Description
a)pH7.5未満で自己組織化し始めるペプチドを含有する溶液であって、前記ペプチドが自己組織化を始めるpHを0.1ないし0.5pH単位超えるpHを有する前記溶液を準備すること;
b)前記溶液のpHを8.0以上に増加させ、そして、凍結乾燥の間に除去されるのに十分な揮発性を有している化合物を添加すること;
c)前記溶液を凍結乾燥すること;および
d)所望により、凍結乾燥物を水溶液に溶解させ、前記ペプチドが自己組織化を始めるpHを0.1ないし0.5pH単位超えるpHを有し、前記ペプチドをそのモノマー状態で含有する溶液を得ること;
を含む、カリエス障害等の歯の障害を治療するのに適した組成物の製造方法を提供するものである。
実施例1:ペプチド溶液の調製
SEQ ID NO:2に示されているシーケンスを有する変性P11−4ペプチドを5+/−3℃の温度で、使用24時間前に解凍した。4mMトリスを、WFI(注入用水(water for injection))グレードの水に、室温で少なくとも10分間撹拌下、添加する。10mg/gの最終濃度になるまで、トレハロース二水和物を、撹拌を続けながら添加する。続いて、ペプチドP11−4を、2 mg/gの最終濃度になるまで添加し、透明になるまで撹拌し溶解する。このペプチド溶液は、最初、約8.2のpHを有している。
ペプチド溶液は、溶解後、ネマチック粘弾性流体であった。pHを8.5に調整後、ペプチド溶液は、アイソトロピック流体(図2A)のようなクリアーで非粘性となった。該溶液をバイアルに充填した後も、該溶液はクリアーで非粘性であった(図2B:溶液の視覚化のためだけにブルーに着色された実施例アリコート)。凍結乾燥により無傷の「ケーキ」、すなわち凍結乾燥物(図2C)が得られ、それは、良好な貯蔵安定性を示した。
いろいろな濃度を有するペプチド溶液の調製
上記したように、いろいろなバッファ(トリス)濃度、ペプチド(P11−4)濃度および増量剤(トレハロース二水和物)濃度を有するペプチド溶液を調製した。pHは上記したように調製した。
該溶液をバイアルに充填後、全3種類の溶液は、クリアーで非粘性であった。凍結乾燥により無傷の「ケーキ」、すなわち凍結乾燥物が得られ、それらは、良好な貯蔵安定性を示した。
P11−4ペプチド溶液の粘度
ペプチド溶液におけるアセンブリーを評価するために、いろいろな充填ニードルを通じてP11−4を通過させてテストした。
P11−4バルク溶液を調合し、DMA300ポータブル密度計(Anton Paar Germany GmbH, オストフィルデン, ドイツ)を使用して、溶液の密度を求めた。シリンジを使用して、バルク溶液を密度計に充填した。25℃への温度補償は、密度計により自動的に行われた。
EDM 3295計量装置(Bausch + Stroebel Maschinenfabrik GmbH+Co.ケージー(KG), イルショーフェン(Ilshofen)、ドイツ)を、250mgの充填重量に調整した。テストインレーバイアルを空で秤量し、バルク溶液で充填し、充填後、再度、秤量した。
Claims (14)
- a)pH7.5未満で自己組織化し始めるペプチドを含有する溶液であって、凍結乾燥の間不揮発性であり、かつ前記ペプチドが自己組織化を始めるpHを0.1ないし0.5pH単位超えるpHを付与するバッファを含む溶液を準備すること;
b)前記溶液のpHを8.0以上に増加させ、そして、凍結乾燥の間に除去されるのに十分な揮発性を有している化合物を添加すること;
c)前記溶液を凍結乾燥すること、ここで、得られた凍結乾燥物は、水に溶解して、前記ペプチドが自己組織化を始めるpHを0.1ないし0.5pH単位超えるpHを有し、前記ペプチドをそのモノマー状態で主に含有する溶液を得るのに適している;および
d)所望により、凍結乾燥物を水に溶解させ、前記ペプチドが自己組織化を始めるpHを0.1ないし0.5pH単位超えるpHを有し、前記ペプチドをそのモノマー状態で主に含有する溶液を得ること;
