウスは欧州標準ケージII型のケージに入れた。床敷Jeluxyl HW 300/500 (Jelu, J.Ehrler GmbH & Co KG, Ludwigsmuehle, D-73494 Rosenberg, Germany)。床敷は層流ユニット内で週1回交換した。動物舎では1時間に約12回換気した。温度は20℃から24℃とし、外界換気系によって制御した。光サイクルは12時間の暗期および12時間の明期(6:00に点灯)とした。飼料はAltromin 1324、Altromin(Im Seelenkamp 20, 32791 Lage, Germany)製。水はUV殺菌し、水ボトルは馴化および実験の間、必要なときに補充した。
15匹のマウス(グループ1-3)は無作為に、5匹からなる3群とした。マウスの処理は以下の表に示すように行う。マウスは異常な臨床症状についてモニターした。各投与回の前に体重を測定する。マウスは14日間の投与後に終了した。終了時に動物は頸椎脱臼によって安楽死させた。300μl血液を各動物から採取し、100-150 μl血清を調製する。腎臓および精巣を摘出して秤量する。サンプルは次のように保存した:腎臓サンプル:1つは急速凍結し、1つは緩衝化したホルマリンもしくはPFA中に入れる;精巣サンプル:1つはホルマリンもしくはPFAに入れ、1/2精巣はブアン固定液に入れ、また1/2の精巣は急速凍結する。精巣上体は急速凍結。
すべての動物は、全身状態について毎日検査する。いかなる臨床的兆候も行動異常も記録する。
軽い痛みや中等度の苦痛を越える臨床的兆候を示す動物はいずれも、苦痛を抑えて安楽死させる。これには、動物愛護的(人道的)エンドポイントの限界を超える臨床的兆候を示す動物を含める。動物が研究中に死んでいるのが見つかったり、時期を早めて安楽死させたりした場合、死亡時期をできるだけ正確に記録する。安楽死の前、または死んだ動物を発見した時点で、体重を測定する。できれば、その動物について剖検を実施する。動物の外観および内観、ならびに重要臓器は、剖検計画(‘Post Mortem Examination Schedule’)にしたがって検査して記述し、臓器の重量を記録する。重要臓器は動物ごとに1つのバイアル中でホルマリンに入れて保存する。対照群の動物の終了時に、同じ剖検を対照群の動物に実施する。これはさらなる検討を可能にするはずである。
体重および苦痛
溶媒、RANKL、およびOPGによる処理後に体重の大きな相違はなかった。動物は苦痛の兆候を示さなかった。
男性受精能力の総合評価は行われなかったが、生殖器官(精巣および精巣上体)の重量は、溶媒およびRANKLでそれぞれ処理したマウスよりOPG処理マウスにおいて有意に重かった(図8および9)。それに加えて、RANKLおよび溶媒で処理された10週齢マウスよりOPG処理マウスにおいて、いっそう多くの精細管がより太く見えて(図10上図)、生殖細胞上皮の幅は、OPG処理マウスにおいて精子形成段階と無関係に有意に広かった(図10下図)。OPG処理細胞における生殖細胞の増加は、精巣上体の重量の相違を説明することができるが、それはRANKL処理マウスと比べてOPG処理マウスにおいて多数の精子が蓄積することによって引き起こされると考えられる。
これを確認するために、溶媒処理マウスおよびOPG処理マウスの両方から得られた精巣上体尾部から精子を放出させたが、それは、これらの精子が完全に成熟していて、すぐに射精できる状態であるからであり、したがってこうした精子はその受精能力の間接的なマーカーとなる。本発明者らは、精子濃度に顕著な相違を見いだした。OPG処理マウスは、溶媒処理マウスの13.4 x 106/mlと比べて有意に高い(p=0.02)平均精子濃度25. 3 x 106/mlを示した(図11)。OPG処理マウスと溶媒処理マウスの精子数の著しい相違は、精巣および精巣上体の臓器重量および精巣における生殖細胞層の厚さの有意な相違と合致し、それは、OPG処理マウスの精液の質がより良好であり、その結果としてすぐれた受精能力を有することを強く裏付ける。
