Description
i)全ての、または実質的に全てのhTERTのエピトープを任意の順序でコードする、および、

[定義]
テロメラーゼ複合体は、RNAテンプレートと、「テロメラーゼ逆転写酵素」(TERT)と呼ばれ、テロメラーゼ活性の主要な決定因子である逆転写酵素を含むタンパク質構成要素とから成る。特記されない限り、本明細書において、「テロメラーゼ」という用語はTERTを指し、野生型のヒトテロメラーゼ、またはその変異型を含む。野生型ヒトテロメラーゼ(すなわちhTERT)は知られており(GeneBank アクセッション番号 NM_198253)、配列番号2のアミノ酸配列を持つ(cDNAは配列番号1に記載)。

[プラスミドDNAベクター]
[INVAC-1]
INVAC-1は7120bpのプラスミド発現ベクターであり、127.4kDaのタンパク質に対応する、1158AA(Ubi-hTERT)のヒトユビキチン−テロメラーゼ融合構築物をコードしている(図1Aおよび16)。INVAC-1はヒトでの使用を意図されているため、テロメラーゼ逆転写酵素の酵素活性は安全上の理由から不活性化されている。実際に、INVAC-1がコードするヒトTERT配列は、3つのアミノ酸、バリン-アスパラギン酸-アスパラギン酸(867-869AA)をコードする9bpの欠失によって触媒部位が改変されている(図2A)。加えて、テロメラーゼの細胞内局在化に必要な核小体局在化配列(NoLS)(Yang、2002)を含む、タンパクのN末端部位の47AAは、ユビキチン(Ubi)コード配列(1-76AA)によって置換されている。
Ubi-hTERT遺伝子は、重複する40塩基長のオリゴヌクレオチドアセンブリープロセス(GeneCust、Luxembourg)によって新規合成した。制限酵素部位を削除し、GCリッチ配列を弱めるために、いくつかの保守的な塩基改変を行った。インサートは、HindIII-XbaIクローニングサイトを用いて発現ベクターpcDNA3.1(+)(Invitrogen、Carlsbad、USA)にクローニングし、配列決定により確認した。
ユビキチン-テロメラーゼインサートを、NTCによって作成されたNTC8685-eRNA41H-HindIII-XbaI発現ベクターに導入した。しかし、最適なベクターNTC8685-eRNA41H(NTC-DV8685-41HLV参照)はUbi-hTERTインサートと適合する制限酵素サイトを持っていなかった。従って、当該ベクターをSalIおよびBglIIで切断し、Ubi-hTERTをサブクローニングするのに適切な制限酵素サイト、すなわちHindIII-XbaIを含む合成二本鎖オリゴヌクレオチドを連結させた:

INVAC-1は、研究グレードの品質条件下で、NTCによって最初に作られた。プラスミドDNAは、NTC4862大腸菌(E.coli)細胞に、エレクトロポレーションを用いて形質転換した。細胞は、製造業者によって推奨された通り(NTC Instruction Manual、June2011)、6%スクロース培地に蒔き、培養した。抽出後、プラスミドDNAはエンドトキシンフリーの1×PBSに終濃度2mg/mlで再懸濁した。
全てのINVAC-1誘導体構築物は、約8.9kbの二本鎖DNAプラスミドで、酵素機能が不活性化したヒトユビキチン−テロメラーゼ融合タンパク質をコードしている(図2A)。Ubi-hTERT導入遺伝子は、哺乳類の細胞で高レベルに安定かつ一過的な発現をするように作成された、pcDNA3.0由来のInvitrogen pcDNA3.1(+)ベクター(5.4kb)に導入した。当該ベクターは、ヒトサイトメガロウイルス最初期(CMV-IE)プロモーターおよびウシ成長ホルモンポリアデニル化(BHG-polyA)を、終止配列として含む。
hTERTコード配列は、pcDNA3.1プラスミド主鎖のヌクレオチド923-4492bpに位置する。pUTD10Notは、分子量約130.8kDaに相当する1189AAヒトユビキチン−テロメラーゼ融合タンパク質(Δ10Not)をコードしている(図2A)。hTERTは、最初の23アミノ酸(1-23AA)を、ユビキチンポリペプチド(76AA)によって置換することで欠失させた。触媒部位のドメインでは、野生型hTERT(アクセッション番号NM_198253)のAA860-869(DGLLLRL「VDD」_配列番号21)に対応するアミノ酸912-913(*マーク;図17)の間にさらなる欠失を導入した。この10アミノ酸の欠失は、hTERTの酵素活性の不活性化をもたらす3AAの欠失(ΔVDD)、およびVDD配列の7AA上流のさらなる欠失を含む。Ubi-hTERTのC末端配列にある14アミノ酸はV5エピトープタグをコードしている(図2A)。

全てのINVAC-1誘導体は、RD Biotech(Besancon、France)によって、大腸菌(E.coli)5-アルファ細胞(fhuA2Δ(argF-lacZ)U169 phoA glnV44 Φ80Δ(lacZ)M15 gyrA96 recA1 relA1 endA1 thi-1 hsdR17)(Lucigen Corporation、Middleton、USA、60602-2参照)に形質転換され、製造された。無菌の1×PBSに再懸濁した、濃縮されたエンドトキシンフリーギガプレッププラスミドストック(2mg/mL)を調製した。ベクターは、制限酵素マッピングによって確認した(HindIII-XbaI;図2B)。