を含む、歯の障害を治療するための組成物の製造方法。 - ステップb)におけるpHを、pH8−9、好ましくはpH8.5に増加させる、請求項1に記載の方法。
- ステップc)の前に、安定化剤および/または増量剤を前記溶液に添加する、請求項1〜2いずれかに記載の方法。
- 安定化剤または増量剤が、マンニトール、ソルビトール、スクロース、トレハロースおよびグリシンを含むグループから選択される非還元性の糖である、請求項3に記載の方法。
- ステップb)における前記化合物が、アンモニア、2−アミノエタノール、4−メチルモルホリン、およびピリジンからなるグループから選択される、請求項1〜4いずれかに記載の方法。
- 90%を超えるステップb)の前記化合物が、凍結乾燥時に前記溶液から除去される、請求項1〜5いずれかに記載の方法。
- ステップc)またはd)において得られた組成物が、歯の表面に適用するのに適したビヒクル、好ましくはハイドロゲルとさらに混合される、請求項1〜6いずれかに記載の方法。
- ステップa)における前記溶液が、40mM以下の濃度でトリス、好ましくは20mM以下のトリス、より好ましくは10mM以下のトリスを含有する、請求項1〜7いずれかに記載の方法。
- 7.5未満のpH値で自己組織化するペプチド
非還元性糖、および
凍結乾燥の間不揮発性のバッファ
を含有する組成物であって、前記組成物を水中に溶解した時、前記ペプチドが自己組織化し始めるpHより0.1ないし0.5pH単位超えるpHを有し、かつ0.15未満のイオン強度を有する溶液が得られ、前記ペプチドがモノマー形態で主に存在し、
ここで、前記ペプチドがSEQ ID NO:1またはSEQ ID NO:2に示されるシーケンスを含み、かつ
凍結乾燥の間不揮発性のバッファが、TAPS、ビシン、トリス、トリシン、TAPSO、HEPES、TES、MOPS、PIPES、カコジレート、SSC、MES、およびそれらの任意の組合せからなる群から選択される、または、水酸化カリウム、水酸化アンモニウム、リン酸カリウムジ-およびトリ-塩基、トリス、トリエタノールアミン、ポリエチレンイミンならびにそれらの任意の組合せ、と組み合わせたクエン酸またはリン酸からなる群から選択される、
組成物。 - 7.5未満のpH値で自己組織化するペプチド
非還元性糖、および
凍結乾燥の間不揮発性のバッファ
を含有する、凍結乾燥形態の組成物であって、前記組成物を水中に溶解した時、前記ペプチドが自己組織化し始めるpHより0.1ないし0.5pH単位超えるpHを有し、かつ0.15未満のイオン強度を有する溶液が得られ、前記ペプチドがモノマー形態で主に存在し、
ここで、前記ペプチドがSEQ ID NO:1またはSEQ ID NO:2に示されるシーケンスを含み、かつ
凍結乾燥の間不揮発性のバッファが、TAPS、ビシン、トリス、トリシン、TAPSO、HEPES、TES、MOPS、PIPES、カコジレート、SSC、MES、およびそれらの任意の組合せからなる群から選択される、または、水酸化カリウム、水酸化アンモニウム、リン酸カリウムジ-およびトリ-塩基、トリス、トリエタノールアミン、ポリエチレンイミンならびにそれらの任意の組合せ、と組み合わせたクエン酸またはリン酸からなる群から選択される、
組成物。 - 非還元性糖が、マンニトール、ソルビトール、スクロース、トレハロース、グリシンおよびそれらの誘導体からなる群から選択される、請求項9または10に記載の組成物。
- 前記ペプチドが、SEQ ID NO:1に示されるシーケンスを含み、非還元性糖が、トレハロースまたはトレハロース二水和物であり、かつ凍結乾燥の間不揮発性のバッファがトリスである、請求項9−11のいずれかに記載の組成物。
- 前記ペプチドの、少なくとも70%、好ましくは少なくとも80%、より好ましくは少なくとも90%が、モノマー形態で存在する、請求項9−12いずれかに記載の組成物。
- 歯の障害、好ましくは表面下カリエス障害を治療する方法に使用するための、または歯の表面および/またはカリエス障害、好ましくは表面下カリエス障害にミネラルを補充する方法に使用するための、請求項9−13いずれかに記載の組成物。