理論にとらわれることなく、外来RANKL、OPG、もしくは溶媒で2週間処理されたマウスの生殖器官に関するこの包括的な研究において、本発明者らは、OPG処理によるRANKLシグナル伝達の阻害が精巣および精巣上体の重量および組織に有意な変化をもたらすことを示した。これは、RANKLが精巣において発現されていることを示す上記の先行結果と合致する。それに加えて、上記のin vivoおよびex vivoデータは、デノスマブおよびOPGによるRANKL阻害が結果として生殖細胞の増殖をもたらすことを示す。同様に、OPG処理マウスにおける生殖細胞層は、溶媒処理マウスおよびRANKL処理マウスと比べて有意に厚くなっており、このことは、精巣重量の増加が生殖細胞数の増加に起因することを示している。特に精巣上体重量は、RANKL処理マウスとOPG処理マウスの間で有意差があり、それは、OPG処理マウスと比べてRANKL処理マウスの精巣上体にある精子数が減少していることに起因する可能性がある。こうした結果はやはり、溶媒処理マウスよりOPG処理マウスにおいて精子数が有意に高いことを示す結果と合致し、OPG処理がマウスにおいて受精能力を向上させることを強く裏付ける。

[28] 前記組成物が医薬組成物である、27に記載の組成物。
[29] 前記組成物が、1つもしくは複数の生理的に許容される基剤、賦形剤、および/または希釈剤を含有する、27または28に記載の使用のための組成物。
[30] 前記組成物が、1つもしくは複数の安定化剤、および/または1つもしくは複数の緩衝剤を含有する、27〜29のいずれか1つに記載の使用のための組成物。
[31] 少なくとも1つの安定化剤が界面活性剤である、30に記載の使用のための組成物。
[32] 界面活性剤が、ポリソルベートおよびポリオキシプロピレン-ポリエチレンエステル(Pluronic(登録商標))から選択される、31に記載の使用のための組成物。
[33] 界面活性剤が、ポリソルベート、たとえば、ポリソルベート20およびポリソルベート80から選択されるポリソルベートである、31に記載の使用のための組成物。
[34] 界面活性剤が、0.001-1%の濃度、たとえば0.002-0.5%、たとえば0.004%、または0.01%の濃度で存在する、31〜33のいずれか1つに記載の使用のための組成物。
[35] 前記組成物が、ソルビトール、酢酸塩、水酸化ナトリウム、および注射用水を含有する、27〜34のいずれか1つに記載の組成物。
[36] 前記組成物のpHが、5.2から6.5までの間、たとえば5.5から6.5までの間、たとえば4.5から5.5までの間、たとえば6.3、または、たとえば5.2である、27〜35のいずれか1つに記載の組成物。
[37] 前記組成物が、カルシウムおよびビタミンDから選択される1つもしくは複数の成分を含有する、27〜36のいずれか1つに記載の組成物。
[38] 哺乳動物において男性不妊を治療および/もしくは予防する、ならびに/または、哺乳動物において男性受精能を改善するための方法であって、その方法が、治療上有効な量の、1〜18のいずれか1つに記載の化合物、または26〜36のいずれか1つに記載の組成物を投与することを含む、前記方法。
[39] 前記哺乳動物が、26に記載の被験者である、38に記載の方法。
[40] 前記化合物または組成物が静脈内もしくは皮下に投与される、38または39に記載の方法。
[41] 前記化合物が、皮下に、たとえば4週間に1回、60-120 mgの用量で投与される、38〜40のいずれか1つに記載の方法。
[42] 前記化合物が精巣内に投与される、38〜40のいずれか1つに記載の方法。
[43] 精巣において(生殖)細胞増殖の刺激に使用するための、ならびに/または哺乳動物において男性不妊の治療および/もしくは予防および/もしくは軽減に使用するための、ならびに/または哺乳動物において男性受精能の誘導もしくは改善に使用するための、OPG(オステオプロテジェリン)、OPGを含む融合タンパク質、たとえばFc-OPG、およびヒト免疫グロブリンG1。