pTRIP-CMV-hTERTは、1232AA(約124.5kDaに相当する)の触媒活性を持つ野生型ヒトtTERT(hTERT)タンパク質をコードする。当該プラスミドは、インビトロアッセイのポジティブコントロールとして使用した。構築物は特許出願WO2007/014740に最初に記載された。pTRIP-CMV-hTERTは、初めに、pBABE-hygro-hTERTプラスミド(Dr.Robert Weinbergのご厚意で譲渡された)由来のEcoRI-SalIhTERTインサートをpSP73ベクター(Promega Life Science、Wisconsin、USA)にサブクローニングして、pSPhTERT構築物を作成することで構築した。その後、BglII-SalIフラグメントをBamHIおよびXhoIで切断したレトロウイルス由来のpTRIP-CMVベクターに挿入し、pTRIP-CMV-hTERTを作成した。hTERTの発現は、ヒトサイトメガロウイルス(CMV)プロモーターによって開始される。
pNTC-hTERTは、触媒活性を持つ1132AA野生型ヒトTERT(hTERT)タンパク質をコードする(配列番号2)。当該プラスミドは、hTERT特異的なインビボでのT細胞応答の幅を、INVAC-1構築物と比較して調査するために使用した。
pNTC-hTERT-ΔVDDは、バリン-アスパラギン酸-アスパラギン酸をコードする9bpを欠失(ΔVDD;867-869AA)することによって触媒部位を改変した1129AAヒトTERT(hTERT)配列をコードする。当該プラスミドは、hTERT特異的なインビボでのT細胞応答の幅を、INVAC-1構築物と比較して調査するために使用した。
CrFK細胞(クランデルリース(Crandell Rees)ネコ腎細胞)、HEK293T細胞(ヒト胚性腎細胞)およびHeLa細胞(ヘンリエッタラックスヒト子宮頸部腺癌細胞)は、10%熱失活ウシ胎児血清(PAA、Velizy-Villacoublay、France)および1%ペニシリン/ストレプトマイシン(Life Technologies、Saint-Aubin、France)を追加したダルベッコ変法イーグル培地(DMEM)で培養した。
ウエスタンブロット解析のため、遺伝子導入したCrFKおよびHEK293T細胞は、プロテアーゼ阻害剤混合物(Roche Diagnostic、Indianapolis、USA)を追加したRIPAバッファー(Sigma-Aldrich、St.Louis、USA)で、10-20分氷上にて溶解した。溶解液は14,000rpmで15分、4℃にて遠心分離することによって清澄にした。上清を回収し、ブラッドフォード比色法を用いて、タンパク質濃度を測定した。タンパク質サンプルを95℃で5分変性させ、Nu-PAGE(登録商標)Novex4-12%Bis-Trisゲル(Invitrogen、Carlsbad、USA)で分離し、iBlot(登録商標)装置(Invitrogen、Carlsbad、USA)を使ってPVDF膜(iBlot(登録商標)transfer stack、Invitrogen、Carlsbad、USA)にエレクトロブロットした。Novex(登録商標)Sharp Prestained Protein Ladder (Invitrogen、Carlsbad、USA)を、分子量決定のために使用した。PVDF膜は約60kDaで切断し、1×PBS、0.05%Tween(登録商標)20、3%ミルクでブロックした。PVDF膜の上部は、ブロッキングバッファーで1/2000に希釈した抗hTERTウサギモノクローナル抗体(Abcam、Cambridge、UK)、または1/5000に希釈した抗V5マウスモノクローナル抗体(Invitrogen、Carlsbad、USA)で探索した。PVDF膜の下部は、1/5000に希釈した抗-β-アクチンマウスモノクローナル抗体(Sigma Aldrich SARL、Saint-Quentin Fallavier、France)で探索した。最後に関連タンパク質を、適切な西洋ワサビペルオキシダーゼ(HRP)結合型二次抗体、すなわち1/5000に希釈した抗マウスHRP結合抗体(GE Healthcare、Velizy、France)またはブロッキングバッファーで1/1000に希釈した抗ウサギHRP結合抗体(GE Healthcare、Velizy、France)で室温にて1時間染色することにより、可視化した。免疫ブロットシグナルは、ECL HRP化学発光基質試薬キットを用いた高感度化学発光アッセイによって検出した。フィルムおよび対応するカセットは、GE Healthcare(Buckinghamshire、UK)から購入した。
テロメラーゼ活性は、製造者の説明書に従い、TeloTAGGG Telomerase PCR ELISAPLUS kit(Roche Diagnostic GmbH Mannheim、Germany)を用いるテロメア反復増幅プロトコル(TRAP)の手法(Kim et al.1994)を用いて評価した。上述したように遺伝子導入後24時間後に、CrFK細胞を回収した。細胞は1×PBSで洗浄した後、1600rpmで5分、4℃にて遠心分離した。細胞は、0.2mlの溶解バッファーに再懸濁し、氷上で30分間培養した。溶解液は14,000rpmで20分、4-8℃にて遠心分離することによって清澄にした。上清を回収し、ブラッドフォード比色法を用いて、タンパク質濃度を測定した。上清はテロメラーゼが媒介するテロメア配列の伸長に使用し、伸長産物を、ビオチン化プライマーを使用したPCRによって増幅した。各細胞上清はTRAPアッセイを行う前に、事前に二つに分割した。一方は、85℃10分のテロメラーゼの熱失活によってネガティブコントロールを調製するために使用し、他方は、テロメラーゼが媒介するテロメア配列の伸長を評価するために使用した。さらに、反応混合液中に存在する216bpの長さの内部標準を、溶解液中に存在するであろうTaqDNA-ポリメラーゼ阻害剤による偽陰性の結果を排除するために、同時に増幅した。溶解バッファーをネガティブコントロールとして使用した。全ての反応混合液を25℃で20分加温した後、94℃で5分加温した。テロメラーゼ産物はPCRにより、94℃30秒、50℃30秒、72℃90秒、72℃10分を30サイクル、最後に72℃10分および4℃保持を1サイクル行い、増幅した。

