Description
<1> ポリアミンを有効成分とする、軟骨組織の再生を促進するための組成物。
<2> ポリアミンが、スペルミン及びスペルミジンから選択される少なくとも1の化合物である<1>に記載の組成物。
<3> 軟骨組織の再生を介して、非炎症性の変形性関節症、椎間板損傷、半月板損傷、離断性骨軟骨症及び外傷性軟骨損傷からなる群から選択されるいずれかの軟骨損傷に関連する疾患を治療又は予防するための、<1>又は<2>に記載の組成物。
<4> 軟骨組織の再生を介して、非炎症性の変形性関節症を治療又は予防するための、<1>又は<2>に記載の組成物。
<5> 経口投与用である、<1>〜<4>のいずれか一に記載の組成物。
ラット(Lewisラット、チャールスリバー社製、メス、8〜10週齢、体重200〜250g)をイソフルレン吸引により麻酔した後に、該ラットの膝関節正中の皮膚を1cm切開した。次いで、膝蓋骨外側より関節包を切離し、膝蓋骨を内側へ脱臼させて、大腿骨顆部軟骨を露出させた。そして、大腿骨顆部正中に大腿骨長軸に向かってキルシュナーワイヤー(瑞穂医科工業株式会社製、径1.5mm)を1mmの深さにて刺入した。次いで、関節包、皮膚を4−0ナイロンで縫合し、軟骨損傷モデルラット(大腿骨顆部軟骨損傷モデルラット)を調製した。
前記軟骨損傷モデルラットに摂取させた試験試料は以下のようにして調製した。
前記試験試料を摂取させた軟骨損傷モデルラットから、軟骨を単離した。単離した軟骨を0.1N HClにて245倍希釈し、1時間振とうした後、10,000g、5分間の条件にて遠心し、上清を回収した。回収した上清を蒸留水にて2倍希釈し、0.45μmフィルターにてろ過したものを、高速液体クロマトグラフィー(株式会社島津製作所製、製品名;プロミネンス)にて解析し、スペルミジン及びスペルミンの量を測定した。そして、得られた測定量に基づき、軟骨1gあたりのスペルミジン及びスペルミンの量を各々算出した。
前記損傷を与えてから2週間、軟骨損傷モデルラットに、ポリアミン非含有試験飼料、0.01質量%ポリアミン含有試験飼料、0.05質量%ポリアミン含有試験飼料又は0.1質量%ポリアミン含有試験飼料を、自由に摂取させた。そして、これらラットの大腿骨を摘出し、筋肉等の付随組織を除去した後に、10%ホルマリンにて固定した。ホルマリン固定した大腿骨を用い、正中縦断面で標本を作製した。作製した標本にヘマトキシリン・エオジン(HE)染色又はサフラニンO染色を施し、軟骨の損傷度又は再生の程度を顕微鏡下にて観察した。得られた結果を図1〜3に示す。
Claims (5)
- ポリアミンを有効成分とする、軟骨組織の再生を促進するための組成物。
- ポリアミンが、スペルミン及びスペルミジンから選択される少なくとも1の化合物である請求項1に記載の組成物。
- 軟骨組織の再生を介して、非炎症性の変形性関節症、椎間板損傷、半月板損傷、離断性骨軟骨症及び外傷性軟骨損傷からなる群から選択されるいずれかの軟骨損傷に関連する疾患を治療又は予防するための、請求項1又は2に記載の組成物。
- 軟骨組織の再生を介して、非炎症性の変形性関節症を治療又は予防するための、請求項1又は2に記載の組成物。
- 経口投与用である、請求項1〜4のいずれか一項に記載の組成物。