[44] 精巣において(生殖)細胞増殖の刺激に使用するための、ならびに/または哺乳動物において男性不妊の治療および/もしくは予防および/もしくは軽減に使用するための、ならびに/または哺乳動物において男性受精能の誘導もしくは改善に使用するための、デノスマブ。
Claims (36)
- 精巣において生殖細胞増殖の刺激に使用するための、ならびに/または哺乳動物において男性不妊の治療および/もしくは予防に使用するための、ならびに/または哺乳動物において男性受精能の誘導もしくは改善に使用するための組成物であって、
ここで前記組成物は化合物を含み、
該化合物はオステオプロテジェリン結合タンパク質(OPGbp)/NFκB活性化受容体リガンド(RANKL)のNFκB活性化受容体(RANK)との結合のアンタゴニストもしくは阻害剤となる化合物であり、
前記化合物は、RANKLもしくはRANKペプチドと免疫反応する抗体、当該抗体の抗原結合ドメイン、当該抗体の抗体断片、もしくは当該抗体の抗原結合断片からなる1群から選択されるものであるか、または、OPG(オステオプロテジェリン)、OPGを含む融合タンパク質、もしくはFc-OPGであり、
前記抗体断片は、RANKLまたはRANKペプチドと免疫反応するものである、
前記組成物。 - 前記化合物が、OPG、OPG(オステオプロテジェリン)を含む融合タンパク質、Fc-OPG、およびヒト免疫グロブリンG1からなる1群から選択される、請求項1に記載の組成物。
- 前記化合物が、ポリクローナル抗体、モノクローナル抗体、重鎖および軽鎖が可動性リンカーによって連結された抗体、Fv分子、抗原結合断片、Fabフラグメント、Fab'フラグメント、F(ab')2 分子、完全ヒト抗体、ヒト化抗体、およびキメラ抗体からなる1群から選択される、請求項1または2に記載の組成物。
- 前記化合物がRANKLシグナル伝達の阻害剤である、請求項1または2に記載の組成物。
- 前記の抗体、抗原結合ドメイン、抗体断片、または抗原結合断片が、
i) 配列番号2または配列番号4に記載のアミノ酸配列;
ii) 配列番号3のヌクレオチド1から951に示すヌクレオチド配列を含有するDNA分子によってコードされるアミノ酸配列;
iii) 配列番号11に記載のアミノ酸配列;
iv) i)-ii)に記載の配列のうちいずれか1つの部分配列であって、配列番号11に記載のRANKポリペプチドに結合することができる前記部分配列;ならびに
v) i)-iii)に記載の配列のいずれかに対して少なくとも90%の配列同一性を有する配列; からなる1群から選択されるアミノ酸配列を含むか、またはそれらで構成される、ペプチドもしくはポリペプチドと免疫反応する、請求項4に記載の組成物。 - 前記部分配列のアミノ末端アミノ酸残基が、配列番号4のアミノ酸残基番号1-162のいずれか1つである、請求項5に記載の組成物。
- 前記部分配列のカルボキシ末端アミノ酸残基が、配列番号4のアミノ酸残基番号313-317のいずれか1つである、請求項5または6に記載の組成物。
- 前記部分配列が、配列番号3のヌクレオチド484-951に示すヌクレオチド配列を含むDNAによってコードされるアミノ酸配列である、請求項5または6に記載の組成物。
- 前記の抗体もしくは抗原結合ドメインが、
i) 配列番号5に記載のアミノ酸配列;および
ii) i)のアミノ酸配列の部分配列であって、当該部分配列もしくはバリアントが配列番号6に記載の配列を含んでなる、前記部分配列
からなる1群から選択されるアミノ酸配列と免疫反応する、請求項3〜5のいずれか1項に記載の組成物。 - 前記の抗体もしくは抗原結合ドメインが、ヒトモノクローナル抗体もしくは抗原結合ドメインである、請求項3〜9のいずれか1項に記載の組成物。
- 前記抗体が全長ヒトモノクローナルIgG2である、請求項3〜10のいずれか1項に記載の組成物。
- 前記抗体が、
i) 配列番号7に記載のアミノ酸配列;
ii) 配列番号7に記載のアミノ酸配列の部分配列;ならびに
iii) i)およびii)に記載のアミノ酸配列のいずれかに対して少なくとも90%の配列同一性を有するアミノ酸配列、