CrFK、HEK293T、HeLaおよびQT6細胞を8穴のLab-Tek(登録商標)チャンバースライド(Sigma-Aldrich、St.Louis、USA)に、200μlの培養培地中で、各ウェル当たり2×104個細胞を播種し、37℃、5%二酸化炭素の環境で一晩加温した。翌日、培養培地を廃棄し、新しい培地を200μl添加した。0.2μgのINVAC-1、pTRIP-CMV-hTERTまたはコントロールのエンプティープラスミドpNTC8685-eRNA41Hを含む混合溶液10μlと、OptiMEM(Life Technologies、Saint-Aubin、France)のFugeneHD(Promega France、Charbonnieres-les-bains、France)0.5μlとを、対応するチャンバーに加えた。チャンバー当たり2x104個の未処理の細胞をネガティブコントロールとして使用した。チャンバースライドは24時間、および48時間、37℃、5%二酸化炭素の環境で加温した。形質導入した細胞は、1×PBSで丁寧に洗浄し、細胞を固定化し透過処理するために、各ウェルに200μlの2%PFAを4℃で10分添加した。その後、ウェルを1×PBS 0.05% Tween(登録商標)20で2回洗浄し、200μlのブロッキング溶液(1×PBS 0.05% Tween(登録商標)20に溶解した、0.5% Triton X100;Sigma-Aldrich、3% BSA;Sigma-Aldrich、10%ヤギ血清;Invitrogen)と室温で30分加温した。ブロッキングバッファーで1/100に希釈した抗hTERTウサギモノクローナル一次抗体(Abcam、Cambridge、UK)を、細胞に添加し、室温で1.5時間攪拌した。1×PBS 0.05% Tween(登録商標)20で3回洗浄後、ブロッキング溶液に1/500に希釈したヤギ由来抗ウサギAlexaFluor488(登録商標)二次抗体(Life Technologies、Saint-Aubin、France)を添加し、室温で45分攪拌した。ウェルは1×PBS 0.05% Tween(登録商標)20で3回洗浄し、DAPIを含むVECTASHIELD(登録商標)をマウントさせた培地に乗せた。カバーガラスは、画像処理および画像解析システム(Axiovision、Carl Zeiss MicroImaging GmbH、Jena、Germany)を備えた蛍光顕微鏡(Axio observer Z1、Carl Zeiss MicroImaging GmbH、Jena、Germany)で解析した。
雌のC57BL/6マウス(6〜8週齢)は、Janvier研究室(Saint-Berthevin、France)から購入した。
HLA-B*0702、HLA-A*0201またはHLA-DR拘束性のhTERTペプチドは以前に報告されている(表1の参考文献を参照)。H2-DbおよびH2-Kb拘束性のhTERTペプチドは、4つのオンラインで利用可能なアルゴリズム、Syfpeithi(http://www.syfpeithi.de/)、Bimas(http://www-bimas.cit.nih.gov/)、NetMHCpanおよびSMM(http://tools.immuneepitope.org/main/)を用いて、マウスMHCクラスI分子と結合させるために、インシリコのエピトープ予測によって決定した。全ての合成ペプチドはProimmune(Oxford、United Kingdom)から、凍結乾燥(90%を超える純度)の状態で購入した。凍結乾燥ペプチドは滅菌水に2mg/mLとなるよう溶解し、使用前は−20℃で保管した。ペプチド配列およびMHC拘束性の詳細は、表1に記載した。
凍結乾燥hTERTペプチド(70%を超える純度)は、GenScript(USA)から購入した。当該セットは、重複する11AAを含み、INVAC-1 hTERTのタンパク質全ての配列をカバーする、15AAから成る269のペプチドによって構成されている。各ペプチドは、供給元の推奨に従い使用前に2mg/mlの滅菌水に再懸濁し、使用まで−20℃で凍結させた。9-10のhTERT重複ペプチドから成る27プール(表2)を、IFNγELIspotアッセイにてhTERT特異的T細胞応答の幅を調べるために用いた。
INVAC-1が媒介する抗腫瘍効果を評価するために用いるSarc-2腫瘍細胞株は、自然発症の線維肉腫であるHLA-A2/DR3マウスから取得した。腫瘍マウスは、無菌状態に分離され、初代細胞の懸濁液を産生した。細胞株は、HLA-A*0201分子の発現を示した。細胞は、10%FBS(Life Technologies)および1%ペニシリン/ストレプトマイシンを添加したRPMIグルタマックス培地(Life Technologies)で培養した。
皮内(ID)免疫化は、剃毛後、マウス側腹部下部に、インスリンシリンジおよび特異的針(U-100、29GX1/2’’−0.33×12mm、Terumo、Belgium)を用いて行った。剃毛後、免疫化の間および免疫化後に、赤斑は観察されなかった。筋肉内免疫化(IM)もまた、インスリンシリンジおよび特異的針U-100を使用して、前脛骨筋(anterior tribialis cranialis)に行われた。皮下免疫化(SC)もまた、インスリンシリンジおよび特異的針U-100を使用して、尾底部に行った。各マウスは、経験に基づき、12.5、25、50、100、200、400、800または1200μgのDNAまたは1×PBSに相当する、プラスミド(INVAC-1、NTC、pUTD10Not、pUTD10CogまたはpUTD23Tyn)による、予備刺激のIM、IDまたはSC注射を受けた。ワクチンレジメンに従い、マウスは同様のDNAまたは1×PBSによる二次注射または三次注射を受けることができた。
