Description

本開示は、全体として、無細胞の酵素的転換(transformation)または代謝工学によって作り出された株を用いた発酵(fermentation)のいずれかによるエルゴチオネインの生産に関する。
エルゴチオネインは、1909年にTanretによって麦角から単離された、チオールを含むアミノ酸である。これは、独特の酸化還元特性があるため、最も優れた天然抗酸化剤の1つとなっている。さらに、多くのヒト組織(例えば、肝臓、腎臓、中枢神経系、および赤血球)によって食物から特異的に濃縮される(specific enrichment)ため、エルゴチオネインは、ヒト生理学において多くの有益な役割を有していると提唱された。現在、エルゴチオネインは、10億ドルの市場を有する抗老化製品の重要な成分として広く使用されている。
無細胞酵素系または細胞培養系のいずれかにおいてエルゴチオネインの生産を増加させるか、増大させるか、または最大化するための新規な酵素的方法および代謝工学方法が、本明細書において提供される。本明細書において開示する無細胞酵素系および代謝工学によって作り出される操作された微生物を用いた細胞培養系を用いて、より高いレベルでエルゴチオネインを生産することができる。本明細書において説明するこれらの方法は、発明者らの以下の発見に基づいている:1)大腸菌(E. coli)におけるEgtEの上首尾な生産およびEgtB活性を向上させるための条件の同定。これにより、無細胞転換または発酵のいずれかにより現在公知のエルゴチオネイン生合成経路を用いてエルゴチオネインを生産することが可能になる。2)オボチオール生合成経路(OvoA)に由来する酵素を用いて、Cysとトリメチル-ヒスチジンとの直接的な酸化的結合を触媒して、エルゴチオネイン生合成に必要とされる重要な中間体を生産することができる。OvoAはまた、CysとHisの酸化的結合を触媒して構造的に完全に異なる化合物も生成するため、このような発見は意外で予想外である。理論に拘束されることを望むものではないが、新規に発見されたOvoA活性により、エルゴチオネイン生合成経路を2段階短縮することができ、エルゴチオネイン生合成とグルタチオネイン(glutathioneine)生合成の競合を解消することができる。新規に発見されたOvoA化学反応に基づく無細胞酵素系または細胞培養系のいずれかによってエルゴチオネインを生産することができる。3)また、別の酵素NcEgt1も、Cysとトリメチル-ヒスチジンの直接的結合を触媒することができた。新規に発見されたNcEgt1化学反応に基づく無細胞酵素系または細胞培養系のいずれかによってエルゴチオネインを生産することができる。

エルゴチオネイン生合成遺伝子クラスターの同定
エルゴチオネインおよびオボチオール(5および7、図1)は、1909年にTanretによって麦角から単離された2種のチオール-イミダゾール含有代謝産物である[1]。ヒトにはエルゴチオネイン特異的輸送体が存在するため[2]、我々は食事に由来するエルゴチオネインを少数の器官(例えば、肝臓、腎臓、中枢神経系、および赤血球)においてミリモル濃度に濃縮することができる[3]。他のチオールとは違って、エルゴチオネインおよびオボチオールのチオール-イミダゾール側鎖のチオラート型とチオン型の平衡は、チオン型(例えば、図1Aの5)に優勢に傾き(favors predominantly)[3f、4]、これにより、他のチオール(例えばグルタチオン)と比べて酸化に対してはるかに安定になっている[4a]。エルゴチオネインの独特の酸化還元特性[4a、5]のおかげで、エルゴチオネインは赤血球においてヘモグロビンが酸化されないように守り、水晶体に白内障が形成するのを防ぐことができる[6]。さらに、エルゴチオネインは、細胞の金属解毒のための金属キレート剤としても機能することができる。現在、エルゴチオネインは、数十億ドルの市場規模を有する多くの市販製品(抗老化化粧品、保存剤、栄養補助食品、毛髪および爪の成長促進製品など)の重要な成分の1つとして幅広く使用されている。
(i)EgtBの活性が低い。EgtBは単環式の非ヘム鉄酵素であると提唱されている。現在の活性が低い。本発明者らは、活性を向上させるか、またはより優れた活性を有する酵素を発見する必要がある。
(ii)EgtEタンパク質を過剰発現させることができない。これまで、EgtE過剰発現の成功に関する文献報告はない。エルゴチオネイン生合成経路を再構成するためには、この問題を解決しなければならない。