からなる1群から選択されるアミノ酸配列を含む重鎖を有する、請求項3〜11のいずれか1項に記載の組成物。 - 前記の、配列番号7に記載のアミノ酸配列の部分配列が、配列番号8に記載の配列を含んでなる、請求項12に記載の組成物。
- 前記抗体が、
i) 配列番号9に記載のアミノ酸配列;
ii) i)に記載のアミノ酸配列の部分配列;ならびに
iii) i)およびii)に記載のアミノ酸配列のいずれかに対して少なくとも90%の配列同一性を有するアミノ酸配列、
からなる1群から選択されるアミノ酸配列を含む軽鎖を有する、請求項3〜13のいずれか1項に記載の組成物。 - 前記の、配列番号9に記載のアミノ酸配列の部分配列が、配列番号10に記載の配列を含んでなる、請求項14に記載の組成物。
- 前記化合物がデノスマブである、請求項1に記載の組成物。
- 前記組成物が、精子減少症の予防および/または治療のための組成物である、請求項1〜16のいずれか1項に記載の組成物。
- 前記組成物が、無精子症の予防および/または治療のための組成物である、請求項1〜17のいずれか1項に記載の組成物。
- 前記化合物が、静脈内または皮下に投与される、請求項1〜18のいずれか1項に記載の組成物。
- 前記化合物が、皮下に、投与される、請求項1〜19のいずれか1項に記載の組成物。
- 前記化合物が、皮下に、4週間に1回、60-120 mgの用量で投与される、請求項20に記載の組成物。
- 前記化合物が、精巣内または性器部に投与される、請求項1〜21のいずれか1項に記載の組成物。
- 前記化合物が陰嚢部に投与される、請求項1〜22のいずれか1項に記載の組成物。
- 当該被験者の精巣組織サンプルの組織学的分析によって検出することができる、いかなるステージの精巣がんにも罹っていない男性被験者、または当該被験者の精巣組織サンプルの組織学的分析によって検出することができる、いかなるステージの精巣がんにも罹っていない年齢15-55歳のヒト男性被験者に投与されるためのものである、請求項1〜23のいずれか1項に記載の組成物。
- 前記組成物が医薬組成物である、請求項1〜24のいずれか1項に記載の組成物。
- 前記組成物が、1つもしくは複数の生理的に許容される基剤、賦形剤、および/または希釈剤を含有する、請求項1〜25のいずれか1項に記載の組成物。
- 前記組成物が、1つもしくは複数の安定化剤、および/または1つもしくは複数の緩衝剤を含有する、請求項1〜26のいずれか1項に記載の組成物。
- 前記1つもしくは複数の安定化剤が界面活性剤である、請求項27に記載の組成物。
- 界面活性剤が、ポリソルベートおよびポリオキシプロピレン-ポリエチレンエステルから選択される、請求項28に記載の組成物。
- 界面活性剤が、ポリソルベート、または、ポリソルベート20およびポリソルベート80から選択されるポリソルベートである、請求項28に記載の組成物。
- 界面活性剤が、0.001-1%の濃度、0.002-0.5%、0.004%、または0.01%の濃度で存在する、請求項28〜30のいずれか1項に記載の組成物。
- 前記組成物が、ソルビトール、酢酸塩、水酸化ナトリウム、および注射用水を含有する、請求項1〜31のいずれか1項に記載の組成物。
- 前記組成物のpHが、5.2から6.5までの間である、請求項1〜32のいずれか1項に記載の組成物。
- 前記組成物が、カルシウムおよびビタミンDから選択される1つもしくは複数の成分を含有する、請求項1〜33のいずれか1項に記載の組成物。
- 精巣において生殖細胞増殖の刺激に使用するための、ならびに/または哺乳動物において男性不妊の治療および/もしくは予防および/もしくは軽減に使用するための、ならびに/または哺乳動物において男性受精能の誘導もしくは改善に使用するための組成物であって、OPG(オステオプロテジェリン)、またはOPGを含む融合タンパク質を含む前記組成物。
- 精巣において生殖細胞増殖の刺激に使用するための、ならびに/または哺乳動物において男性不妊の治療および/もしくは予防および/もしくは軽減に使用するための、ならびに/または哺乳動物において男性受精能の誘導もしくは改善に使用するための組成物であって、デノスマブを含む前記組成物。