免疫化したマウスの脾臓を採取してすりつぶし、細胞懸濁液を70mmのナイロンメッシュ(Cell Strainer、BD Biosciences、France)に通して脾細胞を単離した。免疫化したマウスの血液を、麻酔下で後眼窩穿刺によって回収し、末梢血単核球(PBMC)を単離した。脾細胞またはPBMCは、フィコール(リンパ球分離培地、Eurobio、France)を用いて精製した。血液または脾臓からフィコール精製したリンパ球はCellometer(登録商標)Auto T4 Plus counter(Ozyme、France)を用いて数えた。
標的細胞を調製するために、高(5μM)、中(1μM)または低(0.2μM)濃度のCFSE(Vybrant CFDA-SE cell-tracer kit;Life Technologies、Saint-Aubin、France)を含む1×PBS溶液によって、無感作のHLA-B7マウス由来の脾細胞を標識した。5μMおよび1μMのCFSEで標識した無感作の脾細胞を、5μg/mLの2つの異なるHLA-B7ペプチド、すなわち1123および351で、室温で1.5時間パルスした。低濃度のCFSEによって標識した脾細胞は、パルスしなかった。すでにINVAC-1または1×PBSによってワクチン投与された各マウスは、初回注射の14日後または追加注射の10日後に、後眼窩の血管を通して、各フラクションから、同じ数の細胞を含む107CFSE標識細胞の混合物を受け取った。15-18時間後、脾臓由来単一細胞懸濁液を、MACSQUANT(登録商標)フローサイトメーター(Miltenyi、Germany)を用いたフローサイトメトリーによって解析した。

ワクチン投与したHLA-A2/DR1マウス由来の脾細胞(6x105細胞)を、HLA-DR拘束性hTERT由来ペプチド(578、904、および1029)5mg/mlと、37℃で24時間培養した。サイトカイン培養上清を回収し、試験まで−20℃で凍結させた。市販のキットである、マウスTh1/Th2/Th17 Cytometric Beads Array(CBA、BD Biosciences)キットを使用して、IL-2、IFNγ、TNFα、IL-4、IL-6、IL-17aおよびIL-10の各濃度を定量した。CBA免疫測定法は、製造元の説明書に従って実施した。フローサイトメトリーの取得は、FACScan LSRII フローサイトメトリー(BD Biosciences)を使用して行った。解析は、FCAP Array TM Software version 3.0(BD Biosciences)を用いて行った。
治療的ワクチン投与実験のために、24週齢のHLA-A2/DR1マウスに2.104個のSarc-2細胞を、右腹側部に皮下移植した。その後、移植4、21、35日後に、上述したようにマウスをDNAワクチンによってID経路で免疫化した後、エレクトロポレーションした。2〜3日ごとに、腫瘍の成長を、カリパスを使ってモニターした。マウスの体重もまた、2〜3日ごとにモニターした。マウスは、腫瘍が2000mm3に達した時点で、安楽死させた。癌研究における動物の愛護と使用のためのガイドライン、特に、直ちに治療介入を必要とする臨床徴候のモニターのためのガイドライン(Workman et al.2010、BJC)に従った。腫瘍の体積は、次式を用いて計算した。:(L*l2)/2。結果はmm3で表した(L=長さ;l=幅)。
Prism5ソフトウエアを、データ処理、解析およびグラフ化に使用した。データは、平均値±標準偏差または中央値として表した。ELIspotアッセイの統計解析は、マン・ホイットニーのノンパラメトリック検定および/またはダンの多重比較検定を伴うクラスカル・ワリス検定を用いて行った。有意差は、危険率<0.05で設定した。
[INVAC-1プラスミドDNAの特徴および配列解析]
様々な制限酵素を用いた制限酵素マッピングによって示されるように、Ubi-hTERT導入遺伝子をpNTC8685-eRNA41H-HindIII-XbaIに挿入することに成功した(図1Aおよび1B)。結果として得られたpNTC8685-eRNA41H-HindIII-XbaI-Ubi-hTERT(INVAC-1)ベクターはまた、pVAC5’およびpVAC3’プライマーを用いて、接合部位において部分的に配列決定した。当該配列により、クローニング過程が成功したことが確認された。
異なるUbi-hTERT融合タンパク質を発現する3つのINVAC-1誘導体DNAプラスミドを、合成してクローン化した(図2A)。全てのUbi-hTERT導入遺伝子は、HindIIIおよびXbaIによる切断と電気泳動(図2B)によって示されるように、pcDNA3.1(+)Invitrogen発現ベクターに、正常に連結された。インサートおよび連結部は、当該DNAインサートと隣接するベクター配列を照合するPEGFP-N5’およびBGHプライマーを用いて、配列決定した。配列決定の結果によって、導入遺伝子が正常にクローニングされたことを確認した(配列番号13、15、17および図17〜19)。
インビトロでのHEK293TおよびCrFK細胞株への一過性の遺伝子導入の18時間から96時間後に、野生型hTERT、INVAC-1およびINVAC-1誘導体タンパク質の全体的な発現に関する情報を提供するために、ウエスタンブロットアッセイを行った。野生型hTERTタンパク質のバンドは、未修飾のhTERTのサイズである124.5kDa(図3Aおよび3Bの左部分)に一致した。野生型hTERTタンパク質の発現は、特にHEK293T細胞において、全時間帯に渡って安定であると思われた。対照的に、INVAC-1(図3Aおよび3Bの右部分、および図3Cの上部分)およびINVAC-1誘導体タンパク質(図3Cの下部分)は全時間帯に渡って速やかに分解された。