(iii)現在のエルゴチオネイン生合成経路を最適化する必要がある。天然のエルゴチオネイン生合成経路、特に酸化的C-S結合形成段階(EgtB触媒)を最適化する必要がある。さらに、図1Aの経路に従うと、エルゴチオネインとグルタチオンの生合成の間に競合がある。
単環式の非ヘム鉄酵素(例えば、EgtB、OvoA)を改良するのに不可欠な段階として、本発明者らは、少なくとも2種の異なる活性アッセイ法を開発した。したがって、これらのアッセイ法は、これらの酵素の活性を向上させるにあたっての本発明者らの取り組みを目標へと導くのに使用される。
EgtB触媒およびOvoA触媒の両方に関する2つの異なるアッセイ法を考案した(図3)。EgtBおよびOvoAは両方とも酸化酵素であることが提唱されており[9〜10]、HisとCysの間の酸化的結合を触媒してC-S結合を形成させる。同時に、硫黄を酸化してスルホキシドにする。全体的なプロセスは、イソペニシリンN合成酵素の場合に類似した4電子酸化プロセス(図1)である[12]。Seebeckによる最初の報告では、彼らは260nmにおける吸収変化をモニターすることによって反応をモニターした。これは、酸化生成物(3または6、図1)がこの領域に吸収を有するためであった。しかしながら、それはあまり感度が高くない。嫌気条件下では検出可能な生成物が形成されなかったため、酸素が必要である。この特徴に基づいて、本発明者らは、NeoFoxy酸素電極(図3-I-A)を用いて酸素消費速度(図3-I-B)をモニターすることによるEgtB活性アッセイ法およびOvoA活性アッセイ法を考案した。このアッセイ法(図3-I-B)をルーチンのアッセイ法として使用して、EgtB活性およびOvoA活性のための条件を最適化した。
EgtBおよびOvoAは単環式の非ヘム鉄酵素であると提唱されているため、それらが鉄を含む酵素である場合、触媒的代謝回転過程の間に不活性化される可能性がある。酸素消費アッセイ法を用いて、本発明者らは、Seebeckによって報告されている活性と比べて300倍にOvoA活性を向上させた(kcatが572±20/分)[10]。これは、次のいくつかの因子を組み合わせた結果である:1)本発明者らは、好気的ではなく嫌気的にOvoAを精製して、精製過程の間にOvoAが不活性化される機会を最小限にした;2)精製したOvoAをFe2+を用いて嫌気的に再構成;3)触媒的代謝回転過程の間にFe2+をFe3+に酸化することによって一部のOvoAが実際に不活性化される場合、Fe3+を還元してFe2+に戻すために反応緩衝液中にアスコルビン酸を含める。理論に拘束されることを望むものではないが、これらの戦略は、EgtBタンパク質およびNcEgt1タンパク質にも応用することができる。
Seebeckは、インビトロで活性を実証することによって、EgtA、EgtB、EgtC、およびEgtDの機能を確認した。しかしながら、EgtE生産に関する様々な方法の検討に彼らが多大な努力を費やしたにもかかわらず、EgtEタンパク質は、過剰発現させることも精製することもできなかった[9]。したがって、合成生物学によるエルゴチオネイン生産は不可能である。本発明者らの予備研究において、本発明者らは、EgtEを過剰発現させ精製することに成功した(図4A)。精製EgtEのUV可視スペクトルを図4Bに示しており、400nm付近での吸収特徴は、PLP補助因子の存在と一致している。これらの試みから得られた最も重要な発見の内の1つは、EgtEが別の遺伝子、例えばFAD合成酵素と呼ばれる遺伝子と共に同時発現される必要があり得るということである。
上記のセクションにおいて、本発明者らはEgtEの問題を解決することに成功した。これにより、代謝工学によるエルゴチオネイン生産が可能になる。しかしながら、もともとのエルゴチオネイン生合成経路(図1A)は効率的ではない。この問題を解決するために、本発明者らは、図2に概説したように、ヘルシニン(2)とCysを直接的に酸化的結合することができる酵素の探索を開始した。バイオインフォマティクス解析と酸素アッセイ法および1H-NMRアッセイ法を用いた活性解析との組合せによって、このような酵素の内の2種(図1BのOvoAおよびアカパンカビに由来するNcEgt-1[13])が同定された。
