Description
<1> プトレスシン、スペルミジン及びスペルミンを有効成分とする、肝臓組織の再生を促進するための医薬組成物。
大豆胚芽から酸抽出にてポリアミン組成物を調製するために、先ず、大豆(品種名;銀河、WONDER等)の胚芽を採取した。そして、得られた胚芽を水又は弱塩基性溶液(pH12〜13の水酸化ナトリウム水溶液)にて洗浄し、ポリアミンの抽出効率を上げるために、該胚芽に付着した夾雑物を除去した。
ポリアミン非含有試験飼料は、標準的なげっ歯類用飼料(オリエンタルバイオ株式会社製)製造の際に、該飼料の成分である大豆油(ポリアミンを豊富に含有する原材料)の代わりに、コーン油を混合することにより、調製した。
実験には、本発明者らが以前に樹立した、ホタルルシフェラーゼ発現トランスジェニックLewisラット(Luc−Tg LEWラット、雄、体重260〜310g、10〜12週齢)を用いた。なお、このラットにおいて、ジーントラップ ROSA 26プロモーター制御下、全身においてルシフェラーゼが発現している。ルシフェラーゼはルシフェリンを酸化させることにより、光を生じさせることができるが、この発光反応においてアデノシン三リン酸(ATP)は必須であるため、生じる発光を通して、当該ラット体内のATP量を評価することができる(Hakamata Y,ら、Transplantation、2006年、81巻、1179〜1184ページ、Teratani T,ら、Islets、2011年5−6月、3巻、3号、111〜117ページ 参照)。 これらラットは、温度・湿度制御環境下、12時間毎の明/暗サイクルにて、標準的な実験室用飼料又は前記試験飼料、及び水を不断に与えて飼育した。また、本実施例における全ての動物実験は、自治医科大学動物実験指針に従って行った。
生体肝臓移植のドナーにおける、ポリアミンによる肝臓組織の再生促進効果について、モデル動物を用いて検証した。すなわち、ポリアミン非含有試験飼料、0.01質量%ポリアミン含有試験飼料、0.05質量%ポリアミン含有試験飼料又は大豆由来ポリアミン含有試験飼料を3日間、自由摂取させたLuc−Tg LEWラットに、古典的70%肝切除術を施した。そして、術後も術前と同じ飼料を各々与え、術後7日目にこれらラットを安楽死させた。次いで、肝臓を摘出し、それらの重量を測定した。得られた測定値から、肝再生率(平均値±標準偏差)を算出した。なお、肝再生率は下記式にて算出した。
肝再生率(%)=古典的70%肝切除後7日目に摘出した肝重量(g)/推定全肝重量(g)×100
また、前記「推定全肝重量(g)」は、「古典的70%肝切除時に摘出した肝重量(g)+推定残肝重量(g)」であり、「推定残肝重量」は、「(古典的70%肝切除時に摘出した肝重量(g)/0.667)×0.333」より求めた。さらに、連続的データに対してはスチューデントt検定を適用し、非連続低データに対してはマン・ホイットニー検定を適用し、統計解析を行い、P値が<0.05である場合に、統計的に有意であると判断した。得られた結果を図1及び表2に示す。
Luc−Tg LEWラットに、ポリアミン非含有試験飼料、0.01質量%ポリアミン含有試験飼料、0.1質量%ポリアミン含有試験飼料、大豆由来ポリアミン含有試験飼料を自由摂取の条件化で4週間与え、当該ラットの全身のATP代謝を1週間おきに観察した。