INVAC-1誘導体hTERTタンパク質の分解を促進させるために当該タンパク質を非局在化させる考えで、核小体局在化シグナル(hTERTのN末端部分)を除去した。従って、INVAC-1がコードするhTERTの細胞内局在化を、CrFK、HEK293、HeLa、QT6細胞株への遺伝子導入後に、免疫蛍光解析によって評価した(図4)。

ヒトテロメラーゼは、腫瘍細胞の不死化に関与し、老化を防ぐことによって、腫瘍細胞の増殖において重要な働きを果たしている。従って、野生型テロメラーゼのワクチン製品としての利用は、安全への懸念につながり得る。
hTERT特異的T細胞応答の強度を、皮内経路でINVAC-1を事前に免疫化した後に皮膚エレクトロポレーションを行った、または行わなかったC57BL/6マウスにおいて評価した(図6)。免疫注射の14日後に、マウスの脾臓を回収し、H2拘束性のhTERTペプチドを用いたIFN-γELIspotアッセイによって、誘発された免疫応答をモニターした。IFNγ+CD8T細胞の出現頻度の顕著な違いが、INVAC-1のID注射後にエレクトロポレーションを行ったマウス群と、受けなかった群(p<0.05)の間で見られた。従って、本結果は、INVAC-1のワクチン皮内投与後のhTERT特異的CD8T細胞応答を顕著なレベルに誘発する上で、エレクトロポレーションが有利であることを示す。
従来のワクチンは、一般的にSCまたはIM経路で投与されている。しかしながら、皮内経路の免疫化は、近年ワクチン投与の分野で再度注目を集めている(Combadiere and Liard、2011)。従って、ID経路をINVAC-1の投与に試し、また従来のSCおよびIM経路と比較した。
試験した別の重要なパラメーターは、hTERT特異的CD8T細胞応答における、ワクチン投与量の影響であった。C57BL/6マウスを、INVAC-1の投与量を増やして、両側腹部下部に、エレクトロポレーションを伴って、ID経路で免疫化した。ワクチンの容量は、50μL/部位の一定のままであった。マウスは、最後に受けたワクチン投与量に応じて、2カ所または4カ所の部位でワクチン投与した。ワクチン投与/エレクトロポレーションの14日後に、マウスの脾臓を回収し、H2拘束性のhTERTペプチドを用いたIFN-γELIspotアッセイによって、特異的細胞免疫応答をモニターした。
従来のワクチン(BCG、麻疹、インフルエンザなど)に推奨されるたいていのワクチン投与プロコトールは、ワクチン特異的免疫応答の出現頻度を改善する目的で、プライムブーストレジメンを含む。従って、hTERT特異的CD8T細胞応答の発生におけるプライムブーストレジメンの影響を、NVAC-1ワクチンID投与およびエレクトロポレーションで試験した。当該目的のために、遺伝子導入HLA-B7マウスをINVAC-1によるID投与で免疫化し、皮膚のワクチン投与部位をワクチン投与直後にエレクトロポレーションした。最初の免疫化の21日後に、マウスは、同じワクチン投与手順を用いて、二回目のINVAC-1注射を接種した。HLA-B7拘束性のhTERTペプチドを用いたIFN-γELIspotアッセイにより、hTERT特異的CD8T細胞応答をモニターする目的で、プライムブースト免疫化の後いくつかの時点で、末梢血液を採取した(図9)。hTERT特異的CD8T細胞応答のピークが初回投与の14日後に観察された。しかしながら、ワクチン投与したマウスのグループにおけるhTERT特異的CD8T細胞の出現頻度の中央値は、相対的に低く(11.3スポット/200,000個のPBMC)、ワクチンに応答しないマウスが5匹中2匹存在した。追加免疫の後、hTERT特異的CD8T細胞のピークが注射の10日後に観察された。当該時点(初回投与の31日後、追加投与の10日後)のワクチン投与したマウスのグループにおけるhTERT特異的CD8T細胞の出現頻度の中央値は、免疫前のサンプルにおけるhTERT特異的CD8T細胞の出現頻度の中央値と顕著に異なった(p<0.05)。追加投与後は、5匹中4匹の応答マウスが存在した。
INVAC-1の開発と共に、他の3つのDNAプラスミド構築物(INVAC-1誘導体)を設計した:Δ10Not(pUTD10Not)、Δ10Cog(pUTD10Cog)またはΔ23(pUTD23Tyn)。3つの欠失は、hTERT酵素の触媒部位で行われた。これらの欠失は10-23アミノ酸残基に渡り、バリン−アスパラギン酸−アスパラギン酸(Val-Asp-Asp、または1文字表記ではVDD)の3つの決定的な残基に及んだ(図2A)。当該構築物は、酵素活性を取り除くいずれの欠失も、免疫原性を保持し得ることを示すために、設計された。
INVAC-1構築物内で操作されたhTERT配列の修飾、すなわち、(1)核小体局在化シグナルの欠失、(2)ユビキチン配列の付与、(3)触媒部位内の欠失、の影響を、hTERTに対するT細胞免疫応答のレパートリーにおいて、評価した。INVAC-1のhTERT特異的細胞免疫応答を、INVAC-1によるID免疫化/エレクトロポレーションの後に評価し、天然/野生型のヒトTERT配列をコードするDNA(pNTC-hTERT)およびVDD領域のみ欠失したhTERT配列をコードするDNA(pNTC-hTERT-ΔVDD)によって誘発された応答と比較した。コントロールマウスは、25μgのpNTCエンプティーベクターでID注射を接種した後、エレクトロポレーションした。