方法I:図2に概説した新しいエルゴチオネイン生合成経路に従うと、2段階しか必要とされない:OvoAまたはNcEgt-1(2→4変換)によって触媒される、ヘルシニン(2)とCysの酸化的結合によって化合物4になる段階、およびEgtE触媒によるC-S溶解(lysis)段階(4→5変換、図2)。EgtE生産は、EgtEが別の遺伝子(例えばFAD合成酵素遺伝子)と同時発現される場合にのみ達成可能であるため、したがって、最小限のエルゴチオネイン生合成遺伝子クラスターは、3種の遺伝子(OvoAまたはNcΔEgt-1、EgtE、およびFAD合成酵素)を用いて組立てられ得る。本発明者らは、pETDuetベクターのマルチクローニング部位IにEgtEおよびFADを、マルチクローニング部位IIにNcEgt-1遺伝子をクローニングして、図6に示すような構築物を作製し、(Ego-1)と名付けた。

Claims (28)
- (i)組換えによって発現させた、(a)EgtAタンパク質または酵素活性を保持しているその機能的断片、(b)EgtBタンパク質または酵素活性を保持しているその機能的断片、(c)EgtCタンパク質または酵素活性を保持しているその機能的断片、(d)EgtDタンパク質または酵素活性を保持しているその機能的断片、および(e)EgtEタンパク質または酵素活性を保持しているその機能的断片およびFAD合成酵素または酵素活性を保持しているその機能的断片を含む反応混合物と共に、ヒスチジンまたはヘルシニンをインキュベートする段階であって、EgtEタンパク質が、EgtE生産を促進するために、FAD合成酵素をコードする遺伝子と共に組換えによって同時発現される、段階;ならびに(ii)酵素的混合物からエルゴチオネインを単離する段階を含む、エルゴチオネインを調製するための方法。
- (i)組換えによって発現させた、(a)OvoAタンパク質、NcEgt1タンパク質、または酵素活性を保持しているその機能的断片、および(b)EgtEタンパク質または酵素活性を保持しているその機能的断片およびFAD合成酵素または酵素活性を保持しているその機能的断片を含む反応混合物と共に、ヘルシニンをインキュベートする段階であって、EgtEタンパク質が、EgtE生産を促進するために、FAD合成酵素をコードする遺伝子と共に組換えによって同時発現される、段階;ならびに(ii)酵素的混合物からエルゴチオネインを単離する段階を含む、エルゴチオネインを調製するための方法。
- (i)組換えによって発現させた、(a)EgtDタンパク質または酵素活性を保持しているその機能的断片、(b)OvoAタンパク質、NcEgt1タンパク質、または酵素活性を保持しているその機能的断片、および(c)EgtEタンパク質または酵素活性を保持しているその機能的断片およびFAD合成酵素または酵素活性を保持しているその機能的断片を含む反応混合物と共に、ヒスチジンをインキュベートする段階であって、EgtEタンパク質が、EgtE生産を促進するために、FAD合成酵素をコードする遺伝子と共に組換えによって同時発現される、段階;ならびに(ii)酵素的混合物からエルゴチオネインを単離する段階を含む、エルゴチオネインを調製するための方法。
- EgtDタンパク質が、S-アデノシルメチオニン(SAM)合成酵素をコードする遺伝子と共に組換えによって同時発現される、請求項1または3記載の方法。
- 酵素的混合物がメチオニンまたはシステインをさらに含む、請求項1〜4のいずれか一項記載の方法。
- 酵素的混合物が鉄塩をさらに含む、請求項1〜5のいずれか一項記載の方法。
- 反応混合物が細胞溶解物を含む、請求項1〜6のいずれか一項記載の方法。
- (i)egtA遺伝子、egtB遺伝子、egtC遺伝子、egtD遺伝子、egtE遺伝子、およびegtE遺伝子によってコードされるEgtEタンパク質の生産を促進するためのFAD合成酵素をコードする遺伝子を組換えによって発現する細胞を、適切な代謝条件下で適切な培地において培養する段階;ならびに
(ii)培地中または細胞集団中にエルゴチオネインを蓄積させ、その後、それらからエルゴチオネインを回収する段階、
を含む、エルゴチオネインを調製する方法。 - (i)(a)ovoA遺伝子またはNcEgt-1遺伝子ならびに(b)egtE遺伝子およびegtE遺伝子によってコードされるEgtEタンパク質の生産を促進するためのFAD合成酵素をコードする遺伝子を組換えによって発現する細胞を、適切な代謝条件下で適切な培地において培養する段階;ならびに

- 細胞が、細菌細胞、真菌細胞、昆虫細胞、哺乳動物細胞、または植物細胞である、請求項8〜14のいずれか一項記載の方法。
- 細菌細胞が大腸菌細胞である、請求項15記載の方法。
- 培地に鉄塩がさらに補充される、請求項8〜16のいずれか一項記載の方法。
- 培地にメチオニン、ヘルシニン、ヒスチジン、またはシステインの内の他の1種または複数種がさらに補充される、請求項8〜17のいずれか一項記載の方法。