抗腫瘍免疫応答に関連している免疫細胞の中で、細胞傷害性CD8Tリンパ球(CTL)およびTh1CD4T細胞が、最も強力なエフェクター細胞(Vesely et al.、2011)(Braumuller et al.、2013)として同定されている。
ここまでの結果で、エレクトロポレーションを伴うINVAC-1のID注射が、マウスにおいて細胞傷害性CD8T細胞およびTh1CD4T細胞を誘発できたことが示された。従って、次のステップは、INVAC-1ワクチン皮内投与およびエレクトロポレーションによって与えられた遺伝子導入HLA-A2/DR1マウスの防御作用を、Sarc-2(線維肉腫)腫瘍細胞接種後に評価することであった。最初の試みとして、遺伝子導入HLA-A2/DR1マウスに、プライムブースト戦略で、INVAC-1によってエレクトロポレーションを伴うIDワクチン投与を、あるいは、PBSでニセのワクチン投与を行った。予防的なワクチン投与の一ヶ月後に、50,000個のSarc-2細胞をSC経路でマウスに負荷した。腫瘍体積は2〜3日ごとに計測した。図13Aは、マウスの処置に従った腫瘍細胞負荷後の腫瘍体積の中央値の動態を示している。次いで、腫瘍成長遅延(TGD)は500mm3で計算した。当該判断基準は、コントロールグループと処置グループとで、規定の腫瘍に達する時間を比較することにより、腫瘍成長におけるワクチン処置の効果を計ることを可能にする。11日の腫瘍成長遅延が、INVAC-1をワクチン投与したマウスグループと、PBSを投与したマウスグループとの間で観察された。従って、INVAC-1による予防的なワクチン投与は、腫瘍成長を遅らせる原因であった。腫瘍接種は、最後のワクチン投与の1ヶ月後に行われたため、抗腫瘍効果は、ある程度、hTERT特異的記憶T細胞の存在に起因しているかもしれない。
、21日目と35日目に、同様の手順で、二回の追加ワクチン投与を行った。500mm3において腫瘍成長遅延を計算した。INVAC-1でワクチン投与したマウスグループと、NTCエンプティープラスミドを投与したマウスグループとの間で、4日の腫瘍成長遅延が観察された。結論として、INVAC-1による治療的ワクチン投与は、比較的弱いが、それにもかかわらず繰り返し観察される、腫瘍成長の遅延を可能にした。
様々なサイトカインが、これまで、動物モデルおよびヒトの両方の、抗癌ワクチン投与の研究において、抗原認識およびT細胞増殖を促進させる免疫調製物質として使用されてきた。最も頻繁に使用されるサイトカインの一つは、GM-CSF(顆粒球マクロファージコロニー刺激因子)である。当該サイトカインは、抗原提示細胞の成熟を助け、Th1細胞免疫応答を支持することが知られている(Parmiani et al.、2007)。抗腫瘍ワクチンに関してGM-CSFが果たす主な役割に関して、INVAC-1ワクチンID投与およびエレクトロポレーション後のhTERT特異的T細胞応答に対するマウスGM-CSF(mGM-CSF)の追加の影響を試験した。当該目的のために、C57BL/6マウスに、エレクトロポレーションを伴うID経路でのINVAC-1ワクチン投与の18時間前に、mGM-CSFのID注射を接種した(図14A)。別のマウスのグループは、mGM-CSFを接種することなく、INVAC-1/エレクトロポレーションでIDワクチン投与した。コントロールマウスはPBSによってニセのワクチン投与行い、エレクトロポレーションした。注射の14日後に、マウス脾臓を回収し、H2拘束性のhTERTペプチドを用いたIFN-γELIspotアッセイによって、誘発された免疫応答をモニターした。INVAC-1のID注射の前にmGM-CSFを接種したマウスのグループと、mGM-CSFを接種しなかったマウスのグループの間には、IFNγ+CD8T細胞の出現頻度において、有意な違いが観察された(p<0.001)。従って、mGM-CSFの接種は、hTERT特異的CD8T細胞の出現頻度の大幅な増加を可能にした。第二のステップは、hTERT特異的CD4T細胞の質、および、特にTh1特異的T細胞の産生に対する免疫調整物質の影響を調査することにあった。当該目的のために、INVAC-1またはINVAC-1/mGM-CSFをワクチン投与したHLA-A2/DR1遺伝子導入マウス由来の脾細胞を、DR1拘束性のhTERTペプチドのプールで、24時間、インビトロで刺激するか、あるいは、刺激しないままとした。上清を回収し、hTERT特異的CD4T細胞が分泌したTh1、Th2およびTh17サイトカインの濃度を評価するために、サイトカイン結合アッセイ(CBA)によって試験した。図14Bに示したように、INVAC-1のみでワクチン投与したマウス由来の上清と比較して、INVAC-1/mGM-CSFでワクチン投与したマウスから回収した脾細胞の上清では、Th1サイトカイン、IL-2、TNFαおよびIFNγの有意な濃度が見られた。mGM-CSFを接種すると、Th1抗腫瘍サイトカインであるTNFα(p<0.01)、IFNγ(p<0.05)およびIL-2(p<0.05)の濃度において、大幅な増加が見られた。
INVAC-1ワクチン皮内投与およびエレクトロポレーション後のhTERT特異的CD8T細胞応答におけるIL-12サイトカインの影響もまた、調べた。当該目的のために、HLA-A2/DR1マウスに、エレクトロポレーションを伴うINVAC-1のID投与と共に、IL-12のID注射を接種した(図15)。別のグループのマウスには、IL-12の接種を行わずに、INVAC-1/エレクトロポレーションによるIDワクチン投与を行った。コントロールのマウスは、PBSとIL-12、またはPBS単独で、エレクトロポレーションを伴うニセのワクチン投与を行った。注射の14日後に、マウスの脾臓を回収し、A2拘束性のhTERTペプチドを用いたIFN-γELIspotアッセイによって、誘発された免疫応答をモニターした。応答マウスの出現頻度は、IL-12を注射すると増加した。実際、INVAC-1によってワクチン投与したグループおよびINVAC-1/IL-12によってワクチン投与したグループのそれぞれにおいて、5匹中2匹、および5匹中4匹の応答マウスが存在した。

pUTScramおよびpUTInv構築物は、酵素的に不活性な、ヒトユビキチン−テロメラーゼに基づく融合タンパク質をコードする約8.9kbの二本鎖DNAプラスミドである。Scrambledの、およびInvertedの導入遺伝子は、哺乳類細胞において安定かつ一過性の発現を高レベルで行うように設計されたpcDNA3.0由来のInvitrogen pcDNA3.1(+)ベクター(5.4kb)に挿入した。導入遺伝子の発現は、広範囲の哺乳類細胞において効率的で高レベルな発現を可能にする、ヒトサイトメガロウイルス最初期(CMV)プロモーターから行われる。ベクターは、クローニングを可能にするマルチクローニングサイト(MCS)を含む。効率的な転写終結は、ウシ成長ホルモン(BGH)ポリアデニル化シグナルによって行われる。

当該遺伝子は、ユビキチン-テロメラーゼに基づく融合構築物として、重複する40塩基長のオリゴヌクレオチドアセンブリープロセス(GeneCust、Luxembourg)によって、新規合成した。制限酵素部位を削除し、かつGCリッチ配列を弱めるために、いくつかの保守的な塩基改変を行った。遺伝子合成により、望ましい発現システムに当該遺伝子をサブクローニングできるようにする、特有の隣接した制限酵素サイトHindIII/XbaIを含めた。合成した遺伝子は、pcDNA3.1(+)発現ベクター(Invitrogen、Carlsbad、USA)の制限酵素サイトHindIIIとXbaIとの間にクローニングした。プラスミドの配列は、PEGFP-N5’CGGTGGGAGGTCTATATAAG(配列番号27)およびBGH CAGGGTCAAGGAAGGCAC(配列番号28)プライマーを用いた配列決定によって、確認した。
GeneCustによって合成したINVAC-1組換え誘導体は、RD Biotech(Besancon、France)の大腸菌(E.coli)5-α細胞(fhuA2Δ(argF-lacZ)U169 phoA glnV44 Φ80 Δ(lacZ)M15 gyrA96 recA1 relA1 endA1 thi-1 hsdR17)(Lucigen Corporation、Middleton、USA、ref.60602-2)に形質転換して製造した。細胞はアンピシリン(#EU04000D、Euromedex)を含有するLeno×Broth培地に蒔き、増殖させた。抽出および精製した後、1×の滅菌PBSに再懸濁した、濃縮されたエンドトキシンフリーギガプレッププラスミドストック(2mg/mL)を調製した。ベクターは、制限酵素マッピングによって確認した(HindIII-XbaI;図21)。
当該DNAプラスミドは、すでに実施例1に記載した。
CrFK細胞(クランデルリース(Crandell Rees)ネコ腎細胞)、HEK293T細胞(ヒト胚性腎細胞)は、10%熱失活ウシ胎児血清(PAA、Velizy-Villacoublay、France)および1%ペニシリン/ストレプトマイシン(Life Technologies、Saint-Aubin、France)を添加した、ダルベッコ改変イーグル培地(DMEM)で培養した。
ウエスタンブロット解析は、遺伝子導入したHEK293T細胞を用いて行った。ウエスタンブロットの手順は実施例1に記載した通りである。

本実験では、HLA-B*0702遺伝子導入マウス系統を使用した。
HLA-B*0702拘束性のhTERTペプチドは、実施例1に既に記載した。凍結乾燥したペプチドは、2mg/mLで滅菌水に溶解し、使用するまで−20℃で保管した。
皮内(ID)免疫化は、マウス側腹部の下部に、インスリンシリンジおよび特殊な針(U-100、29GX1/2’’-0.33x12mm、Terumo、Belgium)で剃毛後に行った。剃毛後、免疫化手順の間および後に赤斑は観察されなかった。各マウスは、プラスミド(INVAC-1、pUTScramまたはpUTInv)による初回刺激のID注射を、100μgのDNAまたは1×PBSで受けた。ワクチンレジメンに従って、マウスは、DNAまたは1×PBSの二回目の注射を同様に受けることができた。
ELIspotアッセイは、実施例1に記載の方法に従って行った。HLA-B*0702拘束性の三つの特異的hTERTペプチド(p277、p351およびp1123)から成る一つのプールのみ、実施例2では使用した。
インビボ溶解アッセイは、実施例1に記載した手順に従って行った。HLA-B*0702拘束性の二つの特異的hTERTペプチド(p351およびp1123)のみ、それぞれ免疫優勢のペプチドおよび準優位のペプチドとして、実施例2では使用した。
GraphPadPrism5ソフトウエアを、データ処理、解析およびグラフ化に使用した。データは、平均値±標準偏差または中央値として表している。ELIspotアッセイの統計解析は、マン・ホイットニーのノンパラメトリック検定および/またはダンの多重比較検定を伴うクラスカル・ワリス検定を用いて行った。有意差は、危険率<0.05で設定した。
[INVAC-1プラスミドDNAの特徴および配列解析]
INVAC-1プラスミドDNAの特徴および配列解析は、すでに実施例1に記載した。
二つのINVAC-1組換え(shuffled)誘導体遺伝子を合成してクローニングした(図20)。当該構築物は、実施例1に記載されたINVAC-1ヌクレオチド配列、および国際特許出願WO2007/014740に記載された野生型hTERTアミノ酸配列に基づいている。
インビトロでのHEK293T細胞株への一過性の遺伝子導入の18時間から96時間後に、野生型hTERT、INVAC-1、pUTScramおよびpUTInvタンパク質の全体的な発現に関する情報を提供するために、ウエスタンブロットアッセイを行った。野生型hTERTタンパク質のバンドは、未修飾のhTERTのサイズである124.5kDa(図23Aおよび23Cの左部分)に一致した。実施例1において、INVAC-1タンパク質は、安定なレベルに発現された野生型hTERTタンパク質とは反対に、全時間帯に渡って速やかに分解されることが分かった。Scrambledの、およびInvertedの組換え(shuffled)タンパク質の特異的なバンドが全時間帯に渡って検出された(図23Aおよび23Cの右側)。両タンパク質において、特異的なバンドはタンパク質全体の予想サイズ(130.2kDa)よりも小さいサイズ(<110kDa)で観察された。Scrambledの、およびInvertedのタンパク質のこれらの形態は、分解産物に相当する。実際、Scrambledの、およびInvertedの発現した未分解の産物は、ウエスタンブロット解析においては検出不可能だった。当該構築物は、生産後直ちにタンパク質の分解が高速で行われたこと示唆する1つから3つまでの特異的なバンドを、それぞれ提示した。INVAC-1のような、Scrambledの分解産物の全時間帯に渡る同様の分解パターンは、βアクチンローディングコントロールへの標準化後に、証明した(ImageJ解析:図23B)。Invertedの分解産物は、他のINVAC-1誘導体タンパク質に、より近いパターンを持っている(図23C、23Dおよび図3C:pUTD10Not、pUTD10CogおよびpUTD23Tyn、実施例1参照)。
INVAC-1およびINVAC-1誘導体(pUTD10Not、pUTD10CogおよびpUTD23Tyn、実施例1参照)で示したように、pUTScramおよびpUTInvがコードするScrambledの、およびInvertedの組換えタンパク質は、核小体排除パターンによって、核と細胞質との間に分布した(データ不掲載)。
テロメラーゼ陰性CrFK細胞株中の、Ubi-hTERT組換え(shuffled)構築物のテロメラーゼ活性を評価するために、TRAPアッセイを行った。テロメラーゼ活性は、pTRIP-CMV-hTERTプラスミドを用いて野生型hTERTを遺伝子導入したCrFK細胞でのみ見られた。
INVAC-1の特徴の範囲を広げる多様なhTERTエピトープの抗原提示を誘発するよう、pUTScramおよびpUTInv構築物を設計した。pUTScram、pUTInvおよびINVAC-1の免疫原性の比較は、各種構築物によって皮膚エレクトロポレーションを伴ってID免疫化されたHLA-B7マウスにおいて、二回の免疫化(プライムブーストレジメン)の後、行った。マウスは、二回目のワクチン投与/エレクトロポレーションの10日後に屠殺した。マウスの脾臓を回収し、誘発されたCD8T細胞応答を、HLA-B7MHCクラスI拘束性のhTERTペプチド(3つのペプチドp277、p351およびp1123のプール)を用いたIFN-γELIspotアッセイによって、モニターした。コントロールマウスと比較して、INVAC-1、pUTScram(Scrambled)およびpUTInv(Inverted)によってワクチン投与したマウスで、hTERT特異的CD8T細胞の出現頻度の有意な差が見られた(図25)。
抗腫瘍免疫応答に関連している免疫細胞の中で、細胞傷害性CD8Tリンパ球(CTL)およびTh1CD4T細胞が、最も強力なエフェクター細胞(Vesely、Kershaw et al.2011)(Braumuller、Wieder et al.2013)として同定されている。

- hTERTタンパク質がアミノ酸867-869(VDD)の1-12アミノ酸上流および/または下流のさらなる欠失によりテロメラーゼ触媒活性を欠く、請求項1に記載の核酸構築物。
- hTERTタンパク質が少なくともアミノ酸1-23の欠失により核小体局在化シグナルを欠く、請求項1または2に記載の核酸構築物。
- hTERTタンパク質がアミノ酸1-47の欠失により核小体局在化シグナルを欠く、請求項3に記載の核酸構築物。
- DNAである、請求項1〜4のいずれか1項に記載の核酸構築物。
- DNAプラスミドである、請求項5に記載の核酸構築物。
- 配列番号12のアミノ酸配列をコードする、請求項5に記載の核酸構築物。
- 配列番号11または配列番号11のヌクレオチド3488-6961を含む、請求項7に記載の核酸構築物。
- 配列番号14、16または18のアミノ酸配列をコードする、請求項5に記載の核酸構築物。
- 配列番号13、15、または17のヌクレオチド配列を含む、請求項9に記載の核酸構築物。
- テロメラーゼを過剰発現する細胞に対して、対象者の免疫応答を誘発するのに用いられる、請求項1〜10のいずれか1項に記載の核酸構築物。
- 異形成細胞、腫瘍細胞、またはオンコウイルスに感染した細胞に対して、対象者の免疫応答を誘発するのに用いられる、請求項11に記載の核酸構築物。
- 患者の腫瘍の予防または処置に用いられる、請求項1〜12のいずれか1項に記載の核酸構築物。
- ヒトテロメラーゼ逆転写酵素(hTERT)由来の配列を含む核酸構築物であって、hTERT由来の配列が
i)hTERTの全てのまたは少なくとも80%のエピトープを任意の順序でコードする、および
ii)テロメラーゼ触媒活性および核小体局在化シグナルを欠くタンパク質であって、アミノ酸867-869(VDD)の欠失によりテロメラーゼ触媒活性を欠き、かつ、ユビキチンとN末端で融合するタンパク質をコードする、

- テロメラーゼを過剰発現する細胞に対して、対象者の免疫応答を誘発するのに用いられる、請求項14〜20のいずれか1項に記載の核酸構築物。
